Titanium dioxide (TiO 2 ) nanoparticle has good photo-/sono-catalytic features, the reunion of this particle in solution-phase generally limits the extensive biomedical application. In the present study, the aggregation of TiO 2 nanoparticles was alleviated by facile fabrication under different pH conditions. A novel TiO 2 nanocomposite was further synthesized by properly conjugation with trace amount of DVDMS sensitizer (named DFT). The characterization, sonoactivity, as well as the antibacterial efficiency were specially evaluated. The results showed that the sonochemical activity of DFT was greatly improved as compared with the simple surface modification of TiO 2 (F-TiO 2 ) and free DVDMS, regarding to the hydroxyl radicals and singlet oxygen yields using the same ultrasound exposure. Moreover, ultrasonic stimulation of DFT exhibited excellent bacterial eradication, with up to 92.41% of killing efficiency in S. aureus . The flow cytometry analysis indicated an increased intracellular ROS and membrane disturbance by combination of DFT and ultrasound. The findings suggest that the proper fabrication and DVDMS incorporation greatly improved the sonocatalytic process of TiO 2 , and the ultrasound based biomedical applications of DFT deserve future deep investigation.

Upon harnessing low-intensity ultrasound to activate sonosensitizers, sonodynamic therapy (SDT) induces cancer cell death through the reactive oxygen species (ROS) mediated pathway. Compared with photodynamic therapy (PDT), SDT possesses numerous advantages, including deeper tissue penetration, higher accuracy, fewer side effects, and better patient compliance. Sinoporphyrin sodium (DVDMS), a sonosensitizer approved by the FDA, has drawn abundant attention in clinical research, but there are some deficiencies. In order to further improve the efficiency of DVDMS, many studies have applied self-assembly nanotechnology to modify it. Furthermore, the combined applications of SDT/chemodynamic therapy (CDT) have become a research hotspot in tumor therapy. Therefore, we explored the self-assembly of nanoparticles based on DVDMS and copper to combine SDT and CDT. A cost-effective sonosensitizer was synthesized by dropping CuCl2into the DVDMS solution with the assistance of PVP. The results revealed that the nanostructures could exert excellent treatment effects on tumor therapy and perform well for PET imaging, indicating the potential for cancer theranostics. In vitro and in vivo experiments showed that the nanoparticles have outstanding biocompatibility, higher ROS production efficiency, and antitumor efficacy. We believe this design can represent a simple approach to combining SDT and CDT with potential applications in clinical treatment and PET imaging. Keywords:sonodynamic therapy;chemodynamic therapy;theranostics;DVDMS;self-assembly

Sonodynamic therapy (SDT) has attracted great interest in the field of cancer therapy because of its non-invasiveness, deep penetration, and spatiotemporal controllability. However, the sonodynamic effect is severely hindered by hypoxia and a high glutathione (GSH) level in tumor microenvironment. In this work, a new type of nanohybrid sonosensitizer is designed, which incorporates several prominent advantages of organic, inorganic, and natural sonosensitizers for imaging-guided SDT and ferroptosis induction. As an endogenous transporter, hemoglobin (Hb) has dual functions of oxygen and iron supplement. Apparently, Hb can transport iron to the tumor site for Fe-dependent ferroptosis, yet this oxygen carrier is able to enhance the sensitivity of tumor cells to oxygen-driven SDT. We innovatively hybridized Hb, sinoporphyrin sodium (DVDMS), and titanium dioxide (TiO 2 ) to prepare an integrated nano-oxygen carrier (designated as HDT) for multifarious cancer theranostics, which not only attained satisfactory sonosensitization of DVDMS/TiO 2 but also achieved oxygen-boosted SDT and potent ferroptosis via Hb. Under ultrasound irradiation, the oxygenation of HDT markedly amplified the generation of reactive oxygen species (ROS) in hypoxic tumors, promoted the valence transition of Hb to accelerate Fenton reaction, and led to ferroptosis. This strategy virtually eradicated tumors in situ and exhibited immunoregulatory potential for augmenting anti-tumor effects in vivo and in vitro through ROS formation, O 2 self-supplement, GSH depletion, and lipid peroxidation. Collectively, HDT demonstrated excellent dual-modal imaging performance, providing a valuable platform for photoacoustic/fluorescence-guided SDT and ferroptosis induction in tumor cells.
