Graphical Two imidazole-based color-changing fluorescent probes 4-(4,5-bis(4-chlorophenyl)-1 H -imidazol-2-yl)- N , N -diphenylaniline (CIN) and 4-(4,5-di(furan-2-yl)-1 H -imidazol-2-yl)- N , N -diphenylaniline (FIN) were synthesized and characterized. The new probes CIN and FIN exhibit excellent selectivity and sensitivity towards Fe 3+ with relatively low detection limits and strong anti-interference properties. Both the probes CIN and FIN complexed 1 : 1 with Fe 3+ ion. Two imidazole-based color-changing fluorescent probes 4-(4,5-bis(4-chlorophenyl)-1 H -imidazol-2-yl)- N , N -diphenylaniline (CIN) and 4-(4,5-di(furan-2-yl)-1 H -imidazol-2-yl)- N , N -diphenylaniline (FIN) were synthesized and characterized. The new probes CIN and FIN exhibit excellent selectivity and sensitivity towards Fe 3+ with relatively low detection limits (0.26 μM and 0.10 μM) and strong anti-interference properties. Both the probes CIN and FIN complex 1 : 1 with Fe 3+ ion, and the complexation constants were 1.165×10 5 M −1 (CIN) and 1.899×10 5 M −1 (FIN), respectively. Moreover, CIN and FIN could be recycled by EDTA and could effectively monitor Fe 3+ ions at pH 4–10, which ensures the sensor‘s potential for detecting Fe 3+ in actual aqueous solutions. The probes were further applied to the detection of Fe 3+ in the actual reaction of Fe 2+ oxidation, and the filter paper and the probe solution are combined to make an indicator to detect Fe 3+ more conveniently and quickly.

Hygroscopic film integrated with the fin structure: a long-term cooling solution for fin heat exchanger

Chengzhi Hu,Yanhui Jiang,Haiyuan Zhang,Yong Cai,Yubai Li,Dawei Tang

Integrating the hygroscopic heat dissipation layer with the fin represents an innovative approach to enhance the heat transfer coefficient, while simultaneously reducing the overall volume, weight, and material requirements for the fin heat exchanger. However, designing a hygroscopic layer that seamlessly integrates with the fin structure, has an excellent hygroscopic heat dissipation effect, and long-term enhanced heat dissipation after the liquid has dissipated poses a significant challenge. Here, we report a flexible, self-adhesive, adjustable thickness, high heat transfer coefficient desorption radiation film (DRF) for long-term heat dissipation enhancement through integrating desorption cooling and radiation-enhanced cooling, along with controlling the thermal resistance for conduction. Using a hydrophilic sodium alginate-CaCl2 crosslinked adsorption medium and polyvinyl alcohol to enhance radiation and adhesion properties, a porous hygroscopic film is fabricated with fast moisture adsorption, no leakage, high emissivity, controllable thermal resistance, and strong adhesion. The flexible film can be directly attached to the fin surface, maintaining the original advantages of the fin structure with minimal modification, making it excellent in adhering effectively. Compared to an aluminum plate, the system reduces the temperature rise rate by 21.3 times and lowers the steady-state temperature by 9℃ even staying lower than the graphene-coated surface over time. After three cycles (1 hour for desorption and 1 hour for adsorption in each cycle), the cooling efficiency only decreased by 4%, showing minimal impact from the short adsorption time. After applying to the real fins and optimizing the corresponding structure, the cooling down of the same volume reaches 8.5℃, and the heat transfer coefficient is increased by 18%. In photovoltaic panel tests, the system reduced temperature by 3.5℃. Our strategy offers a novel, simple, cost-effective solution for long-term enhanced heat dissipation of fin heat exchangers, while also providing insights into desorption cooling for reducing heat transfer resistance.

