Olfactory behavioral responses of two Drosophila species and their pupal parasitoid to volatiles from bananas

Yaluan Bi,Xuan Zhang,Xuefei Chang,Jiaxin Li,Shan Xiao,Bo Zhang,Cong Dang,Linlin Sun,Hongwei Yao,Qi Fang,Fang Wang,Gongyin Ye

Background Behavior-based manipulation is an essential part of Drosophila integrated pest management (IPM). Effective compounds are useful for improving the efficiency of baits and the development of attract-and-kill or push–pull strategies to manage Drosophila populations. Here, we investigated the olfactory behavior of two Drosophila species, as well as their pupal parasitoid, to volatiles from bananas, for the identification of effective compounds to control fly populations. Results The results showed that overripe bananas were most attractive to both flies, with a higher attraction index (AI) in Drosophila melanogaster than in Drosophila suzukii . The profiles of volatile organic compounds (VOCs) from bananas with three different ripening stages were compared, and six VOCs were selected for behavioral tests. D. suzukii showed significantly different responses to isoamyl alcohol with D. melanogaster . The effects of ethyl butyrate and ethyl isovalerate on the two flies were dose-dependent, with lower concentrations acting as repellent and higher concentrations acting as attractant. Isoamyl acetate, isoamyl butyrate and isoamyl isovalerate (0.005–0.05% v/v) were attractive to both flies. The parasitoid wasp Pachycrepoidus vindemmiae was repelled by isoamyl alcohol and ethyl butyrate, and attracted by ethyl isovalerate, but showed no significant response to isoamyl acetate, isoamyl butyrate and isoamyl isovalerate at the tested concentration . Conclusion This study indicated that the behavioral response of Drosophila to the odor of chemical compounds is dose-dependent. Isoamyl alcohol and isoamyl acetate at certain concentrations could be used as repellents, whereas ethyl isovalerate as an attractant in control strategy design for D. suzukii. © 2023 Society of Chemical Industry.

5-Hydroxymethylfurfural mediated developmental toxicity in Drosophila melanogaster

Xunlin Chen,Qinghui Tu,Wenzheng Zhao,Xiaorong Lin,Zhongzheng Chen,Bin Li,Yuanyuan Zhang

5-hydroxymethylfurfural is a common byproduct in food. However, its effect on growth and development remains incompletely understood. This study investigated the developmental toxicity of 5-HMF to Drosophila larvae. The growth and development of Drosophila melanogaster fed with 5–50 mM 5-HMF was monitored, and its possible mechanism was explored. It was found that 5-HMF prolonged the developmental cycle of Drosophila melanogaster (25 mM and 50 mM). After 5-HMF intake, the level of reactive oxygen species in the third instar larvae increased by 1.23–1.40 fold, which increased the level of malondialdehyde and caused changes in antioxidant enzymes. Moreover, the nuclear factor erythroid-2 related factor 2 antioxidant signaling pathway and the expression of heat shock protein genes were affected. At the same time, 5-HMF disrupted the glucose and lipid metabolism in the third instar larvae, influencing the expression level of key genes in the insulin signal pathway. Furthermore, 5-HMF led to intestinal oxidative stress, and up-regulated the expression of the pro-apoptotic gene, consequently impacting intestinal health. In short, 5-HMF causes oxidative stress, disturbs glucose and lipid metabolism and induces intestinal damage, damaging related signaling pathways, and ultimately affecting the development of Drosophila melanogaster .

Pharyngeal mechanosensory neurons control food swallow in Drosophila melanogaster

Jierui Qin,Tingting Yang,Kexin Li,Ting Liu,Wei Zhang

As the early step of food ingestion, the swallow is under rigorous sensorimotor control. Nevertheless, the mechanisms underlying swallow control at a molecular and circuitry level remain largely unknown. Here, we find that mutation of the mechanotransduction channel genes nompC, Tmc, or piezo impairs the regular pumping rhythm of the cibarium during feeding of the fruit fly Drosophila melanogaster. A group of multi-dendritic mechanosensory neurons, which co-express the three channels, wrap the cibarium and are crucial for coordinating the filling and emptying of the cibarium. Inhibition of them causes difficulty in food emptying in the cibarium, while their activation leads to difficulty in cibarium filling. Synaptic and functional connections are detected between the pharyngeal mechanosensory neurons and the motor circuit that controls swallow. This study elucidates the role of mechanosensation in swallow, and provides insights for a better understanding of the neural basis of food swallow.

