Six new polyketides named paraverrucsins A–F (1–6) with oxabicyclic and dioxatricyclic skeletons, together with eight known metabolites (7–14), were discovered and isolated from the fermentation medium of Paraphaeosphaeria verruculosa. Paraverrucsin A–C possessed a novel decarboxylated skeleton compared with that of trichocladinols. Their structures were elucidated by extensive spectral analysis and DP4+ calculations. Paraverrucsins B/C and D/E were isolated as a mixture for the mutarotation occurred at C-2. Paraverrucsins B/C, D/E, F/trichocladinol B, 8, and 9 displayed antifeedant activities against silkworm larvae, with antifeedant index percentages ranging from 62.5 to 93.0%, at a concentration of 50 μg/cm2. Among them, Paraverrucsins B/C and 9 had EC50 values at 13.9 and 18.2 μg/cm2. Most compounds showed antifungal activities against phytopathogenic fungi with minimum inhibitory concentration (MIC) values of 16–64 μg/mL. Coculture of P. verruculosa and host plant Dendrobium officinale leads to the enhancement of antifeedant and antiphytopathogenic activities. Compounds 1, 2/3, 4/5, 6/14 were tested for cytotoxicity against five human carcinoma cell lines, HL-60, A549, MCF-7, SW480, and SMMC-7721, while they exhibited selected cytotoxicity against SW480 with inhibition ratios of 32–38% at a concentration of 40 μM.

Salt stress severely constrains plant growth and productivity, so understanding the underlying mechanisms of salt stress is crucial for the genetic improvement of plants. Aquaporins (AQPs), which facilitate the transport of water or nutrients, exert an indispensable role in plants' response to salt stress. The AQP family in Dendrobium officinale , a commercially important medicinal and edible orchid, has not been identified or functionally characterized, while an understanding of this plant's tolerance to salinity remains unexplored. In this study, a genome-wide identification of AQP genes in D. officinale was performed using a salt-induced transcriptome. The 28 AQP members were mined and classified into four subfamilies (NIPs, PIPs, SIPs, and TIPs), and their physicochemical and bioinformatic properties were analyzed. Overexpression of DoTIP1-1 in D. officinale significantly enhanced salt tolerance via the accumulation of proline and water-soluble polysaccharides that improved water retention. Furthermore, salt tolerance was fortified by the improved activity of antioxidant enzymes (peroxidase and superoxide dismutase), allowing reactive oxygen species to be scavenged, and augmenting photosynthetic efficiency due to the upregulated expression of stress-responsive genes. This study provides the first comprehensive analysis of the AQP family in D. officinale and demonstrates that DoTIP1-1 is a key genetic determinant for enhancing salt tolerance, offering significant potential for the genetic improvement of this plant to resist abiotic stress.

Photodynamic therapy (PDT) eradicates tumors via the generation of toxic reactive oxygen species (ROS) by activation of a photosensitizer (PS) with appropriate light. Local PDT toward tumors can trigger the immune response to inhibit distant tumors, but the immune response is usually insufficient. Herein, we used a biocompatible herb polysaccharide with immunomodulatory activity as the carrier of PS to enhance the immune inhibition of tumors after PDT. The Dendrobium officinale polysaccharide (DOP) is modified with hydrophobic cholesterol to serve as an amphiphilic carrier. The DOP itself can promote dendritic cell (DC) maturation. Meanwhile, TPA-3BCP are designed to be cationic aggregation-induced emission PS. The structure of one electron-donor linking to three electron-acceptors endows TPA-3BCP with high efficiency to produce ROS upon light irradiation . And the nanoparticles are designed with positively charged surfaces to capture antigens released after PDT, which can protect the antigens from degradation and improve the antigen-uptake efficiency by DCs. The combination of DOP-induced DC maturation and antigen capture-increased antigen-uptake efficiency by DCs significantly improves the immune response after DOP-based carrier-mediated PDT. Since DOP is extracted from the medicinal and edible Dendrobium officinale , the DOP-based carrier we designed is promising to be developed for enhanced photodynamic immunotherapy in clinic.

