Interaction Between Hydrophobic Au Nanoparticles and Pulmonary Surfactant (DPPC) Monolayers

Zhang Kangzhen,Liu Lianke,Bai Tingting,Guo Zhirui

Hydrophobic nanoparticles from the air get into the lungs through inhalation and firstly interact with pulmonary surfactant, which is essential for normal lung function. 1,2-dipalmitoyl-sn-glycerol-3-phosphocholine (DPPC) is the major component of pulmonary surfactant. Interaction between Langmuir monolayer of DPPC and hydrophobic Au nanoparticles (Au NPs) were investigated in this study by atomic force microscope. The experimental results indicated that incorporation of hydrophobic Au NPs into the monolayer hindered the phase transition, reduced the compressibility of monolayer and disrupted the large micro-domains in liquid-condensed (LC) phase into smaller ones. Moreover, Au NPs that modulated by DPPC domains formed wandering wires, which were enclosed LC phase domains, at low concentration and relatively large islands at high concentration. The large islands still remained on the water surface after five cycles of compression–expansion at high concentration of Au NPs, but single NPs or wires with 1–2 particle width escaped into the water during the compression through the help of DPPC coating. The interaction between hydrophobic NPs and pulmonary surfactant provides an insight into the toxicological effects of inhaled NPs from the atmosphere to the function of pulmonary surfactant in the lung.

Impact of carbon quantum dots on dynamic properties of BSA and BSA/DPPC adsorption layers

Lu Lai,Xiao-Qian Wei,Wei-Hua Huang,Ping Mei,Zhao-Hua Ren,Yi Liu

The effects of carbon quantum dots (CQDs) on the dynamic properties of bovine serum albumin (BSA) were investigated using pendant drop profile analysis method. Moreover, the effects of CQDs on the competitive adsorption of BSA and dipalmitoyl phosphatidylcholine (DPPC) were examined. CQDs reduce the fluorescence intensity of BSA and cause a red shift in fluorescence emission . The quenching constant at pH 4.3 is almost twice as large as that of the value obtained at pH 6.0. A small amount of CQDs does not influence the dynamic surface adsorption properties of BSA molecules. As the CQD concentration increases, a gradual increase in adsorption rate of BSA molecules is observed. Moreover, the addition of CQDs results in a significant transition of kinetic dependencies of surface elasticity of BSA solution when the CQD concentration exceeds a critical value. The appearance of the maximum surface elasticity value is probably attributed to the formation of tails and loops. When the dynamic surface properties are dominated by BSA molecules, the effects of CQDs on the surface properties of BSA/DPPC mixture are similar to those of BSA alone. However, when the surface film mainly consists of DPPC, CQDs can obviously change the interfacial properties of DPPC monolayer.

Real-time monitoring of the effect of carbon nanoparticles on the surface behavior of DPPC/DPPG Langmuir monolayer

Jiajie Hu,Xinrui Li,Meng Li,Yazhuo Shang,Yifan He,Honglai Liu

Air pollution has become increasingly serious. Fine particulate matter (PM2.5) is the most well-known air pollutant, which leads to some common respiratory diseases when inhaled into the lungs to certain concentration. However, there is a lack of research on the process of dynamically monitoring the real-time effect of nanoparticles on the pulmonary surfactant monolayer. In this study, the DPPC/DPPG monolayer is prepared by the Langmuir method to simulate the lung surfactant monolayer during respiration and the carbon nanoparticles are introduced to the monolayer under different surface pressures to simulate the real dynamic process of inhaling nanoparticles during breathing. The effect of carbon nanoparticles on the surface behavior of DPPC/DPPG monolayer in real-time was examined in details by a combination of surface pressure (π)-area (A) isotherms, compressibility modulus ( C s - 1 Cs-1 )-surface pressure (π) isotherms and the Brewster angle microscopy (BAM). The results have shown that the introduction of carbon nanoparticles under different surface pressures affects the properties of lipid monolayers. The added carbon nanoparticles under lower surface pressure are easy to penetrate the lipid molecules to inhibit monolayer phase transition. When the carbon nanoparticles are introduced to the monolayer under higher surface pressure, they tend to self-aggregate to reduce the monolayer stability rather than interact with lipid tail chains. This work not only confirms the exotic hydrophobic carbon nanoparticles retain in the DPPC/DPPG monolayer irreversibly and affect the surface behavior of monolayer during respiration, but also opens a new idea for real-time monitoring of the effects of PM2.5 on lung health.

