Toxicological analysis of Eisenia fetida in soil under the coexistence of rockwool substrate andtricyclazole

Shengyang Zheng,Chenzhe Wang,Haitao Zhao,Qigen Dai,Wei Mao,Ping Liu,Jianbing Lu,Jing Ju,Manhong Huang

This study investigated the combined effects of rockwool, a novel seedling substrate, and tricyclazole (TCA) on the bioavailability of TCA to Eisenia fetida . The single addition of rockwool and TCA alone to the soil inhibited the growth of E. fetida . A high concentration (300 mg·L −1 ) of TCA significantly decreased the biomass of E. fetida . The addition of 20-mesh rockwool reduced this effect on earthworm biomass by decreasing the soil TCA through adsorption, effectively mitigating TCA bioaccumulation in earthworms. A mechanistic analysis showed that the Mg–O functional group on the rockwool surface combined with the C C functional group in TCA to generate Mg–O–C, and the adsorption process was dominated by chemisorption. Toxicology experiments demonstrated that malondialdehyde and cellulase could be used as biomarkers of inhibitory effects of combined rockwool and TCA in soil on E. fetida . Macrogenomic analyses revealed that small particle sizes and high concentrations of rockwool caused co-stress effects on earthworms when TCA was present. When the particle size of rockwool increased, the toxic effect of TCA on earthworms instead decreased at higher rockwool concentrations. Therefore, in practical agricultural production, the particle size of rockwool can be controlled to realize the adsorption of TCA and reduce the toxic effects of TCA and rockwool on earthworms.

Different Nanomaterials for Soil Remediation Affect Avoidance Response and Toxicity Response in Earthworm (Eisenia fetida)

Liu Yaxin,Xu Kun,Cheng Jiemin

The application of nano-level passivating agents in the remediation of soil heavy metal pollution has received widespread attention, but its harm to soil animals should also be addressed. This study explored the effect of three nanomaterials—nanohydroxyapatite apatite (n-HAP), nano zeolite (n-zeolite), and nanometer iron oxide (n-Fe 3 O 4 ), on catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD) activity and malondialdehyde (MDA) content through filter paper contact test. The effects of nanomaterials spiked at 1.5%wt of soils on earthworm avoidance behavior were also be studied, and the crystallinity and surface charge of three nanomaterials were characterized. The results showed that the activities of CAT, SOD and POD and the content of MDA have been changed at different level. And earthworms have obvious avoidance behavior to the three kinds of nanomaterials. Therefore, nanomaterials do have adverse effects on earthworms, and their biological toxicity should be considered when selecting passivating agents for soil heavy metal pollution remediation.

The protective layer formed by soil particles on plastics decreases the toxicity of polystyrene microplastics to earthworms (Eisenia fetida)

Jinbo Liu,Jianjun Qin,Lang Zhu,Kecheng Zhu,Ze Liu,Hanzhong Jia,Eric Lichtfouse

The recent discovery of microplastics contaminants in most ecosystems has raised major health issues, yet knowledge on their impact on soil organisms is limited, especially their toxicity evolution with aging. Herein, the toxicity of polystyrene microplastic (PS-MP) to earthworm ( Eisenia fetida ) along with aging was investigated. Results showed that the 28 d-LC 50 (50% lethal concentration) of PS-MP was 25.67 g kg −1 , whereas that increased to 96.47 g kg −1 after PS-MP initially aged in soil for 28 days, indicating the toxicity of PS-MP decreased with aging. Laser scanning confocal microscope and scanning electron microscope (SEM) found that the toxicity of PS-MP to earthworm may be due to the ingestion of PS-MP by earthworms and the physical damage (e.g., epidermis abrasion and setae loss) of PS-MP to earthworms. Similarly, the levels of reactive oxygen species, antioxidant enzyme activities and malondialdehyde content increased with PS-MP concentrations from 0.1 to 1.5 g kg −1 , but decreased with aging from 7 to 28 days. The integrated biomarker response index also confirmed that the toxicity of PS-MP decreased with aging. SEM found that PS-MP were progressively covered by soil particles during soil aging, inducing the formation of protective layer and increasing the particle size of PS-MP, which prevented direct contact with earthworms and decreased the ingestion of PS-MP, in turn decreased PS-MP toxicity. Overall, our study provides valuable insights for elucidating the effect of aging on the toxicity of microplastics.

