Decabromodiphenyl ethane (DBDPE) can pose a potential toxic threat to human beings and the environment. P. ostreatus , as one of the typical white-rot fungi, can effectively degrade various refractory pollutants. The biodegradable characteristics of DBDPE by P. ostreatus , as well as the mechanisms, and toxicological response were investigated in this study. The removal rate reached 47.73% and 43.20%, respectively, for 5 and 20 mg/L DBDPE after 120-h degradation by P. ostreatus . As a coexisting substance, Pb could inhibit the biodegradation. It is found that both the intracellular enzyme (P450) and extracellular enzymes (manganese peroxidase (MnP), lignin peroxidase (LiP), and laccase (Lac)) played a very important role in the biodegradation of DBDPE, of which Lac dominated the degradation. The toxic response was monitored during the degradation. The activities of SOD and CAT were enhanced to eliminate excess ROS in P. ostreatus triggered by DBDPE. In addition, debromination, hydroxylation, and oxidation were inferred as the main degradation pathways preliminarily. The findings provide a theoretical basis for the application of microbial degradation of DBDPE contamination.

Decabromodiphenyl Ethane (DBDPE), a kind of new brominated flame retardants (NBFRs) used to replace DecaBDE, has been frequently detected in the environment and human samples. In this study, we explored its toxic effects on male mouse offspring after perinatal exposure to DBDPE. During the perinatal period, pregnant ICR mice were exposed to DBDPE (100 μg/kg body weight) via oral gavage. After weaning, male offspring were fed on a low-fat diet and a high-fat diet, respectively. We measured and recorded body weight, liver weight, and epididymis fat mass, blood biochemical markers, metabolites changes in liver, and gene expression involved in lipid and glucose homeostasis . The results showed that perinatal exposure to DBDPE increased the risk of obesity in mouse offspring and affected triglyceride synthesis, bile secretion, purine synthesis, mitochondrial function and glucose metabolism, furthermore, the use of HFD feeding may further exacerbate these effects. All of these results show that early-life exposure to low doses of DBDPE can promote the development of metabolic dysfunction, which in turn induces obesity.

Decabromodiphenyl ethane (DBDPE), a chemical compound widely detected in aquatic ecosystems, has shown a range of toxicological effects in zebrafish ( Danio rerio ). However, its effects on zebrafish reproduction remain underexplored. In this study. Adult zebrafish were exposed to 0, 1, 10, and 100 μg/L of DBDPE for 21 days. DBDPE accumulated in the gonads, leading to a marked decrease in both cumulative spawning and gonadosomatic index in female zebrafish. To investigate the mechanisms underlying reproductive abnormalities induced by DBDPE, we measured serum hormone levels, oocyte development, and gene expression in the hypothalamic–pituitary–gonadal (HPG) axis. Our results showed that exposure to DBDPE significantly reduced estradiol (E2) levels in female zebrafish, along with abnormal oocyte development and disrupted gene expression in the HPG axis. Further metabolomic and lipidomic analyses revealed considerable changes in taurine and hypotaurine metabolism, glutathione metabolism, and the FoxO signaling pathway, along with significant disturbances in lipid metabolism. These disruptions in metabolic pathways may be an important underlying cause of delayed oocyte development. Our findings show that exposure to DBDPE delays oocyte development by altering lipid metabolism, reducing sex hormone levels, and impairing HPG axis regulation, ultimately leading to reduced fecundity in female zebrafish. This study highlights the repercussions of DBDPE exposure on the reproductive health of zebrafish, emphasizing the need for stricter regulation of its production and use, a well as a comprehensive reassessment of the risks it poses to fish.

