Cloning, overexpression, and characterization of a novel organic solvent-tolerant lipase from Paenibacillus pasadenensis CS0611

Jiaxin Gao,Xiaoyang Ou,Pei Xu,Minhua Zong,Wenyong Lou

We found a novel lipase gene in the Paenibacillus pasadenensis CS0611 strain. The lipase gene sequence was cloned into the pET-28a expression vector to construct a recombinant lipase protein containing 6 × His tags at the C- and N-termini, respectively. High-level expression of the lipase in E.coli BL21 (DE3) was obtained upon induction with IPTG at 20 °C. The recombinant lipase activity was approximately 1631-fold higher than the wild type. His-tagged recombinant lipase was purified rapidly and efficiently by using Ni-charged affinity chromatography with 63.5% recovery and a purification factor of 10.78. The purified lipase was stable in a broad range of temperatures and pH values, with the optimal temperature and pH being 50 °C and 7.0, respectively. Its activity was stimulated to different degrees in the presence of metal ions such as Ca 2+ , Mg 2+ , and some non-ionic surfactants. In addition, the purified lipase was activated by a series of water-miscible organic solvents such as some short carbon chain alcohols and was highly tolerant to some water-immiscible organic solvents.

Defect engineering-mediated OER-enhanced ECL for ultrasensitive detection of E.Coli

Yiting Shao,Xuemei Wang,Hanxi Liu,Jianfei Xia,Feifei Zhang,Zonghua Wang

The electrochemiluminescence (ECL) system of Luminol is frequently constrained by its sluggish reaction kinetics and strong dependence on dissolved oxygen in practical applications. Herein, we report an innovative strategy for enhanced oxygen evolution reaction (OER) catalysis mediated by defect engineering, which provides a new insight into boosting the kinetic process of ECL performance in the Luminol/H 2 O system. In this study, Ferrocene monocarboxylic acid (FcCA) was coordinatively linked to the topological framework of nickel-based metal-organic frameworks (NiBDC MOFs) to construct Fc-NiBDC MOFs, achieving a significant improvement in the OER performance, leading to a 4-fold increase in the ECL intensity of Luminol. This is mainly because FcCA induces coordination defects at adjacent Ni sites, creating a high density of oxygen vacancies to facilitate the conversion of reactive oxygen species (ROS). At the same time, by utilizing its strong adsorption ability for hydroxyl groups, it can significantly increase the local concentration of ROS at the active sites. Furthermore, the introduced Fc/Fc + redox cycle continuously accelerates the generation and transformation of ROS, thereby constructing an efficient ROS cyclic regeneration mechanism. This triple synergistic enhancement effect can effectively promote the sustained generation and efficient enrichment of ROS, thereby significantly improving the ECL efficiency of Luminol immobilized on NiBDC MOFs through amide bonds. The biosensor constructed on this basis was applied for the highly sensitive detection of E.coli , with a limit of detection (LOD) as low as 0.33 CFU mL −1 . This strategy pioneers a new pathway for the defect engineering in MOF-based ECL systems, offering a versatile platform for pathogen diagnostics.

Green synthesis and antimicrobial study on the catechin-functionalized Fe3O4-Ag magnetic nanocomposites

Shengkai Liu,Cheng Chang,Boyu Chen,Han Song,Jiashen Fan,Yujie Fu,Zhiguo Liu

In this study, catechin-functionalized Fe3O4-Ag nanocomposites were synthesized by a hydrothermal method. The motivation for this study was to develop a novel antibacterial agent with enhanced stability and biocompatibility. The objective was to create a nanocomposite combining the antimicrobial properties of silver with the antioxidant and bioactive characteristics of catechin. We hypothesized that the synergistic effect of catechin and Fe3O4-Ag would yield a highly effective antibacterial material against common pathogens. The obtained nanocomposites were characterized by TEM, SEM, AFM, XPS, XRD, FTIR and physical property measurement system (PPMS). TEM images indicated that catechin-functionalized Fe3O4-Ag nanocomposites have a spherical morphology with an average size of 25.7 nm. The SEM and AFM imaging revealed that the nanocomposites appear as a number of large particles with average diameter of 581 nm. XPS and XRD and FTIR measurement confirmed the presence of catechin components, Fe3O4 and Ag in the nanocomposites. Taken together, we conclude that the catechin-Fe3O4-Ag nanocomposites in this study have a jujube cake structure in which the Fe3O4-Ag alloy nanoparticles serve as the jujube and the condensed catechin form into the cake substrate. The antimicrobial test indicated the catechin-functionalized Fe3O4-Ag nanocomposites have obvious inhibitory effects on E.coli, S.aureus, and C.albicans.

