Dalbergia odoriferais widely used to treat cardiovascular diseases. Our research group found thatDalbergia odoriferavolatile oil has a good anti-myocardial ischemic effect, and its main pharmacodynamic components are trans-nerolol and its oxides. However, the exact mechanisms underlying this effect have not yet been elucidated. This study aimed to explore the potential myocardial protective effects of trans-nerolol and its underlying molecular mechanisms. Molecular docking was used to predict and visualize the possible mechanism of the anti-apoptotic myocardial protection by trans-nerolol. The myocardial protective effect of trans-nerolol was evaluated by observing pathological injury, myocardial enzyme levels, oxidation, antioxidant levels, and the expression of related proteins. Molecular docking results showed that trans-nerolol binds closely to cytochrome C (Cytc) and apoptosis-related proteins, suggesting that it may play a role in interacting with these target proteins. The results showed that pre-treatment with dose-dependent trans-nerolol significantly mitigated the myocardial histological damage; decreased lactate dehydrogenase (LDH), creatinine kinase (CK), alanine transaminase (ALT), and aspartate transaminase (AST) levels; reduced nitric oxide (NO) production, hydrogen peroxide (H2O2), and lipid peroxide (LPO); and increased the total antioxidant content (T-AOC), glutathione (GSH), catalase (CAT), and superoxide dismutase (SOD) activities compared with the model group. In addition, dose-dependent trans-nerolol significantly increased the Na+-K+-ATPase and Ca2+-Mg2+-ATPase levels. Moreover, trans-nerolol markedly reduced the endogenous and external apoptotic pathways; downregulated the protein expression of Cytc, apoptotic protease activating factor-1 (Apaf1), Fibroblast-associated (Fas), Cysteine-aspartate protease 3 (Caspase3), Cysteine-aspartate protease 8 (Caspase8), and Cysteine-aspartate protease 9 (Caspase9); and upregulated the expression of Heat shock protein 70 (Hsp70) and B-cell lymphoma-2 (Bcl-2). These data indicate that trans-nerolol exerts protective effects against myocardial ischemia (MI), and its mechanism is associated with the suppression of the Cytc- and caspase-signaling pathways. Trans-nerolol has a therapeutic effect on MI, and its mechanism of action is related to its anti-apoptotic effect. These results suggest thatDalbergia odoriferahas a potential role to be developed as an MI-promoting therapeutic agent.

Dalbergia odorifera, a critically endangered tree species, produces heartwood containing a vast variety of flavonoids. This heartwood, also known as Chinese rosewood, has high economic and medicinal value, but its formation takes several decades. In this study, we showed that discolored wood induced by pruning displays similar color, structure, and flavonoids content to those of natural heartwood, suggesting that wounding is an efficient method for inducing flavonoid production inD. odorifera. Transcriptome analysis was performed to investigate the mechanism underlying wounding-induced flavonoids production inD. odoriferaheartwood. Wounding upregulated the expression of 90 unigenes, which covered 19 gene families of the phenylpropanoid and flavonoid pathways, including PAL, C4H, 4CL, CHS, CHI, 6DCS, F3’5’H, F3H, FMO, GT, PMAT, CHOMT, IFS, HI4’OMT, HID, IOMT, I2’H, IFR, and I3’H. Furthermore, 47 upregulated unigenes were mapped to the biosynthesis pathways for five signal molecules (ET, JA, ABA, ROS, and SA). Exogenous application of these signal molecules resulted in the accumulation of flavonoids in cell suspensions ofD. odorifera, supporting their role in wounding-induced flavonoid production. Insights from this study will help develop new methods for rapidly inducing the formation of heartwood with enhanced medicinal value. Keywords:Dalbergia odorifera;flavonoid biosynthesis;heartwood;mechanical wounding;signals;transcriptome

