Effects of polysaccharide from hot-compressed steamed Rehmannia glutinosa on the immune system and gut microbiota in an immunosuppressed mice model

Yahui Jia,Zhiqiang Zhang,Siming Zhang,Xia Ma,Yuan Ruan,Bingji Ma,Li Wang

In this study, the immunomodulatory effects of polysaccharide obtained by hot-compressed steaming of Rehmannia glutinosa Libosch (HRP) were investigated using both in vitro and in vivo methods. It was found that HRP activated the TLR4/NF-κB signaling pathway, up-regulated the intracellular expression of TNF-α, IL-6 and IL-1β, and induced of innate immune memory in macrophages. We then investigated the effect of HRP on immunosuppressed mice induced by cyclophosphamide (CTX). Surprisingly, HRP improved CTX-induced weight loss and increased the splenic index, alleviated intestinal mucosal damage and hematopoietic insufficiency caused by CTX, as demonstrated by H&E staining. In addition, HRP promoted the expression of key proteins in the TLR4/NF-κB and autophagy pathways in intestinal tissues, thereby enhancing intestinal immune function. Bacterial 16S rRNA gene sequences of colon contents suggested that HRP may alleviate gut microbiota disruption by increasing the populations of Lachnospiraceae and Erysipelotrichaceae while inhibiting Lactobacillaceae . The results of this study show the potential use of HRP as an immunomodulator in functional foods or pharmaceuticals.

Naoxintong Capsule Inhibits the Development of Cardiovascular Pathological Changes in Bama Minipig Through Improving Gut Microbiota

Zhang Wei-jian,Su Wei-wei,Li Pei-bo,Rao Hong-yu,Lin Qing-wei,Zeng Xuan,Chen Tao-bin,Yan Zeng-hao,Liu Hong,Yao Hong-liang

Naoxintong capsule (NXT), a Chinese medicine, has performed excellent effects on the prevention and treatment against cardiovascular diseases. NXT is a fine powder mixture without any herb extraction, and there must be lots of ingredients hard to be absorbed. However, little is known about the correlation between the NXT’s cardioprotective effects and gut microbiota. Herein, we report the effect of NXT on the development of cardiovascular diseases and clarify the correlation between NXT’s cardioprotective effects and gut microbiota. In the current study, minipigs were selected and fed with high-fat diet and NXT daily for successive 8 months. During the process, up to 18 biomedical parameters were monthly determined to observe the dynamic changes after NXT treatment. At the end of experimental process, pathological examinations of heart, coronary artery, carotid artery, thoracic aorta, and abdominal aorta were conducted by HE staining and 16SrDNA sequencing, and analyzing of gut microbiota were conducted. Our results showed that NXT’s effects against cardiovascular diseases were through regulating blood lipid profiles, inhibiting vascular inflammation, enhancing antioxidant capacity, and alleviating myocardial injury, without damages on liver and kidney particularly. Concurrently, we also found that long-term administration of NXT increased the diversity of gut microbiota, influenced the microbiome structure and composition stably, and revered the increase of the ratio of the Firmicutes to Bacteroidetes (F/B ratio) in relative abundance. Specifically, our results revealed some key bacterium of Caproiciproducens (enhanced), Sutterella (enhanced), Erysipelotrichaceae (enhanced), and Romboutsia (decreased) that were closely involved in NXT’s effects. Taken together, our study demonstrates that NXT can inhibit the development of cardiovascular diseases by ameliorating high-fat diet–induced metabolic disorders and partly through improving gut microbiota.

Wallace melon juice fermented with Lactobacillus alleviates dextran sulfate sodium-induced ulcerative colitis in mice through modulating gut microbiota and the metabolism

Junwei Zhang,Yu Zhong,Danfeng Wang,Jiangxiong Zhu,Yun Deng,Yuncheng Li,Cong Liu,Ji-Li-Te Wang,Minyan Zhang

Fermented foods have shown promise in preventing or treating ulcerative colitis (UC) via regulating intestinal flora and correcting metabolic disorders. However, the prevention effect of fermented Wallace melon juice (FMJ) on UC is unclear. In this study, the effects of FMJ on dextran sodium sulfate (DSS)-induced UC were investigated via 16S rRNA sequencing and non-targeted metabolomics. The results showed that FMJ was effective in alleviating the symptoms of UC, reducing histological damage and oxidative stress, decreasing the levels of pro-inflammatory cytokines. After FMJ treatment, the level of propionic acid, butyric acid, and valeric acid increased by 14.1%, 44.4%, and 52.4% compared to DSS-induced UC mice. Meanwhile, the levels of harmful bacteria such as Oscillospira , Bacteroidetes , and Erysipelotrichaceae and Clostridium decreased, while the levels of beneficial bacteria such as Akkermansia , Lactobacillus , and Bifidobacterium increased. Fecal metabolomics analysis identified 31 differential metabolites, which could regulate metabolic disorders in UC mice by controlling the primary bile acid biosynthesis, purine metabolism, and pantothenate and CoA biosynthesis pathway. Additionally, the abundances of butyric acid, bile acids, and pantothenic acid were positively correlated with Allobaculum, Bifidobacterium , and other beneficial bacteria ( R 2 > 0.80, p < 0.01). The results indicated that FMJ played a role in regulating the structure of intestinal flora, which in turn helped in repairing metabolic disorders and alleviated colitis inflammation.