Fin-like CdSeS Nanoplatelets for Pesticide Sensing

Fan Gao,Lanlan Yang,Jianguo Tang,Zhonglin Du,Yao Wang,Zhongyi Hu,Dongni Han,Linjun Huang,Laurence A. Belfiore

Pesticide residues in farm products create human health hazards. Hence, sensors that detect their existence are advantageous to address this critical problem. In this research contribution, fin-like CdSeS nanoplatelets were synthesized from solution by adjusting the selenium-to-sulfur molar ratio. Structural and photophysical properties were characterized via XPS, XRD, UV absorption, and fluorescence emission. CdSeS nanoplatelets exhibit a regular fin-like structure when the molar ratio of selenium-to-sulfur is 1:1. Fluorescence emission from these nanoplatelets occurs at two distinct wavelengths, characteristic of quantum dots. CdSeS fin-like nanoplatelets generate red-shifted fluorescence to 530 nm, which is useful to detect the popular organophosphorus pesticide dimethoate effectively at concentrations that range from 10–5 to 10–7 mol/L. Upon increasing dimethoate concentrations in solution, fluorescence intensity of CdSeS nanoplatelets decreases. Hence, direct sensing of dimethoate is feasible. Another important significance of this work is the use of low-toxicity and recyclable cadmium acetate instead of cancer-causing and nonrecyclable cadmium oxide, which will be helpful to reduce environmental pollution.

The Fin-Improving Effects of Fucosylated Chondroitin Sulfate from Green and Purple Apostichopus japonicus on Caudal Fin Regeneration of Zebrafish Larvae

Tingting Hou,Zaizhao Wang,Kui Tang,Shuai Zhang,Shilin Liu,Jialiang Liu,Xiaoteng Fan,Xiaolin Liu

This study investigated the capability for fin regeneration in zebrafish larvae with the mechanism of FCS isolated from green and purple sea cucumbers Apostichopus japonicus (G-FCS and P-FCS). HPGPC determined that mean molecular weight of purified G-FCS and P-FCS is 19.5 and 18.8 kDa, and the FT-IR spectrum displayed similar characteristic absorption peaks. AFM images presented that G-FCS was shown as a spherical polysaccharide, while P-FCS molecules exhibited elongated chains. For the regeneration examination, FCS increased the regrowth area of the larval fin at 48 hours postamputation with enhanced locomotor behaviors, and the transcription levels of regeneration-related genes (Wnt10a, msx1b, fgf20a, bmp2a, bmp4, and igf2b) were upregulated. Moreover, FCS promoted the mRNA expression of inflammatory factors (iNOS, TNF-α, IL-6, IL-10, and IFN-γ) and TLR/NF-κB pathway-related genes (TLR3, TLR4a, NF-κb, and MyD88). The Western blot analyses of TNF-α and IL-1β supported that FCS enhanced immune response. ELISA results indicated that FCSs increased the contents of NO, CAT, SOD, and G6PD. Summarily, the present results suggested that FCS was a potential regeneration enhancer by elevating immunomodulatory and antioxidative processes in zebrafish.

6PPD impairs immune responses and fin regeneration in zebrafish

Xiaoyu Mao,Dashuang Mo,Yuqin Cheng,Mengzhu Lv

N -(1,3-Dimethylbutyl)- N ′-phenyl- p -phenylenediamine (6PPD), a commonly used antioxidant in tire manufacturing, has been widely detected in the environment and shown to exhibit acute toxicity in several organs. However, the effects of 6PPD on immune responses, particularly following injury, remain poorly understood. In this study, we investigated the impact of 6PPD exposure on immune responses using zebrafish as a model. 6PPD exposure disrupted caudal fin regeneration at various stages of the regenerative process. Further analysis revealed that 6PPD impaired immune responses following fin amputation, as evidenced by the reduced number of lyz + /mpx + neutrophils and the downregulation of key immune-related genes. Besides, the morphology of neutrophils was changed upon 6PPD exposure, indicating the defective migration of immune cells. The incubation of zebrafish larvae with lipopolysaccharide (LPS), which induces global immune responses, also exhibited impaired immune function when combined with 6PPD exposure. Additionally, the injection of LPS into the egg yolk or trunk exacerbated immune responses at the injury site, yet 6PPD exposure significantly reduced neutrophil accumulation and downregulated the expression of immune-related genes, confirming the toxicity of 6PPD in immune responses. These findings provide new insights into the toxic effects of 6PPD on immune responses during injury, highlighting its potential to impair immune function in animals and human.