Antiaging effects of rice protein hydrolysates on Drosophila melanogaster

Yang Yue,Mengting Wang,Zhangping Feng,Yanyun Zhu,Jianchu Chen

Rice protein hydrolysates (RPH) prepared by enzymatic hydrolysis have plenty of bioactive functions. Herein, we investigated the antiaging effect of RPH on Drosophila melanogaster (fruit fly) and its mechanisms. According to the results, fruit flies reared on 0.2% and 3.2% RP-supplement diet prolonged their average lifespan, 50% survival days, and the maximum lifespan, together with increasing superoxide dismutase, manganese superoxide dismutase, and catalase activity compared to those reared on basal diet. Further studies showed the lifespan extending effect of RPH was regulated by the cooperation with the intrinsic stress protection system (Nrf2/Keap1), age-related signaling pathway (TOR, S6K) and the expression of longevity genes (methuselah). In conclusion, the lifespan extending effect of RPH makes it possible to be applied in food and healthcare industry. Practical applications In previous studies, rice protein hydrolysates (RPH) have been found to have strong antioxidant properties. But so far, most researches focused on the preparation, identification and in vitro antioxidant experiments of RPH, and there is still a lack of researches on its effect on the antioxidant system of fruit flies and the antiaging of fruit flies. This report showed that RPH enhanced the antioxidant system and prolonged the lifespan of Drosophila, which might help us rationally use rice peptides in functional foods.

Puerarin extends the lifespan of Drosophila melanogaster by activating autophagy

Ai-Wen Kang,Chi Sun,Hai-Tao Li,Kun Zhong,Xu-Hui Zeng,Zhi-Feng Gu,Bing-Qian Li,Xiao-Ning Zhang,Jian-Lin Gao,Tian-Xing Chen

Lifespan longevity has attracted increasing attention with societal development. To counter the effects of aging on longevity, we focused on the natural chemicals of plants. In this study, we investigated the effects of puerarin supplementation on the lifespan of Drosophila melanogaster. Puerarin supplementation significantly extended the lifespan of D. melanogaster at 60 μM and 120 μM by upregulating proteasome subunit beta 5 (prosbeta5) and sirtuin-1 (Sirt1). However, puerarin-induced longevity of male flies (F0 generation) may not be passed on to descendants. Additionally, a puerarin diet for 10 and 25 days did not influence the body weight and food intake of male Canton-S flies. Puerarin significantly improved the climbing ability, starvation resistance, and oxidation resistance of male flies by upregulating the expression of Shaker, catalase (CAT), superoxide dismutase 1 (SOD1), and Methuselah, and downregulating poly [ADP-ribose] polymerase (PARP-1) and major heat shock 70 kDa protein Aa (HSP70). Moreover, 120 μM puerarin supplementation for 25 days significantly increased adenosine 5′ triphosphate (ATP) content by increasing adenosine 5′-monophosphate (AMP)-activated protein kinase (AMPK) levels. Additionally, the puerarin diet for 25 days suppressed male fecundity in male flies by decreasing the levels of Bam and Punt. Mechanistically, puerarin enhanced lysosome-involved autophagy by promoting the expression of lysosome markers [β-galactosidase and lysosomal associated membrane protein 1 (LAMP1)], and elevating the levels of autophagy-related genes, including autophagy-associated gene 1 (ATG1), ATG5, and ATG8b. However, puerarin decreased the phosphorylation of the target of rapamycin (TOR) protein. In conclusion, puerarin is a promising compound for improving the longevity of D. melanogaster by activating autophagy.

Chlordane exposure causes developmental delay and metabolic disorders in Drosophila melanogaster

Qifang Wu,Xueting Du,Xucong Feng,Huimin Cheng,Yingjun Chen,Chenying Lu,Mingjiang Wu,Haibin Tong

The incidence of metabolic diseases is increasing every year, and several studies have highlighted the activity of persistent organic pollutants (POPs) in causing hyperlipidemia and diabetes, and these compounds are considered to be endocrine disrupting chemicals (EDCs). Chlordane is classified as an endocrine disruptor, but the mechanism of how it functions is still unclear. This study investigates the effects of chlordane exposure on Drosophila larvae. Drosophila was cultured in diet containing 0.01 μM, 0.1 μM, 1 μM, 5 μM, and 10 μM chlordane, and the toxicity of chlordane, the growth and development of Drosophila , the homeostasis of glucose and lipid metabolism and insulin signaling pathway, lipid peroxidation-related indicators and Nrf2 signaling pathway were evaluated. We here found that exposure to high concentrations of chlordane decreased the survival rate of Drosophila and that exposure to low concentrations of chlordane caused disruption of glucose and lipid metabolism, increased insulin secretion and impairment of insulin signaling. Notably, it also led to massive ROS production and lipid peroxidation despite of the activation of Nrf2 signaling pathway, an important pathway for maintaining redox homeostasis. Collectively, chlordane causes lipid peroxidation and disrupts redox homeostasis, which may be a potential mechanism leading to impaired insulin signaling and the metabolism of glucose and lipid, ultimately affects Drosophila development.