Toxic responses of metabolites, organelles and gut microorganisms of Eisenia fetida in a soil with chromium contamination

Ronggui Tang,Xiaogang Li,Yongliang Mo,Yibing Ma,Changfeng Ding,Junsong Wang,Taolin Zhang,Xingxiang Wang

The toxic sensitivity in different physiological levels of chromium (Cr) contaminated soils with environmentally equivalent concentrations (EEC) was fully unknown. The earthworm Eisenia fetida was exposed to a Cr-contaminated soil at the EEC level (referred to as Cr-CS) to characterize the induced toxicity at the whole body, organ, tissue, subcellular structure and metabolic levels. The results showed that the survival rate, weight and biodiversity of the gut microorganisms (organ) had no significant difference (p > 0.05) between control and Cr-CS groups. Qualitative histopathological and subcellular evaluations from morphology showed earthworms obvious injuries. The organelle injuries combined with the metabolic changes provided additional evidence that the Cr-CS damaged the nucleus and probably disturbed the nucleic acid metabolism of earthworms. 2-hexyl-5-ethyl-3-furansulfonate, dimethylglycine, betaine and scyllo-inositol were sensitive and relatively quantitative metabolites that were recommended as potential biomarkers for Cr-CS based on their significant weights in the multivariate analysis model. In addition, the relative abundance of Burkholderiaceae, Enterobacteriaceae and Microscillaceae of the earthworm guts in the Cr-CS group significantly increased, particularly for Burkholderiaceae (increased by 13.1%), while that of Aeromonadaceae significantly decreased by 5.6% in contrast with the control group. These results provided new insights into our understanding of the toxic effects of the EEC level of Cr contaminated soil from different physiological levels of earthworms and extend our knowledge on the composition and sensitivity of the earthworm gut microbiota in Cr contaminated soil ecosystems. Furthermore, these toxic responses from gut microorganisms to metabolites of earthworms provided important data to improve the adverse outcome pathway and toxic mechanism of the Cr-CS if the earthworm genomics and proteomics would be also gained in the future.

Effects of combined exposure to perfluoroalkyl acids and heavy metals on bioaccumulation and subcellular distribution in earthworms (Eisenia fetida) from co-contaminated soil

Zhao Shuyan,Yang Qiao,Wang Bohui,Peng Yihong,Zhan Jingjing,Liu Lifen

The effects of combined exposure to perfluoroalkyl acids (PFAAs) and heavy metals (HMs) including cadmium (Cd), copper (Cu), zinc (Zn), nickel (Ni), and lead (Pb) on earthworms ( Eisenia fetida ) were investigated. The results have demonstrated that the concentrations of labile acid exchangeable Cd, Zn, Ni, Pb, and Cu in soil were enhanced in addition of PFAAs. With PFAAs, the uptake of Cd, Zn, Ni, Pb, and Cu in earthworms was increased compared to those without PFAAs with the order of Cd > Zn > Pb > Ni > Cu. In the presence of HMs, the average biota-to-soil accumulation factors (BSAFs) of PFAAs in earthworms were decreased by 0.498–0.729 times for perfluorooctane sulfonate (PFOS) and 0.606–0.978 times for perfluorooctanoic acid (PFOA), indicating decrease rates of PFOS were higher than those of PFOA. And different levels of HMs led to insignificant different responses on the inhibiting effects of PFAAs uptake in earthworms. The increase of Cd in fraction C (associated with cytosol) and decrease of PFAAs in fraction C and fraction P (associated with tissue fragments, cell membranes, and intact cells) especially for fraction C were revealed when they were combined, suggesting cytosolic PFAAs and Cd were susceptibly mutual effected. This study indicated that PFAAs and metals mutually affected their bioaccumulation and subcellular distribution in earthworms, which will help to understand the fate and risks of PFAAs and metals in co-contaminated soil.