A comprehensive painkillers screening by assessing zebrafish behaviors after caudal fin amputation

Gilbert Audira,Jong-Chin Huang,Kelvin H.-C. Chen,Kevin Adi Kurnia,Ross D. Vasquez,Marri Jmelou M. Roldan,Yu-Heng Lai,Chung-Der Hsiao,Cheng-Yo Yen

Recently, the usage of zebrafish for pain studies has increased in the past years, especially due to its robust pain-stimulated behaviors. Fin amputation has been demonstrated to induce a noxious response in zebrafish. However, based on the prior study, although lidocaine, the most used painkiller in zebrafish, has been shown to ameliorate amputated zebrafish behaviors, it still causes some prolonged effects. Therefore, alternative painkillers are always needed to improve the treatment quality of fin-amputated zebrafish. Here, the effects of several analgesics in recovering zebrafish behaviors post-fin amputation were evaluated. From the results, five painkillers were found to have potentially beneficial effects on amputated fish behaviors. Overall, these results aligned with their binding energy level to target proteins of COX-1 and COX-2. Later, based on their sub-chronic effects on zebrafish survivability, indomethacin, and diclofenac were further studied. This combination showed a prominent effect in recovering zebrafish behaviors when administered orally or through waterborne exposure, even with lower concentrations. Next, based on the ELISA in zebrafish brain tissue, although some changes were found in the treated group, no statistical differences were observed in most of the tested biomarkers. However, since heatmap clustering showed a similar pattern between biochemical and behavior endpoints, the minor changes in each biomarker may be sufficient in changing the fish behaviors.

Radix Rehmanniae Praeparata promoted zebrafish fin regeneration through aryl hydrocarbon receptor-dependent autophagy

Fengyan Chen,Shiming Pu,Li Tian,Huan Zhang,Huixian Zhou,Yijing Yan,Xiaolin Hu,Qiong Wu,Xueping Chen,Shuk Han Cheng,Shisan Xu

Headings ethnopharmacological relevance Rehmanniae Radix Praeparata (RRP), a staple in traditional Chinese medicine, is derived from Rehmannia glutinosa Libosch and is renowned for its wound-healing properties. Despite its clinical prevalence, the molecular mechanisms underlying RRP's wound-healing effects have not been fully elucidated. Aim of the study This research endeavored to delineate the molecular and cellular mechanisms underlying the beneficial effects of RRP on wound healing, utilizing a zebrafish model. Materials and methods Zebrafish larvae at 3 days post-fertilization were amputated at the fin and subsequently treated with RRP. The pro-wound healing and regenerative effects of RRP were evaluated through morphological analysis, assessment of cell proliferation and apoptosis, Additionally, mechanistic insights were gained through a comprehensive approach encompassing network pharmacology analysis, cell tracing, RNA-sequencing, CRISPR/Cas9 gene editing, and pharmacological inhibition. Results Our findings demonstrate that RRP significantly accelerates caudal fin regeneration in zebrafish following injury by suppressing cell apoptosis, promoting cell proliferation, and upregulating the expression of regenerative-related genes. Furthermore, RRP triggers autophagy signals during the regenerative process, which is attenuated by the autophagy inhibitor chloroquine (CQ). Notably, the administration of RRP enhances the expression of ahr1 and ahr2 in the regenerating fin. Genetic knockout of ahr1a , ahr1b , or ahr2 using CRISPR/Cas9, or pharmacological blockade of AHR signals with the antagonist CH-223191, diminishes the regenerative potential of RRP. Remarkably, zebrafish lacking ahr2 completely lose their fin regeneration ability. Additionally, inhibition of AHR signaling suppresses autophagy signaling during fin regeneration. Conclusions This study uncovers that RRP stimulates fin regeneration in zebrafish by inducing AHR signals and, at least partially, activating the autophagy process. These findings provide novel insights into the molecular mechanisms underlying the wound-healing effects of RRP and may pave the way for the development of novel therapeutic strategies.

Fin Cells as a Promising Seed Cell Source for Sustainable Fish Meat Cultivation

Zongyun Du,Jihui Lao,Yuyan Jiang,Jingyu Liu,Shili Liu,Jianbo Zheng,Fei Li,Yongyi Jia,Zhimin Gu,Jun Chen,Xiao Huang