Bisphenol S induces developmental neurotoxicity by disrupting Lar function in Drosophila melanogaster

Ke Zhang,Xing Zhang,Yaopeng Zheng,Binquan Wang,Xiaoyan Zhang,Yuanyuan Song,Xiaoxiao Luo,Mingkuan Sun

Bisphenol S (BPS), a widely used substitute for Bisphenol A (BPA), has been implicated in neurodevelopmental disorders by numerous studies. While BPS has been known for neurodevelopmental disturbance, the analysis and molecular mechanisms underlying the neurotoxicity are not completely understood. This study utilizes a Drosophila model to investigate the developmental neurotoxicity of BPS and its underlying molecular mechanisms. The results demonstrate that BPS exposure significantly induces abnormal behaviors in Drosophila , including social interaction deficits, increased self-grooming, and impaired locomotor activity. Moreover, BPS disrupts axonal development, leading to abnormal α and β lobes in the mushroom body (MB) and structural damage in the neuromuscular junction (NMJ). NMJ defects are marked by reductions in area, length, perimeter, bouton number, branching, and overall morphological integrity. A genome-wide association study (GWAS) identified Lar as a key mediator of BPS-induced neurotoxicity. Molecular docking simulations and Western blot analyses further confirmed that BPS interferes with the cytoplasmic functions of Lar . Notably, neuronal overexpression of Lar ’s cytoplasmic domain rescued MB and NMJ defects and mitigated BPS-induced behavioral abnormalities. These findings suggest that BPS impairs Lar function, thereby disrupting MB and NMJ morphology and ultimately inducing abnormal behaviors in Drosophila .

Characterization of azadirachtin resistance in a laboratory-selected strain of Drosophila. melanogaster

Kuitun Liu,Cong Rao,Chongwei Li,Wenlin Jiang,Bo Hu,Jianya Su

Azadirachtin, a natural sesquiterpenoid compound, exhibits broad-spectrum insecticidal activity, yet its mechanism of action remains incompletely understood. This study aimed to elucidate its toxicological mechanisms by developing an azadirachtin-resistant Drosophila strain through continuous laboratory selection. After five years of directional screening, we successfully established the Aza-Sel strain with a resistance ratio of 86.7-fold. Genetic analysis revealed that the resistance trait is incompletely recessive and polygenic in nature. Detoxification enzyme and synergism assays demonstrated that elevated activities of glutathione-S-transferases and esterases play crucial roles in the metabolic resistance of this strain. Cross-resistance spectrum analysis showed that the Aza-Sel strain exhibited 12.4-fold and 6.7-fold cross-resistance to emamectin benzoate and dinotefuran, respectively, while displaying negative cross-resistance to chlorpyrifos and indoxacarb, with no significant cross-resistance to other tested insecticides. Notably, the resistant strain exhibited substantial fitness costs: significant developmental delays across all stages (egg, larva, pupa), prolonged developmental duration, markedly reduced fecundity and egg hatch rate, and significantly shortened male longevity. These findings indicate that the azadirachtin-resistant strain bears severe fitness costs, which may account for the slow development of resistance under laboratory selection conditions. The results confirm the significant advantages of azadirachtin in integrated pest management, particularly its negative cross-resistance characteristics with emamectin benzoate and indoxacarb, making it an ideal option for managing resistant pests.

