Sugarcane ( Saccharum spp.) is a major sucrose and bioenergy crop in the world. The fungal pathogen Sporisorium scitamineum causes sugarcane smut, a devastating disease that destroys stalks and reduces sugar content in sugarcane. This disease can be controlled most effectively by applying smut-resistant sugarcane varieties. Previous studies have shown that Dirigent (DIR ) genes are involved in the synthesis of the lignin precursor pinoresinol, which plays a crucial role in plant resistance to biotic stresses. However, the immune response of the DIR homologs in sugarcane ( ScDIR ) has not been reported yet. In this study, we found that the lignin content of smut-resistant sugarcane varieties (ZZ1, ZZ6, and ZZ9) was significantly higher than that of smut-susceptible varieties (GT42, ROC22, and FN41), and the lignin content of sugarcane increased after smut infection. The smut-resistant and smut-susceptible clones derived from the same genetic population (ROC25 × YZ89-7) showed similar patterns. Quantitative real-time PCR assays revealed that among the 64 DIR genes in sugarcane, ScDIR5 , ScDIR7 , ScDIR11 , and ScDIR40 showed elevated expression after S. scitamineum infection. In vitro coupling reactions showed that the four corresponding ScDIR proteins could mediate the coupling of coniferyl alcohol and its conversion into the lignin precursor pinoresinol. Overexpression of the four ScDIR genes in Nicotiana benthamiana enhanced disease resistance to the fungal pathogens Sclerotium rolfsii , Rhizoctonia solani , and Botrytis cinerea . Moreover, transgenic sugarcane overexpressing these ScDIR genes showed enhanced resistance to smut disease. Taken together, our findings provide evidence that sugarcane ScDIR genes can improve the resistance of plants to fungal pathogens and highlight their potentials in sugarcane breeding for disease resistance.

Image-guided chemo-photothermal therapy based on near-infrared (NIR) theranostic agents has found promising applications in treating tumors. In this multimodal treatment, it is of critical importance to image real-time distribution of photothermal agents in vivo and to monitor therapeutic outcomes for implementing personalized treatment. In this study, an optimally synthesized dextran-polylactide (DEX-PLA) copolymer was assembled with doxorubicin (DOX) and DiR, a kind of NIR dye, to construct desirable micelles ((DiR + DOX)/DEX-PLA) for performing image-guided chemo-photothermal therapy. These (DiR + DOX)/DEX-PLA micelles had good physical and photothermal stability in aqueous media and showed high photothermal efficiency in vivo. Based on the H22-tumor-bearing mouse model, (DiR + DOX)/DEX-PLA micelles were found to accumulate inside tumors sustainably and to emit strong fluorescence signals for more than three days. The (DiR + DOX)@DEX-PLA micelles together with NIR laser irradiation were able to highly inhibit tumor growth or even eradicate tumors with one injection and two dose-designated 5-minute laser irradiations at the tumor site during 14 days of treatment. Furthermore, they showed almost no impairment to the body of the treated mice. These (DiR + DOX)@DEX-PLA micelles have confirmative translational potential in clinical tumor therapy on account of their persistent image-guided capacity, high antitumor efficacy and good in vivo safety.

Imaging-guided photothermal therapy (PTT) has promising application for treating tumors. Nevertheless, so far imaging-guided photothermal drug-delivery systems have been developed with limited success for tumor chemo-photothermal therapy. In this study, as the proof-of-concept, a stimuli-responsive tumor-targeting rapamycin/DiR loaded lipid-polyaniline nanoparticle (RDLPNP) for dual-modal imaging-guided enhanced PTT efficacy is reported for the first time. In this system, polyaniline (PANI) with π-π electronic conjugated system and effective photothermal efficiency is chosen as the appropriate model receptor of fluorescence resonance energy transfer (FRET), and loaded cyanine probe (e.g., 1,1-dioctadecyl-3,3,3,3-tetramethylindotricarbocyanine iodide, DiR) acts as the donor of near-infrared fluorescence (NIRF). In addition, rapamycin (RAPA), which is used as the antiangiogenesis chemotherapeutic drug, can cutdown the tumor vessels and delay tumor growth obviously. After intravenous treatment of RDLPNPs into Hela tumor bearing mice, fluorescent (from DiR) and enhanced photoacoustic (from DLPNPs) signals were found in tumor site over time, which reached to peak at the 6 h time point. After irradiating with an NIR laser , a good anti-tumor effect was observed owing to the enhanced photothermal and antiangiogenic effect of RDLPNPs. These results show that the multifunctional nanoparticle can be used as a promising imaging-guided photothermal drug delivery nanoplatform for cancer therapy.

