Efficient optical clearance is fundamental for whole brain imaging. In particular, clearance of the brain without membrane damage is required for the imaging of lipophilic tracer-labeled neural tracts. Relying on an ascending gradient of fructose solutions, SeeDB can achieve sufficient transparency of the mouse brain while ensuring that the plasma membrane remains intact. However, it is challenging to extend this method to larger mammalian brains due to the extremely high viscosity of the saturated fructose solution. Here we report a SeeDB-derived optical clearing method, termed FRUIT, which utilizes a cocktail of fructose and urea. As demonstrated in the adult mouse brain, combination of these two highly water-soluble clearing agents exerts a synergistic effect on clearance. More importantly, the final FRUIT solution has low viscosity so as to produce transparency of the whole adult rabbit brain via arterial perfusion, which is impossible to achieve with a saturated fructose solution. In addition to good compatibility with enhanced yellow fluorescent protein, the cocktail also preserves the fluorescence of the lipophilic tracer DiI. This work provides a volume-independent optical clearing method which retains the advantages of SeeDB, particularly compatibility with lipophilic tracers.

Programed cell death ligand 1 (PD-L1) is a protein biomarker overexpressed on exosomes derived from tumor cells. It plays an important role in tumor diagnosis, screening, evaluation of therapeutic efficacy, and prognosis. In this study, a facile method is presented to detect PD-L1-overexpressing cancer exosomes with high specificity and sensitivity. First, gold nanospheres (GNSs) were attached to the bottom of an eight-well chambered slide by electrostatic adsorption, forming the detection substrate. Then, Cy5-labeled CD63 aptamers (i.e., the capture probes) were modified on the GNSs by Au–S bond. After adding samples containing target exosomes which were stained by membrane dyes DiI in advance, FAM-labeled PD-L1 aptamers (i.e., the immunoprobes) were added to recognize PD-L1 on the target exosomes. By triple-color fluorescence co-localization (TFC) of the Cy5, DiI, and FAM channels, highly sensitive and reliable detection of the PD-L1-overexpressing exosomes was achieved in the concentration range 7.78 × 10 1 to 7.78 × 10 4 particles/mL with a detection limit down to 6 particles/mL. The advantages of the proposed detection method include the following; first, the detection substrate is easy to prepare and convenient to clean. Second, the TFC strategy can completely exclude nonspecific reaction sites and thus significantly improves the accuracy. Such a facile and reliable detection method holds a great potential in exosome-based cancer theranostics. Graphical abstract In this paper, we proposed a triple-color fluorescence co-localization (TFC) strategy to significantly improve the reliability of exosome detection and the detection substrate is easy to prepare and convenient to clean. In addition, the LOD is down to 6 particles/mL, which is quite low compared with other detection methods

Fluorescent probes are extensively used for tracking the in vivo behavior of nanocarriers. However, the impact of fluorescent probes on the in vivo fate of nanomedicines remains uncertain. Potential interference from the probes may lead to misleading conclusions. This study investigated the influence of a commonly used fluorescent probe, DiI, on the in vivo fate of paclitaxel (PTX)-loaded polymeric micelles (PMs) in rats. The drug release kinetics of PTX-loaded PMs (PTX-PM) with DiI/PTX co-loaded PMs (DiI/PTX-PM) in rat plasma was quantitatively compared. The results indicate that co-encapsulation with DiI significantly exacerbated drug release and altered the protein binding of PTX through competitive binding, resulting in an increased proportion of free PTX. DiI enhanced diffusion-driven drug leakage without affecting the micelle dissociation kinetics. Notably, DiI inhibited elimination and increased the systemic exposure of both the free micelle material and the nanocarrier. This study provides evidence that DiI can potentially interfere with the in vivo fate of both the therapeutic payload and the nanocarrier, highlighting the necessity for rigorous validation when employing it for nanocarrier tracking.
