The potato rot nematode (Ditylenchus destructor) is one of the most destructive pests in the production of tuber crops, resulting in severely decreased yields and inferior product quality. In 2021, a great number of nematodes were detected in the roots ofMazus japonicus, a weed that is harmful to crop growth, in Qingdao, Shandong Province, China. The present study was undertaken to characterize and identify the nematodes isolated fromM. japonicusthrough morphological identification and molecular approaches. Their morphological characteristics were highly consistent with the descriptions ofD. destructorThorne, 1945. The nematodes collected fromM. japonicuswere identified asD. destructorhaplotype B using D1/D2 and sequence characterized amplified region (SCAR) primers. PCR-ITS-RFLP analysis was conducted to monitor intraspecific variations. In addition, the phylogenetic analysis of the internal transcribed spacer (ITS) demonstrated that thisD. destructorpopulation was clustered in haplotype B, supported by a 100% bootstrap value. Another assay, in whichM. japonicuswas inoculated with a mixture of the life stages ofD. destructor, was performed. This assay showed thatM. japonicusexhibited a high susceptibility toD. destructorin pots. This is the first record ofD. destructorparasitizingM. japonicusin China, and it is of great importance becauseM. japonicuscould be a potential reservoir forD. destructorin the field. Keywords:Ditylenchus destructor;Mazus japonicus;morphological characterization;molecular analysis

The present study investigated the chemical composition of essential oil extracted from Elsholtzia fruticosa, and evaluated the biological activities of the essential oil and its main components against Ditylenchus destructor. The essential oil of E. fruticosa was obtained by hydrodistillation and analyzed by GC/ MS. Twenty-three components of essential oil were identified. The most abundant compounds were eucalyptol (40.1%), γ-terpinene (15.8%), limonene (9.3%) and β-pinene (5.2%). Essential oil and three major constituents (eucalyptol, γ-terpinene, and limonene) were tested for their biological activities against D. destructor. The results showed that the essential oil, eucalyptol, γ-terpinene, and limonene exhibited a strong effect of suppression against D. destructor (EC50/24h = 0.16, 0.27, 0.36 and 0.23 mg/ml, respectively). The essential oil, eucalyptol, ã-terpinene, and limonene also possessed the obvious nematicidal activities against D. destructor (LC50/24h = 0.18, 0.46, 0.66 and 0.38 mg/ml, respectively). The essential oil and main components described herein merit further study as potential botanical nematicides against D. destructor.

Plant-parasitic nematodes pose a severe threat to global agricultural production, creating an urgent demand for green and highly efficient nematicides. In this study, we investigated the nematicidal activity and underlying action mechanism of 4-(diethylamino)salicylaldehyde (DSA) against Ditylenchus destructor . DSA exhibited significant nematicidal activity with a 72 h LC₅₀ of 1.951 μg/mL. At 10 μg/mL, DSA completely inhibited nematode infection, comparable to fluopyram. DSA damaged the body wall, induced reactive oxygen species accumulation and lipid peroxidation, and inhibited antioxidant enzymes. Multi-omics analysis revealed that DSA disrupted glycolysis and the TCA cycle, suppressed pyruvate dehydrogenase activity, and reduced ATP levels. Molecular docking and site-directed mutagenesis confirmed that DSA directly targets the PDH E1α subunit via Arg93 and Tyr92. Notably, DSA showed low toxicity to mammals, birds, and silkworm, with only moderate toxicity to zebrafish. Collectively, this study reveals a PDH-targeted, multi-pathway synergistic nematicidal mechanism of DSA, providing a theoretical basis and a promising lead compound for green nematicide development.

Essential oils (EOs) extracted from botanical resources have greatly been appraised as insecticides against a broad spectrum of pre- and postharvest pests and pathogens. This study aimed to investigate the chemical constituents of the EO from Rhododendron anthopogonoides and the biological activity of EO and its main compound against Tribolium castaneum and Ditylenchus destructor . A total of 14 compounds were identified, and the main components of EO from R. anthopogonoides were 4-phenyl-2-butanone (47.73%), eudesma-3,7(11)-diene (14.52%), curzerene (9.5%), 4-phenylbutan-2-ylacetate (6.43%), β -cadinene (5.79%), and germacrone (5.73%). The results of bioassays indicated that the EO oil of R. anthopogonoides possessed obvious contact activity (LD 50 = 9.83 μg/adult) and repellent activity against T. castaneum . The indoor toxicity indicated that the R. anthopogonoides EO and its main constituent 4-phenyl-2-butanone possessed strong nematicidal activity against D. destructor (LC 50 = 0.21 and 0.04 mg/mL). These results provided the theoretical basis for controlling T. castaneum and D. destructor using the EO from R. anthopogonoides .

This study investigated chemical compositions and biocidal activities from the aerial parts of Rhododendron thymifolium against Tribolium castaneum , Lasioderma serricorne , and Ditylenchus destructor . In total, 13 components were isolated from the aerial parts of R. thymifolium by chromatographic separation techniques. Based on the results of MS, 1 H NMR, and 13 C NMR spectrometry, their structures were determined as ent-kaurane-16β-ol ( 1 ), pentacosane ( 2 ), undecanoic acid, ethyl ester ( 3 ), 1-Tridecanethiol ( 4 ), 1-Dodecanol ( 5 ), 4-Methyl-7-hydroxycoumarin ( 6 ), tertradecan-1-ol ( 7 ), apocynin ( 8 ), 3-Methoxy-4-methylbenzaldehyde ( 9 ), scopoletin ( 10 ), hyperin ( 11 ), quercetin ( 12 ), acetophenone ( 13 ). In addition, ent-kaurane-16β-ol ( 1 ) was isolated from R. thymifolium for the first time. Then, the insecticidal toxicities against T. castaneum , L. serricorne , and D. destructor . of compounds 1–13 were evaluated, and the results showed that apocynin ( 8 ) and acetophenone ( 13 ) exhibited obvious contact activity against the storage pests with LD 50 values of 6.46 and 8.72 μg/adult respectively. Acetophenone ( 13 ) showed excellent contact activity against D. destructor with LC 50 values of 0.08 mg/mL. The study clearly showed the active fractions from R. thymifolium have might potential to be developed into potential compounds on storage insect prevention.
