The toxicology of microplastics in combination with other pollutants has attracted widespread attention. In this study, zebrafish were exposed to 3 mg/L polystyrene microplastic, 0.2 mg/L phenanthrene, and a combination of both. Zebrafish microplastic uptake, phenanthrene accumulation, antioxidant-associated enzyme activity and related gene expression, immune-associated gene expression, and the gut microflora were measured after 12 and 24 days of exposure. Phenanthrene and microplastic accumulation increased with exposure time and was also greater in the combined exposure group than in the single exposure group. Combined analysis of antioxidant enzyme activity and immune and antioxidant-related genes shows that exposure alone causes oxidative stress in zebrafish, ultimately increasing immunity and the expression of oxidative stress genes, while combined exposure exacerbates these changes. Fusobacteria decreased and Proteobacteria and Bacteroidetes increased in the three exposure groups of gut microorganisms. Overall, our study demonstrates that microplastics enhance the toxicity of phenanthrene and that the two have a synergistic effect.

Monosultap (Mon) is a broad-spectrum insecticide used in agricultural production to control stem borers in rice fields . Currently, little evidence shows how Mon affects notochord development in zebrafish ( Danio rerio ). In our study, zebrafish embryos were exposed to 0.25, 0.5, and 0.75 mg/L Mon to determine the effects of different concentrations of Mon on notochord development. Mon exposure reduced the body length, decreased the heart rate and hatchability, and induced notochord deformity in zebrafish. The effects of Mon exposure on the internal organization of the notochord and the structural abnormalities were determined based on histological staining of paraffinized tissue sections. Quantitative polymerase chain reaction (qPCR) and in situ hybridization findings revealed that the expression levels of genes related to notochord development ( shha, col2a, and ptch2 ) showed an increasing trend in a concentration-dependent manner. An abnormal increase of apoptosis and cell proliferation in some parts of the notochord suggested that Mon exposure could cause developmental abnormality of the notochord. This study revealed the toxicity of Mon in notochord development. Our findings provide information in assessing the risk of Mon to the ecological environment and human health.

Triclosan (TCS) exposure has widely adverse biological effects such as influencing biological reproduction and endocrine disorders. While some studies have addressed TCS-induced expression changes of miRNAs and their related down-stream target genes, no data are available concerning how TCS impairs miRNA expression leading us to study up-stream regulating mechanisms. Four miRNAs (miR-125b, miR-205, miR-142a and miR-203a) showed differential expression between TCS-exposure treatments and the control group; their functions mainly involved fatty acid synthesis and metabolism. TCS exposure led to the up-regulation of mature miR-125b that was concomitant with consistent changes in pri-mir-125b-1 and pri-mir-125b-3 among its 3 pri-mir-125bs. Up-regulation of miR-125b originated from direct shear processes involving the two up-regulated precursors, but not pri-mir-125b2. Increased expression of pri-mir-125b-1 and pri-mir-125b-3 resulted from nfe2l2- and c/ebpα-integration with positive control elements of promoters for the two precursors. The overexpression of transcriptional factors, nfe2l2 and c/ebpα, initiated the promoter activity for the miR-125b precursor. CpG islands and Nfe2l2 were involved in constitutive expression of mir-125b-1 and mir-125b-3 . The activities of two promoter regions, −487 to −1 bp for pri-mir-125b1 and −1327 to +14 bp for pri-mir-125b-3 having binding sites for NFE2 and Nfe2l2/MAF:NFE2, were higher than other regions, further demonstrating that the transcriptional factor Nfe2l2 was involved in the regulation of pri-mir-125b1 and pri-mir-125b-3. TCS’s estrogen activity resulted from its effects on GPER, a novel membrane receptor, rather than the classical ERα and ERβ. These results explain, to some extent, the up-stream mechanism for miR-125b up-regulation, and also provide a guidance to future mechanistic study on TCS-exposure.

