Toxicity Response and Swimming Speed Regularity in Daphnia magna After Short-Term Exposure to Diuron

Feihu Qin,Nanjing Zhao,Gaofang Yin,Yunfei Luo,Tingting Gan

The agricultural production process contributes to the global issue of pesticide pollution. Based on the static toxicity test of diuron (DCMU) onDaphnia magna(D. magna) for EC50-48 h, a concentration range of 0.2 to 1 mg/L was set as sublethal concentrations, while lethal concentrations were set at 2 mg/L and 4 mg/L. This study analyzes the toxic response patterns of the swimming behavior indicators ofD. magnaexposed to different concentrations of DCMU. The results showed that the average speed (V) ofD. magnadecreased step by step with exposure time, regardless of exposure to sublethal concentration or lethal concentration. However, during the same short-term exposure period, the V ofD. magnaat lethal concentration was higher than that at sublethal concentration, which indicates that the swimming behavior ofD. magnaexposed to DCMU may be stimulated and accelerated. Compared to the control group, there is a statistically significant difference in the V ofD. magnaafter short-term exposure, especially showing an extremely significant difference after 5 min of exposure. Evidently, compared to the traditional 48 h static toxicity testing method, the swimming behavior indicators ofD. magnashow a more sensitive response to DCMU after 5 min of exposure, making it more suitable for rapid toxicity detection. By expanding the range of exposure concentrations, it was found that the V indicator ofD. magnaresponded significantly to a DCMU concentration of 0.05 mg/L after only 5 min of exposure, and a high degree of correlation was observed between the indicator and the exposure concentration. Through nonlinear fitting, the relationship between V and the dose–effect of DCMU toxicity presents an S-shaped curve, with R2> 0.9. Consequently, it becomes possible to study the dose–effect relationship between the changes in the swimming behavior indicators ofD. magnaand the stress concentration based on this theory. This further establishes a foundation for the development of comprehensive aquatic toxicity rapid detection technology based on the toxic response of swimming behavior indicators.

Daphnia magna and mixture toxicity with nanomaterials – Current status and perspectives in data-driven risk prediction

Diego Stéfani T. Martinez,Laura-Jayne A. Ellis,Gabriela H. Da Silva,Romana Petry,Aline M.Z. Medeiros,Hossein Hayat Davoudi,Anastasios G. Papadiamantis,Adalberto Fazzio,Antreas Afantitis,Georgia Melagraki,Iseult Lynch

The aquatic ecosystem is the final destination of most industrial residues and agrochemicals resulting in organisms being exposed to a complex mixture of contaminants. Nanomaterials (NMs) are being increasingly applied in many technologies and industrial sectors, so there is an increasing concern about the negative impacts of NMs in the environment after their interaction with co-contaminants. Consequently, mixture toxicology has been gaining attention in nanotoxicology recently. Usually, mixture toxicity or combined toxicity is estimated from the individual effects of the chemicals using the mathematical models of concentration addition (CA) or independent action (IA), however these models do not account for metabolic interactions between the chemicals, when they act in related metabolic pathways and molecular targets . As NMs unique physico-chemical properties make them highly reactive with a high surface area for adsorption, those models may not realistically estimate the toxicological effects of mixtures containing NMs. The co-exposition of NMs and other environmental contaminants (e.g., organic pollutants and heavy metals) may cause different mixture effects such as addition, synergism, antagonism, or even other complicated responses, including altered toxicokinetics/toxicodynamics, which vary according to the individual components properties, environmental exposure conditions, and the biological system. Therefore, the large number of factors that may influence the toxicity of a NM and contaminant mixture makes NMs mixture risk assessment a complex task. Daphnia magna are one of the most commonly used model species in nanotoxicology, including in mixture studies. It’s advantages include short generation time, small body sizes, ability to produce large populations rapidly, coupled with its completely mapped genome which allows the use of a multitude of omics techniques to understand the stress responses of daphnids to NMs and chemicals. Here, we analyse the toxicological effects of NMs and contaminant mixtures using Daphnia as a model organism, and discuss future perspectives for NMs-mixtures risk assessment focusing on harmonization of methodologies and application of data-driven science in mixture ecotoxicology.

