The jasmonic acid (JA) signaling pathway has been demonstrated to play a crucial role in plant defense against herbivorous insects. However, the relationship betweenEctropis obliqua-induced defensive metabolites and the JA signaling pathway in tea plants remains poorly understood. In this study, we investigated seven key special metabolites, includingp-coumaroylputrescine, feruloylputrescine, prunin, naringenin, and three monolignols, to address this knowledge gap. Epicatechin was selected as a positive control based on its well-documented regulation through the JA signaling pathway. Notably, the content of all selected compounds was significantly increased byE. obliquainfestation. Furthermore, exogenous application of high-dose methyl jasmonate (MeJA) induced the accumulation of six of the eight compounds, excludingp-coumaryl alcohol and sinapyl alcohol, whereas low-dose MeJA failed to elicit their accumulation. To confirm the results, we screened two bioactive molecules, D-allose and L-theanine, which significantly increased the endogenous JA levels at low concentrations. Interestingly, neither D-allose nor L-theanine triggered the biosynthesis of these defensive compounds. Additionally, D-allose-treated tea leaves had no significant effect on the performance ofE. obliqualarvae. These findings demonstrate that the metabolic accumulation induced byE. obliquais mediated through a high-threshold JA signaling cascade. This study provides novel insights into the relationship between plant resistance and JA signaling pathway, advancing our understanding of special metabolites mediated plant-insect interactions.

(Z)-3-Hexenyl acetate (3-HAC) is an herbivore-induced plant volatile (HIPV) that plays a crucial role in plant herbivore resistance. In this study, we report the mechanisms underlying the anti-herbivore activity of 3-HAC. Stable-isotope-labeled [2H2]-3-HAC was applied to tea plants and was effectively absorbed by the leaves, subsequently inducing resistance against the destructive herbivore Ectropis obliqua. An integrated omics approach revealed that 3-HAC accelerates jasmonic acid (JA) biosynthesis. A lipoxygenase gene (CsLOX2), an early committed gene in JA biosynthesis, was significantly induced by exogenous 3-HAC exposure, and its suppression attenuated 3-HAC-induced JA biosynthesis and impaired the acquired resistance of tea plants to E. obliqua. Furthermore, two transcription factors, CsNAC2 and CsMYB306, which were co-expressed with CsLOX2 under 3-HAC induction, positively regulated CsLOX2 transcription. Suppression of either one attenuated 3-HAC-induced JA biosynthesis and tea plant resistance to E. obliqua. Simultaneous suppression of both genes showed stronger effects than the suppression of either gene alone. These findings provide insights into the mechanisms of HIPV-induced herbivore resistance in plants and emphasize the agricultural significance of 3-HAC as an environmentally friendly anti-herbivore agent.

Jasmonates (JAs), the group of lipid-derived hormones, were found to control the defense responses in a myriad of plants. Meaningfully, the macrolactones of 12-hydroxy jasmonate isoleucine (12OH-JA-Ile) were reported to induce the defensive response of wild tobacco. However, little to nothing has been known about the elicitation effect of JA-Ile-macrolactones on woody plants to harmful organisms, let alone its underlying mechanisms. Here, we first optimized the synthetic routine using mild toxic reagent isobutyl chloroformate instead of ethyl chloroformate for conjugation, and we used acetonitrile (MeCN) instead of ethyl alcohol for the better dissolution ofp-toluenesulfonic acid (p-TsOH) to gain JA-Ile-macrolactones. JA-Ile-macrolactone5b-treated tea plants significantly inhibited the larvae weight gain ofEctropis obliqualarvae and the lesions caused byColletotrichum camelliae. Furthermore, the expression level ofCsOPR3was significantly upregulated in5b-treated leaves. Meanwhile,5breduced the accumulation of eriodictyol 7-O-glucuronide (EDG) in tea plants, which was confirmed to promote the growth rate ofE. obliqualarvae by artificial diet assay. In conclusion, our study proved that the exogenous application of5bcould induce the tea plant resistance both to herbivoreE. obliquaand pathogenC. camelliae, and EDG was identified as one of the secondary metabolites that could influence the growth rate ofE. obliqua, but it did not directly influence the infection ofC. camelliaein vitro. Further research should be carried out to clarify the mechanism through which5binduces tea plant resistance toC. camelliae.Keywords:JA-Ile-macrolactone;induced resistance;Camellia sinensis;Ectropis obliqua;Colletotrichum camelliae

Insect-induced plant volatile organic compounds (VOCs) may function as either direct defence molecules to deter insects or indirect defence signals to attract the natural enemies of the invading insects. Tea ( Camellia sinensis L.), an important leaf-based beverage crop, is mainly infested by Ectropis obliqua which causes the most serious damage. Here, we report a mechanistic investigation of tea plant-derived VOCs in an indirect defence mechanism against E. obliqua . Parasitoid wasp Parapanteles hyposidrae , a natural enemy of E. obliqua , showed strong electrophysiological response and selection behaviour towards S -linalool and β-ocimene, two monoterpenes with elevated emission from E. obliqua -damaged tea plants. Larvae frass of E. obliqua , which also released S -linalool and β-ocimene, was found to attract both mated female or male Pa. hyposidrae according to gas chromatography-electroantennogram detection and Y-tube olfactometer assays. In a field setting, both S -linalool and β-ocimene were effective in recruiting both female and male Pa. hyposidrae wasps. To understand the molecular mechanism of monoterpenes-mediated indirect defence in tea plants, two novel monoterpene synthase genes, CsLIS and CsOCS-SCZ , involved in the biosynthesis of S -linalool or β-ocimene, respectively, were identified and biochemically characterised. When the expression of these two genes in tea plants was inhibited by antisense oligodeoxynucleotide, both volatile emission and attraction of wasps were reduced. Furthermore, gene expression analysis suggested that the expression of CsLIS and CsOCS-SCZ is regulated by the jasmonic acid signalling pathway in the tea plant.
