Bark beetles mainly rely on detoxification enzymes to overcome the defensive oleoresin terpenes of their host. Insect Carboxylesterases (CarEs) play an important role in the detoxification of plant allelochemicals and insecticides. Nevertheless, the functions of specific CarE genes in metabolization of exogenous substances are still unclear in the Chinese white pine beetle, Dendroctonus armandi . In this study, we sequenced and characterized the full-length cDNAs of six CarE ( DaEST1 - DaEST6) genes from D. armandi . Spatiotemporal expression profiling showed that the expression of DaEST1, DaEST3 and DaEST4 in D. armandi was higher in the larval and adult stages, and these genes mainly expressed in fat body, Malpighian tubules and midgut. Moreover, the expression of DaEST1, DaEST3 and DaEST4 was significantly induced after treatment with four terpenoids. Importantly, disruption of DaEST3 significantly inhibited the CarE activity and reduced the tolerance of adults to host allelochemicals in D. armandi . Additionally, treatment with triphenyl phosphate to adults also reduced the tolerance after terpenoids exposure, which resulted in a significant decrease of CarE activity, leading to a significant improvement of mortality in D. armandi . These results indicate that DaEST3 plays an important role in determining the tolerance to host allelochemicals in D. armandi , which might be of great significance in developing new therapeutic methods to pest control.

Bark beetles mainly rely on detoxification enzymes to resist the host tree’s defense against oleoresin terpenes. Cytochrome P450 enzymes (CYPs) play an important role in the detoxification of plant allelochemicals and pesticides in insect. One P450 gene (DaCYP4BQ1) is associated with the response of (+)-α-pinene inDendroctonus armandi. However, the regulatory mechanism of this P450 gene response to (+)-α-pinene is still unknown. In this study, spatiotemporal expression profiling indicated thatCYP4BQ1was highly expressed in adult and larval stages ofD. armandi, and it was predominantly expressed in fat body, midgut, and Malpighian tubules of adults. Moreover, the expression ofCYP4BQ1significantly increased after exposure to (+)-α-pinene, and depletion of it decreased the tolerance of adults to (+)-α-pinene. In addition, (+)-α-pinene treatment induced the expression of the transcription factors cap ‘n’ collar isoform C (CncC) and its binding factor muscle aponeurosis fibromatosis (Maf), elevated the level of hydrogen peroxide (H2O2), and increased the activities of antioxidant enzymes. SilencingCncCsuppressedCYP4BQ1expression and enhanced the susceptibility of beetles to (+)-α-pinene. Similarly, application of the reactive oxygen species (ROS) scavenger N-acetylcysteine reduced the production and accumulation of H2O2, suppressed the expression ofCncC, Maf, andCYP4BQ1and led to decreased tolerance of adults to (+)-α-pinene. In contrast, ingestion of the CncC agonist curcumin elevatedCYP4BQ1expression and enhanced (+)-α-pinene tolerance. The results demonstrate that, inD. armandi, (+)-α-pinene inducesCYP4BQ1via activation of the ROS/CncC signaling pathway. Keywords:Dendroctonus armandi;phytochemical tolerance;cytochrome P450;oxidative burst;CncC pathway

The Chinese white pine beetle (Dendroctonus armandi) is undoubtedly one of the most important pests causing ecological damage in the Qinling Mountains. When bark beetles invade conifers, they must overcome host tree defenses, including primary resistance and induced resistance responses. Moreover, this induced resistance occurs following herbivory by bark beetles. Bark beetles have a corresponding defense mechanism for degrading toxic compounds, and glutathione S-transferases (GSTs) can catalyze the binding of endogenous substances that reduce glutathione (GSH) to various harmful electrophilic substrates, increasing their solubility and facilitating their excretion from cells. In this experiment, we successfully obtained sixteen full-length sequences of D. armandi, which belonged to four GST categories (delta, epsilon, sigma, and theta). The transcript levels of sixteen GSTs in D. armandi were compared at four developmental stages (larvae, pupae, teneral adults, and adults), three different tissues (antennae, gut, and reproductive organs), and under various levels of terpenoid stimuli during feeding on phloem tissue to evaluate the various relevant modes of action. This study aids in the understanding of the interaction between monoterpenes and beetles, and beetles’ detoxification through GSTs.