Imaging-guided diagnosis and chemo-photothermal combination therapy have promising applications for the treatment of cancer. Nevertheless, the accurate diagnosis and efficient treatment of tumors are not yet satisfactory. Herein, a tumor targeting DiR loaded cisplatin-icodextrin prodrug nanoparticle, with selective drug release, was fabricated as a multifunctional theranostic nanoplatform for chemo-photothermal combination therapy. By loading DiR into the hydrophobic domain of folic acid-icodextrin-polycaprolactone (FA-ICO-PCL, FIP) and cisplatin-icodextrin-polycaprolactone (Pt-ICO-PCL, PtIP) co-assembly, the resultant DiR@(PtIP + FIP) (DPtFIP) NPs had a diameter of around 70 nm and showed excellent tumor targeting ability and negligible side effects. Moreover, the DPtFIP NPs achieved real-time NIR fluorescence imaging of solid tumors with high contrast. By the accurate tumor imaging, local laser irradiation dramatically enhanced the chemotherapy for triple-negative breast cancer. Such a biocompatible nanotherapeutic holds great potential for tumor diagnosis and imaging-guided combinational cancer therapy.

Vivianite is an important bio-induced product of dissimilated iron reduction (DIR), which exists widely in activated sludge and digested sludge of wastewater treatment plants (WWTPs). Extracellular polymeric substances (EPS) surrounding dissimilatory iron-reducing bacteria (DIRB) were reported to be transient media for microbial extracellular electron transfer (EET) and DIR. However, what happens when a layer of EPS envelops microbial cells during vivianite recovery? This study tried to assess the relationship between iron reduction and electron transfer by stripping EPS layer-by-layer, then clarified the delamination of different EPS layers on the performance of vivianite recovery. After stripping total EPS (PC-EPS batches) of DIRB, the Fe (III) reduction efficiencies ( R Fe ), Fe (III) reduction rate ( V Fe ), and vivianite formation efficiency ( R V ) decreased by 33%, 37%, and 30%, respectively. Tightly bound-EPS (TB-EPS) was confirmed to play a crucial role in vivianite formation, which contributed 28–51% to vivianite recovery and 34–55% to iron reduction. Electronic shuttle substances were contained in TB-EPS, which enhanced the EET rate and efficiency between cells and Fe (III). The polysaccharide content of TB-EPS was positively related to adhesiveness between cells and mineral, as well as the iron reduction efficiency. Furthermore, redox proteins (PpcA, OmcB) were embedded in EPS, which shortened the distance of electron hopping to across the EPS layer and further accelerated vivianite recovery from wastewater. Long-term operation of anaerobic reactors confirmed that TB-EPS played critical roles in bio-induced vivianite formation.

Due to the high risk of tearing and rupture, vulnerable atherosclerotic plaques would induce serious cardiovascular and cerebrovascular diseases. Despite the available clinical methods can evaluate the vulnerability of plaques and specifically treat vulnerable plaques before a cardiovascular event, but the efficiency is still low and undesirable. Herein, we rationally design and engineer the low-intensity focused ultrasound (LIFU)-responsive [email protected] nanomedicine for the highly efficient treatment of vulnerable plaques by facilely loading phase transition agent perfluorohexane (PFH) into biocompatible PLGA-PEG-PLGA nanoparticles (PPP NPs) and then attaching dextran sulphate (DS) onto the surface of PPP NPs for targeting delivery. DS, as a typical macrophages-targeted molecule, can achieve the precise vaporization of NPs and subsequently controllable apoptosis of RAW 264.7 macrophages as induced by acoustic droplet vaporization (ADV) effect. In addition, the introduction of DiR and Fe 3 O 4 endows nanomedicine with near-infrared fluorescence (NIRF) and magnetic resonance (MR) imaging capabilities. The engineered [email protected] nanomedicine that uses macrophages as therapeutic targets achieve the conspicuous therapeutic effect of shrinking vulnerable plaques based on in vivo and in vitro evaluation outcomes. A reduction of 49.4% of vascular stenosis degree in gross pathology specimens were achieved throughout the treatment period. This specific, efficient and biosafe treatment modality potentiates the biomedical application in patients with cardiovascular and cerebrovascular diseases based on the relief of the plaque rupture concerns.