Decabromodiphenyl ethane (DBDPE), a key alternative to deca-BDE (BDE-209), has been ubiquitous in the receiving ecosystem. However, little is known about its uptake process and fate in plants. Here, the plant absorption, distribution, and metabolism of 14C-DBDPE under two distinct exposure pathways (i.e., soil-root and airborne dust-leaf) were investigated with three vegetables (cherry radish, water spinach, and eggplant). DBDPE was absorbed and primarily accumulated in directly exposed roots and leaves, with translocation factors to other tissues of 0.04–0.77 and 0.73–12.80 × 10–3, respectively. DBDPE in exposed leaves was more difficult to transport as most DBDPE did not enter the interior of the leaves but was stored in waxes (>90%). Furthermore, DBDPE within leaves was found to enter mostly through waxy layer penetration (>97%). The majority of 14C-DBDPE was localized in cell walls (52.6 and 75.6%, respectively) and organelles (22.7 and 45.5%, respectively) of exposed tissues, substantially restricting its in-plant translocation. Debrominated products were detected in exposed roots, but barely found in exposed leaves, as most DBDPE was blocked by the waxy layer. Moreover, DBDPE was stable in soils and dust without degradation. Our findings contribute to a deeper understanding of the environmental fate of DBDPE in soil-plant and airborne dust-plant systems.

Decabromodiphenyl ethane is a novel brominated flame retardant , that has always been dissolved in organic solvents to explore its activities on aquatic organisms. In this study, the influences of decabromodiphenyl ethane on the microalga Chlorella sorokiniana ( C. sorokiniana ) were studied, and three microalgae treatments, including decabromodiphenyl ethane dissolved in dimethyl sulfoxide solvent (DBDPE treatment), dimethyl sulfoxide alone (control II) or untreated (control I) were used in the experiment, respectively. The results showed that the growth of C. sorokiniana was remarkably enhanced in the DBDPE treatment compared with the control I and II groups. Conjoint analysis of transcriptomics and quantitative proteome displayed that the upregulated differentially expressed genes and proteins of DBDPE:control I were enriched in 6 pathways, and downregulated genes/proteins of DBDPE:control I were enriched in 3 pathways. The upregulated differentially expressed genes and proteins of DBDPE:control II were enriched in 4 pathways, and downregulated genes/proteins of DBDPE:control II were enriched in 6 pathways. In addition, decabromodiphenyl ethane changed the fatty acid concentration in C. sorokiniana cells. The activities of superoxide dismutase were enhanced when C. sorokiniana were treated by decabromodiphenyl ethane. The data highlighted that the mRNA and protein expression relating to the fatty acid production, of C. sorokiniana were significantly affected by decabromodiphenyl ethane, and decabromodiphenyl ethane pollution changed the physiological metabolism of microalgae and had harmful effects on natural environments.

Decabromodiphenyl ethane (DBDPE) is an efficient novel brominated flame retardant (NBFR) widely used in electronics, plastics, textiles, and other industries. DBDPE tends to permeate into surrounding environments during its synthesis, utilization, and disposal processes, accumulating in organisms and causing biomagnification effects. In this study, sodium persulfate (Na 2 S 2 O 8 , PS) was employed for the degradation of DBDPE in soil and indicated that elevated reaction temperature and increased initial alkalinity could both enhance the degradation efficiency of DBDPE. Under the optimal conditions (60°C, pH 11), DBDPE degradation efficiency reached 83.01% within 24 h. It was found that the system’s initial alkalinity exhibited negligible effects on the enhancement of activation by temperature, while chloride ions and weakly acidic conditions significantly inhibited DBDPE degradation. This study showed that hydroxyl radicals (HO•) played a dominant role in the degradation process, supplemented by sulfate radicals (SO 4 • − ). ECOSAR predictions revealed a marked reduction in the ecotoxicity of the seven potential degradation products relative to DBDPE, some of which were characterized by lower bromine content and oxygen incorporation. This study provides insights into the mechanisms of DBDPE degradation and a potential remediation strategy for contaminated soil.