The aryl hydrocarbon receptor (AhR) and aryl hydrocarbon receptor nuclear translocator (ARNT) belong to the bHLH-PAS (basic Helix–Loop–Helix–Period/ARNT/Single-minded) family of transcription factors, which participate in the sensing and transmitting stimuli of exogenous and endogenous chemical substances, and subsequently activates genes transcription involved in various detoxification and physiological functions. However, they have not been identified inDendroctonus armandi, and their roles in the detoxification metabolism are unclear. In the present study,AhRandARNTofD. armandiwere characterized. Spatiotemporal expression profiling indicated thatDaAhRandDaARNTwere highly expressed in the adult and larval stages ofD. armandiand mainly expressed in the midgut and Malpighian tubules of adults. Additionally, the expression ofDaAhRandDaARNTsignificantly increased after exposure to (−)-?-pinene, (+)-3-carene, and (±)-limonene. SilencingDaAhRandDaARNTincreased the susceptibility ofD. armandito (−)-?-pinene, (+)-3-carene, and (±)-limonene, and the activities of detoxification enzyme were also remarkably reduced. Moreover,DaCYP6DF1andDaGSTs2were significantly down-regulated after injections ofdsAhRanddsARNTin the male and female adults, with the expression ofDaCYP6DF1decreasing by higher than 70%. The present study revealed that the transcription factorsAhRandARNTofD. armandiwere induced by terpenoids and participated in the regulation ofDaCYP6DF1 expression, which was associated withD. armandi’s susceptibility to (−)-?-pinene and (±)-limonene. These results may provide a theoretical basis for the integrated control ofD. armandiand improve our comprehension of insect toxicology. Keywords:Dendroctonus armandi;AhR;ARNT;detoxification metabolism;transcriptional regulation

Bark beetles oxidize the defensive allelochemicals of their host trees both to detoxify them and convert them into components of their pheromone systems which were catalyzed by cytochrome P450 enzymes (CYPs) and occur in different tissues of the insect. We study P450 genes in the Chinese white pine beetle ( Dendroctonus armandi ), and some bio-information analysis was done for the full-length deduced amino acid sequences. The tissue specificity of these P450 genes was determined in three tissues (antenna, gut and reproductive organs). Differential expression of the P450 genes was observed between sexes, and within these significant differences exposed to stimuli (α-pinene (1:1 racemic mix), (S)-(−)-α-pinene, (S)-(−)-β-pinene, (+)-3-carene, (±)-limonene and turpentine oil) at 24 h. Increased expression of P450 genes suggested that they play a role in the detoxification of monoterpenes released by the host trees. The different transcript accumulation patterns of these bark beetle P450 genes provided insight into ecological interactions of D . armandi with its host pine.

Bark beetles rely on detoxifying enzymes to resist the defensive oleoresin terpenes of the host tree. Insect cytochrome P450 (CYPs) plays a key role in the detoxification of plant allelochemicals and pesticides. CYP6 family is unique to Insecta, and its biochemical function is basically related to catabolize heterologous substances. In this study, two Dendroctonus armandi CYP6 genes, CYP6DF1 and CYP6DJ2, were characterized. Spatiotemporal expression profiling revealed that CYP6DF1 and CYP6DJ2 expressions were higher in larvae and adult stages of D. armandi than in egg and pupae stages, and that two genes predominantly expressed in brain , midgut, fat body, or Malpighian tubules . Moreover, CYP6DF1 and CYP6DJ2 expressions were significantly induced after exposure to (+)- α -pinene. Importantly, silencing CYP6DF1 and CYP6DJ2 significantly inhibited the CYP activity and increased the mortality in the adults fumigated with (+)- α -pinene. Additionally, piperonyl butoxide exposure to adults also increase the sensitivity after treatment with (+)- α -pinene, which led to a significant reduction of the CYP activity, resulting a significant increase in adult mortality. These results suggest that the CYP6 family plays a key role in determining the susceptibility of D. armandi to (+)- α -pinene, which may have implications for the development of novel therapeutics to control this important pest.