Introduction and aims Temporomandibular joint osteoarthritis (TMJ-OA) is characterized by cartilage destruction and pain. Although mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) have shown therapeutic efficacy, their clinical translation is hampered by low yield and batch-to-batch variability. Recent studies indicate that intracellular vesicles (IVs) can be harvested at markedly higher quantities than EVs while displaying comparable biological profiles. Therefore, this study directly compared high-yield dental pulp stem cell-derived intracellular vesicles (DPSC-IVs) with conventional dental pulp stem cell-derived extracellular vesicles (DPSC-EVs) in terms of their ability to regenerate cartilage matrix and relieve pain in TMJ-OA. Methods Following characterization of DPSC-IVs and DPSC-EVs, we first evaluated their effects on the proliferation and migration of rat mandibular condylar chondrocytes (MCCs) using endocytic tracing, CCK-8 assays, and Transwell migration assays. Subsequently, an in vitro inflammation model was established using IL-1β, and Western blot analysis and RT-qPCR were employed to investigate their regulatory effects on cartilage matrix synthesis and inflammatory pathways. Next, an in vivo TMJ-OA model was constructed using sodium monoiodoacetate (MIA). The therapeutic efficacy of the two vesicles on subchondral bone remodelling and condylar cartilage matrix regeneration was multidimensionally validated through micro-CT, RT-qPCR, Western blotting, immunofluorescence and immunohistochemistry staining. Furthermore, their underlying molecular mechanisms for alleviating joint pain were elucidated. Results At the same therapeutic concentration, both DPSC-IVs and DPSC-EVs markedly enhanced MCCs proliferation and migration and suppressed IL-1β-induced cartilage matrix breakdown and inflammatory gene expression. In a rat TMJ-OA model, they equivalently repaired MIA-induced subchondral bone and condylar cartilage damage and significantly attenuated pain via inhibition of the ERK-CREB-CGRP pathway. Conclusion DPSC-IVs, with higher yield and simpler extraction, deliver therapeutic efficacy comparable to DPSC-EVs, offering a more clinically feasible strategy for TMJ-OA treatment.

Chemotherapy has been established as a standard therapeutic regimen for triple-negative breast cancer (TNBC). However, the efficacy of current therapeutic approaches remains suboptimal, primarily attributed to insufficient drug accumulation and the rapid development of chemoresistance. Thus, the rational design of drug delivery systems capable of triggering multimodal cell death pathways represents a promising strategy to combat TNBC. Ferroptosis emerges as a potent form of nonapoptotic cell death, showing great potential for tumor inhibition. The effective codelivery of chemotherapeutic drugs and ferroptosis activators provides potential ideas for the treatment of TNBC. Prodrug-based nanoassemblies have attracted tremendous attention in drug delivery, especially homodimeric prodrugs with ultrahigh drug loading. However, due to their symmetric structures, most homodimeric prodrugs exhibit poor self-assembly ability. Herein, two doxorubicin (DOX) homodimeric prodrugs were designed and coassembled with a GPX4 inhibitor (ML210). The incorporation of ML210 significantly promotes the stability of DOX nanoassemblies and improves the intracellular breakage of the DOX prodrug by regulating the GSH level. As a result, the ML210-driven doxorubicin homodimeric prodrug nanoassemblies exhibit high drug loading capacity, high self-assembly stability, and synergistic chemotherapy-ferroptosis effects. Our results present a viable approach to promote the self-assembly of homodimeric prodrugs while offering an efficient nanoplatform for TNBC treatment.

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal malignancies, marked by extensive stromal fibrosis and a profoundly immunosuppressive, immune-excluded tumor microenvironment (TME) that hampers the efficacy of immune checkpoint blockade. Although photodynamic therapy (PDT) can induce immunogenic cell death (ICD) and stimulate anti-tumor immunity, its effectiveness against PDAC is limited by insufficient immune activation and persistent stromal-mediated immunosuppression. To address these challenges, we develop a liposomal nanodrug that co-encapsulates a mitochondrial-targeted photosensitizer (MP) and a TGF-β receptor inhibitor (LY2109761) to synergize PDT with PD-1 checkpoint blockade. MP selectively accumulates in mitochondria and, upon light activation, amplifies mitochondrial reactive oxygen species production, inducing mitochondrial damage. This damage triggers the release of mitochondrial DNA and damage-associated molecular patterns, activating the STING pathway and eliciting potent ICD and anti-tumor immune responses. Simultaneously, liposome-mediated delivery of LY2109761 mitigates stromal desmoplasia and reverses TGF-β-driven immune suppression, enhancing effector T cell infiltration and activity. In murine PDAC models, this dual-action strategy transforms the immune-cold TME into an immune-inflamed phenotype, sensitizing tumors to PD-1 therapy and leading to pronounced tumor regression and prolonged survival. Our findings present a promising nanodrug-based approach to remodel the fibrotic and immunosuppressive TME of PDAC and enhance immunotherapeutic outcomes.