The effect of microplastics on the depuration of hydrophobic organic contaminants in Daphnia magna: A quantitative model analysis

Wei Lin,Yu Li,Xiaoying Xiao,Fuqiang Fan,Jiakun Jiang,Ruifen Jiang,Yong Shen,Gangfeng Ouyang

Microplastics are emerging pollutants that can absorb large amounts of hydrophobic organic contaminants (HOCs). However, no biodynamic model has yet been proposed to estimate their effects on HOC depuration in aquatic organisms, where the HOC concentrations are time-varying. In this work, a microplastic-inclusive biodynamic model was developed to estimate the depuration of HOCs via ingestion of microplastics. Several key parameters of the model were redefined to determine the dynamic HOC concentrations. Through the parameterized model, the relative contributions of dermal and intestinal pathways can be distinguished. Moreover, the model was verified and the vector effect of microplastics was confirmed by studying the depuration of polychlorinated biphenyl (PCB) in Daphnia magna ( D. magna ) with different sizes of polystyrene (PS) microplastics. The results showed that microplastics contributed to the elimination kinetics of PCBs because of the fugacity gradient between the ingested microplastics and the biota lipids, especially for the less hydrophobic PCBs. The intestinal elimination pathway via microplastics would promote overall PCB elimination, contributing 37–41 % and 29–35 % to the total flux in the 100 nm and 2 μm polystyrene (PS) microplastic suspensions, respectively. Furthermore, the contribution of microplastic uptake to total HOC elimination increased with decreasing microplastic size in water, suggesting that microplastics may protect organisms from HOC risks. In conclusion, this work demonstrated that the proposed biodynamic model is capable of estimating the dynamic depuration of HOCs for aquatic organisms. The results can shed light on a better understanding of the vector effects of microplastics.

Micro- and nanoplastic-mediated phototransformation and bioaccessibility of fluorinated liquid crystal monomer in aquatic environments

Yiping Feng,Jingyi Wu,Wenhao Lao,Weibiao Ye,Danni Guo,Zhu Wang,Xiaowei Wu,Racliffe Weng Seng Lai

Micro- and nanoplastics are emerging pollutants that have attracted significant attention due to their potential to concentrate and transport coexisting organic pollutants in aquatic environments. Fluorinated liquid-crystal monomers (FLCMs) have also emerged as contaminants of concern, given their frequent occurrence, potential toxic effects, and propensity to co-occur with plastics in the environment. However, the influence of plastics on the environmental fate of FLCMs remains unclear yet. To address this knowledge gap, we investigated the accumulation of a key FLCM, 4-cyano-3-fluorophenyl 4-ethylbenzoate (CEB-F), on three common plastics, and examined the effects of nanoplastics on the phototransformation of CEB-F and its acute toxicity to Daphnia magna (D. magna). Our findings revealed that the adsorption capacity of CEB-F on different plastic materials followed the order: polystyrene (PS) < mixed cellulose ester (MCE) < polyamide (PA). The adsorption processes of CEB-F on the three plastics aligned more closely with the pseudo-first-order kinetic model and the Langmuir isotherm model, suggesting that the adsorption is primarily governed by physical diffusion. Theoretical calculations indicated that the adsorption of CEB-F on PS plastics is mainly driven by hydrophobic interactions. Additionally, PS nanoplastics (PSNPs) significantly enhanced the UV degradation of CEB-F, although the types of degradation intermediates did not change substantially, suggesting a limited impact on the degradation process and mechanism. Acute toxicity tests showed that PSNPs increased the toxicity of CEB-F to D. magna at lower concentrations, while the toxicity was reduced at higher concentrations. The obtained findings are of great significance to unraveling the plastic-mediated environmental fate and aquatic toxicity of FLCMs in natural waters.

