Extraction of diosgenin from the rhizome of Dioscorea by catalytic hydrolysis over recyclable solid acids is one of the most enviromentally friendly ways for the conversion of biomass into chemicals. In this paper, a magnetic solid acid, Fe 3 O 4 @SiO 2 @NH-(CH 2 ) 2 -NH 2 @SO 3 H with the particle size of about 80 nm, was synthesized by using Fe 3 O 4 (~ 20 nm) as a magnetic core and then coated orderly with tetraethyl orthosilicate (TEOS) and N-{3-(Trimethoxysilyl) propyl} ethylenediamine (TMPED) by sol-gel reactions in water/ethanol solution, followed by the sulfonation of chlorosulfonic acid. The solid acid was characterized by Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), scanning electron microscope (SEM), X-ray diffraction (XRD), vibrating sample magnetometer (VSM), and X-ray photoelectron spectroscopy (XPS). The content of acidic sites on the surface of the solid acid was 1.01 mmol/g, measured by back titration method. The prepared solid acid was used to hydrolyze and extract diosgenin from Dioscorea nipponica Makino (DNM). The results show that the magnetic solid acid has higher hydrolysis activity than 2.5 M hydrochloric acid under same hydrolysis conditions, at 110 °C for 5 h. In addition, the magnetic solid acid can be easily separated from the reaction mixture by the application of a magnet and reused several times without significant activity loss. This work has a potential application value for the extraction of diosgenin from plants.

To obtaining diosgenin from Dioscorea nipponica Makino ( D. nipponica ) was usually subjected to the multiple processes such as extraction, hydrolysis, transformation and separation etc. In this study, a novel method for rapid production and recovery of diosgenin from D. nipponica was developed by integrating microwave-assisted extraction (MAE), microwave-assisted hydrolysis (MAH) and two-phase extraction (TPE) in one-step procedure, that named MAEH-TPE. Using a two-phase system of ethanol aqueous solution-sulfuric acid-petroleum ether, the MAEH-TPE process was optimized by single-factor experiments and response surface methodology (RSM). Under optimum conditions of ethanol aqueous solution of 60 % (v/v), sulfuric acid concentration of 1.4 mol/L, liquid-to-material ratio of 20 mL/g, extraction temperature of 90 ℃, and extraction time of 35 min, the yield of diosgenin reached 17.09 ± 0.32 mg/g which conformed very closely to the predicted value of 17.185 mg/g, and validation experiments confirmed effectiveness and reliability of the RSM model. Compared with the conventional methods, MAEH-TPE exhibited higher extraction yield, shorter extraction time and simpler operation. The mechanism in MAEH-TPE process was explored in detail by chromatographic analysis and multi-phase equilibria. Enzyme inhibition assay demonstrated that the inhibition rate of the extracted diosgenin on porcine pancreatic lipase (PPL) reached 56.18 ± 1.38 % with IC 50 of 191.42 µg/mL, further proving that diosgenin as the main parent structure of the saponins in D. nipponica dominantly contributed to PPL inhibitory effect. Also, the results proved that MAEH-TPE had excellent extraction performance and simplified the complex processes, reducing energy consumption and saving solvents and resources.

Traditional maceration extraction methods for dioscin typically involve substantial consumption of organic solvents and long extraction times. The present study aimed to develop a green and sustainable extraction procedure for the extraction of Dioscin from Dioscorea nipponica Makino (DNM). A series of switchable natural deep eutectic solvent (SNADES) based on medium chain fatty acids were prepared to achieve high extraction efficiency of Dioscin from DNM. As a result, hexanoic acid and heptanoic acid were selected as the optimal solvents. The optimal extraction conditions were as follows: a molar ratio of 1:1, a liquid-solid ratio of 50 mL·g −1 , an ultrasonic duration of 50 min, and a polarity switch phosphoric acid dosage of 800 μL without additional salt. The SNADES-UALLME method established in this study minimizes the use of organic reagents and provides a new insight for development of green extraction methods for active ingredients from traditional Chinese medicine.

Dioscoreae nipponica Makino ( D. nipponica ) as the rhizome of dioscoreaceae rich in steroidal saponins, has been reported to have the hypolipidemic effects etc. However, it is still unclear which exact active components are primary responsible for the beneficial effects. This study was conducted to fish out the lipase inhibitors from D. nipponica , and evaluate the inhibitory activity on porcine pancreatic lipase (PPL) through in vitro kinetic assay using p -nitrophenyl palmitate as substrate. Accordingly, the ethanolic extract was subjected to D101 macroporous resin purification for spectrophotometric screening, high performance liquid chromatography (HPLC) separation and structural characterization by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry. Through orlistat validation, the PPL inhibitory activity and IC 50 value of the extract were respectively 68.34 ± 1.47 % and 107.05 μg/mL under the optimized inhibition conditions. From 6 steroidal saponins identified, the inhibitory components named the protodioscin, protogracillin, dioscin and gracillin were fished out by grouping separation and HPLC analysis. Furthermore, dioscin and gracillin with the parent structure of diogenin were confirmed as the major inhibitors by virtue of stability tests based on transformation of protodioscin and protogracillin. Finally, the inhibitory mechanism of the major inhibitors toward PPL was further clarified by kinetic analysis and molecular docking analysis. The proposed method not only revealed the PPL inhibitory components in D. nipponica , but also provided an effective approach to hierarchical screening of PPL inhibitors from natural plants.

The rhizome of Dioscorea nipponica Makino (RDN) is a widely used herbal medicine, which has significant anti-inflammatory activities on various inflammatory diseases. However, the bioactive compositions responsible for the anti-inflammatory activity of RDN are still unknown. This study aimed to identify the anti-inflammatory bioactive compounds in RDN using high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (HPLC-Q/TOF-MS), quantitative analysis of multiple components by single marker (QAMS) and chemometric methods. Firstly, an HPLC-Q/TOF-MS method was employed for identification of bioactive steroidal saponins in RND, and a total of twelve steroid saponins were identified. Then, QAMS method was employed to determine the contents of seven bioactive steroidal saponins, including protodioscin, protogracillin, methyl protodioscin, pseudoprotodioscin, pseudoprogracillin, dioscin and gracillin in RND samples using dioscin as the reference analyte. The anti-inflammatory effects of RDN samples were then evaluated by inhibition of NO production in LPS-induced RAW264.7 cells. Furthermore, chemometric methods, including Pearson correlation analysis and partial least squares regression (PLSR) were employed to investigate the correlations between chemical components and anti-inflammatory activities, and explore the potential anti-inflammatory bioactive compounds of RDN. The results indicated that protodioscin, dioscin and gracillin were selected as the major anti-inflammatory compounds in RND. The further verification experiments showed that protodioscin, dioscin and gracillin exhibited great inhibition on NO production with IC 50 values (the half maximal inhibitory concentration) of 0.712 μM, 0.469 μM and 0.815 μM, respectively. They also significantly reduced the levels of TNF-α, IL-1β, and IL-6 in LPS-induced RAW264.7 cells. The present study provided evidences for the anti-inflammatory activity of RND and identification of the anti-inflammatory components in RDN.