Atherosclerosis, a leading pathological basis of cardiovascular diseases, is closely associated with oxidative stress, inflammation, and lipid metabolism disorders. Resveratrol has demonstrated anti-atherosclerotic activity, but its clinical application is limited due to poor solubility, short circulation time, and lacking targeting. To overcome these limitations and establish a multi-pathway synergistic strategy for treating atherosclerosis, we designed a stealth lipid nanosphere system with macrophage-targeted and pH-responsive properties. The complete lipid nanospheres were evaluated for biocompatibility, cellular uptake, antioxidant and anti-inflammatory activities, as well as the inhibitory effects on foam cell formation. Pharmacokinetics and targeting efficiency were analyzed using both high-performance liquid chromatography and an in vivo imaging system. Therapeutic efficacy and biosafety were assessed via serum biochemistry and histopathological examination. Results showed that the complete lipid nanospheres exhibited uniform spherical morphology, high encapsulation efficiency, and pH-responsive release. In vitro evaluations demonstrated excellent biocompatibility of the complete lipid nanospheres, along with selective uptake by activated macrophages. Furthermore, the complete lipid nanospheres exhibited potent antioxidant and anti-inflammatory effects, as well as significant inhibition of foam cell formation. In vivo studies demonstrated that the treatment significantly extended the plasma half-life of resveratrol while enhancing its targeted accumulation within atherosclerotic plaques. In ApoE −/− mice with atherosclerosis, this intervention exhibited multifaceted therapeutic effects, including attenuation of oxidative stress, suppression of inflammatory responses, and reduction of lipid deposition. The study concludes that the complete formulation, stealth folic acid-modified pH-responsive lipid nanospheres, can specifically target resveratrol to the activated macrophages in plaques and inhibit plaque progression with minimal side effects, primarily due to resveratrol's antioxidant, anti-inflammatory, and lipid metabolism regulatory activities.

Acute liver injury (ALI), often triggered by an acetaminophen (APAP) overdose, is characterized by severe oxidative stress, inflammation, and hepatocyte apoptosis. Current therapies, such as N-acetylcysteine (NAC), are limited by narrow treatment windows, highlighting the need for more effective antioxidant strategies. In this study, cerium oxide nanoparticles (CeO2 NPs, nanoceria) were synthesized and comprehensively characterized using the transmission electron microscopy (TEM), dynamic light scattering method (DLS), X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS) to confirm their branched morphology, high crystallinity, and mixed Ce3+/Ce4+ valence states. Their enzyme-mimetic antioxidant activities were evaluated through superoxide, hydrogen peroxide, and hydroxyl radical scavenging assays. Nanoceria exhibited excellent cytocompatibility and effectively suppressed the generation of lipopolysaccharide (LPS)-induced reactive oxygen species (ROS) and lipid peroxidation and caspase-3-mediated apoptosis in macrophages. They also downregulated pro-inflammatory mediators (nitric oxide synthase (iNOS), TNF-α, IL-1β, and NLRP3) while enhancing anti-inflammatory markers (Arg1 and IL-10). In an APAP-induced ALI mouse model, nanoceria preferentially accumulated in the liver, alleviated oxidative stress and inflammation, and significantly reduced aspartate aminotransferase (AST) levels, showing hepatoprotective efficacy comparable to NAC. Nanoceria protect against APAP-induced ALI via synergistic antioxidative and anti-inflammatory mechanisms based on reversible Ce3+/Ce4+ redox cycling. These findings underscore nanoceria’s potential as a next-generation nanotherapeutic for oxidative stress-related liver diseases.

Occlusive thrombi within blood vessels are a primary cause of life-threatening cardiovascular diseases, which continue to pose significant global health challenges. Here, we report a polymer nanoplatform that integrates near-infrared II (NIR-II) light-triggered photothermal thrombolysis with intrinsic antithrombotic functionality. A novel semiconducting polymer, featuring intense NIR-II absorption and exceptionally high photothermal conversion, is rationally engineered to enable rapid and localized light-to-heat thrombolysis. A thermosensitive hydrogen sulfide (H 2 S) donor is also incorporated, generating multifunctional “nanosweepers” that combine synergistic photothermal and gas-mediated thrombolytic activity. The formulation is further functionalized with bis-serotonin groups, which confer myeloperoxidase- and hydrogen peroxide-responsive thrombus-targeting capability, ensuring selective accumulation and site-specific therapy. This agent possesses robust NIR-II absorption, high photothermal conversion efficiency (77.2%), and pronounced thrombus-specific enrichment. Importantly, the combination of localized photothermal heating and on-demand H 2 S release enable rapid, near-complete thrombolysis in both carotid and lower-limb arterial thrombosis models, achieving almost full restoration of blood flow. This work establishes a versatile and highly efficient strategy that integrates precise NIR-II photothermal conversion with H 2 S-mediated redox regulation, providing a promising platform for site-specific multimodal synergistic treatment of life-threatening thrombotic disorders. The findings offer new insights into the design of multifunctional, precision therapeutics for vascular disease management.