Dendrobium officinale Kimura et Migo has been used as a traditional Chinese medicine (TCM) and a functional food for thousands of years. Carbohydrate is one of the most important effective substances and indicative components in D. officinale. However, since the qualitative and quantitative analysis of polysaccharides in D. officinale remains a challenge and limitation, herein, an oligosaccharide-quality marker approach was newly developed for quality assessment of D. officinale by spectrum–effect relationships between high performance liquid chromatographic (HPLC) fingerprints and anti-inflammatory effects. The HPLC fingerprints of 48 batches of oligosaccharides from D. officinale (DOOS) were developed and analyzed with similarity analysis (SA) and hierarchical cluster analysis (HCA), and eight common peaks were identified. In vitro screening experiment indicated that DOOS potentially inhibited nitric oxide (NO) production and effectively reduced the release of inflammatory cytokines, such as TNF-α, IL-6, and IL-1β in RAW 264.7 cells, thereby reducing the inflammatory response of cells. Finally, the HPLC fingerprint of different batches of DOOS was combined with in vitro anti-inflammatory activity to assess the spectrum–effect relationships of DOOS by gray correlation analysis (GCA), in addition, the purified oligosaccharide components were identified and validated for NO inhibitory activity. Our results showed four DOOS (maltotetraose, maltopentaose, maltohexaose, and mannohexaose) were relevant to anti-inflammatory effects and could be as quality markers for the quality control of D. officinale. It suggests that the “oligosaccharide-spectrum-effect” relationships approach is a simple and reliable method for the quality control of herb medicines or nutritious foods.

Background Dendrobium officinale flowers (DOF) have the effects of antiaging and nourishing yin, but it lacks pharmacological research on skin aging. Objective Confirming the role of DOF in delaying skin aging based on the “in vitro animal–human” model. Methods In this experiment, three kinds of free radical scavenging experiments in vitro, D-galactose-induced aging mouse model, and human antiaging efficacy test were used to test whether DOF can improve skin aging through anti-oxidation. Results In vitro experiment shows that DOF has certain scavenging effect on 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical, hydroxyl free radical, and superoxide free radical, and its IC 50 is 0.2090 μg/mL, 15.020, and 1.217 mg/mL respectively. DOF can enhance the activities of T-AOC, SOD, CAT, and GSH Px in the serum of aging mice, increase the content of GSH, and reduce the content of MDA when administered with DOF of 1.0, 2.0, and 4.0 g/kg for 6 weeks. In addition, it can enhance the activity of SOD in the skin of aging mice, increase the content of Hyp, and decrease the content of MDA, activated Keap1/Nrf2 pathway in the skin of aging mice. Applying DOF with a concentration of 0.2 g/mL on the face for 8 weeks can significantly improve the skin water score and elasticity value, reduce facial wrinkles, pores, acne, and UV spots, and improve the facial brown spots and roughness. Conclusion DOF can significantly improve skin aging caused by oxidative stress, and its mechanism may be related to scavenging free radicals in the body and improving skin quality.

An extraction-purification process for Dendrobium officinale polysaccharides was established, with a yield of 35.03%, content of 49.34%, and protein removal rate of 78.70%. Two polysaccharides, DOP‑1 (linear) and DOP‑2 (branched), protect against photoaging via the TGF‑β/Smad pathway, with DOP‑2 showing stronger activity. Objective: To optimize the extraction-purification process of Dendrobium officinale polysaccharides (DOPs) and characterize the fine structures of two homogeneous fractions (DOP-1, DOP-2), as well as to evaluate their anti-photoaging activity on HFF cells. Methods: An integrated process including petroleum ether defatting, cellulase hydrolysis, water extraction-ethanol precipitation, trypsin-Sevag deproteinization, activated carbon decolorization, and dialysis was used. DOP-1 and DOP-2 were separated by DEAE-52 and Sephadex G-100 chromatography. Their structures were analyzed by FT-IR, methylation-GC/MS, NMR, and SEM. Anti-photoaging effects were assessed via CCK-8, SA-β-Gal staining, and qPCR of TGF-β/Smad pathway genes. Results: The optimized process increased crude polysaccharide yield from 30.34% to 35.03% and polysaccharide content from 31.23% to 49.34%. DOP-1 (Mn 989 Da, PDI 1.15) was a linear oligosaccharide composed of →4)-β-D-Manp (72.32%) and →4)-β-D-Glcp (23.78%) in an alternating pattern. DOP-2 (Mn 2970 Da, PDI 1.67) was a branched polysaccharide with a backbone of →6)-α-D-Glcp (58.06%) and branches at →2,6)-Manp and →4,6)-Glcp. Both DOP-1 and DOP-2 (62.5 μg/mL) protected HFF cells from UVA-induced photoaging, upregulated TGF-β1, Smad3, Smad4, and Collagen I, and downregulated Smad7 and MMP-1, with DOP-2 showing dose-dependent effects. Conclusion: The optimized process enables efficient preparation of DOPs. DOP-1 and DOP-2 possess distinct structural features and exert anti-photoaging effects via regulation of the TGF-β/Smad signaling pathway. The purpose of this study was to screen the optimum extraction of polysaccharides (DOP) from Dendrobium officinale stem. Firstly, different methods, including hot water extraction (HWE), cold-pressing (CP), freeze-thawing cold-pressing (FTCP), ultrasonic-assisted hot water extraction (UHWE), microwave-assisted hot water extraction (MHWE) and enzyme-assisted hot water extraction (EHWE), were employed to extract DOP under their respective best parameters. Then, the extraction yield, structure and antioxidant activity of the polysaccharides from different extraction methods were compared under the same condition. The data implied that UHWE and FTCP possessed higher extraction yield than the other extraction methods. Besides, DOP CP and DOP FTCP had higher molecular weight than the other polysaccharide samples. More importantly, DOP FTCP had the highest antioxidant activity. Overall, DOP FTCP exhibit high extraction yield, well-preserved molecular chains and best antioxidant activity, all these indicated FTCP was the most suitable method to extract DOP.

