Enrichment of EPA and DHA by enzymatic ethanolysis: Effects of lipases from Thermomyces lanuginosus, Rhizomucor miehei, Rhizopus oryzae and Candida antarctica

Miao Qiao,Xin Hua,Yongjun Yuan,Xi Cao

This work attempts to achieve the enrichment of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in acylglycerols as well as biodiesel production by investigating the performance of various lipases in ethanolysis. The ethanolysis activities of five immobilized lipases in decreasing order were as follows: Lipozyme TL IM, Novozym 40086/Lipozyme RM IM, Novozym 435 and Rhizopus oryzae lipase immobilized on polypropylene (RO PP). EPA could be enriched to a certain content by Lipozyme TL IM, Novozym 40086, Lipozyme RM IM, and RO PP. The enrichment of DHA was achieved well by those four sn -1,3 regioselective lipases, and DHA contents in acylglycerols were the highest when Lipozyme TL IM and Novozym 40086 were used, reaching 2.87-fold and 2.81-fold of the initial content. However, Novozym 435 could not enrich EPA and DHA effectively. Fatty acid ethyl esters produced in this work contained abundant palmitic acid and palmitoleic acid, which could be ideal biodiesel considering their chain length. The competitive factors of five lipases towards DHA were all significantly higher than those towards EPA. When Lipozyme TL IM and Novozym 40086 achieved the maximum DHA contents, their DHA recoveries in acylglycerols reached 91.87% and 89.00%, respectively. The DHA recoveries were all significantly higher than the corresponding EPA recoveries in the reactions catalyzed by five lipases. Moreover, Lipozyme TL IM could be a potential lipase for the separation of EPA and DHA. This study may promote the full use of fish oil and broaden the application of lipases.

Efficient synthesis of DHA/EPA-rich phosphatidylcholine using immobilized phospholipase A1 on a novel microflow support

Qian Zhou,Neng-Bing Long,Rui-Feng Zhang

A new epoxy-based polymerizing/compositing synchronic process was carried out in 3D structure of a melamine sponge. The cured polymer formed a porous and wave-shaped coating on the surface of skeleton in a uniform way. The obtained composite sponge was used as a microflow support for the immobilization of phospholipase A 1 (PLA 1 , from Thermomyces lanuginosus/Fusarium oxysporum ) by co-adsorption/cross-linking with polyethylenimine (PEI). The loading amount and the specific activity of immobilized PLA 1 under the optimal immobilization conditions were 45.7 mg/g support and 9.1 U/mg protein . The PLA 1 -immobilized microflow supports were placed in a columnar reactor, in which transesterification was operated in continuous circulation way. The reaction produced DHA/EPA-incorporated phosphatidylcholine , the hydrolysis side reaction could be inhibited by removal of free water in the reaction medium. Under optimized conditions the incorporation percentage of DHA/EPA and PC yield of 54.3% and 82.4% within 18 h at 55 ℃, respectively. After 10 cycles, the immobilized PLA 1 retained 71.2% of the original activity. The use of microflow support has an advantage on easier scaling up the reactor size towards much larger productivity and higher efficiency.

DHA-phospholipids (DHA-PL) and EPA-phospholipids (EPA-PL) prevent intestinal dysfunction induced by chronic stress

Wanxiu Cao,Chengcheng Wang,Yaoxian Chin,Xin Chen,Yuan Gao,Shihan Yuan,Changhu Xue,Yuming Wang,Qingjuan Tang

Stress exposure can increase the appearance of intestinal dysfunction. DHA and EPA have been shown to possess significant anti-inflammatory and immuno-enhancement bioactivities. The aim of the study was to investigate whether different forms of DHA or EPA would affect intestinal barriers (including intestinal epithelium integrity and immunity responses, gut microbiota and its metabolites) in mice under chronic stress, and might therefore prevent stress induced intestinal dysfunction. Chronic stress caused a series of anomalies in the intestine, including decreased faecal water content, increased pro-inflammatory cytokines (IFN-γ, TNF-α, IL-1β and IL-6), reduced expression levels of ZO-1, occludin and E-cadherin, and aberrant microbiota composition (especially Roseburia spp., Prevotella spp., bifidobacteria and lactobacilli) and its metabolites, mainly LPS, acetic acid, propionic acid and butyric acid. Our data indicated that both DHA-PL and EPA-PL counteracted these adverse effects effectively. In conclusion, DHA-PL and EPA-PL may effectively protect mice against intestinal dysfunction under chronic stress exposure as potential ingredients for functional food.