Bis(zinc(II)-dipicolylamine)-functionalized sub-2 μm core-shell microspheres for the analysis of N-phosphoproteome

Hu Yechen,Jiang Bo,Weng Yejing,Sui Zhigang,Zhao Baofeng,Chen Yuanbo,Liu Lukuan,Wu Qiong,Liang Zhen,Zhang Lihua,Zhang Yukui

Protein N-phosphorylation plays a critical role in central metabolism and two/multicomponent signaling of prokaryotes. However, the current enrichment methods for O-phosphopeptides are not preferred for N-phosphopeptides due to the intrinsic lability of P-N bond under acidic conditions. Therefore, the effective N-phosphoproteome analysis remains challenging. Herein, bis(zinc(II)-dipicolylamine)-functionalized sub-2 μm core-shell silica microspheres (SiO 2 @DpaZn) are tailored for rapid and effective N-phosphopeptides enrichment. Due to the coordination of phosphate groups to Zn(II), N-phosphopeptides can be effectively captured under neutral conditions. Moreover, the method is successfully applied to an E.coli and HeLa N-phosphoproteome study. These results further broaden the range of methods for the discovery of N-phosphoproteins with significant biological functions.

Electrochemical biosensing for E.coli detection based on triple helix DNA inhibition of CRISPR/Cas12a cleavage activity

Jiuying Cui,Qisheng Luo,Cheng Wei,Xiandong Deng,Hongqu Liang,Jihua Wei,Yuanxun Gong,Qianli Tang,Kai Zhang,Xianjiu Liao

Background Escherichia coli ( E.coli ) is both a commensal and a foodborne pathogenic bacterium in the human gastrointestinal tract , posing significant potential risks to human health and food safety. However, one of the major challenges in E.coli detection lies in the preparation and storage of antibodies. In traditional detection methods, antibodies are indispensable, but their instability often leads to experimental complexity and increased false positives. This underscores the need for new technologies and novel sensors. Therefore, the development of a simple and sensitive method for analyzing E.coli would make significant contributions to human health and food safety. Results We constructed an electrochemical biosensor based on triple-helical DNA and entropy-driven amplification reaction (EDC) to inhibit the cleavage activity of Cas12a, enabling high-specificity detection of E.coli . Replacing antibodies with nucleic acid aptamers (Apt) as recognition elements, we utilized the triple-helical DNA generated by the binding of DNA2 and DNA5/DNA6 double-helical DNA through the entropy-driven amplification reaction to inhibit the collateral cleavage activity of clustered regularly interspaced short palindromic repeats gene editing system (CRISPR) and its associated proteins (Cas). By converting E.coli into electrical signals and recording signal changes in the form of square wave voltammetry (SWV), rapid detection of E.coli was achieved. Optimization of experimental conditions and data detection under the optimal conditions provided high sensitivity, low detection limits, and high specificity. Significance With a minimal detection limit of 5.02 CFU/mL and a linear range of 1 × 10 2 - 1 × 10 7  CFU/mL, the suggested approach was successfully verified to analyze E.coli at various concentrations. Additionally, after examining E.coli samples from pure water and pure milk, the recoveries ranged between 95.76 and 101.20%, demonstrating the method's applicability. Additionally, it provides a feasible research direction for the detection of pathogenic bacteria causing other diseases using the CRISPR/Cas gene editing system.

Qazi Iqra Shafi,Haseena Ihsan,Yufan Hao,Xin Wu,Nehar Ullah,Mohammad Younas,Benqiao He,Mashallah Rezakazemi