Contributions of enzymes and gut microbes to biotransformation of perfluorooctane sulfonamide in earthworms (Eisenia fetida)

Shuyan Zhao,Bohui Wang,Zhe Zhong,Tianqi Liu,Tiankun Liang,Jingjing Zhan

Perfluorooctane sulfonamide (FOSA) is known as a key intermediate of perfluorooctane sulfonic acid (PFOS) precursors, which can be frequently detected in the environment and biota. FOSA could be bioaccumulated in earthworms from soil, but the contributions of enzymes and gut microbes involved in the biotransformation of FOSA in earthworms have not been identified. Therefore, the effects of enzyme inhibitors and intestinal microflora on biotransformation of FOSA in earthworms were investigated in the present study. FOSA was biotransformed to form PFOS by earthworms obtained from in vivo and in vitro tests. The addition of FOSA had significantly positive effects on cytolchrome P450 (CYP450) and glutathione- s -transferase (GST) activities, suggesting CYP450 and GST are likely involved in the enzymatic transformation. In addition, both 1-Aminobenzotriazole (ABT) and ezatiostat hydrochloride (TLK199), which were selected to inhibit the CYP and GST enzymes, respectively, demonstrated inhibition effects on biotransformation of FOSA in earthworms with a dose-dependent relationship. However, the concentrations of FOSA weren't changed by the bacteria isolated from worm gut, suggesting that gut bacteria did not contribute to FOSA biotransformation in earthworms. The results of this study confirm that the transformation of FOSA in earthworms is mediated mainly by enzymes rather than by gut microbes.

Toxicological effects of microcystin-LR on earthworm (Eisenia fetida) in soil

Wen Hong-Fei,Li Yan-Wen,Xiang Lei,Zhao Hai-Ming,Chen Lei,Cai Quan-Ying,Li Hui,Mo Ce-Hui,Zhou Dong-Mei,Wong Ming-Hung

Microcystin-LR (MC-LR) is a cyclic heptapeptide toxin produced by cyanobacteria in eutrophic water. It can be transferred into soil–crop systems via irrigation and cyanobacterial paste fertilization. No studies have examined the potential toxicity of MC-LR to soil animals. Therefore, in the present study, the toxicological effects of MC-LR on earthworm ( Eisenia fetida ), including survival, growth, reproduction, oxidative stress, and cell viability, were investigated. The LC 50 of MC-LR was 0.149 μg cm −2 at 72 h based on a filter paper test and 0.460 mg kg −1 at 14 days based on an acute soil test. MC-LR seriously affected the reproduction of earthworms. Based on hatchability, the EC 50 of MC-LR was 0.268 mg kg −1 , similar to environmentally relevant concentrations of microcystins. The changes in activities of superoxide dismutase, guaiacol peroxidase, catalase, and glutathione peroxidase, together with the levels of glutathione and malondialdehyde, indicated that oxidative damage and lipid peroxidation played significant roles in MC-LR toxicity. In addition, the toxicity of MC-LR in earthworms increased despite degradation of MC-LR in soil over time, possibly due to the formation of toxic metabolites of MC-LR or the bioaccumulation of MC-LR in earthworms. A reduction in the neutral red retention time along with an increase in coelomocyte apoptosis with increasing MC-LR concentrations indicated a severe damage to viability. These results suggest that environmentally relevant MC-LR concentrations in agricultural soil may cause reproductive, biochemical, and cellular toxicity to Eisenia fetida . This information can be used in ecological risk assessments on MC-LR in soil.