The spatial arrangement of astaxanthin in bilayers greatly influenced the structural stability of DPPC liposomes

Lijun Ding,Jie Yang,Kangrui Yin,Hao Cheng,Jing Li,Changhu Xue

Liposomes are regarded as the ideal nanocarrier for concurrent or separate delivery of nutraceuticals in the food industries. Precise control of the structural stability is essential for the processing, storage, and nutrition delivery of liposomes. Astaxanthin was found to significantly affect the membrane stability of liposomes by inserting into the phospholipid bilayers in a similar way to cholesterol. Compared with cholesterol, astaxanthin could significantly improve the phase transition temperature, membrane fluidity , and membrane compactness of liposomes. Additionally, the membrane stability was well modulated by controlling the distribution patterns of astaxanthin (monomers, H- and J-aggregates) in bilayers. For instance, astaxanthin H-aggregates could endow the liposomal membrane with highest rigidity and compactness. Additionally, astaxanthin aggregates, especially J-aggregates could greatly improve storage stability of liposomes, thus providing a novel strategy to regulate and optimize the stability of liposomes for their diversified applications.

Synergistic effect of thermal and pulsed electric field (PEF) treatment on the permeability of soya PC and DPPC vesicles

Zhi-Wei Liu,Xin-An Zeng,Da-Wen Sun,Zhong Han,Rana Muhammad Aadil

The permeability change of vesicles composed of soya phosphatidylcholine (PC) and dipalmitoyl phosphatidylcholine (DPPC) encapsulated with 5(6)-carboxyfluorescein (CF) suspended in isotonic sucrose solution under separate or combined thermal and continuous pulsed electric field (PEF) treatments was investigated, respectively. The electric field intensity applied was in the range from 20 to 40   kV/cm with total treatment time of 1.2   ms, and thermal treatments were conducted at temperatures from 15 to 65   °C. Results indicated that the permeability behavior between DPPC vesicles and soya PC vesicles were quite different under the experimental conditions. Compared to soya PC vesicles, DPPC vesicles were more sensitive to thermal treatments. As the temperature increased to 41   °C, the 5(6)-CF release percentage (R) reached almost 100% abruptly. On the other hand, the synergetic effects of PEF and temperature on permeability of two vesicles were apparently different. For soya PC vesicles, the increment degree of R value was almost the same under various combination treatment conditions. However, for DPPC vesicles, it was demonstrated that the lipid bilayers membrane was almost impermeable to CF when the PEF strength was lower than 30   kV/cm at 20   °C and 20   kV/cm at 30   °C. As the temperature increased to 38   °C, the permeability of DPPC vesicles was significantly increased with increasing PEF intensity, reaching 25.25% at 40   kV/cm. Results from the current study could be helpful for better understanding of the underlying mechanism on PEF inactivation as affected by the lipid composition of cell membrane and its phase transition temperature.