Effects of Addition of Tea Polyphenol Palmitate and Process Parameters on the Preparation of High-Purity EPA Ethyl Ester

Xuyang Ding,Fujun Liu,Rui Zheng,Xuechen Pei,Ziye Wang,Dayong Zhou,Fawen Yin

High-purity eicosapentaenoic acid (EPA) ethyl ester (EPA-EE) can be produced from an integrated technique consisting of saponification, ethyl esterification, urea complexation, molecular distillation and column separation. In order to improve the purity and inhibit oxidation, tea polyphenol palmitate (TPP) was added before the procedure of ethyl esterification. Furthermore, through the optimization of process parameters, 2:1 (mass ratio of urea to fish oil, g/g), 6 h (crystallization time) and 4:1 (mass ratio of ethyl alcohol to urea, g/g) were found to be the optimum conditions in the procedure of urea complexation. Distillate (fraction collection), 115 °C (distillation temperature) and one stage (the number of stages) were found to be the optimum conditions for the procedure of molecular distillation. With the addition of TPP and the above optimum conditions, high-purity (96.95%) EPA-EE was finally obtained after column separation.

Global acetylome profiling indicates EPA impedes but OA promotes prostate cancer motility through altered acetylation of PFN1 and FLNA

Chao He,Xiuyuan Chen,Ying Chen,Jianying Sun,Manting Qi,Sonia Rocha,Mu Wang

Prostate cancer (PCa) is one of the leading causes of cancer morbidity and mortality in men. Metastasis is the main cause of PCa-associated death. Recent evidence indicated a significant reduction in PCa mortality associated with higher ω-3 polyunsaturated fatty acids (PUFAs) consumption. However, the underlying mechanisms remained elusive. In this study, we applied global acetylome profiling to study the effect of fatty acids treatment. Results indicated that oleic acid (OA, monounsaturated fatty acid, MUFA, 100 µM) elevates while EPA (eicosapentaenoic acid, 100 µM) reduces the acetyl-CoA level, which alters the global acetylome. After treatment, two crucial cell motility regulators, PFN1 and FLNA, were found with altered acetylation levels. OA increased the acetylation of PFN1 and FLNA, whereas EPA decreased PFN1 acetylation level. Furthermore, OA promotes while EPA inhibits PCa migration and invasion. Immunofluorescence assay indicated that EPA impedes the formation of lamellipodia or filopodia through reduced localization of PFN1 and FLNA to the leading edge of cells. Therefore, perturbed acetylome may be one critical step in fatty acid-affected cancer cell motility. This study provides some new insights into the response of ω-3 PUFAs treatment and a better understanding of cancer cell migration and invasion modulation.

Efficient synthesis of DHA/EPA-rich phosphatidylcholine by inhibition of hydrolysis reaction using immobilized phospholipase A1 on macroporous SiO2/cationic polymer nano-composited support

Jia-Qin Wu,Ding-Lin Wang,Xiao-Mei Xu,Neng-Bing Long,Rui-Feng Zhang

Novel nano-composites were prepared by coating epoxy resin-based cationic polymer in nano-thickness via in-situ curing on the wall of macroporous SiO 2 with pore size of 0.5∼1 μm. By changing the thickness of polymer coating the porosity and specific surface area varied in range of 90.4∼83.9 % and 115∼74 m 2 /g, respectively. Through electrostatic adsorption phospholipase A 1 (PLA 1 ) was efficiently immobilized on the nano-composited support, the loading amount of PLA 1 on the nano-composited support reached to a maximum of 138.6 mg/g support , the specific activity of the immobilized PLA 1 reached as high as 21,300 U/g protein . The immobilized PLA 1 was applied for the highly efficient synthesis of DHA/EPA-rich phosphatidylcholine . The incorporation of DHA/EPA reached as high as 74.8 % and the yield of phosphatidylcholine (PC) was 83.1 % within 20 h at 60 °C using PC/ethyl esters mass ratio of 1:6. After 12 cycling reuses the immobilized PLA 1 still retained 57.8 % of the incorporation of DHA/EPA.

N-3 PUFA Deficiency Aggravates Streptozotocin-Induced Pancreatic Injury in Mice but Dietary Supplementation with DHA/EPA Protects the Pancreas via Suppressing Inflammation, Oxidative Stress and Apoptosis

Hong-Yu Zou,Hui-Juan Zhang,Ying-Cai Zhao,Xiao-Yue Li,Yu-Ming Wang,Tian-Tian Zhang,Chang-Hu Xue

