Sensitive Electrochemical Detection of DR1 Based on Electrodeposit AuNPs-Poly(thiophene-3-acetic acid) and Copolymer PLGA

Yaping Zhang,Gao Si,Yilong Wang,Zhendong Wang,Zutian Li,Haiyan Wei,Huaixia Yang,Fuchun Si,Xiaofei Li

The sensitive detection of down-regulator of transcription 1 (DR1) provides important information for early noninvasive diagnosis and treatment of Hashimoto thyroiditis (HT). In this study, a label-free electrochemical biosensor for DR1 detection was constructed based on electrodeposited gold nanoparticles (AuNPs) and poly(lactic-co-glycolic acid) (PLGA). The AuNPs were first modified to the electrode surface by electrodeposition to increase the specific surface area of the electrode, thus loading more target molecules to increase the sensitivity of the sensor. Then, a sandwich structure was formed on the electrode surface based on the specific recognition of antigen and antibody. PLGA was fixed on the electrode surface by binding with antibody 2 (Ab2) through amide bond to further improve the sensitivity of the sensor. Induced by the dual signal amplification, the sensor has a linear detection range of 50 fg·ml−1 to 50 ng·ml−1, with a detection limit as low as 19.4 fg·ml−1. Moreover, the sensor has good selectivity, specificity, and anti-interference capability, which holds great potential for the early diagnosis of HT and analysis of other disease markers.

Sensitive electrochemical detection of DR1 based on gold nanoparticle-modified MoS2 and hyaluronic acid-based thionine

Yaping Zhang,Gao Si,Zhendong Wang,Yilong Wang,Xiaojing Cui,Huaixia Yang,Fuchun Si,Yanjiu Liu

The analysis of down-regulator of transcription 1 (DR1) offers significant information for the rapid and non-invasive diagnosis of Hashimoto's thyroiditis (HT). In this study, we report a novel dual-signal amplification electrochemical biosensor for the sensitive detection of DR1. Gold nanoparticle (AuNP)-modified molybdenum disulfide (MoS2@AuNPs), which has extremely strong electron transfer ability and abundant binding sites, is first modified on an electrode surface as a substrate material to implement the first signal amplification. After the formation of the sandwich structure based on the specific recognition of antigens and antibodies, the electroactive molecules hyaluronic acid-based thionine (HA@Thi) are introduced to achieve the second signal amplification. Using this dual-signal amplification strategy, the proposed biosensor achieves a linear range of 1 × 10−4–1 × 102 ng mL−1 with a low detection limit of 10.99 fg mL−1. In addition, the electrochemical biosensor has high selectivity and good stability, and is applicable to the assay of DR1 in the presence of complex biological matrices, which is expected to provide a scientific approach for the clinical application of serum DR1 monitoring. More importantly, our method may extend the application of protein-based biosensors in disease diagnosis techniques.

A novel electrochemical biosensor based on WO3@AuNPs and HBP@BIBB macromolecule-triggered ATRP for DR1 detection

Xiaojing Cui,Yaping Zhang,Hanmeng Zhang,Huaixia Yang,Yanju Liu,Fuchun Si

The sensitive detection of down-regulator of transcription 1 (DR1) plays an important role in the early diagnosis and treatment of Hashimoto's thyroiditis (HT). Herein, we report a novel electrochemical sensor based on the nanocomposite of tungsten oxide–gold nanoparticles (WO3@AuNPs) and hyperbranched polymer–2-bromo-isobutyryl bromide (HBP@BIBB) initiated atom transfer radical polymerization (ATRP) for the ultrasensitive assay of DR1. Monolayer WO3 nanosheets, which have high conductivity, were first compounded with AuNPs to obtain the WO3@AuNP nanocomposite. WO3@AuNPs can not only increase the conductivity of the electrode but also provide more active sites for biological molecules, thereby effectively amplifying the output signal. By introducing macroinitiator HBP@BIBB into the ATRP reaction, more electroactive molecules were introduced into the sensor, further amplifying the output signal and making the sensor ultra-sensitive. The optimized sensor demonstrated a wide linear range of 1 × 10−4 –1 × 102 ng mL−1 with a low detection limit of 2.91 fg mL−1. Moreover, the electrochemical biosensor has high selectivity, good stability, and strong anti-interference ability, and is suitable for the determination of DR1 in complex biological matrices. In conclusion, this electrochemical biosensor shows great potential in biomarker detection.

A novel electrochemical biosensor based on dual signal amplification of CMK-3@AuNPs and ATRP for DR1 detection

Fuchun Si,Xiaojing Cui,Yaping Zhang,Yifan Li,Huaixia Yang,Yanju Liu

Down-regulator of transcription 1 (DR1) is a crucial biomarker closely related to Hashimoto's Thyroiditis (HT). In this study, a highly selective electrochemical biosensor for ultrasensitive detection of DR1 was prepared by combining nanomaterials CMK-3 @AuNPs and atom transfer radical polymerization (ATRP) signal amplification strategy for the first time. Based on the specific recognition of antigen and antibody, the sensor has strong selectivity. CMK-3 @AuNPs can not only increase the conductivity of the electrode, but also provide more active sites for biological molecules, so as to effectively amplify the output signal. The FMMA monomers were polymerized on the electrode surface through ATRP reaction, which made the electrochemical signal increased significantly. Under optimal conditions, the fabricated electrochemical biosensor showed a wide detection range of 5 × 10 -4 ng/mL ∼ 5 × 10 2 ng/mL with a detection limit as low as 0.159 pg/mL. In addition, the electrochemical biosensor has good stability and anti-interference ability. Therefore, the sensor has broad prospects in the detection of biomarkers.