Decabromodiphenyl ethane (DBDPE), a newly emerging brominated flame retardant (BFR), has garnered increasing attention due to its high production volumes and widespread usage, prompting worries about its possible impacts on human well-being. Prior investigations have highlighted the substantial toxicity of DBDPE to the thyroid, liver, and cardiovascular systems, yet its effects on fetal growth and development remain inadequately understood. This investigation aims to elucidate the underlying mechanisms and consequences of DBDPE exposure on fetal growth and development through both in vivo and in vitro models. Pregnant mice were administered DBDPE orally at doses of 0, 0.05, 0.5, and 5 mg/kg bw/day. Results revealed that gestational DBDPE exposure caused placental damage, resulting in fetal growth restriction (FGR). A significant reduction in the phosphorylation level of AIFM1 Ser116 in placental trophoblasts was observed, specifically correlating with the activation of oxeiptosis. Metabolomic and transcriptomic analyses further suggested that DBDPE exposure disrupts the oxidative phosphorylation (OXPHOS) pathway, thereby impairing mitochondrial function. Notably, treatment with MitoQ, a mitochondria-targeted antioxidant, effectively reversed DBDPE-induced oxeiptosis in placental trophoblasts, alleviating the negative effects of DBDPE on placental damage and FGR. Mechanistically, the mitochondrial dysfunction induced by gestational DBDPE exposure initiates oxeiptosis in placental trophoblasts, exacerbating placental injury and ultimately leading to FGR. In summary, this study integrates the roles of environmental pollutants, oxeiptosis, and mitochondrial dysfunction, offering new insights into the toxicological mechanisms by which DBDPE and other emerging pollutants impact fetal growth and development.

Purpose Decabromodiphenyl ethane (DBDPE) and lead (Pb), two prevalent contaminants in e-waste dismantling areas, pose significant threats to soil ecosystem health. However, the ecotoxicological effects of their co-exposure on soil invertebrates remain poorly characterized. Methods Avoidance behavior, survival rate, and growth-inhibition assays were performed to assess the combined toxic effects of DBDPE and Pb on earthworm ( Eisenia fetida ) in an artificial soil system over exposure periods of 7–21 d. Responses from multiple biochemical biomarkers, including superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA), glutathione S-transferase (GST), and acid phosphatase (ACP) were also measured, and the data were integrated by the biomarker response index (BRI). Interaction between Pb and DBDPE on BRI was further analyzed by calculation of effect addition index (EAI). Results Avoidance behavior emerged as the most sensitive endpoint and exhibited concentration-dependent escalation under co-exposure conditions. While individual exposure to DBDPE (0–0.75 mg/kg) induced negligible impacts on growth and mortality, its combination with Pb (1000 mg/kg) resulted in significant synergistic effects. Similarly, biochemical biomarkers—including SOD, CAT, MDA, GST, and ACP—showed markedly greater alterations in co-exposure groups compared to single DBDPE treatments. The BRI revealed only slight alterations in biological health across most treatments. The EAI values indicated that antagonistic effects dominated the interaction between DBDPE and Pb. Conclusion Our findings demonstrated significant variation in biomarker sensitivity to pollutant exposure. Notably, short-term sub-lethal co-exposures may elicit synergistic effects on avoidance, growth, and mortality, revealing a time-concentration toxicity dependency. These results provide novel insights into the interaction between heavy metals and organo-pollutants and highlight the ecological hazards posed by emerging brominated flame retardants (e.g., DBDPE) in heavy metal-contaminated soil systems.

In this paper, the hepatocytotoxicity and aryl hydrocarbon receptor (AHR) activity of decabromodiphenyl ethane (DBDPE), decabromodiphenyl ether (BDE209) and other 18 analogues were evaluated in vitro using human normal liver cell L02. These dioxin-like compounds showed differential hepatocytotoxicity (EC 50 = 0.38–17.87 mg/L) and AHR activity (EROD activity = 4.53–46.35 U/μg). In silico study indicated the distance of π-π bonds between the benzene ring of compounds and residue Phe234 of AHR played a key role in the binding of AHR, and the substituents on the benzene ring also influenced the activity. Combining molecular biology and bioomics, the comprehensive investigations on the hepatotoxic mechanisms have demonstrated the AHR signaling pathway was the key mediation mechanism for the hepatotoxicity of DBDPE/BDE209. The cytochrome P450s (CYP2 family) mediated formation of reactive oxygenated intermediates might be the dominant toxic mechanism, which could produce oxidative stress or cause genotoxicity. Although the experimental toxicity of DBDPE was smaller relative to BDE209, the health risk of DBDPE may be much greater than we expected, due to the high potential to form a variety of dioxin-like intermediates by microbial oxidation of ethyl group. Therefore, whether it is really safe to replace BDE209 with DBDPE is a debatable question, and more ecotoxicological and health data are needed to clarify this issue.