It has been reported that dietary n-3 polyunsaturated fatty acids (n-3 PUFAs) exert therapeutic potential for the preservation of functional β-cell mass. However, the effect of dietary n-3 PUFA deficiency on pancreatic injury and whether the supplementation of n-3 PUFA could prevent the development of pancreatic injury are still not clear. In the present study, an n-3 PUFA deficiency mouse model was established by feeding them with n-3 PUFA deficiency diets for 30 days. Results showed that n-3 PUFA deficiency aggravated streptozotocin (STZ)-induced pancreas injury by reducing the insulin level by 18.21% and the HOMA β-cell indices by 31.13% and the area of islet by 52.58% compared with the STZ group. Moreover, pre-intervention with DHA and EPA for 15 days could alleviate STZ-induced pancreas damage by increasing the insulin level by 55.26% and 44.33%, the HOMA β-cell indices by 118.81% and 157.26% and reversed the area of islet by 196.75% and 205.57% compared to the n-3 Def group, and the effects were significant compared to γ-linolenic acid (GLA) and alpha-linolenic acid (ALA) treatment. The possible underlying mechanisms indicated that EPA and DHA significantly reduced the ration of n-6 PUFA to n-3 PUFA and then inhibited oxidative stress, inflammation and islet β-cell apoptosis levels in pancreas tissue. The results might provide insights into the prevention and alleviation of pancreas injury by dietary intervention with PUFAs and provide a theoretical basis for their application in functional foods. Keywords:streptozotocin;polyunsaturated fatty acid;pancreas injury;oxidative stress;inflammation;apoptosis;mouse

Eicosapentaenoic acid (EPA) exhibits antioxidant activity via mitochondrial modulation

Baoping Xiao,Yuanyuan Li,Yanqi Lin,Jingyu Lin,Lingyu Zhang,Daren Wu,Jun Zeng,Jian Li,Jing wen Liu,Guiling Li

Reactive oxygen species (ROS) are mitochondrial respiration byproducts, the accumulation of which may cause oxidative damage and is associated with several chronic health problems. As an essential unsaturated fatty acid, eicosapentaenoic acid (EPA) provides various physiological functions; however, its exact regulatory role remains elusive. The current study aimed to address how EPA regulates cellular antioxidant capacity and the possible mechanisms of action. Upon 48 h of EPA treatment, the ROS levels of HepG2 cells were reduced by at least 40%; the total cellular antioxidant capacity was increased by approximately 50–70%, accompanied by enhanced activities and expression of major antioxidant enzymes. Furthermore, the mitochondrial membrane potential and the mitochondrial biogenesis were dramatically improved in EPA-treated cells. These data suggest that EPA improves cellular antioxidant capacity by enhancing mitochondrial function and biogenesis, which sheds light on EPA as a dietary complement to relieve the oxidative damage caused by chronic diseases.

A comprehensive study of lipid profiles of round scad (Decapterus maruadsi) based on lipidomic with UPLC-Q-Exactive Orbitrap-MS

Chen He,Zexin Sun,Xingchen Qu,Jun Cao,Xuanri Shen,Chuan Li

Round scad ( Decapterus maruadsi ) is rich in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). However, there is no comprehensive information covering its lipidomic profile. In this study, lipid profiles including fatty acid composition, distribution and detailed structure information were explored using gas chromatography-flame ionization detector (GC-FID) and ultra-high-performance liquid chromatography coupled with hybrid quadrupole-orbitrap mass spectrometry (UPLC-Q-Exactive Orbitrap-MS). The results showed that triacylglycerol (TG) was the dominant class, and the EPA and DHA were concentrated as glycerophospholipid (GP). The saturated fatty acids and monounsaturated fatty acids were mainly esterified in positions 1 and 3 of TG and sn -1-position of GP, while the polyunsaturated fatty acids (PUFA) were mainly esterified in sn -2-position of TG and GP. A total of 1282 species from six classes (21 subclasses) including glycerolipids (GL), sphingolipids (SP), GP, fatty acyls (FA), saccharolipids (SL) and prenol lipids (PR) were identified. Several molecular species were characterized with PUFA, especially EPA and DHA in GP. Considering the superior fatty acids composition and distribution of round scad, it is deserved for further exploitation of its marine lipid source on account of the healthy and nutritional functions.

Bio-imprinted lipase-catalyzed production of medium- and long-chain structured lipids rich in n-3 polyunsaturated fatty acids by acidolysis

Xiaoqiang Zou,Mudassar Hussain,Imad Khan,Yanxi Wang,Bangzhi Jiang,Lei Zheng,Yuechao Pan,Jijie Hu,Azqa Ashraf

Medium- and long-chain structured lipids (MLSLs) rich in docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were synthesized by acidolysis of fish oil (Omax 1812, a mixture of sardine and anchovy oils) with caprylic acid (CA) using a lipase bio-imprinted with fatty acids as a catalyst. A bio-imprinted lipase for the acidolysis reaction was first prepared under optimal conditions, and its activity was 1.43 times higher than that of the non-bioimprinted. The bio-imprinted lipase was used as a catalyst to produce such MLSLs, and the reaction conditions were optimized. Under the optimal conditions, equilibrium was achieved in 8 h, and the total and the sn -1, 3 CA contents in the obtained product were 28.72% and 42.26%. Additionally, the total and the sn -2 DHA contents were 14.23% and 21.10%, respectively, and the total and the sn -2 EPA contents were 16.43% and 10.14%, respectively. These obtained MLSLs, rich in sn -1, 3 CA, and sn -2 DHA, EPA can deliver significant health benefits.