A functional ruthenium(II) complex for imaging biothiols in living bodies

Zhiqiang Ye,Quankun Gao,Xin An,Bo Song,Jingli Yuan

A unique ruthenium(II) complex, [Ru(bpy)2(DNS-bpy)](PF6)2 [bpy: 2,2′-bipyridine, DNS-bpy: 4-(2,4-dinitrophenylthio)-2,2′-bipyridine], that can act as a probe for the recognition and luminescence sensing of biothiols has been designed and synthesized. Due to the presence of effective photo-induced electron transfer (PET) from the potent electron donor (Ru-bpy centre) to the strong electron acceptor (2,4-dinitrophenyl moiety), the Ru(II) complex itself is weakly luminescent. Reaction of [Ru(bpy)2(DNS-bpy)](PF6)2 with biothiols leads to the replacement of the 2,4-dinitrophenyl moiety by biothiols, which results in the loss of PET within the complex, to allow recovery of the MLCT-based emission of the Ru(II) complex with an 80-fold increase in luminescence intensity. Taking advantage of the high specificity and sensitivity, and the excellent photophysical properties of Ru(II) complexes, [Ru(bpy)2(DNS-bpy)](PF6)2 was successfully applied to the luminescence imaging of biothiols in living Daphnia magna. The results demonstrated the practical applicability of [Ru(bpy)2(DNS-bpy)](PF6)2 as a luminescent probe for the monitoring of biothiols in living bodies.

Combined effect of polystyrene nanoparticles and chlorpyrifos to Daphnia magna

Yu-Hang Zhang,Rong-Yao Gao,Few Zhu,Li-Min Fu,Jian-Ping Zhang

Microplastics and nanoplastics (MPs/NPs) are emerging contaminants ubiquitous in the environment. These particles can act as carriers of hydrophobic organic compounds (HOCs), such as chlorpyrifos (CPF), an organophosphorus insecticide. This study investigates the acute toxicity of CPF combined with model polystyrene nanoplastics (PS-NPs) using Daphnia magna as a model organism. The uptake and accumulation of luminescence-labelled 240 nm PS-NPs were visualized and quantified during co-exposure tests. Heart rates were measured using a video-based method, and IC 50 values for heart rate were calculated to assess the acute toxicity of CPF. The results demonstrate that PS-NPs alone have no acute toxicity, while CPF exhibits high toxicity, with an IC 50 of 50.8 μg/L. However, in the presence of 50 μg/L PS-NPs, the IC 50 increased to approximately 400 μg/L, indicating an antagonistic effect of PS-NPs on CPF toxicity. The adsorption of CPF onto PS-NPs can reduce the concentration of free CPF, leading to aggregation and sedimentation of CPF-PS-NP clusters, thereby decreasing the uptake of CPF by D. magna . This interaction between CPF and PS-NPs in D. magna exemplifies the toxicity-attenuating effects and potential load capacity of nanoplastics for HOCs.

Effects of Spontaneous Fluorescent Polystyrene on Nostocaceae and Daphnia Magna

Shaolin Qiu,Wenjiao Yuan,Yujie Qin,Xin Feng,Meitong Li,Yuhong Xie

Microplastic pollution is widespread around the world and inevitably comes into contact with organisms. With the accumulation of microplastics in the environment, the negative impact of microplastics on organisms has become the main focus in the field of microplastics. In this study, the different particle and concentration effects of fluorescent polystyrene microplastics (PS-MPs) onNostocaceaeandDaphnia Magnawere researched. The results indicate that PS-MPs adhered toNostocaceaethrough static electricity, which hindered the absorption of photons and CO2byNostocaceae, resulting in a decrease in chlorophyll, a low growth rate and high mortality forNostocaceae. PS-MPs with very small particles may be integrated into the blood ofDaphnia Magna, leading to an increasing trend of mortality and a decrease in spawning rate. The research provides basic data and a reference for the effect of PS-MPs on freshwater organisms and has implications for the further study of microplastics. Keywords:microplastics;fluorescent polystyrene;Nostocaceae;Daphnia Magna;photosynthesis;reproductive effect

