Novel direct and indirect competitive fluorescence-linked immunosorbent assays ( c FLISA and ic FLISA) for detection of ochratoxin A (OTA) were described using CdTe quantum dots (QDs) as fluorescent label. CdTe QDs were successfully synthesized, which had an emission wavelength of 615 nm. The high purity monoclonal antibody against OTA was prepared through cell thawing and the octylic acid-ammonium sulfate method. The OTA MAbs were successfully coupled with CdTe QDs, and which also retained the original biological activity. The 50% inhibition values (IC 50 ) of the c FLISA and ic FLISA were 0.630 ng/mL, 0.234 ng/mL, the limits of detection (LOD) were 7.06 × 10 –3 and 4.15 × 10 –3 ng/mL, and detection ranges were 7.06 × 10 –3 − 18.34 ng/mL and 4.15 × 10 –3 − 4.88 ng/mL, in-order. The recoveries were 96.0–104.7% along with coefficients of variation (CVs) below 10%. The FLISA provided novel method for determination of OTA and the potential of MAb–CdTe QDs for the establishment of fluorescent immunochromatographic test strip.

Florfenicol (F), an antimicrobial agent exclusive to veterinary use within the chloramphenicol class, is extensively applied as a broad-spectrum remedy for animal diseases. Despite its efficacy, concerns arise over potential deleterious residues in animal-derived edibles, posing threats to human health. This study pioneers an innovative approach, introducing a quantum dot fluorescence-based immunoassay (FLISA) for the meticulous detection of F residues in animal-derived foods and feeds. This method demonstrates heightened sensitivity, with a detection limit of 0.3 ng/mL and a quantitative detection range of 0.6–30.4 ng/mL. Method validation, applied to diverse food sources, yields recoveries from 90.4 % to 109.7 %, featuring RSDs within 1.3 % to 8.7 %, the results showed high consistency with the national standard HPLC-MS/MS detection method. These findings underscore the method's accuracy and precision, positioning it as a promising tool for swift and reliable F residue detection, with substantial implications for fortifying food safety monitoring.

Mycotoxins are secondary products produced primarily by fungi and are pathogens of animals and cereals, not only affecting agriculture and the food industry but also causing great economic losses. The development of rapid and sensitive methods for the detection of mycotoxins in food is of great significance for livelihood issues. This study employed an amino-functionalized zirconium luminescent metal–organic framework (LOF) (i.e., UiO-66-NH2). Click chemistry was utilized to assemble UiO-66-NH2in a controlled manner, generating LOF assemblies to serve as probes for fluorescence-linked immunoassays. The proposed fluoroimmunoassay method for Zearalenone (ZEN) and Fumonisin B1 (FB1) detection based on the UiO-66-NH2assembled probe (CLICK-FLISA) afforded a linear response range of 1–20 μmol/L for ZEN, 20 μmol/L for FB1, and a very low detection limit (0.048–0.065 μmol/L for ZEN; 0.048–0.065 μmol/L for FB1). These satisfying results demonstrate promising applications for on-site quick testing in practical sample analysis. Moreover, the amino functionalization may also serve as a modification strategy to design luminescent sensors for other food contaminants.

Background Colorectal cancer (CRC) requires accurate diagnostic approaches for effective clinical management. Serum biomarkers PD-L1, SORL1, and GDF-15 offer valuable diagnostic information, but their simultaneous detection is hindered by the limited sensitivity and multiplexing capacity of conventional immunoassays. This work addresses the critical need for a highly sensitive multiplexed platform for precise CRC diagnosis. Results We constructed a multicolor QDs-K@FLISA platform employing K24 polylysine-conjugated CdTe quantum dots with distinct emissions (543/587/647 nm). The K-peptide enabled multi-QD loading, significantly amplifying fluorescence signals and facilitating ultrasensitive triplex detection. The assay achieved exceptional sensitivity with detection limits of 0.996 pg/mL (PD-L1), 1.101 pg/mL (SORL1), and 1.035 pg/mL (GDF-15), along with wide linear ranges (7.8-500 pg/mL for PD-L1/SORL1; 6-1600 pg/mL for GDF-15). High reproducibility was demonstrated (RSD < 2.3%). Clinical validation using 120 serum samples revealed strong correlation with reference methods (R 2 > 0.99) and effective discrimination between CRC patients and healthy controls. Significance and Novelty This work establishes a novel peptide-mediated multi-QD loading strategy that enables ultrasensitive multiplexed detection of CRC biomarkers. The platform presents significant potential for clinical applications in CRC diagnosis and biomarker profiling.
