Effective Determination of Diosmin Using Nitrogen Doped Carbon Dots as Probe Based on Internal Filtering Effect

Chen Xian,Zhang Menghan,Zheng Lang,Deng Xiaoqin,Chen Qiang,Han Wendi,Huang Zhengjun,Weng Shaohuang

The establishment of a convenient and efficient testing method is crucial and needed for the monitoring of diosmin. In this study, nitrogen doped carbon dots (N-CDs) with the particle size distribution of 2.5–5.7 nm and the average diameter of 4.1 nm were successfully synthesized using a simple strategy. N-CDs exhibited excellent and stable fluorescence performance with the quantum yield of 22.33%. Correspondingly, a fluorescence analytical method was developed for diosmin determination using N-CDs as probe. UV-vis absorbance spectroscopy and the evaluation of internal filtering parameters verified that the mechanism causing the quenching of N-CDs was an internal filtering effect (IFE). The concentration of diosmin can be directly evaluated based on the quenched fluorescence intensities. After optimizing experimental conditions, it was found that the fluorescence quenching efficiency ((F 0 -F)/F 0 ) of N-CDs exhibited a good linear relationship with the concentration of diosmin (C Diosmin ) in the range of 3.0–50 µg/mL. The limit of detection (LOD) was 0.86 µg/mL based on 3σ/slope ( n  = 13). This method was successfully applied to accurately determine the content of diosmin in diosmin tablets and human plasma samples with good reproducibility. It stands out for its simplicity, speed, and acceptable determination performance.

Preparation of monoclonal antibody against rhoifolin and its application in enzyme-linked immunosorbent assay of rhoifolin and diosmin

Yifan Liu,Zihui Jin,Di Sun,Jiexin Zheng,Bo Xu,Tianyu Lan,Qiyang Zhao,Yue He,Jing Li,Yaohai Zhang,Yongliang Cui

Both rhoifolin and diosmin belong to flavonoids, which are widely present in citrus. Diosmin is not only used in the medical field in the world, but also used as a dietary supplement in the United States. Rhoifolin has a similar structure to diosmin and also exhibits antioxidant and anti-inflammatory properties. In this study, an anti-rhoifolin monoclonal antibody was prepared and an indirect competitive enzyme-linked immunosorbent assay (icELISA) method was established. The half-maximal inhibitory concentration (IC 50 ) of icELISA was determined to be 4.83 ng/mL, and the detection range was 0.97–33.87 ng/mL. The results of UPLC-MS/MS and icELISA generally demonstrate consistency. Moreover, by exploiting the cross-reactivity of the antibody, diosmin in tablets can be detected by icELISA. The results demonstrate that the developed method has good accuracy, reproducibility, and broad application prospects.

Preparation of hapten and monoclonal antibody of diosmin and establishment of enzyme-linked immunosorbent assay

Yifan Liu,Bo Xu,Jiexin Zheng,Zihui Jin,Di Sun,Tianyu Lan,Qiyang Zhao,Yue He,Jing Li,Yongliang Cui,Yaohai Zhang

Diosmin, as one of the research hotspots among flavonoids, has been used medicinally in many countries and as a dietary supplement in the United States. It has been confirmed to be effective in the treatment of hemorrhoids, varicose veins, protecting blood vessels, and managing chronic venous diseases. In this study, an artificial antigen of diosmin was prepared using diosmetin, the aglycone of diosmin. Through screening, specific antibodies against diosmin were obtained. The half maximal inhibitory concentration (IC 50 ) of an indirect competitive enzyme-linked immunosorbent assay (icELISA) was 2.62 ng/mL, with a detection range of 0.52 to 26.39 ng/mL. When applied to the detection of diosmin in limons and tablets, the results obtained by icELISA were consistent with those of Ultra Performance Liquid Chromatography - Tandem Mass Spectrometry (UPLC-MS/MS), demonstrating the good accuracy and reproducibility of this method.