Toxicological effects and defense mechanisms induced by beta-cypermethrin in Drosophila melanogaster

Guangxia Xu,Xiaoyu Long,Junjie Zhai,Yunhao Liu,Chaowei Wang,Jinbu Li,Xing Wang

Widespread use of the pyrethroid insecticide beta-cypermethrin (beta-CYP) has led to adverse effects on nontarget populations within agroecosystems. Despite the efficacy of beta-CYP in pest control, its toxicological and defense mechanisms remain incompletely understood. In the present study, we explored the toxicological effects, antioxidant mechanisms and immune response against beta-CYP using Drosophila melanogaster , a well-established model organism for the study of insect biology, to represent the broader class of nontarget organisms. We exposed Drosophila larvae to 0.667 μg/mL beta-CYP and revealed that delayed development and caused intestinal epithelial damage in larvae. To gain insights into the molecular underpinnings of these effects, RNA sequencing analysis and quantitative polymerase chain reaction validation were performed. These analyses revealed that the messenger RNA levels of glutathione S-transferase were increased, third instar larvae exhibited an increase in reactive oxygen species content and a corresponding increase in antioxidant enzyme activity in response to beta-CYP exposure, indicating an upregulated response to oxidative stress. Beta-CYP also activated Hippo pathway to resist apoptosis and promote cell proliferation. Moreover, beta-CYP induced melanization and Toll immune pathways involved in immune response in Drosophila larvae, specifically the Toll pathway gene Drs . This activation suggests that Drosophila increases antioxidant defenses and promotes mitosis in damaged tissues as compensatory mechanisms to mitigate the cytotoxic effects of beta-CYP. These findings provide new insight into the mechanisms of beta-CYP–induced toxicity and the defense mechanisms in insects; they may also inform strategies for the sustainable use of insecticides and the development of mitigation measures to protect nontarget species in agroecosystems.

Developmental neurotoxic effects of bisphenol A and its derivatives in Drosophila melanogaster

Jie Wang,Chunyan Wu,Xing Zhang,Yuanyuan Song,Binquan Wang,Ke Zhang,Mingkuan Sun

As a result of the ban on bisphenol A (BPA), a hormone disruptor with developmental neurotoxicity, several BPA derivatives (BPs) have been widely used in industrial production. However, there are no effective methods for assessing the neurodevelopmental toxic effects of BPs. To address this, a Drosophila exposure model was established, and W 1118 was reared in food containing these BPs. Results showed that each BPs displayed different semi-lethal doses ranging from 1.76 to 19.43 mM. Exposure to BPs delayed larval development and affected axonal growth, resulting in the abnormal crossing of the midline of axons in the β lobules of mushroom bodies, but the damage caused by BPE and BPF was relatively minor. BPC, BPAF, and BPAP have the most significant effects on locomotor behavior, whereas BPC exhibited the most affected social interactions. Furthermore, exposure to high-dose BPA, BPC, BPS, BPAF, and BPAP also significantly increased the expression of Drosophila estrogen-related receptors. These demonstrated that different kinds of BPs had different levels of neurodevelopmental toxicity, and the severity was BPZ > BPC and BPAF > BPB > BPS > BPAP ≈ BPAl ≈ BPF > BPE. Therefore, BPZ, BPC, BPS, BPAF, and BPAP should be evaluated as potential alternatives to BPA.

Toxicity assessment of SiO2 nanoparticles on developmental and reproductive traits in Drosophila melanogaster

Junyue Wang,Yang Jiang,Yán Wāng

Silicon dioxide nanoparticles (SiO2 NPs) are widely utilized in industrial and biomedical applications owing to their unique physicochemical properties; however, their potential biological effects require comprehensive evaluation. In this study, the model organism Drosophila melanogaster was employed to investigate the impacts of dietary exposure to SiO2 NPs of different sizes and concentrations on developmental and reproductive outcomes. The assessed parameters included egg-laying rate, pupation time, adult emergence time, pupation rate, adult emergence rate, larval weight, and sex ratio. The results revealed that at concentrations of 0.2% or lower, neither nanoparticle size produced significant effects on development or reproductive capacity. In contrast, exposure to 2% SiO2 NPs (both 15 nm and 30 nm) led to reduced body weight in third instar larvae. Notably, 30 nm SiO2 NPs exposure significantly decreased pupation and adult emergence rates and was associated with delayed pupation and emergence times. Although total egg production remained unchanged, flies exposed to 30 nm SiO2 NPs exhibited an earlier oviposition peak. These findings suggest that exposure to SiO2 NPs at the national standard concentration of 0.2% does not cause notable developmental effects in Drosophila, whereas a tenfold increase in concentration may induce developmental delays. Considering that the 0.2% standard is based on human exposure and accounting for interspecies extrapolation, the 2% concentration may still represent a relevant dose range. Overall, these results indicate that excessive intake of SiO2 NPs could pose toxicological risks and provide a theoretical foundation for further studies on the mechanisms underlying SiO2 NPs-induced toxicity.