This work presents the experimental investigation of flat sheet composite nanofiltration membrane synthesized with chitosan nanoparticles through interfacial polymerization of piperazine with trimesoyl chloride on polyethersulfone/sulfonated polysulfone substrates. The synthesized membrane was tested in wastewater treatment containing inorganic salts and E.Coli . Single binary electrolyte solution of KCl, MgCl 2 , MgSO 4, and Na 2 SO 4 , ternary electrolyte solution, containing a combination of MgCl 2 and MgSO 4 , KCl and MgCl 2 and quaternary electrolyte solution of KCl, MgCl 2, and MgSO 4 as feed were treated in crossflow membrane cell for the water flux and species rejection in the permeate under operating pressure up to 0.5 MPa. The rejection of Na 1+ , K 1+ , Mg 2+ , Cl 1− , and SO 4 2− was observed to be 81, 28, 87, 96, and 98%, respectively with an average water flux up to 214 ± 10 L m⁻ 2 .hr⁻ 1 in the permeate for the binary electrolyte solution. Similarly, the rejection for K 1+ , Mg 2+ , Cl 1− and SO 4 2− was noted to be 33, 94, 97, and 99%, respectively, for ternary electrolyte solution with an average water flux up to 211 ± 10 L m −2 .hr −1 . The quaternary ion system in the feed resulted in an average water flux up to 198 ± 12 L m⁻ 2 .hr⁻ 1 with the rejection of K + , Mg +2 , Cl − and SO 4 −2 as 35, 87, 96, and 99%, respectively. The model feed solution of E. coli after passing through the membrane achieved an E. coli rejection (99%) with water flux up to 220 L m −2 .hr −1 .

Entropy-driven amplification reaction and the CRISPR/Cas12a system form the basis of an electrochemical biosensor for E.coli-specific detection

Longjian Huang,Wenzhao Zhang,Mingxuan Liu,Yuanxun Gong,Qianli Tang,Kaihua Wang,Xianjiu Liao,Kai Zhang,Jihua Wei

We present an innovative biosensor designed for the precise identification of Escherichia coli ( E.coli ), a predominant pathogen responsible for gastrointestinal infections. E.coli is prevalent in environments characterized by substandard water quality and can lead to severe diarrhea, especially in hospital settings. The device employs entropy-driven reactions to synthesize copious amounts of double-stranded DNA (dsDNA), which, upon binding with crRNA, triggers the CRISPR/Cas12a system’s cleavage mechanism. This process results in the separation of a ferrocene (Fc)-tagged DNA strand from the electrode, enhancing the electrochemical signal for E.coli ’s rapid and accurate detection. Our tests confirm the biosensor’s ability to quantify E.coli across a dynamic range from 100 to 10 million CFU/mL, achieving a detection threshold of just over 5 CFU/mL. The development of this electrochemical biosensor highlights its exceptional selectivity, high sensitivity, and user-friendly interface for E.coli detection. It stands as a significant step forward in pathogen detection technology, promising new directions for identifying various bacterial infections through the CRISPR/Cas mechanism.

Biodegradable polyurethane based clay composite and their anti-biofouling properties

Abid Ali,Yue Xiao,Lina Song,Jiankun Hu,Qingqing Rao,Muhammad Shoaib,Bilal Ul Amin,Xiaoli Zhan,Qinghua Zhang

The colonization of tiny marine organisms on ship's hull which are connected directly from seawater , is a costly issue. Biofouling contributes to enhanced hydrodynamic drag, which leads to higher consumption of fuel and greenhouse gas emissions . Therefore, the environment-friendly solution is very necessary for anti-biofouling activity. For this purpose, we have synthesized biodegradable polyurethane (CL-PU) composed of ε-CL and 4,4′-methylenebis(cyclohexyl isocyanate) (H 12 MDI) and 1,4 butanediol (1,4 BD). The synthesized CL-PU was further modified with 4, 5-dicholoro-2-octyl-isothiazolone (DCOIT) and clay by mixing of the solution to make composites. Our study showed that CL-PU/DCOIT/clay composite degraded in the artificial seawater (ASW), enzymatic solution and seawater (through laboratory test) due to the contents of clay. Because the existence of clay reduced the size of the spherulite of polycaprolactone (PCL) in the composite, therefore remarkably improved the crystallinity as determined via the polarizing optical microscope (POM) and differential scanning calorimetry (DSC). The composite acted as carrier of antifoulant (DCOIT) and controlled their release rate. The anti-bacterial and anti-diatom experiments exhibited that the composite with DCOIT contents were effective in preventing the accretion of Escherichia coli ( E.coli) (region inhibition 14 mm) and marine diatom Navicula incerta (82.5% reduction). The degradable CL-PU/DCOIT/clay composite will have tremendous durability and excellent anti-fouling activity for marine biofouling owning to sustainability and tunability without being toxic to aquatic biota.