Magnetic MOF derivatives from Tunable MIL-88A for enhanced lipase immobilization and phosphatidyl EPA/DHA synthesis

Yuhan Li,Lingmei Dai,Dehua Liu,Wei Du

Metal-organic frameworks (MOFs) with tunable porosity are promising carriers for enzyme immobilization, but their practical application is limited by structural instability and difficult recovery. We previously developed an oxidation-reduction (O-R) pyrolysis strategy for mesoporous MIL-88 A, yielding a magnetic carrier (O-R-Meso) with excellent lipase immobilization and recyclable catalysis. Herein, we verify the applicability of this O-R pathway for Fe-based MIL-88 A with tailored pore structures (micro-, meso-, macroporous) by extending it to microporous C-MIL-88 A and macroporous SOM-MIL-88 A. The O-R derivatives (O-R C, O-R-Meso and O-R-SOM) retained the original morphology and pore channel integrity, with improved specific surface area and sufficient magnetization (13–23 emu/g) for magnetic separation. Notably, after O-R pyrolysis, MIL-88 A with various pore sizes essentially maintained or even enhanced the specific activity of immobilized lipase ( Candida antarctica lipase B, CalB), successfully deriving into magnetic carriers with enhanced catalytic efficiency and recyclability. This superiority originates from the preserved pore structure and biocompatible γ-Fe 2 O 3 surface of the O-R derivatives, which effectively retain the native conformation of CalB. Besides, this work systematically compares the secondary structure of free and immobilized CalB on MOF-derived carriers with distinct pore architectures, providing direct structural evidence for the superior enzyme-carrier compatibility of mesoporous materials. In the transesterification synthesis of phosphatidyl EPA/DHA, CalB@O-R-Meso showed the highest EPA/DHA total incorporation, prominent sn -1 positional selectivity, and optimal reusability, outperforming CalB@O-R-C and CalB@O-R-SOM. Furthermore, kinetic studies on the CalB@O-R-Meso-catalyzed system confirmed a Ping-Pong Bi Bi mechanism and revealed competitive product inhibition by phosphatidyl EPA/DHA ( K i  = 29.8 mM) with negligible internal mass transfer resistance (η ≈ 1). This work establishes a O-R pyrolysis strategy for tunable porosity Fe-MOFs, clarifies pore size-dependent enzyme -carrier compatibility, and provides a fundamental kinetic basis for phosphatidyl EPA/DHA synthesis.

Enzymatic synthesis of phosphatidyl-EPA/DHA using Candida antarctica lipase B immobilized on mesoporous MIL-88 A

Li Yuhan,Wu Guowei,Liu Zeqing,Dai Lingmei,Liu Dehua,Du Wei

Mesoporous MIL-88A (Meso-MIL-88A) shows significant potential as an effective carrier for immobilizing large molecules such as lipases. This study investigates Meso-MIL-88A immobilized lipase for the catalysis of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) incorporation into soybean phosphatidylcholine. Phosphatidyl-EPA and phosphatidyl-DHA are known for their superior health benefits and have garnered significant global attention. Firstly, we developed a novel and green water-washing strategy to synthesize Meso-MIL-88A, demonstrating its significant potential as an effective carrier for immobilizing large molecules such as lipases. Nitrogen adsorption/desorption and XPS analyses revealed that water as the eluent yielded larger average pore diameters and lower sulfur residue content compared to ethanol. This optimized Meso-MIL-88A carrier was used to immobilize Candida antarctica lipase B (CalB@Meso-MIL-88A), which was applied to the production of phosphatidyl-EPA and phosphatidyl-DHA. The immobilized CalB@Meso-MIL-88A exhibited exceptional catalytic efficiency, achieving an unprecedented sn -1 positional incorporation rate of 86.8% (44.2% EPA, 42.7% DHA) with 90% EPA/DHA-ethyl ester (EE) donors, while 97% DHA-EE resulted in a record-high 90.1% DHA incorporation at the sn -1 position. Kinetic studies and molecular docking simulations indicated a higher substrate affinity for DHA-EE, attributed to enhanced transfer efficiency of EPA/DHA-EE in organic solvent system. This study presents the first demonstration of the potential of Meso-MIL-88A for industrial lipase immobilization via this optimized route, offering an eco-friendly and highly efficient catalytic application for nutraceutical synthesis.