Simple SummaryThe Chinese white pine beetle (Dendroctonus armandi) is a highly destructive forest pest that inflicts significant damage to coniferous ecosystems in the middle Qinling Mountains of China. Insect ATP-binding cassette (ABC) transporters are critically involved in mediating xenobiotic metabolism. However, the roles ofABCgenes in host terpenoid detoxification remain uncharacterized inD. armandi. In the present study, verapamil (an ABC transporter inhibitor) significantly increased beetle mortality induced by four monoterpenes. Then we characterized threeABCHsubfamily genes. All three genes were upregulated during developmental transitions and predominantly expressed in Malpighian tubules, fat body, and midgut. Notably,DaABCH1was significantly induced by α-pinene and limonene and silencingDaABCH1significantly increased mortality, which indicatedDaABCH1represents a critical candidate gene implicated in plant chemical defense mechanisms.Bark beetles depend on detoxifying enzymes to counteract the defensive terpenoids produced by host trees. Insect ABC transporters play a critical role in the detoxification of insecticides and plant secondary metabolites. However, the specific functions ofABCgenes in the metabolism of host allelochemicals remain unclear inD. armandi. In this study, we observed that verapamil significantly enhanced the mortality of host allelochemicals in beetles, indicating that ABC transporter genes are involved in the metabolism of monoterpenes byD. armandi. We then sequenced and characterized the full-length cDNAs of threeABCHsubfamily genes (DaABCH1–DaABCH3) fromD. armandi. Spatiotemporal expression profiling revealed that all three genes were upregulated during developmental transitions (egg to larva and pupa to adult) and tissue-specific enrichment in detoxification-related organs (Malpighian tubules, fat body, and midgut). Additionally,DaABCH3expression was detected in the hindgut and brain. Furthermore,DaABCH1andDaABCH2were significantly induced by treatment with α-pinene and limonene, whereasDaABCH3was induced by β-pinene and limonene. Importantly, silencingDaABCH1significantly increased mortality in adults fumigated with α-pinene and limonene. These results strongly suggest thatDaABCH1acts as a key regulator modulatingD. armandi’s sensitivity to host plant allelochemicals. This finding provides a conceptual basis for developing novel control strategies against this economically significant forest pest.

Insect cytochrome P450 (CYPs) plays a key role in the detoxification of pesticides and plant allelochemicals . CYP6 family is unique to Insecta, and its biochemical function is basically related to the metabolism of exogenous substances. In this study, we sequenced and characterized the full-length cDNAs of two CYP6 genes from Chinese white pine beetle, Dendroctonus armandi . Spatiotemporal expression profiling revealed that the expression of CYP6CR2 and CYP6DE5 was higher in larval and adult stages of D. armandi than that in other developmental stages, and that two genes predominantly expressed in brain , midgut, fat body, Malpighian tubules or hemolymph. The expression of CYP6CR2 and CYP6DE5 was significantly induced after feeding on the phloem of Pinus armandii and exposure to six stimuli [(±)- α -pinene, (−)- α -pinene, (−)- β -pinene, (+)-3-carene, (±)-limonene and turpentine]. Importantly, silencing CYP6CR2 and CYP6DE5 separately could increase the sensitivity, led to a significant reduction of the activity of P450, resulting a significant increase in adult mortality after treatment with terpenoids. The comprehensive results of this study showed that in the process of host selection and colonization, the functions of CYPs were mainly to hydrolyze the chemical defense of the host and degrade odor molecules. These findings may help to develop new treatments to control this important pest.

BACKGROUND Bark beetles rely on detoxifying enzymes to resist the defensive terpenoids of their host trees. Research on carboxylesterases (CarEs) has focused on their multiple functions in the metabolic detoxification of pesticides and plant allelochemicals, drug resistance, and juvenile hormone and pheromone degradation. RESULT We identified eight new CarE genes in the Chinese white pine beetle ( Dendroctonus armandi ) and carried out bioinformatics analysis on the deduced full-length amino acid sequences. Differential transcript levels of CarE genes were observed between sexes; within these levels, significant differences were found among the different development stages, and between insects fed on the phloem of Pinus armandi and exposed to five stimuli [(−)-α-pinene, (−)-β-pinene, (+)-3-carene, limonene and turpentine] at 8 and 24 h. CONCLUSION Transcription levels of CarE genes suggest some relationship with the detoxification of terpenoids released by host trees. The functions of bark beetle esterase are mainly in hydrolyzing the host chemical defense and degrading odorant molecules during host selection and colonization. © 2018 Society of Chemical Industry