Decabromodiphenyl ethane (DBDPE) as a novel brominated flame retardant is frequently detected in environmental media due to its widespread use. Studies have shown that exposure to environmental pollutants in utero could lead to weight loss in newborns and obesity in adulthood. However, the mechanisms of how the cubs grow rapidly from low birth weight to obesity remain unclear. Although it has been reported that perinatal DBDPE exposure caused obesity in the offspring of mice in adulthood, its metabolic changes in offspring juvenile are unknown. Here, we monitored changes of body weight in cubs following exposure to DBDPE in utero. Furthermore, 1 H NMR-based metabolomics was used to investigate the effects of in utero DBDPE exposure on the metabolic profile of neonates included days 1 and 25. Additionally, the expression levels of key genes in the relevant pathways were quantified. The results showed that exposure to high levels of DBDPE in utero caused cubs to be smaller at birth and grow rapidly during lactation. Metabolomics analysis showed that cubs in the exposed group consistently accelerated glycogenolysis and lipid synthesis to promote appetite and energy absorption, and achieved rapid growth in the subsequent lactation, which was further evidenced by the expression levels of genes related to glycolipid metabolism. These results reveal the mechanism for the first time how the weight loss of newborns in utero environmental pollutant exposure transformed into obesity in adulthood, although the exposures here were much higher than would be anticipated from the environment.

The health risks of Decabromodiphenyl ethane (DBDPE) with its cardiovascular toxicity , liver toxicity and cytotoxicity had been generally acknowledged. However, the influence on gut microbiome and short-chain fatty acids (SCFAs) metabolism caused by DBDPE exposure remained unknown. In this study, three exposure groups (5, 50, 500 mg/L) and control group were used to investigate the effect of DBDPE by using simulator of the human intestinal microbial ecosystem (SHIME). 16S rRNA gene high-throughput sequencing illustrated that high dose DBDPE exposure increased the α -diversity of gut microbiota , while reduced the abundance of Firmicutes and Proteobacteria . In addition, the low dose (5 mg/L) DBDPE inhibited the increasing of SCFAs, but the medium and high dose (50 and 500 mg/L) DBDPE promoted the advancement, especially in ascending colon . Notably, DBDPE exposure lead a similar changing of acetic acid and butyric acid contents in different sections of the colon. This study confirmed the alternation of composition and metabolic function in gut microbial community due to DBDPE exposure, indicating an intestinal damage and appealing for more attention concentrated on the health effects of DBDPE exposure.

Decabromodiphenyl ethane (DBDPE) is a brominated flame retardant widely used for its high flame retardancy and low bioavailability. However, DBDPE exhibits bioaccumulative potential. The placental vasculature, critical for the exchange between maternal and fetal circulation, is susceptible to environmental factors. We aimed to understand the potential toxicity of DBDPE to the placental vasculature. Gestational CD-1 mice and human umbilical vein endothelial cells (HUVECs) were exposed to 0, 0.5, 5, 50 mg/kg·bw·day or 0, 6.25, 12.5, 25, 50 μmol/L DBDPE, respectively. DBDPE exposure impaired placental angiogenesis and inhibited the proliferation and invasion of HUVECs by activating the endothelial-to-mesenchymal transition (EndMT) through the TGF-β1/Smad pathway, in vivo and in vitro . DBDPE upregulated the transcription factor TWIST2 and mRNA acetyltransferase NAT10. Knockdown or overexpression of TWIST2 reversed the vascular dysfunction and EndMT mechanism. DBDPE facilitated the recruitment of NAT10 by TWIST2, promoting N4-acetylcytidine (ac4C) RNA modifications and stabilizing the TGF-β1 mRNA. NAT10 inhibition rescued the effects caused by DBDPE. These findings uncover molecular mechanisms linking DBDPE exposure to adverse pregnancy outcomes, highlighting potential risks to pregnant women.
