Preparation of food-grade EDC/NHS-crosslinked gelatin nanoparticles and their application for Pickering emulsion stabilization

Huan Gong,Ye Zi,Guangyi Kan,Li Li,Cuiping Shi,Xichang Wang,Jian Zhong

Herein, a safe desolvation and crosslinking method was developed to prepare food-grade bovine bone gelatin (BBG) nanoparticles for Pickering emulsion stabilization. The nanoparticle-like structures were formed by adjusting pH 9.0 and adding ethanol, and then stable nanoparticles were formed by using N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide (EDC) and N -hydroxysuccinimide (NHS) as crosslinker. Compared with other pH (2.5, 5.0, 7.0, and 12.0), pH 9.0 was the appropriate pH to prepare BBG nanoparticles. Individual nanoparticles (6.50 nm in height), oligomeric nanoparticles (13.42–22.52 nm in height), and polymeric nanoparticles (obvious liquid-precipitate separation) were formed at EDC·HCl/NHS concentrations of 6, 9–12, and 15–20 mg/mL, respectively. The oligomeric nanoparticles induced the highest emulsion creaming stability. The emulsion creaming ability increased with the increase of BBG nanoparticle concentrations. Low NaCl concentration (e.g., 100 mmol/L) could increase the emulsion creaming stability. Finally, 4 °C was the best storage temperature for fish oil-loaded Pickering emulsions.

Preparation and evaluation of an innovative antibacterial bi-layered composite dressing for skin wound healing

Leilei Sun,Mingbo Li,Tengfei Gong,Jianling Feng

Aim of the study The aim of the current study was to develop collagen-based bi-layered composite dressings with antibacterial property and evaluate the efficiency for wound healing. Materials and methods A bi-layered composite wound dressing was fabricated using two marine biomacromolecules (collagen and chitosan or carboxymethyl chitosan). Non-crosslinked and N-Ethyl-N'-(3-dimethylaminopropyl) carbodiimide/N-Hydroxy succinimide (EDC/NHS) cross-linked collagen sponges fabricated by vacuum freeze-drying technology was used as the inner layer. The medical spun-laced nonwoven coated with chitosan and carboxymethyl chitosan was used as the outer layer. The antibacterial activities against E. coli and S. aureus were evaluated by the inhibition zone assay. Deep second-degree scald model was performed to evaluate the efficiency of bi-layered composite dressings for wound healing. Results In view of comprehensive evaluation of appearance and in vitro antibacterial activity, medical spun-laced nonwoven coated with 3% of chitosan solution was chosen to be used as the optimized preparation conditions to produce the outer layer of composite dressing, which acted as a barrier against microorganisms and provided mechanical support. Furthermore, the results of wound closure and histopathological analysis indicated that EDC/NHS cross-linked collagen-based bi-layered composite dressing was superior to non-crosslinked and commercial products, which stimulated the wound healing process and accomplished deep second-degree scalded skin healing within a time span of 28 days. Conclusion The EDC/NHS cross-linked collagen-based bi-layered composite dressing had immense potential to be applied for an ideal wound dressing for more efficient and faster wound healing. Therefore, the findings provided the essential theoretical basis for great potential of collagen-based composite dressing used in wound healing applications.

Preparation and Optimization of a Biomimetic Triple-Layered Vascular Scaffold Based on Coaxial Electrospinning

Hu Qingxi,Wu Chuang,Zhang Haiguang

Electrospinning is a promising method for preparing bionic vascular scaffolds. In particular, coaxial electrospinning can encapsulate polymer materials in biological materials and provide vascular scaffolds with good biomechanical properties. However, it is difficult to produce a stable Taylor cone during the coaxial electrospinning process. Moreover, glutaraldehyde cross-linked natural biomaterials are cytotoxic. To address these issues, a novel electrospinning process is proposed in this report. A non-ionic surfactant (Tween 80) was added to poly(lactic-co-glycolic acid) electrospinning solution and gelatin-collagen electrospinning solution, which prevented the interfacial effect of coaxial electrospinning due to different core/shell solutions. The as-prepared materials were then cross-linked with the non-toxic coupling agents N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide/N-hydroxysuccinimide (EDC/NHS). By comparing the biomechanical properties of EDC/NHS cross-linked vascular scaffold with glutaraldehyde vapor–cross-linked vascular scaffold, it was found that the fracture strain and biological performance of EDC/NHS cross-linked vascular scaffold were better than those of the glutaraldehyde cross-linked scaffold. Finally, a three-layer bionic vascular scaffold was prepared by the proposed electrospinning process. Biomechanical performance tests were carried out and the prepared scaffold was found to meet the requirements of tissue-engineered blood vessels. The research in this paper provides a useful reference for the preparation and optimization of vascular scaffolds.

Comprehensive assessment of Nile tilapia skin collagen sponges as hemostatic dressings

Leilei Sun,Bafang Li,Wenkui Song,Kai Zhang,Yan Fan,Hu Hou

Nile tilapia skin collagen sponge was fabricated by lyophilization and cross-linked with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide in the presence of N-hydroxysuccinimide (EDC/NHS). The physicochemical properties were examined. The EDC/NHS cross-linked collagen sponge presented an enhanced water absorption capacity. In addition, biocompatibility and hemostatic efficiency were evaluated by acute systemic toxicity assay, dermal irritation test, intradermal reaction test, sensitization test, cytotoxicity, blood clotting assay in vitro , and liver and femoral artery hemorrhage models in vivo . Results showed that the produced collagen sponges before and after EDC/NHS cross-linking had excellent biocompatibility. Furthermore, EDC/NHS cross-linking promoted fibroblast cells viability and proliferation reflected by the MTT reduction assay. Meanwhile, EDC/NHS cross-linked collagen sponge exhibited the best blood clotting ability and hemostatic efficiency in rat femoral artery hemorrhage model in comparison with non-crosslinked and commercial collagen sponges. Our results demonstrated that the fabricated collagen sponges could be used as perfect hemostatic dressings.
