Investigations have shown that storage bugs seriously harm grains during storage. In the interim, essential oils (EOs) have been proven to be a good botanical pesticide. The anti-Lasioderma serricorneproperties ofElsholtzia ciliataessential oil, which was obtained by steam distillation, were evaluated usingDL-limonene, carvone, and their two optical isomer components using contact, repelling, and fumigation techniques. Simultaneously, the fumigation, contact, and repellent activities of carvone and its two optical isomers mixed withDL-limonene againstL. serruricornewere evaluated. The results showed thatE. ciliata, its main components (R-carvone,DL-limonene), andS-carvone exhibited both fumigations (LC50= 14.47, 4.42, 20.9 and 3.78 mg/L) and contact (LD50= 7.31, 4.03, 28.62 and 5.63 µg/adult) activity againstL.serricorne.A binary mixture (1:1) ofR-carvone andDL-limonene displayed an obvious synergistic effect. A binary mixture (1:1) of carvone and its two optical isomers exhibited an obvious synergistic effect, too. Furthermore, the repellent activity of the EO, carvone, and its two optical isomers,DL-limonene, and a combination of them varied. To stop insect damage during storage,E. ciliataand its components can be utilized as bio-insecticides.

Investigations have indicated that storage pests pose a great threat to global food security by damaging food crops and other food products derived from plants. Essential oils are proven to have significant effects on a large number of stored grain insects. This study evaluated the contact toxicity and fumigant activity of the essential oil extract from the aerial parts ofElsholtzia ciliataand its two major biochemical components against adults and larvae of the food storage pest beetleTribolium castaneum. Gas chromatography–mass spectrometry analysis revealed 16 different components derived from the essential oil ofE. ciliata, which included carvone (31.63%), limonene (22.05%), andα-caryophyllene (15.47%). Contact toxicity assay showed that the essential oil extract exhibited a microgram-level of killing activity againstT. castaneumadults (lethal dose 50 (LD50) = 7.79 μg/adult) and larvae (LD50= 24.87 μg/larva). Fumigant toxicity assay showed LD50of 11.61 mg/L air for adults and 8.73 mg/L air for larvae. Carvone and limonene also exhibited various levels of bioactivity. A binary mixture (2:6) of carvone and limonene displayed obvious contact toxicity againstT. castaneumadults (LD50= 10.84 μg/adult) and larvae (LD50= 30.62 μg/larva). Furthermore, carvone and limonene exhibited synergistic fumigant activity againstT. castaneumlarvae at a 1:7 ratio. Altogether, our results suggest thatE. ciliataessential oil and its two monomers have a potential application value to eliminateT. castaneum. Keywords:Elsholtzia ciliata;Tribolium castaneum;essential oil;carvone;limonene;insecticidal activity;synergistic effect

The main objectives of the present study are to introduce a ecologically safe, and natural compound for controlling the red flour beetle , Tribolium castaneum . This study aims to thoroughly examine the effects of Elsholtzia ciliata essential oil ( E. ciliata EO) and its primary components, carvone , on Tribolium castaneum eggs, larvae, pupae, and adults. The study also includes a simulated warehouse test to assess the efficacy of these substances as contact agents, fumigants , repellents, and antifeedants. All of these anti-insect activities were positively correlated with the concentrations of E. ciliata EO and carvone. Firstly, in the contact activity assay, E. ciliata EO and carvone completely inhibited the hatching of eggs and pupae of the T. castaneum at a concentration of 2.5 μL/cm 2 . The LC 50 of E. ciliata EO and carvone were 7.62 and 4.33 μg/adult, respectively, against senior adults (5 weeks old). Secondly, in the fumigant activity assay, 60 mg/L of E. ciliata EO and carvone essentially completely inhibited pupae eclosion . On the other hand, the LD 50 of E. ciliata EO and carvone against senior adults (5 weeks old) were 8.72 and 5.02 mg/L, respectively. Thirdly, for T. castaneum adults, at the highest test concentration, the repellent rate of the E. ciliata EO was above 90%. Fourthly, the larvae and adults of the T. castaneum could reach complete antifeedant activity by E. ciliata EO and carvone at the highest test concentrations. Finally, 3000 ppm of carvone was sufficient to kill all the pests in the warehouse in the simulated warehouse test. In conclusion, the results of this study will contribute to the better control of the storage pest , T. castaneum , and the development of environmentally friendly insecticides based on E. ciliata EO and carvone.

Objective: The aims of the present study were to determine the efficacy of edible traditional Chinese medicines (ETCMs) in treating constipation, verify their laxative effects, and conduct preliminary investigations into their mechanisms of action. Methods: ICR mice were treated with loperamide to induce constipation, and various fecal parameters, including fecal volume, water content, and intestinal transport function, were measured in these constipation model mice to screen for ETCMs with laxative properties. The mechanism of action was preliminarily explored by examining changes in the intestinal mucosal structure, protein expression levels, and alterations in intestinal flora composition. Results: In ICR mice with loperamide-induced constipation, Elsholtzia ciliata aqueous extract (ECAE) and Hovenia dulcis aqueous extract (HDAE) significantly ameliorated constipation symptoms, mitigated colonic pathological tissue damage, significantly increased the expression levels of proteins associated with the promotion of intestinal peristalsis [Stem Cell Factor Receptor (c-Kit) and Stem Cell Factor (SCF)] and the maintenance of the intestinal barrier [Zonula Occludens-1 (ZO-l), Occludin and Claudin-l], and promoted beneficial intestinal bacterial colonization. Conclusion: ECAE and HDAE ameliorated constipation in mice, and their mechanism of action may be related to the increased abundance of intestinal bacteria such as Turicibacter, Olsenella, and Odoribacter, which contribute to higher butyrate production. This increase in butyric acid reduces inflammation, improves intestinal barrier function, and increases the abundance of beneficial intestinal bacteria.