Dibutyl phthalate (DBP) is a commonly used plasticizer that is frequently detected in water samples due to its widespread use. Titanium dioxide nanoparticles (n-TiO 2 ) have been found to enhance the harmful effects of organic contaminants by increasing their bioavailability in aquatic environments. However, the combined toxic effects of DBP and n-TiO 2 on aquatic organisms remain unclear. This study aimed to investigate the neurotoxicity of DBP and n-TiO 2 synergistic exposure during the early life stage of zebrafish. The results of the study revealed that co-exposure of DBP and n-TiO 2 led to an increase in deformities and a significant reduction in the active duration of zebrafish larvae. Furthermore, the co-exposure of DBP and n-TiO 2 resulted in elevated levels of oxidative stress and altered gene expression related to neurodevelopment and apoptosis. Notably, n-TiO 2 exacerbated the oxidative damage and apoptosis induced by DBP alone exposure. Additionally, co-exposure of the 1.0 mg/L DBP and n-TiO 2 significantly affected the expression of genes associated with neurodevelopment. Moreover, disturbances in amino acid metabolism and interference with lipid metabolism were observed as a result of DBP and n-TiO 2 co-exposure. In general, n-TiO 2 aggravated the neurotoxicity of DBP in the early life stage of zebrafish by increasing oxidative stress, apoptosis, and disrupting amino acid synthesis and lipid metabolism. Therefore, it is essential to consider the potential risks caused by DBP and nanomaterials co-existence in the aquatic environment.

Strobilurin fungicides are used globally and have been detected in microgram per liter concentrations in aquatic environments. Here, we determined the potential toxicity of four commonly used strobilurins (azoxystrobin, kresoxim-methyl, pyraclostrobin, trifloxystrobin) on mitochondrial function and locomotor activity of larval zebrafish at an environmentally relevant level. As the mode of action of strobilurins in fungi is binding to cytochrome bc1 in mitochondrial complex III, we evaluated exposure effects on mitochondrial oxidative phosphorylation of zebrafish, by measuring oxygen consumption rates, mitochondria-related enzyme activities , and transcripts levels for genes associated with the electron transfer chain and citric acid cycle. We found that 50 nM pyraclostrobin and trifloxystrobin lowered basal respiration, oligomycin-induced ATP respiration, and maximal respiration of embryos. Dysfunction in mitochondrial bioenergetics was associated with changes in mitochondrial complex III activity and transcripts of oxidative respiration and stress-related genes. Lower activity of complex III, and reduced cytb mRNA levels were hypothesized to contribute to reduced electron supply to complex IV and V. Both coxI and atp6 were up-regulated, suggesting a compensatory response to impaired oxidative respiration. Cluster analysis indicated that strobilurin-induced oxidative stress and cytb transcript were related to impaired oxidative phosphorylation. We also assessed larval behavior responses, as reduced ATP can affect activity. We observed that pyraclostrobin and trifloxystrobin induced hypoactive responses in zebrafish. At 50 nM, azoxystrobin and kresoxim-methyl exerted no effects on mitochondrial function nor locomotion of zebrafish. Studies such as this are important for determining sublethal toxicity to these fungicides, as widespread detection of strobilurins in aquatic environments suggests there is a potential for adverse effects in aquatic organisms .

Dicofol (DCF), a widely used organochlorine pesticide, is classified as a persistent organic pollutant (POP). However, its toxicological profile and the underlying mechanisms of action in vertebrates remain poorly understood. In this study, exposure of zebrafish ( Danio rerio ) to sublethal concentrations of DCF led to notable behavioral abnormalities in both larvae and adults, effects that were accompanied by increased apoptosis in brain tissues. Additionally, molecular docking analysis identified the α3-subunit containing Na + /K + -ATPase (ATP1A3A) as a potential target of DCF; this finding was corroborated by significant inhibition of Na + /K + -ATPase activity observed in brain tissue samples upon DCF treatment. Further investigation demonstrated elevated intracerebral calcium levels, enhanced oxidative stress, downregulation of chaperones involved in protein folding, increased protein ubiquitylation, and accumulation of phosphorylated microtubule-associated tau protein in the brains of DCF-treated adults. Transcriptomic profiling corroborated these findings, revealing activation of apoptotic pathways, dysregulation of calcium homeostasis, impairment of ROS clearance, and aberrant expression of genes related to neuronal function. In summary, our findings demonstrate that DCF exposure induces toxicity in zebrafish by disrupting multiple signaling cascades, culminating in neuronal apoptosis and neurotoxicity. These findings underscore the potential ecological risks and human health implications associated with DCF exposure. Environmental implication Exposure to the persistent organic pollutant dicofol (DCF) induces neurotoxicity in zebrafish by disrupting multiple signaling cascades. Such disruption triggers calcium dysregulation, oxidative stress, protein misfolding, phosphorylated tau protein accumulation, and neuronal apoptosis, which are hallmarks of neurodegeneration. Given the evolutionary conservation of these toxicity mechanisms across vertebrates, DCF may pose potential ecological risks to other aquatic species, suggesting that DCF contamination in water sources could represent an unrecognized environmental risk factor for human neurological health. These findings warrant regulatory attention to this legacy pesticide.