Ethnopharmacological relevance The heart wood of Dalbergia odorifera is a Chinese herbal medicine commonly used for the treatment of various ischemic diseases in Chinese medicine practice. Aim of the study In this study, therapeutic angiogenesis effects of the Dalbergia odorifera extract (DOE) were investigated on transgenic zebrafish in vivo and human umbilical vein endothelial cells (HUVECs) in vitro . Materials and methods The pro-angiogenic effects of DOE on zebrafish were examined by subintestinal vessels (SIVs) sprouting assay and intersegmental vessels (ISVs) injury assay. And the pro-angiogenic effects of DOE on HUVECs were examined by MTT , scratch assay, protein chip and western blot . Results In the in vivo studies, we found that DOE was able to dose-dependently promote angiogenesis in zebrafish SIVs area. In addition, DOE could also restore the injury in zebrafish ISVs area and upregulate the reduced mRNA expression of VEGFRs including kdr , kdrl and flt-1 induced by VEGF receptor kinase inhibitor II (VRI). In the in vitro studies, we observed that DOE promoted the proliferation, migration of HUVECs and also restored the injury induced by VRI. Moreover, protein chip and western blot experiments showed the PI3K/MAPK cell proliferation/migration pathway were activated by DOE. Conclusions DOE has a therapeutic effects on angiogenesis, and its mechanism may be related to adjusting the VEGFRs mRNA and activation of PI3K/MAPK signaling pathway. These results suggest a strong potential for Dalbergia odorifera to be developed as an angiogenesis-promoting therapeutic.

Cutaneous squamous cell carcinoma (cSCC) lacks effective and well-tolerated pharmacological options for long-term management, highlighting the need for multi-target natural agents. In this study, we hypothesized that a flavonoid-rich extract from Dalbergia odorifera exerts (DOE) anti-cSCC effects by regulating apoptosis-related pathways. To test this hypothesis, we employed an integrated strategy combining UPLC-based chemical profiling, network pharmacology prediction, and in vitro and in vivo validation. UPLC analysis identified five major flavonoids in DOE-Luteolin, Naringenin, Butein, Liquiritigenin, and Formononetin. Network pharmacology predicted the involvement of key pathways, including PI3K/AKT, MAPK, and EGFR. Experimental validation, however, was focused on apoptosis-related markers Bax and Bcl-2, providing partial support for the predicted mechanisms. In vitro, DOE significantly reduced cell viability and colony-forming ability. Migration was decreased by more than 80%, and apoptosis increased substantially at higher doses. These findings were confirmed by qRT-PCR and Western blot analyses, which showed a downregulation of Bcl-2 and upregulation of Bax, consistent with the proapoptotic mechanism predicted by network pharmacology. In vivo, using a DMBA/croton oil-induced cSCC mouse model, DOE administration resulted in dose-dependent tumor inhibition and improved body weight loss, thymus, and spleen indices. DOE, as a flavonoid extract, significantly inhibits cell proliferation, migration, and promotes apoptosis in cSCC, positioning it as a promising candidate for further development as a complementary or adjuvant therapeutic strategy.

An efficient microwave-assisted aqueous two-phase extraction (MA-ATPE) technique was developed to simultaneously extract and enrich genistein and biochanin A from Dalbergia odorifera T. Chen leaves. Three salts were investigated, and dipotassium hydrogen phosphate was selected to form aqueous two-phase system (ATPS). Box–Behnken Design combined with response surface methodology was applied to optimize the operating parameters of MA-ATPE. The optimal conditions were as follows: salt concentration 20%, absolute alcohol concentration 25%, extraction temperature 45.5 °C, extraction time 10 min and irradiation power 300 W. Under the optimized condition, the extraction yields of genistein and biochanin A reached 1.034 mg/g DM and 2.023 mg/g DM, respectively. Compared with microwave assisted extraction (MAE) and heat reflux extraction (HRE), the contents of genistein and biochanin A in the extracts reached 12.966 mg/g extract and 25.526 mg/g extract, which were 2.48- and 2.30-fold to that of MAE, 2.64- and 2.80-fold to that of HRE. Furthermore, the antioxidant activities of extracts from MA-ATPE, MAE and HRE were 0.342, 0.389, 0.411 mg/mL (IC 50 ), according to the DPPH radical-scavenging assay. Moreover, the phase-forming salt can be recyclable. In conclusion, MA-ATPE was an efficient, eco-friendly and economical technique, which represented an excellent alternative for the extraction of bioactive compounds from plant.