Oxidative potential of heavy-metal contaminated soil reflects its ecological risk on earthworm

Chunmei Ran,Chunguang Liu,Chu Peng,Xinxin Li,Yu Liu,Ye Li,Wenjun Zhang,Hongming Cai,Lei Wang

The ecological risk of heavy metal-contaminated soil is usually evaluated by its oxidative stress on terrestrial animals , which are vulnerable to the impact of individual differences of animals and environmental conditions. Oxidative potential (OP) is the potential of particles to induce the formation of reactive oxygen species (ROS). In this study, OP of the in situ contaminated soils collected from an industrial site (i.e., 64.5–7783 mg/kg of Cu, 54.9–397 mg/kg of Pb, 278–2085 mg/kg of Zn and 0.615–121 mg/kg of Cd) were measured as 38.0–60.1 pmol/min/mg, and the water extractable Cu, Pb, Zn, and Cd in soils have been proved to contribute 89%–98% to OP. Oxidative stress in earthworms ( Eisenia fetida ) caused by the industrial contaminated soils can be reflected by the biomarkers such as 8-hydroxy-2′-deoxyguanosine, metallothionein , and antioxidant enzymes, and a negative correlation presents between the comprehensive biomarker response index and the OP of soils ( r  = −0.979, p  < 0.01). This is the first time to propose a dose-response relationship between OP and oxidative stress. Metabolomic analysis also verified that the regulation of four ROS-related metabolites (i.e., l-pipecolate, 1-methylhistidine, 5-methoxytryptamine, and xanthosine) in earthworms treated with contaminated soil were directly correlated with OP values. These results indicate that OP can be used as an indicator for ecological risk assessment of heavy metal-contaminated soil, which provides a fast, stable and easily testable chemical method.

Comparative toxicity and bioaccumulation of fenvalerate and esfenvalerate to earthworm Eisenia fetida

Xiaoqing Ye,Kang Xiong,Jing Liu

More attention is being paid to the enantioselective toxicity of chiral pesticides. However, limited investigations have been done to assess the ecological risks of chiral pesticides to soil community. Fenvalerate (FV), an extensively used synthetic pyrethroid, is a typical chiral pesticide. The most insecticidally active enantiomer of FV, esfenvalerate (ESFV), also has been marketed and widely used. In this study, the toxicological sensitivity and bioaccumulation of FV and ESFV in earthworms were assessed. The results showed that FV was less toxic than ESFV, but more accumulated in earthworms. ESFV was at least 4 times more toxic to earthworms than FV according to the filter paper contact toxicity test and the artificial soil test. Enantiospecific induction in oxidative stress was observed in earthworms exposed to FV and ESFV. The bioaccumulation of FV and ESFV in earthworm tissues was also enantioselective, preferentially accumulating FV. The uptake of ESFV by earthworms was lower than that of FV, so that the biota to soil accumulation factor (BSAF) value of ESFV was lower than that of FV. Our findings suggest that the enantioselective toxicity and bioaccumulation of chiral pesticides should be considered for evaluating ecological risks of these compounds to non-target organisms.

Effects of benzotriazole on copper accumulation and toxicity in earthworm (Eisenia fetida)

Yanshuai Xing,Xiaoshuang Meng,Lei Wang,Junjie Zhang,Zijing Wu,Xinying Gong,Chenye Wang,Hongwen Sun