Excessive carbohydrate intake leads to metabolic disorders in fish. However, few literatures have reported the appropriate carbohydrate level for zebrafish, and the metabolic response to dietary carbohydrate remains largely unknown in zebrafish. This study assessed the responses of zebrafish and zebrafish liver cell line (ZFL) to different carbohydrate levels. In vivo results showed that ≥30% dietary dextrin levels significantly increased the plasma glucose content, activated the expression of hepatic glycolysis-related genes, and inhibited the expression of hepatic gluconeogenesis-related genes in zebrafish. Oil red O staining, triglyceride content, and Hematoxylin-Eosin staining results showed that dietary dextrin levels of ≥30% significantly increased lipid accumulation and liver damage, as well as processes related to glycolipid metabolism and inflammation in zebrafish. In ZFL, the transcription factor sterol regulatory element binding protein-1c signal intensity, 4,4-difluoro-1,3,5,7,8-pentamethyl-4-bora-3a,4a-diaza-s-indacene (BODIPY 493/503) signal intensity, and triglyceride content were also significantly increased when incubated in high glucose, along with abnormal glycolipid metabolism and increased inflammation-related genes. In conclusion, we demonstrated that the maximum dietary carbohydrate level in adult zebrafish should be less than 30%. Excess dietary carbohydrates (30%–50%) caused hepatic steatosis and damage to zebrafish, similar to that seen in aquaculture species. Thus, this study assessed responses to different carbohydrate levels in zebrafish and illustrated that zebrafish is an optimal model for investigating glucose metabolism in some aquatic animals.

Zebrafish (Danio rerio) has been a widely used vertebrate animal model in developmental biology and behavioral neuroscience, but knowledge about some of its basic behaviors, for example, light/dark preference, is still controversial. Appropriate preference for light or dark environments can be crucial for an animal's survival, so we hypothesize that zebrafish may have its light/dark preference varied with the circadian clock. In this present work, we tested the hypothesis by recording the light/dark preference in a two-compartment tank continuously for over 60 h. We found that the light avoidance of fish generally increases with time from morning (8:00am) to midnight (2:00am), and then decreases with time from midnight (2:00am) to morning (8:00am), exhibiting a clear circadian-like trend in the light/dark preference. As melatonin can mediate circadian clock output and promote sleep in zebrafish, by adding extra melatonin at around 9:00am on the third day, the mean proportion of time fish spent in the dark area was increased significantly. Our results demonstrate that the circadian clock plays a significant role in regulating the light/dark preference in zebrafish, which provides valuable insights into understanding the metabolism mechanism underlying the neurobehavior, and facilitate further studies related to the neurobiology of normal and pathological behavior.

Despite the known hazards to wildlife, plastic waste to waterways and oceans continues to increase at alarming rates every year. Nanoplastics are a breakdown product of plastic debris in the environment (0.1–100 nm). Their small size renders them bioavailable, and they are likely to accumulate/adsorb persistent organic pollutants, which could increase their toxicity. In this study, we evaluated the effects of nanoplastics and phenanthrene on the early life stages of zebrafish. Zebrafish embryos were exposed to fluorescent nanoparticles and observed with fluorescence microscopy. Polystyrene nanoplastics (80 nm) attached to the embryonic chorionic membrane and were ingested when zebrafish reached 144 h after fertilization, the point at which they first open their mouth, as observed by the fluorescence in their intestines. Exposure alone or in combination with phenanthrene resulted in zebrafish pericardial edema, spinal curvature, increased embryonic heart rate, shortened body length, and increased mortality. Co-exposure to 5 mg/L nanoplastics and 0.1 mg/L phenanthrene caused more negative effects on length, teratogenicity and mortality of zebrafish embryos than the corresponding concentrations of single exposure. This study provides clear evidence that nanoplastics can be ingested by zebrafish larvae, thus future work to assess the toxicological effects is important to marine wildlife management.