BACKGROUND Bark beetles rely on their detoxifying enzymes to resist the defensive terpenoids of host trees. Glutathione S -transferases (GSTs) conjugate xenobiotic compounds with a glutathione moiety (GSH) and often work in tandem with cytochromes P450 or other enzymes that aid in the detoxification, sequestration or excretion of toxic compounds. RESULT We identified nine new GST genes in the Chinese white pine beetle ( Dendroctonus armandi ) and carried out a bioinformatics analysis on the deduced full-length amino acid sequences. These genes belong to four different classes (epsilon, sigma, omega and theta). Differential transcript levels of each class of GST genes were observed between sexes, and, within these levels, significant differences were found among the different adult substages that were fed phloem of Pinus armandi and exposed to six stimuli [(±)- α -pinene, (−)- α -pinene, (−)- β -pinene, (+)-3-carene, (±)-limonene and turpentine] at 8 and 24 h. CONCLUSION The increased transcription levels of GST genes suggested that they have some relationship with the detoxification of terpenoids that are released by host trees. The mediating oxidative stress that is caused by monoterpene might be the main role of the bark beetle GSTs. © 2015 Society of Chemical Industry

Simple SummaryThe bark beetle is the most well-known pest in coniferous trees worldwide. These insects only leave the host pine bark when they disperse to locate a new host. Determining howDendroctonus armandiovercome the trees’ terpene-based defense systems has been the key problem in the study of bark beetles. Therefore, the aim of this study was to discover the molecular mechanism of insect detoxification enzymes’ ability to confer resistance to terpenes. For this purpose, the genes of cytochrome P450s, glutathione S-transferases, and carboxylesterases were studied in beetles given diets containing terpenes. The results suggest that beetles express different genes in response to terpenoids, and the responses of multiple detoxifying enzymes indicate these insects’ adaption to their chemical environment.AbstractBark beetles overcome the toxic terpenoids produced by pine trees by both detoxifying and converting them into a pheromone system. Detoxification enzymes such as cytochrome P450s, glutathione S-transferases, and carboxylesterases are involved in the ability ofDendroctonus armandito adapt to its chemical environment. Ten genes from these three major classes of detoxification enzymes were selected to study how these enzymes helpD. armandito respond to the host defenses. The expression profile of these detoxification enzyme genes was observed in adult beetles after feeding on different types of diet. Significant differences were observed between two types of seminatural diet containing the phloem of pines, and a purely artificial diet containing five monoterpenes ((−)-α-pinene, (−)-β-pinene, (+)-3-carene, (±)-limonene, and turpentine oil) also caused differential transcript levels in the detoxification enzyme genes. The results suggest that monoterpenes enter the beetles through different routes (i.e., respiratory and digestive systems) and cause the expression of different genes in response, which might be involved in pheromone metabolism. In addition, the xenobiotic metabolism in bark beetles should be considered as a system comprising multiple detoxifying enzymes. Keywords:artificial feeding;chemical defense;Chinese white pine beetle;cytochrome P450;real-time qPCR

Bark beetles oxidize the defensive allelochemicals from their host trees to both detoxify and convert these materials into components of their pheromone system. The ability of this insect to adapt to its chemical environment might be explained by the action of major detoxification enzymes such as cytochrome P450s (or CYPs). Sixty-four sequences coding for P450s were identified, and most of the transcripts were found to be expressed in the larvae, pupae and adults of Dendroctonus armandi . To gain information on how these genes help D. armandi overcome the host defense, differential transcript levels of the CYP genes were observed between sexes and within the sexes. Significant differences were observed among developmental stages, in feeding on the phloem of Pinus armandi and in exposure to stimuli ((±)-α-pinene, (S)-(−)-α-pinene, (S)-(−)-β-pinene, (+)-3-carene, (±)-limonene and turpentine oil) for 8 h. We investigated the effect of sex and generations on the survivorship of individual D. armandi that were exposed to host volatiles at concentrations comparable to constitutive and induced levels of defense using fumigant exposure to understand the ability of the beetles to tolerate host defensive chemicals. The differential transcript accumulation patterns of CYP genes of these bark beetle provided insight into the ecological interactions of D. armandi with its host pine.