Diabetes is a metabolic diseases characterized by hyperglycemia resulting from defects in insulin secretion and/or action. The anti-diabetic effects of polysaccharide from Dendrobium officinale stem (DOP) on type II diabetic rats was evaluated in this study. Treatment with DOP significantly decreased the concentrations of fasting blood glucose, insulin, glycated serum protein, serum lipids (TC, TG, LDL-c, NEFA), and increased the concentration of HDL-c, compared with the untreated diabetic rats. In addition, DOP treatment alleviated the oxidative stress injury by increasing the activity of superoxide dismutase and total antioxidant capacity , as well as decreased the level of malonaldehyde. Histopathological analysis revealed that DOP has protective effects on type II diabetes induced pancreatic injury. Serum metabolomics and lipidomics analysis also found DOP treatment could decrease serum fatty acids, glycerolipids, sphingolipids, phospholipids and bile acids, and alleviate the metabolism disorders of lipid, bile acids and amino acids .

The acetylation or deacetylation of polysaccharides can influence their physical properties and biological activities. One main constituent of the edible medicinal orchid,Dendrobium officinale, is water-soluble polysaccharides (WSPs) with substitutedO-acetyl groups. BothO-acetyl groups and WSPs show a similar trend in different organs, but the genes coding for enzymes that transfer acetyl groups to WSPs have not been identified. In this study, we report that REDUCED WALL ACETYLATION (RWA) proteins may act as acetyltransferases. ThreeDoRWAgenes were identified, cloned, and sequenced. They were sensitive to abscisic acid (ABA), but there were no differences in germination rate and root length between wild type and35S::DoRWA3transgenic lines under ABA stress. Three DoRWA proteins were localized in the endoplasmic reticulum.DoRWA3had relatively stronger transcript levels in organs where acetyl groups accumulated thanDoRWA1andDoRWA2, was co-expressed with polysaccharides synthetic genes, so it was considered as a candidate acetyltransferase gene. The level of acetylation of polysaccharides increased significantly in the seeds, leaves and stems of three35S::DoRWA3transgenic lines compared to wild type plants. These results indicate that DoRWA3 can transfer acetyl groups to polysaccharides and is a candidate protein to improve the biological activity of other edible and medicinal plants. Keywords:acetyl groups;Dendrobium officinale;REDUCED WALL ACETYLATION;endoplasmic reticulum

Alkaloids are the main active ingredients in the medicinal plantDendrobium officinale. Based on the published genomic and transcriptomic data, a proposed terpenoid indole alkaloid (TIA) biosynthesis pathway may be present inD. officinale. In this study, protocorm-like bodies (PLBs) with a high-yielding production of alkaloids were obtained by the optimization of tryptophan, secologanin and methyl jasmonate (MeJA) treatment. The results showed that the total alkaloid content was 2.05 times greater than that of the control group when the PLBs were fed with 9 µM tryptophan, 6 µM secologanin and 100 µM MeJA after 36 days. HPLC analysis showed that strictosidine synthase (STR) activity also increased in the treated plants. A total of 78 metabolites were identified using gas chromatography-mass spectrometry (GC-MS) in combination with liquid chromatography-mass spectrometry (LC-MS) methods; 29 differential metabolites were identified according to the multivariate statistical analysis. Among them, carapanaubine, a kind of TIA, exhibiteddramaticallyincreased levels. In addition, a possible underlying process of the metabolic flux from related metabolism to the TIA biosynthetic pathway was enhanced. These results provide a comprehensive view of the metabolic changes related to alkaloid biosynthesis, especially TIA biosynthesis, in response to tryptophan, secologanin and MeJA treatment. Keywords:Dendrobium officinale;metabolomics;alkaloids;tryptophan;secologanin;methyl jasmonate