The succinate dehydrogenase inhibitor (SDHI) fungicides have been extensively used in agriculture, and some of their potential ecological risks to aquatic organisms have been demonstrated recently. Sedaxane (SDX) is a broad spectrum SDHI fungicide. Despite being extensively used in environment, little is known about its potential developmental effect in zebrafish embryo. This study examined the effects of which SDX triggered in zebrafish through embryonic development assessments. Results show that SDX induced mortality, hatch delay and failure in zebrafish embryos, which were concentration dependent. In addition, several developmental abnormalities were observed at 2 mg/L and higher concentrations, including edema, microcephaly, body deformation, and swim bladder not fully inflated. SDX exposure influenced reactive oxygen species , malondialdehyde, peroxidase, glutathione S-transferase, superoxide dismutase and glutathione in live larvae, which indicated that oxidative stress was caused in zebrafish. Furthermore, SDX induced decrease of succinate dehydrogenase activity in zebrafish larvae. These results provide toxicological data of SDX on developing zebrafish embryo, which could be help for further understanding the potential risk on the environment.

Glyphosate (GLY) induces developmental toxicity in fish, but research on the toxicity mechanism is limited. In this study, zebrafish embryos were exposed for 120 hpf to 0.7, 7, and 35 mg L -1 GLY. The results show that GLY treatment induced developmental toxicity in the fish, including premature hatching, reduced heartbeats, pericardial and yolk sac oedema, swim bladder deficiency, and shortened body length, which was possibly due to a significantly decreased triiodothyronine (T3)/thyroxine (T4) ratio and the abnormal expression patterns of hypothalamic-pituitary-thyroid (HPT) ( crh , tshβ , tr α , tr β , and t tr ) and growth hormone/insulin-like growth factor (GH/IGF) axis-related genes ( gh , ghrα , ghrβ , igf1 , igf1rα , and igf1rβ ) in larvae exposed to GLY. In addition, GLY exposure altered the levels of SOD and CAT, increased ROS, promoted malondialdehyde (MDA) content, and significantly altered the levels of endoplasmic reticulum (ER) stress signalling pathway factors ( perk , eif2α , gadd34 , atf4 , ire1α , xbp1 , atf6 , hspa5 , and chop ), suggesting that GLY treatment induced oxidative injury and ER stress in the larvae. Further research showed that treatment with a higher concentration of GLY upregulated the levels of iNOS, IL-1β, and TNF-α while inhibiting the expression of IL-10 and TGF-β, suggesting that GLY causes an inflammatory reaction in the larvae. In addition, we also found that apoptosis was induced in the larvae, which was determined by acridine orange staining and abnormal expression of p53 , caspase-3 , -8 , and -9 . Taken together, our results demonstrate that GLY exposure altered the T3/T4 ratio, disturbed the expression patterns of HPT and GH/IGF axis-related genes, and induced oxidative and ER stress, inflammatory reactions, and apoptosis in the zebrafish larvae. This investigation contributes to improved understanding of the developmental toxicity mechanism of GLY in fish.

The early development of zebrafish (Danio rerio) is a complex and dynamic physiological process involving cell division, differentiation, and movement. Currently, the genome and transcriptome techniques have been widely used to study the embryonic development of zebrafish. However, the research of proteomics based on proteins that directly execute functions is relatively vacant. In this work, we apply label-free quantitative proteomics to explore protein profiling during zebrafish’s embryogenesis, and a total of 5961 proteins were identified at 10 stages of zebrafish’s early development. The identified proteins were divided into 11 modules according to weighted gene coexpression network analysis (WGCNA), and the characteristics between modules were significantly different. For example, mitochondria-related functions enriched the early development of zebrafish. Primordial germ cell-related proteins were identified at the 4-cell stage, while the eye development event is dominated at 5 days post fertilization (dpf). By combining with published transcriptomics data, we discovered some proteins that may be involved in activating zygotic genes. Meanwhile, 137 novel proteins were identified. This study comprehensively analyzed the dynamic processes in the embryonic development of zebrafish from the perspective of proteomics. It provided solid data support for further understanding of the molecular mechanism of its development.