Background and purpose : Cerebral ischemia-reperfusion injury stands as a significant contributor to disability globally, highlighting the urgent requirement for therapeutic interventions aimed at repairing the central nervous system. Worldwide interest in the use of Chinese traditional medicine for health promotion and disease prevention has increased significantly. Recently, Dalbergia odorifera T.C. Chen leaf has been listed in local food safety standard of Hainan Province (standard number: DBS46/003‒2021). Previous research findings from our team have demonstrated Dalbergia odorifera T.C. Chen leaf extract lead the genuine alleviation of brain edema following cerebral ischemia-reperfusion injury, albeit with the underlying mechanism still remaining elusive. Study design and methods : D. odorifera leaf extract were prepared via ethanol extraction. We investigated the protective effects of D. odorifera leaf extract administered via gastric infusion using the middle cerebral artery occlusion/reperfusion model. The assessment covered the impacts on infarction volume in the brain, permeability of the blood-brain barrier, and neurological evaluations, which were determined by means of 2,3,5-triphenyltetrazolium chloride staining and Evans blue dye. Furthermore, to assess cerebral inflammation and neuronal apoptosis, techniques such as hematoxylin and eosin (H&E) staining and Nissl staining were applied. In order to delve deeper into the alterations occurring in brain tissue following treatment with D. odorifera leaf extract, we employed advanced methodologies including single-cell RNA sequencing (scRNA-seq), immunofluorescence analysis, and cerebral blood flow assessments. Assisted cellular experimental data help verify that the performed accuracy of animal experimental results. The energy metabolic capacity on ATP production, oxygen-consuming capacity, and glucose uptake capacity was estimated with intracellular ATP measurement, 2-NBDG uptake, glutamic acid assay, and oxygen consumption test method. Results : Following cerebral ischemia-reperfusion, neutrophils migrate into the blood vessels at the ischemic site, with their presence closely linked to the severity of neuroinflammatory damage. Administration of D. odorifera leaf extract was found to alleviate cerebral edema, augment neurological function, and diminish infarct size after cerebral ischemia-reperfusion injury. scRNA-seq revealed that D. odorifera leaf extract enhance the disturbance of interaction between microglial cell and neutrophil. Further insights from scRNA-seq studies indicate that D. odorifera leaf extract enhances phagocytosis function by reprogramming energy metabolism, shifting it from glycolysis to oxidative phosphorylation. Additionally, D. odorifera leaf extract supports oxidative phosphorylation, leading to increased ATP production, which in turn fuels microglial phagocytosis. Consequently, this mechanism enhances the phagocytic activity and clearance of neutrophils in the ischemic brain. Activation of these processes was linked and dependent on GAS6-AXL, MERTK, TYRO3 signaling pathway. To sum up, our findings reveal that D. odorifera leaf extract modulates energy metabolism pathways to bolster microglial-mediated phagocytosis of neutrophils subsequent to ischemic stroke. These discoveries may pave the way for novel therapeutic approaches in stroke management by focusing on the modulation of microglial metabolic reprogramming. Conclusion : Our results demonstrated that the neutrophil aggregation pathological processes were reduced greatly by D. odorifera leaf extract under on cerebral ischemia reperfusion. The regulation mechanism of D. odorifera leaf extract could be exerted by altering the functional states of microglial differentiation, and concurrently facilitates the phagocytosis of neutrophils by microglia, thereby leading to Simultaneously, it enhances the phagocytic activity of microglia toward neutrophils, thereby contributing to a decreased accumulation of neutrophils within the brain. Additional research has shown that D. odorifera leaf extract reprogramme energy metabolism to promote oxidative phosphorylation, thereby furnishing microglial phagocytosis with ample energy. Above results add to the present stage evidences that D. odorifera leaves has better application prospects in cerebral ischemia‒reperfusion injury.

In this study, green and efficient deep eutectic solvent-based negative pressure cavitation-assisted extraction (DES-NPCE) followed by macroporous resin column chromatography was developed to extract and separate four main isoflavonoids, i.e. prunetin, tectorigenin, genistein and biochanin A from Dalbergia odorifera T. Chen leaves. The extraction procedure was optimized systematically by single-factor experiments and a Box-Behnken experimental design combined with response surface methodology. The maximum extraction yields of prunetin, tectorigenin, genistein and biochanin A reached 1.204, 1.057, 0.911 and 2.448 mg/g dry weight, respectively. Moreover, the direct enrichment and separation of four isoflavonoids in DES extraction solution was successfully achieved by macroporous resin AB-8 with recovery yields of more than 80%. The present study provides a convenient and efficient method for the green extraction and preparative separation of active compounds from plants.