Study transport of hesperidin based on the DPPC lipid model and the BSA transport model

Hong Zhuang,Xiaoliang Zhang,Sijia Wu,Chen Mao,Yaxi Dai,Pang Yong,Xiaodi Niu

Hesperidin (HE), a significant flavonoid polyphenolic compound present in citrus plants, exhibits diverse pharmacological effects. Considering the crucial involvement of biological membranes and transporter proteins in the transportation and biological processes of HE, it becomes essential to comprehend the potential mechanisms through which HE interacts with membranes and transporter proteins. In order to simulate the process of active molecule transport, a cell membrane model consisting of 1,2-dipalmitoyl-n- glycero -3-phosphatidylcholine (DPPC) and a transporter protein model of bovine serum albumin (BSA) were employed for investigation. The present study aimed to investigate the mechanism of action of hesperidin (HE) in DPPC and BSA using fluorescence quenching, Fourier transform infrared spectroscopy (FTIR), and differential scanning calorimetry (DSC). The localization and interaction of HE within liposomes were also elucidated. Furthermore, the binding of BSA and HE was analyzed through UV/Vis absorption spectroscopy, fluorescence spectroscopy, infrared spectroscopy, and computational biology techniques. Computational biology analysis revealed that the binding between HE and BSA primarily occurred via hydrogen bonding and hydrophobic interactions. This study aimed to investigate the role and mechanism of HE in the DPPC cell membrane model and the BSA transporter protein model, thereby offering novel insights into the action of HE in DPPC and BSA.

Understanding the Transepithelial Transport and Transbilayer Diffusion of the Antihypertensive Peptide Asn-Cys-Trp: Insights from Caco-2 Cell Monolayers and the DPPC Model Membrane

Sijia Wu,Ping Jiang,Xiaoliang Zhang,Chen Mao,Yaxi Dai,Hong Zhuang,Yong Pang

Understanding the transport mechanism of the peptide Asn-Cys-Trp (NCW) is crucial to improving its intestinal absorption and bioavailability. This study investigated the absorption of NCW through Caco-2 cell monolayers and its interaction with the DPPC bilayers. Results revealed that after a 3 h incubation, the Papp (AP-BL) and Papp (BL-AP) values of NCW at a concentration of 5 mmol/L were (22.24 ± 4.52) × 10–7 and (6.63 ± 2.31) × 10–7 cm/s, respectively, with the transport rates of 1.59 ± 0.32 and 0.62 ± 0.20%, indicating its moderate absorption. NCW was found to be transported via PepT1 and paracellular transport pathways, as evidenced by the significant impact of Gly-Pro and cytochalasin D on the Papp values. Moreover, NCW upregulated ZO-1 mRNA expression. Further investigation of the ZO-1-mediated interaction between NCW and tight junction proteins will contribute to a better understanding of the paracellular transport mechanism of NCW. The interaction between NCW and the DPPC bilayers was predominantly driven by entropy. NCW permeated the bilayers through electrostatic, hydrogen bonding, and hydrophobic interactions, resulting in increased fluidity, flexibility, and disorder as well as phase transition and phase separation of the bilayers.

DPPC-coated lipid nanoparticles as an inhalable carrier for accumulation of resveratrol in the pulmonary vasculature, a new strategy for pulmonary arterial hypertension treatment

Zerong Li,Wenmei Qiao,Chenghao Wang,Heqiao Wang,Mengchao Ma,Xinyu Han,Jingling Tang

In this study, we investigated the feasibility of dipalmitoylphosphatidylcholine-coated lipid nanoparticles (DPPC-LNs) as a carrier for preferential accumulation into lungs of Resveratrol (Res), a potentially promising drug for the treatment of pulmonary arterial hypertension (PAH). Res-loaded DPPC-LNs were prepared following a thin film hydration-ultrasonic dispersion technique using glyceryl monostearate as lipid core. DPPC can reduce the interactions between nanoparticles and pulmonary surfactant. The optimal formulation was prepared and characterized for physicochemical properties, storage stability and in vitro release profiles. The optimal formulation was evaluated for uptake by pulmonary arterial smooth muscle cells (PASMCs) using fluorescence microscopy. The efficacy of Res-loaded DPPC-LNs in reducing hyperplasia was tested in 5-HT induced proliferated PASMCs. The drug absorption profiles upon intratracheal administration were monitored in healthy rats. Optimized spherical DPPC-LNs – with mean size of 123.7 nm, zeta potential of –19.4 mV and entrapment efficiency of 94.40% – exhibited an 80% cumulative drug release over 48 h. Fluorescence microscopic study revealed an time-dependent enhancement of cellular uptake of Rh123-labeled DPPC-LNs by PASMCs. PASMC proliferation induced by 5-HT was significantly inhibited by Res-loaded DPPC-LNs. Optimized DPPC-LNs appeared to be safe when incubated with PASMCs. Besides, plasma and lung tissue data analysis indicated higher value of accumulation after intratracheal administration of Res-loaded DPPC-LNs in comparison with the intravenously dosed Res solution, indicating longer retention of Res in the lungs and their slower entry to the systemic blood circulation. DPPC-LNs could be a viable delivery system for site-specific treatment of PAH.

