Digital microfluidics (DMF) technology provides high-precision and flexible liquid manipulation capabilities for lab-on-a-chip (LOC) systems, enabling complex biochemical reactions and detection analyses within a compact space. Electrowetting-on-dielectric (EWOD), one of the most widely used actuation mechanisms in DMF devices, is favored for its simplicity, controllability, rapid response, and cost-effectiveness. However, due to challenges in fabricating sensors on hydrophobic surfaces, reports on EWOD chips integrated with biosensors remain limited. In this work, we successfully integrated electrochemical sensors into EWOD chips via plasma treatment for surface modification. The plasma-treated EWOD device maintained excellent droplet actuation performance (movement time <8 s) and achieved glucose detection in the 0-0.6 mM range using the embedded sensor. The limit of detection was determined to be 0.056 mM (S/N = 3). This EWOD chip with integrated printed electrochemical sensors represents a promising tool for diverse biochemical analyses.

The closed-open digital microfluidic (DMF) system offers a versatile and powerful platform for various applications by combining the advantages of both closed and open structures. The current closed-open DMF system faces challenges in scaling up due to electrode structural differences between closed and open regions. Here we developed an adjustable closed-open DMF platform by utilizing the modified slippery liquid-infused porous surfaces (SLIPS) with asymmetric electrowetting on dielectric (AEWOD) as a hydrophobic dielectric layer. The consistent electrode structures of the bottom printed circuit board (PCB) electrode array on both the closed and open regions, and the utilization of a transparent acrylic with floating potential as the top plate allow a low-cost and easily scalable closed-open DMF system to be achieved. The impacts of applied voltage, parallel plate spacing, electrode switching interval, and electrode driving strategies on various droplet manipulations were investigated. The results show that the optimal plate spacings range from 340–510 μm within the closed region. Meanwhile, we also studied the influence of the thickness, geometry, and position of the top plate on the droplet movement at the closed-open boundary. Through force analysis and experimentation, it is found that a thin top plate and a bevel of ∼4° can effectively facilitate the movement of droplets at the boundary. Finally, we successfully achieved protein staining experiments on this platform and developed a customized smartphone application for the accurate detection of protein concentration. This innovative closed-open DMF system provides new possibilities for future applications in real-time biological sample processing and detection.

A dielectric film with high breakdown strength and dielectric constant are crucial for the reliable operation of electrowetting on dielectric (EWOD). In this study, barium titanate (BaTiO3) nanoparticles modified with a silane coupling agent (KH550) were incorporated into poly(vinyl fluoride-trifluoroethylene-chlorotrifluoroethylene) (P(VDF-TrFE-CTFE)) and poly(methyl methacrylate) (PMMA) to prepare a BaTiO3–KH550/P(VDF-TrFE-CTFE)/PMMA composite film to enhance the breakdown strength and dielectric constant of a dielectric layer. Results revealed that BaTiO3–KH550 is uniformly dispersed within the P(VDF-TrFE-CTFE)/PMMA polymer matrix. When the BaTiO3–KH550 content is 10 wt %, the dielectric constant reached 24.6, and the energy density was 2.65 J cm–3, representing increases of 94% and 73%, respectively, compared to the pure P(VDF-TrFE-CTFE)/PMMA film, where was 12.7 and 1.53 J cm–3. Additionally, the breakdown strength of all films remains above 150 kV mm–1 due to the strong interfacial interaction between PMMA and P(VDF-TrFE-CTFE). Furthermore, the breakdown strength can keep at 194.90 kV mm–1. Electrowetting experiments demonstrated that the composite film exhibited a high initial contact angle of 105.04°, with a contact angle variation range of 35.3° under a low DC voltage ranged from 0 to 50 V. The results indicated that this film is potential for applications in low-cost and high-dielectric-performance EWOD dielectric layer materials.

Droplet manipulation is a critical step for microfluidic applications. We present digital microfluidic devices based on electrowetting-on-dielectric (EWOD) for reliable and repeatable droplet manipulation. The EWOD device consists of two flexible conductive films as top and bottom electrodes, and one flexible liquid-infused membrane (LIM) as the hydrophobic insulating layer. LIMs made by infusing various fluids (air, fluorocarbon, silicone and hydrocarbon oils) into polytetrafluoroethylene (PTFE) membranes are evaluated by electrowetting performance of a sessile water droplet. Silicone oil is selected as the filling insulating oil according to its excellent electrowetting performance in good reversibility, low contact angle hysteresis, and negligible volatility. Fast (85 mm⋅s −1 ), repeatable (>5000 times without obvious decay) and continuous droplet (designed routes) moving, merging, mixing and positioning have been demonstrated. The factors affecting droplet transport velocity are investigated, including the infused oil viscosity, the conductive volume and the top-to-bottom gap distance. Both the conductive film and LIM are replaceable and robust, showing reliable and long-term stability. Quantitative chromogenic reactions and a flexible EWOD device are demonstrated. These devices are easy-to-fabricate, replaceable-in-material and robust-in-performance, showing high potential for applications in sample preparation, and flexible sensing and display devices.