Waterborne and Dietary Bioaccumulation of Organophosphate Esters in Zooplankton Daphnia magna

Wenxin Liu,Hong Zhang,Jiaqi Ding,Wanyu He,Lin Zhu,Jianfeng Feng

Organophosphate esters (OPEs) are widely used as an additive in flame retardants, plasticizers, lubricants, consumer chemicals, and foaming agents. They can accumulate in aquatic organisms from water (waterborne exposure) and food (dietary exposure). However, the bioaccumulation characteristics and relative importance of different exposure routes to the bioaccumulation of OPEs are relatively poorly understood. In this study,Daphnia magnawere exposed to fo typical OPEs (tris(2-chloroethyl) phosphate (TCEP), tris(1,3-dichloro-2-propyl) phosphate (TDCIPP), tris(2-butoxyethyl) phosphate (TBOEP), and triphenyl phosphate (TPHP)), and their toxicokinetics under waterborne and dietary exposure routes were analyzed. For the waterborne exposure route, the bioconcentration factors (BCFs) increased in the order of TBOEP, TCEP, TDCPP, and TPHP, which were consistent with their uptake rate constants. TPHP might have the most substantial accumulation potential while TBOEP may have the smallest potential. In dietary exposure, the depuration rate constants of four OPEs were different from those in the waterborne experiment, which may indicate other depuration mechanisms in two exposure routes. The biomagnification factors (BMFs) of fur OPEs were all below 1, suggesting trophic dilution in the transfer of four OPEs fromScenedesmus obliquustoD. magna. Except for TBOEP, the contributions of dietary exposure were generally lower than waterborne exposure inD. magnaunder two exposure concentrations. This study provides information on the bioaccumulation and contribution of OPEs inD. magnavia different exposure routes and highlights the importance of considering different exposure routes in assessing the risk of OPEs.

Linking (Poly)Tungstate Speciation to Toxicity and Bioaccumulation in Daphnia magna

Jing Sun,Jie Yan,Lei Song,Yu Dai,Haohao Luo,Wenjuan Wang,Xin Chen,Benjamin C. Bostick,Chengshuai Liu

Tungsten is an emerging pollutant commonly assumed to occur as the simple oxyanion tungstate, yet it often condenses into a suite of polytungstates in polluted waters. Despite growing recognition that tungstate polymerization modulates tungsten’s environmental fate, its effects on toxicity remain largely unexplored. Here, we tracked the (de)polymerization behavior of three representative (poly)tungstates and examined its associations with multiple measures of acute toxicity to Daphnia magna, including the median lethal concentration (LC50), toxicokinetics, reactive oxygen species (ROS) content, metabolomics, and histology. While tungstate remained stable, W12 metatungstate and phosphotungstate depolymerized to different extents, yielding three distinct polymerization regimes in the exposure media. Monomeric tungstate exhibited high LC50, rapid uptake and efficient excretion, weak ROS signals, and minimal disruption of intestinal integrity. Oligomeric tungstates displayed lower LC50 than tungstate, consistent with slow depuration that generated high internal burdens, ROS accumulation, and suppression-oriented metabolic responses. High-order polytungstates, despite limited uptake and low ROS levels, were associated with structural disruption including irreversible gut barrier damage and mitochondrial collapse and displayed the lowest LC50. Together, these results suggest that tungstate polymerization not only amplifies tungsten toxicity but also alters its mode of action, supporting mechanistic interpretation and prediction of its ecotoxicological dynamics.