The transepithelial transport and membrane diffusion pathways of the walnut meal-derived antioxidant peptide YR-10 were investigated using Caco-2 monolayer cells and DPPC liposome models

Hui Ouyang,Jiaying Huo,Xiaojuan Xin,Ying Gao,Ping Wang,Xiongwei Yu,Shugang Li

The transport and absorption of antioxidant peptides are critical for their biological activity. The walnut meal-derived peptide YWSPNDEQFR (YR-10) has been shown to alleviate oxidative stress–induced liver injury, but its absorption mechanisms remain unclear. In this study, we employed a Caco-2 monolayer, DPPC liposomes, and molecular docking to characterize the transepithelial transport of YR-10 across the intestinal epithelium. YR-10 was absorbed through clathrin-mediated endocytosis and paracellular pathway, with significantly promotes the restoration of ZO-1 and closure protein gene expression, indicating its ability to restore the integrity of the epithelial barrier. Further research revealed that YR-10 binding to lipid bilayers was predominantly entropy-driven, involving electrostatic, hydrogen-bonding, and hydrophobic interactions that regulated membrane fluidity and order. These findings collectively elucidate the intestinal absorption mechanism of YR-10 and provide a theoretical basis for improving the bioavailability of walnut powder-derived bioactive peptides in functional food applications.

Surface Coating Approach to Overcome Mucosal Entrapment of DNA Nanoparticles for Oral Gene Delivery of Glucagon-like Peptide 1

Tianqi Nie,Zhiyu He,Yang Zhou,Jinchang Zhu,Kuntao Chen,Lixin Liu,Kam W. Leong,Hai-Quan Mao,Yongming Chen

Oral delivery of nucleic acid therapy is a promising strategy in treating various diseases because of its higher patient compliance and therapeutic efficiency compared to parenteral routes of administration. However, its success has been limited by the low transfection efficiency resulting from nucleic acid entrapment in the mucus layer and epithelial barrier of the gastrointestinal (GI) tract. Herein, we describe an approach to overcome this phenomenon and improve oral DNA delivery in the context of treating type II diabetes (T2D). Linear PEI (lPEI) was used as a carrier to form complexes with plasmid DNA encoding glucagon-like peptide 1 (GLP-1), a common target in T2D treatments. These nanoparticles were then coated with a mixture of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and 1,2-dimyristoyl-rac-glycero-3-methoxy poly(ethylene glycol)-2000 (DMG-PEG) to render the nanoparticle surface hydrophilic and electrostatically neutral. The surface-modified lPEI/DNA nanoparticles showed higher diffusivity and transport in the mucus layer of the GI tract and mediated high levels of transfection efficiency in vitro and in vivo. Moreover, these modified nanoparticles demonstrated high levels of GLP-1 expression for more than 24 h in the liver, lungs, and intestine in a T2D murine model after a single dose, as well as controlled blood glucose levels within a normal range for at least 18 h with repeatable therapeutic effects upon multiple dosages. Taken together, this work demonstrates the feasibility of an oral plasmid DNA delivery approach in the treatment of T2D through a facile surface modification to improve the mucus permeability and delivery efficiency of the nanoparticles.
