To explore the effect of nitric oxide (NO) on Enterocytozoon hepatopenaei (EHP) invading the shrimp, sodium nitroprusside was used as an exogenous NO donor to investigate the effects of different concentrations (0 μg/L (control group), 0.3 μg/L, 0.6 μg/L, and 0.9 μg/L) of sodium nitroprusside on the EHP copy number and the structure in the hepatopancreas of Exopalaemon carinicauda infected by EHP. The results showed that the EHP copy number in the sodium nitroprusside group was significantly lower than that in the control group, and the hepatopancreas in the sodium nitroprusside injection group had less structure damage than the control group. Additionally, superoxide dismutase (SOD) activity in the control group and 0.3 μg/L groups increased and then decreased, while alkaline phosphatase (AKP) activity decreased and then increased, with the same trend, and, the inducible nitric oxide synthase (iNOS) activity in the haemolymph of the 0.3 μg/L group was significantly higher than that of the control group at 3 days (). The NO content the 0.3 μg/L group was also higher from 1 to 4 days than that in the control group and significantly higher than that in the control group at 3 days (). The injection of NO could reduce the EHP copy number in infected E. carinicauda and slow down the pathological changes in the hepatopancreas structure, which might be related to the increase in NO content.

Hepatopancreatic microsporidiosis (HPM), caused by Enterocytozoon hepatopenaei (EHP), has emerged as a major threat to the shrimp aquaculture industry. However, no effective strategies have yet been developed to control the spread of this disease. In this study, we assessed the anti-EHP activity of ten monomeric compounds in Penaeus vannamei . The results showed that tetrahydropalmatine reduced the EHP copy number by 73.8 % at a concentration of 20 mg/L. In a therapeutic experiment, feeding EHP-infected shrimp a diet containing 0.1 % tetrahydropalmatine significantly lowered the EHP load. Additionally, the growth-related gene juvenile hormone esterase-like carboxylesterase 1 was significantly upregulated, while ecdysteroid-regulated protein and farnesoic acid O-methyltransferase were significantly downregulated, suggesting that tetrahydropalmatine may enhance the growth potential of EHP-infected shrimp. After 30 days of feeding, the EHP-infected shrimp's body weight in the groups fed with tetrahydropalmatine increased by 19.9 % compared to those fed with the normal diet. In a prevention experiment, healthy shrimp fed a diet containing tetrahydropalmatine and subsequently infected with EHP showed an increase in body weight by day 30 post-infection, compared to the control group, although this difference was not statistically significant. Tetrahydropalmatine can inhibit spore germination and electron dense disc formation, which may be an important reason for its suppression of EHP infection. Moreover, tetrahydropalmatine effectively inhibited the horizontal transmission of EHP, suggesting its potential to limit HPM outbreaks. These findings indicate that tetrahydropalmatine may serve as a feed additive for the treatment of HPM caused by EHP.

Effective detection of Enterocytozoon hepatopenaei (EHP) in aquaculture is currently hindered by the lack of field-deployable extraction methods capable of processing complex, inhibitor-rich hepatopancreatic tissue. This study presents a gravity-driven millifluidic platform for the rapid extraction of EHP genomic DNA using an optimized, surfactant-compatible magnetic solid-phase extraction (MSPE) chemistry. Utilizing 5% PEG 8000 and 2.0 M NaCl, the platform facilitates the selectively capture of DNA from inhibitor-rich crustacean lysates. The 3D-printed device employs a tilting rocking plate to generate passive, gravity-driven flow, maintaining homogeneous magnetic bead suspension and maximizing solid-phase capture efficiency without external pumps. The integrated platform achieved a DNA yield of 2804.33 ± 15.31 ng/μL, a 5.9-fold increase over manual magnetic bead extraction. TaqMan quantitative PCR (qPCR) validation targeting the EHP SSU rRNA gene was developed. Using a standard curve spanning 10 1 to 10 7 plasmid copies (Ct = −3.611 log 10 [copy] + 42.309, R 2 = 0.998, amplification efficiency 89.2%), the on-chip MSPE achieved a validated analytical limit of detection (LOD) of 1 spore per reaction (100% detection rate, n = 21), whereas a commercial CTAB-based DNA extraction kit failed to achieve a validated LOD even at 10 spores (85.7%, 18/21). Nested PCR targeting the SWP gene was employed for field evaluation. A pilot study across two cohorts (N = 40) demonstrated consistent detection of confirmed EPH-positive cases; however, the small sample size precludes definitive diagnostic accuracy claims. With a total processing time under 30 min, this platform provides a high-efficiency extraction module. Future work will couple the device with isothermal amplification (e.g., LAMP or RPA) to realize a sample-to-answer system for resource-limited aquaculture.

Enterocytozoon hepatopenaei (EHP) infection poses a significant challenge to shrimp aquaculture because the substantial economic losses it currently causes and the lack of treatment methods . In this study, ten individual compounds were evaluated for their effectiveness in whiteleg shrimp ( Penaeus vannamei ) against EHP. Among these compounds, paeonol exhibited the strongest inhibitory effect, reducing the EHP copy number by 82.2% at 10 mg/L. Both prophylactic and therapeutic administration of paeonol significantly reduced EHP copy number and enhanced shrimp weight gain. A combined prophylactic-therapeutic regimen resulted in superior outcomes compared with either strategy alone. Shrimp receiving a 0.1% paeonol-supplemented diet for 15 d before infection and continuing for 45 d post-infection showed a 28.7% increase in body weight compared with the untreated group. Paeonol inhibited EHP spore germination in a time-dependent manner and disrupted sporogony by decreasing nuclear number and electron-dense disc formation. Furthermore, paeonol upregulated immune-related genes in the JAK/STAT signaling pathway—including Domeless (DOME), JAK , STAT , and c-type lysozyme ( Lyz-c )—indicating an immunomodulatory role in its anti-EHP activity. Collectively, these findings demonstrate that paeonol targets two critical stages of the EHP life cycle—spore germination and developmental progression—and represents a promising candidate for integrated EHP prevention and control strategies in shrimp aquaculture.