Triazole contaminants in water and soil environments can form complexes with metal ions, and therefore affect the bioavailability and toxicity of some heavy metals. In present study, significant increase of copper (Cu) uptake by earthworm ( Eisenia fetida ) was observed when combined pollution of benzotriazole (BTR) presented in soil. For instance, Cu accumulation in earthworms increased 55% approximately when BTR presented at the BTR/Cu molar ratio of 1:2.5. While the single Cu exposure (at 32 mg kg −1 in soil) resulted in increased malondialdehyde (MDA) content in earthworms from 0.319 to 0.668 nmol mg protein −1 , joint exposure to BTR at BTR/Cu molar ratio of 1:10 significantly decreased the MDA content to 0.405 nmol mg protein −1 . This indicates a potential detoxification effect of BTR to Cu induced oxidative damage in earthworms. Varied Cu subcellular distribution can be observed in earthworms of the single and combined exposure treatments. With the combined exposure of BTR, the proportion of Cu associated with granular fraction, the toxically inert fraction in earthworms, increased from 25% to 39%. This phenomenon can be used to explain the protective effects of BTR against oxidative damage.

Multi-omics analysis reveals copper-induced growth inhibition mechanisms of earthworm (Eisenia fetida)

Yanliang Zhang,Jinqi Zhao,Na Sa,Chenyu Huang,Wenyu Yu,Tianxiao Ma,Hongjun Yang,Fang Ma,Siqi Sun,Chaochun Tang,Weiguo Sang

Exposure to high concentrations of copper can cause toxic effects on the growth and development of organisms, but the relevant toxic mechanisms are far from fully understood. This study investigated the changes of metabolites, genes, and gut microorganisms in earthworms ( Eisenia fetida ) exposed to 0 (control), 67.58 (low), 168.96 (medium), and 337.92 (high) mg/kg of Cu in soil for 60 days. Differentially expressed genes (DEGs) and differential metabolites (DMs) at the low-, medium-, and high-level Cu exposure groups were identified and introduced into Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways. Integrated metabolomic and transcriptomic analysis revealed that amino acid metabolism, lipid metabolism , and carbohydrate metabolism are the major metabolic pathways disturbed by Cu exposure. Furthermore, Cu exposure significantly decreased the diversity of the intestinal bacterial community and affected the relative abundance (increased or decreased) of intestinal colonizing bacteria. This resulted in high energy expenditure, inhibited nutrient absorption and fatty acid synthesis, and weakened antioxidant and detoxification abilities, ultimately inhibiting the growth of E. fetida . These findings offer important clues and evidence for understanding the mechanism of Cu-induced growth and development toxicity in E. fetida and provide further data for risk assessment in terrestrial ecosystems .

Soil Matrix Modulates Polystyrene Microplastic Toxicity to Eisenia fetida: From Acute Lethality to Oxidative Genotoxicity

Xiaoling Xu,Chuanxiang Li,Haochen Sun,Jian Wang,Jinbo Liu

Microplastic (MP) contamination in soils has raised widespread concern, yet the extent to which soil matrices regulate MP toxicity remains insufficiently characterized. In this study, the acute and subchronic toxicity of polystyrene (PS) to the earthworm ( Eisenia fetida ) was analyzed in artificial soil and three types of natural soils (lou soil, black soil and latosol soil). The 28 d LC 50 (50% lethal concentration) of PS in artificial soil was 130.86 g kg −1 , whereas it decreased to 39.71 and 39.23 g kg −1 in lou and latosol soil, respectively, and increased to 175.13 g kg −1 in black soil, indicating that soil physicochemical properties may modulate MP toxic potency. Across all soil types, PS exposure triggered reactive oxygen species (ROS) accumulation in earthworms, which in turn activated antioxidant and detoxification enzymes (superoxide dismutase, catalase, and glutathione S-transferase) and ultimately led to lipid peroxidation and DNA damage. Assessment via the integrated biomarker response index suggested divergent PS toxicity across soils, indicating inconsistencies between artificial soil and natural soils. Notably, this study was conducted under controlled laboratory conditions using a single polymer type, particle size, and test organism, and therefore the observed soil-dependent toxicity patterns may not be directly extrapolated to other MP types, sizes, or soil biota. Overall, our study suggested that the toxicity data derived from artificial soil cannot fully represent the real effects in natural soils, providing valuable insights for elucidating the realistic toxicity of MPs.