Investigating the toxicities of different functionalized polystyrene nanoplastics on Daphnia magna

Wei Lin,Ruifen Jiang,Sizi Hu,Xiaoying Xiao,Jiayi Wu,Songbo Wei,Yaxin Xiong,Gangfeng Ouyang

Nanoplastics (NPs) spread widely with water and air current, and they can accumulate in aquatic organisms, even penetrating biofilms, which may cause persistent toxicity and potential hazards. This current study aimed to reveal the toxicological mechanism of different functionalized polystyrene (PS) NPs on Daphnia magna ( D. magna ) by investigating toxicity endpoints in individual level and biochemical level. In this study, acute toxicity, behavioral parameters and biomarker responses of D. magna was measured in the exposure of different functionalized PS NPs (plain PS, PS-p-NH 2 , PS-n-NH 2 and PS-COOH). The results indicated that when exposed to the plain PS, ROS induction would activate MAPKs, thereby causing lethality and adverse behavior effects on D. magna ; while the functionalized PS NPs were less toxic than the plain PS, especially for PS-p-NH 2 which was severely flocculated after exposure, thus showing no immobilization at the investigated concentrations. Also, the antioxidant system was mainly stimulated due to the direct interaction with the cell surface receptor, which was different from the plain PS. Consequently, this work suggests significant effects of functional groups on NPs for environmental toxicity studies, and provides a better understanding of the toxicological mechanism on the toxicity of PS NPs toward D. magna .

Quantifying nanoplastic-bound chemicals accumulated in Daphnia magna with a passive dosing method

Ruifen Jiang,Wei Lin,Jiayi Wu,Yaxin Xiong,Fang Zhu,Lian-Jun Bao,Jing You,Gangfeng Ouyang,Eddy Y. Zeng

Because of their large surface area and high hydrophobicity, nanoplastics (NPs) possess a large sorption capacity for and strong affinity to hydrophobic organic chemicals, which may substantially alter the environmental behavior, especially the bioaccumulation of hydrophobic organic compounds. The present study aims to examine the effects of nano-size polystyrene (PS) on the bioaccumulation of polychlorinated biphenyls (PCBs) in Daphnia magna. A novel passive dosing method was developed to quantify the accumulation of NPs in D. magna in different exposure suspensions with various NP concentrations during 2 h and 24 h exposure. Addition of NPs enhanced the amount of PCBs accumulated in D. magna by 1.4–2.6 times compared to no presence of NPs during 24 h exposure. The relative amounts of NP-bound PCBs accumulated in D. magna were 18% to 81% for different PCB congeners at various NP concentrations. The significance of different uptake pathways in the accumulation of PCBs in D. magna with the presence of NPs was found to relate to the chemical properties of PCBs, the ingestion rate of plastics and the exposure conditions. These results have justified additional research efforts toward investigating the fate and toxic effects of NP-bound contaminants ingested by organisms.

Exposure scenarios modulate the combined toxicity of nanoplastics and Cd to Daphnia magna

Shuang Zhang,Zhongmiao Wang,Caijiao He,Daohui Lin

Nanoplastics (NPs) have extensively accumulated in aquatic environments and coexist with various contaminants including heavy metals. While the adverse effects of NPs and Cd on aquatic organisms have been widely reported, most studies focus on co-exposure scenarios, leaving the effect of different exposure sequences poorly understood. Here, we evaluated the combined toxicity of NPs and Cd to Daphnia magna under different exposure scenarios (single exposure, pre-exposure, and co-exposure). The bioaccumulation of NPs and Cd, intestinal histology, and gut microbiota were further analyzed to elucidate the underlying mechanisms of combined toxicity. Our findings reveal that environmentally relevant concentrations of NPs stimulated D. magna reproduction, increasing total number of offspring (hormesis). Pre-exposure to NPs enhanced tolerance to subsequent stressors, thus reducing the combined toxicity of NPs and Cd. In contrast, slow toxicological recovery of Cd caused persistent residual effects that amplified the toxicity of subsequent combined exposure to NPs and Cd. Specifically, compared to single-exposure groups, Cd pre-exposure led to a 5-fold and 3-fold increase in the bioaccumulation of NPs and Cd, respectively, accompanied by exacerbated intestinal damage and disruption of the gut microbiota. Our results highlighted the importance of evaluating combined toxicity under different exposure scenarios in natural environments, providing new insight into the potential risks of NPs in aquatic environments.
