The successful application of electrochemiluminescence (ECL) in various fields required continuous exploration of novel ECL signal emitters. In this work, we have proposed a pristine ECL luminophor named NiRu MOFs, which owned extremely high and stable ECL transmission efficiency and was synthesized via a straightforward two-step hydrothermal pathway. The foundation framework of pure Ni-MOFs with the initial structure was layered-pillared constructed by the coordinated octahedrally divalent between nickel and terephthalic acid (BDC). The terephthalates were coordinated and pillared directly to the nickel hydroxide layers and the three-dimensional framework was formed, which had a weak ECL response strength. Then, the ruthenium pyridine complex was recombined with pure Ni-MOFs to produce NiRu MOFs and part of the introduced ruthenium was atomically dispersed in the layered-pillared structure through an ion-exchange method, which led to the ECL luminous efficiency being significantly boosted more than pure Ni-MOFs. In order to verify the superiority of this newly synthesized illuminant, an ECL immunoassay model has been designed, and the results demonstrated that it had extremely strong and steady signal output in practical application. This study realized an efficient platform in ECL immunoassay application with the limit of detection of 0.32 pg mL–1 for neuron-specific enolase (NSE). Therefore, the approach which combined the pristine pure Ni-MOFs and the star-illuminant ruthenium pyridine complex would provide a convenient and meaningful solution for exploring the next-generation ECL emitters.

An electrochemiluminescence (ECL) sensor provides a sensitive and convenient method for early diagnosis of diseases; however, it is still a challenge to develop simple and sensitive sensing platforms based on efficient ECL signals and luminophore groups. Porphyrin-based metal–organic frameworks (MOFs) show great potential in ECL sensing; however, the mechanism and structure–activity relationship, as well as application, are rarely reported. Herein, hydrothermal reactions obtained porphyrin Zr-MOFs (PCN-222) with different specific surface areas, pore sizes, structures, and surface charge states by tuning the reaction time were developed, which served both as the ECL luminophore, coreaction promoter for S2O82–, and a connection in the ECL immunoassay. By progressively controlling the condition of the hydrothermal reaction, PCN-222 with large surface area-abundant micropores can be obtained, which has good conductivity and positively charged surfaces, obtaining excellent ECL performance. The ECL performance and the enhancement mechanism were investigated in detail. Using PCN-222-6h with the best ECL intensity as the immobilization matrix for the aptamer, a highly sensitive and selective assay for thrombin was developed. The decrease of the ECL signal was logarithmically linear with the concentration of thrombin in the range from 50 fg mL–1 to 100 pg mL–1 with a low detection limit of 2.48 fg/mL. This proposed strategy provides a brand new approach for tuning of the structures of MOFs as effective ECL signal probes, thus providing wider possibilities for effective ECL immunoassays in the detection of other biomarkers in diagnosis of diseases.

Cyclometalated Ir(III) complexes with high electrochemiluminescence (ECL) efficiency and appropriate bioconjugation sites are urgently needed in ECL immunoassays (ECLIA). Herein, we report the synthesis, photophysics, electrochemistry, and ECL of six new Ir(III) complexes bearing naphthyl (nap) or adamantane phenyl (adap) substitutions, four of which emit cyan, green, or red light and display 1.7- to 7.5-fold increases in ECL intensity. In combination with DFT/TDDFT calculations, this enhancement is rationalized to the augmented radiative rate that arises from both the strengthened spin-orbit coupling (SOC) and the increased transition dipole moment. In addition, the adap-based Ir(III) complex shows high binding affinity with β-cyclodextrin (β-CD) due to the strong hydrophobic interaction, which enables us to develop a modular strategy for the labeling of Ir(III) complexes with biomolecules and to use hydrophobic luminophores in the aqueous-phase detection. As demonstrated, a novel ECLIA is built up and exhibits a wide linear range from 1 ng/mL to 10 μg/mL and a detection limit of 72 pg/mL for the determination of C-reactive protein (CRP). These findings provide new insights into the design, synthesis, and bio-labeling of highly emissive Ir(III) complexes and pave the way for the development of novel ECLIA based on host–guest recognition motifs.