Cell-cultured meat production relies on stable, proliferative seed cells, commonly sourced from muscle satellite cells (MuSCs) and adipose-derived mesenchymal stem cells (AD-MSCs). However, establishing such cell lines in fish species remains technically challenging. While pluripotent stem cells (e.g., ESCs/MSCs) offer alternatives, their differentiation efficiency and predictability are limited. Here, we developed TCCF2022, a novel caudal fin-derived cell line from Topmouth culter (Culter alburnus), which expresses pluripotency markers (AP,Oct4,Sox2,Klf4, andNanog) and aggregated growth to form 3D spheroids. Forskolin supplementation enhanced pluripotency maintenance. The presence of adipogenic and myogenic lineage cells within the 3D spheroids was confirmed, demonstrating their potential as seed cells for cell-cultured meat. Using a small-molecule cocktail 5LRCF (5-Azacytidine, LY411575, RepSox, CHIR99021, and Forskolin), we successfully differentiated TCCF2022 cells into functional myotubes. Additionally, we established a method to induce the differentiation of TCCF2022 cells into adipocytes simultaneously. Thus, the TCCF2022 cell line can be used to improve muscle fiber formation and lipid composition, potentially enhancing the nutritional profile and flavor of cultured fish meat.

Ethyl butyrate inhibits caudal fin regeneration in adult zebrafish by disrupting extracellular matrix remodeling

Sijie Zhang,Hao Wang,Yunlong Meng,Mijia Li,Yang Li,Xinhao Ye,Shiyi Duan,Shimei Xiao,Huiqiang Lu,Keyuan Zhong

Wound healing and tissue regeneration are influenced by a variety of factors. Adverse lifestyle habits, such as excessive alcohol consumption, delay wound healing and increase the risk of secondary infections. Ethyl butyrate is a common food additive widely used to enhance the aroma of alcoholic beverages. This additive is generally considered harmless to human health in both industrial and domestic settings. However, the ecotoxicity and its effects on wound healing have not been elucidated. In this study, we used zebrafish as the experimental animal, and the caudal fins were amputated to explore the effects of ethyl butyrate on wound healing and tissue regeneration. The effect of ethyl butyrate on blastema and bone regeneration and its impact on the transcriptional levels of regeneration-related genes and inflammation-related genes were evaluated. RNA-seq was conducted to determine the differentially expressed genes (DEGs) between the treatment and the control groups. KEGG and GO analysis was conducted to explore the functions of DEGs. Significantly enriched GO terms and KEGG pathways were identified to explore the molecular mechanism underlying the inhibition of zebrafish caudal fin regeneration by ethyl butyrate. The results demonstrated that ethyl butyrate significantly inhibited the regeneration of zebrafish caudal fins, including blastema and bone regeneration. Ethyl butyrate exposure significantly downregulated the expression of genes associated with bone and blastema regeneration and inflammation response. KEGG and GO functional analyses revealed that the DEGs were associated with significant enrichment of extracellular matrix-receptor interactions. Ethyl butyrate treatment downregulated the expression of most extracellular matrix-related genes. These findings indicate that ethyl butyrate potentially modulates pathways associated with the structure, adhesion, modification, and degradation of the extracellular matrix, thereby disrupting extracellular matrix remodeling, inhibiting wound inflammation, impairing blastema and bone regeneration and ultimately hindering caudal fin regeneration. In summary, the findings demonstrate that ethyl butyrate disrupts extracellular matrix remodeling and inhibits the regeneration of zebrafish caudal fins. These results provide valuable insights into the rational use of ethyl butyrate and further investigation of wound healing mechanisms.

Toxicity of silver nanoparticles on wound healing: A case study of zebrafish fin regeneration model

Shaochen Pang,Yue Gao,Fengbang Wang,Yuanyuan Wang,Mengxi Cao,Wenjuan Zhang,Yong Liang,Maoyong Song,Guibin Jiang

Dressings coated with silver nanoparticle (AgNP) are widely used in the management of acute and chronic wounds. However, whether AgNP exerts toxicity on wound healing remains ambiguous. To demonstrate the effects of AgNP on wound healing, we precisely quantified the recovery speed of wound by taking advantage of the fin regeneration of zebrafish. This method also enabled assessment of the adverse effect of AgNP on various steps of wound healing in vivo . We revealed that AgNP treatment at the concentration of 2 μg/ml impaired fin regeneration when exposure was performed at the phases of epithelialization and the beginning of blastema formation. Cell proliferation of regenerative blastema was significantly decreased after AgNP exposure. But the canonical signals including Wingless/Integrated (Wnt), Notch and Fibroblast growth factor (Fgf) which play important roles in cell proliferation during fin regeneration were not modulated at 36 hours post amputation (hpa). Further study showed that AgNP impaired fin regeneration through declining amputation-induced ROS as early as epithelialized phase at 18 hpa, rather than inducing ROS generation. AgNP exposure also promoted recruitment of neutrophils in the early phase of wound healing, which suggests that this event dampened amputation-induced ROS. Overall, this study suggested that application of AgNP-coated dressings should be carefully considered at the beginning stage of wound healing.