Diosmin mitigates high glucose-induced endoplasmic reticulum stress through PI3K/AKT pathway in HK-2 cells

Deng Jiuhong,Zheng Chao,Hua Zhou,Ci Haideng,Wang Guiying,Chen Lijing

Background Diosmin has been reported to treat diabetes, but its role in diabetic nephropathy (DN) remains unclear. This research investigated the mechanism by which diosmin alleviated high glucose (HG)-induced HK-2 cell injury. Methods First, we used CCK-8 to detect the effect of 0.1, 1, or 10 μg/mL diosmin on the viability of HK-2 cells treated with normal glucose or HG. Next, we used flow cytometry, automatic biochemical analyzer, ELISA, immunofluorescence, and colorimetric assay kit to examine the apoptosis, oxidative stress, inflammatory factors, and Caspase-3 expression in HK-2 cells. Thereafter, we used the western blot and qRT-PCR to examine the expression of the endoplasmic reticulum stress-, oxidative stress-, inflammation-, apoptosis-, and autophagy, and PI3K/AKT pathway-related factors. Results Diosmin was non-cytotoxic to normal HK-2 cells and enhanced the HK-2 cell viability suppressed by HG. Meanwhile, diosmin restrained apoptosis, the contents of MDA, pro-inflammatory factors, and Caspase-3 but intensified the contents of SOD and CAT induced by HG. We further confirmed that diosmin blunted oxidative stress-, inflammation-, apoptosis-, and autophagy-related factors expression induced by HG via restraining the CHOP and GRP78 expressions. Further, we also discovered that PTEN level was restrained and the ratios of p-PI3K/PI3K and p-AKT/AKT were enhanced in HK-2 cells induced by HG, which was reversed by co-treatment of HG and diosmin. Conclusion sOur study manifested that diosmin alleviated the HG-mediated endoplasmic reticulum stress injury in HK-2 cells via restraining the PI3K/AKT pathway.

Diosmin as a green depressant for selective flotation separation of chalcopyrite from pyrite: Adsorption mechanism and performance

Youzhen Nie,Bin Huang,Yanyu Tang,Qicheng Feng,Guang Han

Lime and cyanide are frequently employed as depressants in the flotation process of Cu–Fe sulfide minerals; nevertheless, their application often raises environmental issues and poor selectivity. In this research, the introduction of the semi-synthetic flavonoid Diosmin (Dio) was proposed as an innovative and eco-friendly depressant aimed at achieving the selective chalcopyrite-pyrite flotation separation. Micro-flotation experiments demonstrated that Dio exhibited excellent selective depression toward pyrite. When the dosage was set at 40 mg/L, the recovery of pyrite was controlled below 10%, whereas the flotation performance of chalcopyrite remained largely unaffected, with recoveries exceeding 90%. Zeta potential measurements, contact angle analysis, and atomic force microscopy revealed that Dio could adsorb onto the surfaces of both minerals, with a significantly stronger affinity for pyrite. Moreover, Dio pretreatment effectively inhibited the subsequent collector from adsorbing on the pyrite surface, thereby markedly reducing its floatability. Further insights from FT-IR, XPS, and local electrochemical impedance spectroscopy indicated that Dio interacted preferentially with Fe active sites on the pyrite surface through chemisorption, while exerting a negligible influence on the surface properties of chalcopyrite. These findings demonstrate that Dio functions as an efficient depressant for pyrite and provides a promising green alternative for the flotation separation of chalcopyrite and pyrite.

Diosmin ameliorates LPS-induced depression-like behaviors in mice: Inhibition of inflammation and oxidative stress in the prefrontal cortex

Jingwei Zhao,Mingming Zhang,Huamin Zhang,Ying Wang,Bingyu Chen,Jing Shao

Diosmin is a flavone glycoside with a confirmed therapeutic effectiveness on the chronic venous disorders. In this paper, the classical mouse depression model induced by LPS was established to explore the effect of Diosmin on depression. Firstly, we found that Diosmin could inhibit the inflammation and neuronal damage in the prefrontal cortex (PFC) of mice, and thus alleviating the LPS-induced depressive-like behaviors. Specifically, Diosmin treatment significantly suppressed the secretion of pro-inflammatory cytokines (TNF-α, IL-6 and IL-1β), reduced the activation of microglia, and inhibited the expression of NLRP3 inflammasome and its downstream effector caspase-1 in both PFC of mice and BV2 microglial cells exposed to LPS. Then, we demonstrated that pretreatment with Diosmin dramatically suppressed the LPS-induced oxidative stress in the PFC of mice, manifested in the decrease of reactive oxygen species and malondialdehyde while increase of catalase activity. Consistently, Diosmin also alleviated the oxidative stress in BV2 cells exposed to LPS. Finally, we confirmed that Diosmin effectively suppressed the activation of NF-κB signaling pathway in the PFC of LPS-treated mice. Further in vitro experiments also verified that Diosmin could prevent the p65 transposition to nucleus in LPS-treated BV2 cells, suggesting that the antidepressant effects of Diosmin are partially mediated by blocking of NF-κB signaling. Taken together, this study proposes the potential antidepressant effect of Diosmin, which provides useful support to the development of new therapies for depression.
