Acetaminophen (APAP) is widely used for its antipyretic properties and is commonly found in wastewater, potentially threatening aquatic ecosystems. Microbial degradation has been proven effective in removing APAP. However, strains capable of efficiently degrading APAP are rare, and the research on the biodegradation kinetics of APAP is insufficient. In this study, a novel APAP-degrading strain, identified as Delftia sp . PY-12 by morphological characterization and 16S rRNA gene sequencing, was isolated from activated sludge. Experimental conditions were optimized using the Box-Behnken response surface design, revealing that PY-12 degraded 100 mg/L APAP under pH 7.11, 30℃, and 163 rpm within 24 h, outperforming most other strains. Comparative analysis of substrate inhibition models shows that the Aiba and Tiesser were most suitable for the substrate inhibition characteristics of PY-12. The models indicate that PY-12 can still degrade APAP even at concentrations of 446.919 mg/L and 554.215 mg/L. Through potential enzyme activity measurements, the degradation pathway of APAP by PY-12 was determined. APAP is first converted by amidohydrolase into 4-aminophenol (4-AP) and then converted by deaminase into hydroquinone (HQ), which is further converted by hydroquinone 1,2-dioxygenase to form 4-hydroxymuconic semialdehyde, ultimately entering the TCA cycle. This study revealed the potential application value of PY-12 in APAP degradation, which could be a strong candidate strain for microbial bioremediation of APAP pollution in environmental remediation.

In this study, a water–silicone oil biphasic system was developed to enhance the biodegradation of monochlorobenzene (CB) byDelftia tsuruhatensisLW26. Compared to the single phase, the biphasic system with a suitable silicone oil fraction (v/v) of 20% allowed a 2.5-fold increase in the maximum tolerated CB concentration. The CB inhibition onD. tsuruhatensisLW26 was reduced in the presence of silicone oil, and the electron transport system activity was maintained at high levels even under high CB stress. Adhesion of cells to the water–oil interface at the water side was observed using confocal laser scanning microscopy. Nearly 75% of cells accumulated on the interface, implying that another interfacial substrate uptake pathway prevailed besides that initiated by cells in the aqueous phase. The 8-fold increase in cell surface hydrophobicity upon the addition of 20% (v/v) silicone oil showed that silicone oil modified the surface characteristics ofD. tsuruhatensisLW26. The protein/polysaccharide ratio of extracellular polymeric substances (EPS) fromD. tsuruhatensisLW26 presented a 3-fold enhancement. These results suggested that silicone oil induced the increase in the protein content of EPS and rendered cells hydrophobic. The resulting hydrophobic cells could adhere on the water–oil interface, improving the mass transfer by direct CB uptake from silicone oil. Keywords:waste gas;monochlorobenzene;biodegradation;silicone oil;cell adhesion

As a representative polycyclic aromatic hydrocarbon (PAH), pyrene poses a significant threat to the environment and public health due to its persistence and potential toxicity. Extensive research has been conducted on biodegradation of pyrene, highlighting the crucial need to unravel its degradation mechanisms. Coastal and marine sediments are promising sources for discovering microbial candidates capable of degrading pyrene. In this study, three bacterial consortia, designated as BS, YS, and SS, were enriched from sediments collected from the Bohai Sea, Yellow Sea, and Liao River estuarine wetland, respectively. The bacterial consortia BS, YS, and SS demonstrated high pyrene degradation efficiencies, achieving 86.61 %, 80.39 %, and 75.70 % degradation at the concentration of 50 mg/L within 8 days, respectively. High-throughput sequencing of 16S rRNA gene amplicons revealed Pandoraea , Delftia , and Pseudomonas as the dominant populations in bacterial consortia BS, YS, and SS, respectively, with some species potentially being novel pyrene degraders. Further analysis included identifying pyrene degradation metabolites by LC-Q-TOF-MS (high-performance liquid chromatography tandem quadrupole time-of-flight mass spectrometry) and predicting functional genes through PICRUSt (Phylogenetic Investigation of Communities by Reconstruction of Unobserved Stats). Based on these results, the metabolic pathways for pyrene degradation by the bacterial consortia were proposed, which predominantly involved the dioxygenation pathway. Overall, these findings should offer valuable bacterial resources for facilitating the bioremediation of PAH-contaminated environments.
