Characterization of three-spot seahorse (Hippocampus trimaculatus) skin collagen and its fibrillar gel reinforced by EDC: A comparative study

Wei Liao,Yangfeng Liu,Guanghua Xia,Xuan Ri Shen,Zhouwei Duan,Jian Yang

In current study, three-spot seahorse ( Hippocampus trimaculatus ) skin collagen (SC) was extracted for the first time, and its physicochemical properties were characterized. Nile tilapia-skin collagen (TC) and pig-skin collagen (PC) were used to the references. The yields of SC and TC were 40.41 ± 1.23 and 52.6 ± 0.97% (dry weight), respectively. The SDS-PAGE and FTIR confirmed that SC, TC, and PC were preliminarily identified as type I collagen with triple helical structure. The denaturation temperatures of SC, TC, and PC were 35.12, 38.66, and 42.56°C, respectively. Gel-forming studies indicated SC exhibited higher capability than TC and PC under the effect of NaCl. After cross-linking by 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide (EDC), SC showed a significant increase in thermal stability and mechanical strength, and electron microscopic observation corroborated this phenomenon. Therefore, the isolated collagen from three-spot seahorse skin can be used in food packaging and biomaterials as alternatives to mammalian collagen. Practical applications In this study, we evaluated a new source of skin collagen from three-spot seahorse which has been large-scale artificially cultured in recent years. The finding of this study demonstrate that SC exhibited higher gel-forming capability under the effect of NaCl, which suggest that it may be used in photographic film, sausage casings, and food additive as alternatives to mammalian collagen. Furthermore, thermal stability and mechanical strength of SC gel have a significant increase after cross-linking by EDC, so it can be further used for biomaterials, such as food packaging, drug delivery, tissue engineering constructs, and wound dressing systems.

Bioinspired fabrication of EDC-crosslinked gelatin/nanohydroxyapatite injectable microspheres for bone repair

Rui Zhang,Mingyue Lin,Chenxin Wang,Yufan Li,Yubao Li,Qin Zou

The present study focuses on the synthesis and evaluation of the gelatin/nanohydroxyapatite (Gel/n-HA) composite microspheres fabricated by the water-in-oil (W/O) emulsification process as potential substrates for bone repair applications. Herein, 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC)-crosslinked Gel/n-HA (10 wt%) composite microspheres were synthesized. The morphology, physicochemical properties, and cellular behavior of the EDC-crosslinked Gel-HA microspheres (ECM) were characterized. In vitro cell culture assays demonstrated that the ECM could effectively promote the adhesion and growth of MG63 cells and in vivo bone repair experiments suggested that ECM possessed sustainable regenerative effects. This study provided a simple strategy to prepare functionalized microspheres that showed great potential in promoting bone regeneration as injectable microscaffolds.

Fabrication of Photo-crosslinked Chitosan- Gelatin Scaffold in Sodium Alginate Hydrogel for Chondrocyte Culture

Zhao Peng,Deng Cuijun,Xu Hongzhen,Tang Xing,He Hailong,Lin Chao,Su Jiansheng

Photo-crosslinked chitosan-gelatin scaffolds were fabricated and applied for chondrocyte culture in vitro. Photocurable methacryloyl chitosan was synthesized and characterized by FTIR and 1H NMR, respectively. Microstructure and mechanical properties of the chitosan-gelatin scaffold treated with or without EDC as crosslinking agent were analyzed by scanning electronic microscopy (SEM), compression and viscoelastic measurement. It is demonstrated that EDC-treated chitosan-gelatin scaffold possesses better porous structure and improved mechanical properties. Photo-crosslinked chitosan-gelatin scaffold could be further integrated in sodium alginate hydrogel using calcium chloride to support proliferation of chondrocytes for over 21 days and maintain spherical phenotype, as evaluated by AlamarBlue assay and SEM, respectively, implying that the chitosan-gelatin-hydrogel system exhibits great cyto-biocompatibility. Results of this study show that photo-crosslinked chitosan-gelatin scaffold in sodium alginate hydrogel is suited as a scaffold candidate for cartilage tissue engineering.

Preparation of Pickering emulsion gels based on κ-carrageenan and covalent crosslinking with EDC: Gelation mechanism and bioaccessibility of curcumin

Bao Zhang,Ran Meng,Xiao-Long Li,Wen-Jie Liu,Jie-Shun Cheng,Wu Wang

Pickering emulsions stabilized by zein/carboxymethyl dextrin nanoparticles were added to the κ-carrageenan-based gel matrix to prepare emulsion gels via EDC - induced covalent crosslinking. Texture, WHC and freeze–thaw stability of the emulsion gels increased after crosslinking. The Confocal laser scanning microscope and SEM suggested that droplet clusters could be observed in gel with higher concentration of emulsion. The rheological measurements indicated that the viscosity and gel-like structure were relied on crosslinking agent and emulsion concentration. The photothermal stability of curcumin was significantly enhanced after crosslinking. In addition, in vitro digestion study suggested that the bioaccessibility of curcumin in emulsion gel crosslinked was lower compared to emulsion gel without crosslinking agent. These studies might facilitate the preparation of emulsion gels with excellent stability for bioactive compounds delivery in food and pharmaceutical applications.

Lyophilization as a novel approach for preparation of water resistant HA fiber membranes by crosslinked with EDC

Jie Chen,Cheng Peng,Jun Nie,John F. Kennedy,Guiping Ma

The hyaluronic acid (HA) fibers scaffold with an extracellular matrix mimic structure has been prepared via lyophilization. The morphology of the HA fibers varying the concentration from 0.05   wt% to 0.15   wt% was characterized by scanning electron microscope (SEM). The diameter of HA fibers increased and the morphology changed from fiber to sheet-like structure as the concentration of HA solution increased. To enhance the water-resistance, the pure HA fiber membranes were chemical cross-linked by 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC), and confirmed by fourier transform infrared (FT-IR) spectra. In vitro degradation behavior of cross-linked HA fiber membranes in both of water and PBS solutions was investigated, the physical properties were also studied by differential scanning calorimetry (DSC) and thermogravimetry (TG). The results showed that the bonding water capacity increased after crosslinking, and indicated that the crosslinked HA fibers could be used as scaffold in tissue engineering.