Establishment and Application of Kidney Cell Line and Caudal Fin Cell Line of Pseudocaranx dentex

Shun Zhou,Dong Zheng,Jiaxue Song,Lirong Jiang,Jinlong Dong,Yu Zhang,Jinjing Cao,Yunji Xiu

Pseudocaranx dentex , a precious marine economic fish, is widely distributed in tropical and subtropical seas. However, frequent disease outbreaks have hindered the sustainable development of its aquaculture. In this investigation, the kidney cell line (named PDK) and caudal fin cell line (named PDF) of P. dentex were established and characterized. According to our findings, both cell lines thrived best in L-15 media with 20% fetal bovine serum (FBS) at 26°C. Both cell lines have been stably passaged for more than 40 generations and maintained high viability after cryopreservation in liquid nitrogen. Their origin from P. dentex was confirmed by the COI gene sequencing data. Meanwhile, chromosome analysis demonstrated that PDK and PDF cells shared a modal chromosome number of 48. In addition, both cell lines could be transfected with pEGFP-N3 plasmid via non-liposomal method, indicating potential for in vitro exogenous gene function studies. Furthermore, bacterial susceptibility testing revealed that both cell lines exhibited high sensitivity to Vibrio anguillarum and Vibrio vulnificus . However, PDK cells demonstrated reduced susceptibility to Edwardsiella tarda , while PDF cells showed diminished sensitivity to Vibrio alginolyticus . Virus susceptibility results indicated that PDK cells exhibit greater sensitivity to Micropterus salmoides rhabdovirus (MSRV) than to Red sea bream iridovirus (RSIV), while PDF cells are only susceptible to MSRV but show no response to RSIV. In summary, the PDK and PDF cell lines can serve as key tools for studies on the genetics, cell biology, and immune defence mechanisms of P. dentex , and hold significant value for fish genetics, disease research, and aquaculture disease prevention and control.

Isolation and identification of antioxidative peptides from peptic hydrolysates of half-fin anchovy (Setipinna taty)

Ru Song,Rong-bian Wei,Guang-qiang Ruan,Hong-yu Luo

The objective of this study was to identify antioxidative peptides from peptic hydrolysates of half-fin anchovy (HAHp). Furthermore, the possible synergistic effects of identified peptides were evaluated. HAHp were separated using CM FF ion exchange and Sephadex G 25 chromatography. HAHp1-2, one peptide fraction with the highest 1, 1-diphenyl-2-pycrylhydrazyl (DPPH) scavenging activity, was subjected to RP-HPLC coupled with ESI under positive ionization mode. Six peptides, AGDDARPA (771.35 Da), ESAGLHE (741.33 Da), NKVKGELD (901.45 Da), TLWSGAGQTVNT (1233.63 Da), EMSAGLHE (872.37 Da) and WRKKDPLND (1171.60 Da), were identified by ESI-MS/MS analysis. Peptides AGDDARPA (HAHp1-2Ⅰ), ESAGLHE (HAHp1-2Ⅱ), NKVKGELD (HAHp1-2Ⅲ), EMSAGLHE (HAHp1-2Ⅳ) and WRKKDPLND (HAHp1-2Ⅴ) were synthesized to determine in vitro antioxidative activity. Results showed that HAHp1-2Ⅳ and HAHp1-2Ⅴ displayed higher DPPH radical scavenging activities than other three peptides and l -carnosine. Peptide HAHp1-2Ⅲ demonstrated similar reducing power ability as l -carnosine. All synthetic peptides had moderate or negligible metal chelating activities. Additionally, the enhanced DPPH radical scavenging activities or reducing power abilities were observed for the mixture of HAHp1-2Ⅲ, HAHp1-2Ⅳ or HAHp1-2Ⅴ, indicating the possible synergistic effects. These results suggested that the antioxidative peptides derived from HAHp could be used as potential antioxidants in functional foods and nutraceuticals.
