FMT from Exercise and Konjac Glucomannan Preconditioned Donors Rescues Antibiotic-Induced Dysbiosis with Enhanced Ecological Restoration in Mice

Minghan Wang,Yanan Lyu,Jiangyu Zhang,Yanyan Wang,Yan Yang,Yu-Heng Mao

Background: Although antibiotics have a wide range of applications in medical clinical practice and possess significant clinical value, their inevitable contribution to gut microbiome dysbiosis warrants attention. Our previous research has confirmed that the combined intervention of exercise and konjac glucomannan (KGM) has a better regulatory effect on gut dysbiosis in mice compared with individual interventions. Methods: This study aims to further investigate whether this effect can be transmitted through fecal microbiota transplantation (FMT), and to compare the recovery effects of autologous FMT (a-FMT), fecal microbiota transplantation after exercise combined with KGM intervention (EK-FMT), and combinative intervention with exercise and KGM (EXE-KGM) on gut microbiome dysbiosis. Sample sizes ranged from five to six animals. Results: The results showed that the a-FMT group recovered α diversity the fastest, including Chao, Shannon, and Simpson indices( p < 0.05), within 2 weeks after transplantation when compared with the CTL group. At the end of the experiment, the Bray–Curtis distance of the a-FMT group was closest to the CTL group, while the EXE-KGM group had delayed recovery, there was no significant difference between the EK-FMT group and the EXE-KGM group. Metagenomic analysis and metabolomics analysis indicated that the arginine synthesis and metabolism pathways (KEGG: map00471, map00473, arginine biosynthesis) played a core role in the restoration of the microbiota. Conclusions: The results of this experiment indicate that EK-FMT group can partially transfer the regulatory effects of combined exercise and KGM intervention, a-FMT accelerates the recovery speed of the gut microbiome and arginine metabolism may play an important role in it. This finding provides a theoretical basis and practical direction for special populations to receive special donor fecal treatment.

Effects of chitosan oligosaccharides (COS) and FMT from COS-dosed mice on intestinal barrier function and cell apoptosis

Kai Na,Jiani Wei,Li Zhang,Ying Fang,Xiangyu Li,Shuang Lu,Xiaohua Guo

Chitosan oligosaccharides (COS) show the potential to support the intestinal health, but the mechanism and role of COS-derived intestinal microbiota are unknown. We explored the protective effect of direct administration of COS on intestinal barrier function using an in vivo colitis mouse model and an in vitro enterotoxigenic Escherichia coli (ETEC)-challenged IPEC-J2 cell model. COS directly enhanced the intestinal barrier function. COS intervention also promoted the abundance and diversity of intestinal flora. Importantly, FMT intervention with a COS-derived microbiome decreased the disease index level and alleviated histopathological changes, and improved gut barrier function in the colitis model. Both COS and COS-derived microbiota suppressed ETEC-induced cellular apoptosis in IPEC-J2 cells. This study firstly confirms transplantation of COS-modified fecal microbiota can enhance the intestinal barrier function. The mechanism underlying COS benefits is due to a direct intervention by COS supplementation and an indirect improvement of the gut microbiota induced by COS exposure.

Gut microbiota-derived metabolites regulate fat deposition via N6-methyladenosine (m6A) RNA methylation in germ-free and FMT piglets1

Zimeng Xin,Shun Chen,Ruiti Ren,Jiaqi Liu,Shizhe Xiang,Yuchu Han,Liangpeng Ge,Yongming Zhou,Jing Sun,Xinxia Wang

The gut microbiota plays a critical role in regulating host fat deposition, yet the underlying mechanisms remain poorly understood. To investigate this question, this study employed germ-free (GF) and fecal microbiota transplantation (FMT) piglet models to systematically elucidate how microbiota-derived metabolites modulate fat deposition via N 6 -methyladenosine (m 6 A) RNA methylation. Although the FMT group showed no significant change in body weight compared with the GF group, it exhibited markedly increased subcutaneous and intramuscular fat deposition, as evidenced by larger adipocyte size ( P <0.001), elevated serum and longissimus dorsi muscle triglyceride levels ( P <0.05), and upregulated expression of adipogenic genes ( P < 0.05). Non-targeted metabolomics revealed elevated levels of bile acids, including hyodeoxycholic acid (HDCA), 12-ketodeoxycholic acid (12KDCA), and 3b-hydroxy-5-cholenoic acid (3b-h5-CA), as well as tryptophan metabolites indoxyl sulfate (IS) and L-kynurenine (L-Kyn) in the FMT group, and these metabolites were significantly correlated with lipid metabolism parameters. Mechanistic studies showed that these metabolites reduced m 6 A modification levels in mouse stromal vascular fraction (SVF) cells and fibro-adipogenic progenitor (FAPs) cells by upregulating fat mass and obesity-associated protein (FTO) and downregulating methyltransferase-like 3 (METTL3) ( P < 0.05), consistent with the in vivo observations in piglets. Furthermore, Bacteroides , Lactobacillus , Parabacteroides , and Akkermansia were identified as key genera involved in bile acid and tryptophan metabolism. Together, these findings reveal a gut microbiota–metabolite– m 6 A regulatory axis in fat deposition in pigs, providing new insights into host–microbiota interactions and offering potential strategies for improving metabolic health and meat quality in livestock.

An exercise-associated gut microbiota signature enhances endurance performance: A study combining a human cohort and a mice FMT model

Xiongbao Zhang,Yating Fu,Jiahui Chen,Hao Shen,Yumeng Du,Jun Wu,Hongjing Lai,Yu Liu,Jingbo Chen,Xiaomei Hou,Wenjun Chang,Xuefei Hu

Background The gut microbiota is closely related to exercise, but the interrelationship between the two remains elusive. In this study, we aimed to explore differences in the gut microbiota between young adults with exercise and sedentary lifestyles. In addition, we evaluated the effects of gut microbiota from these different lifestyle populations on endurance exercise capacity. Methods The exercise status and nutritional characteristics of young adults were evaluated by PARS-3 and food frequency questionnaires. The gut microbiota of young adults from exercise and sedentary lifestyle groups was analyzed by 16S rRNA analysis. Subsequently, we performed fecal bacteria transplantation (FMT) from the human donors into mice and evaluated the effects on their endurance exercise capacity. Results The exercise group exhibited significantly higher gut microbiota diversity compared to the sedentary group. Several beneficial bacteria, including Veillonella, Faecalibacterium, and Bacteroides, were enriched in the exercise group. The FMT experiment confirmed that mice receiving microbiota from the exercise group showed significantly improved endurance exercise capacity. Conclusion Young people who exercise regularly possess a more diverse gut microbiota enriched with beneficial bacteria. This exercise-associated microbiota has the potential to directly improve exercise capacity.

Pivotal role of the gut microbiota in congenital insensitivity to pain with anhidrosis

Zhang Mi,Hong Yishun,Wu Wenyao,Li Ningbo,Liu Baowen,Sun Jiaoli,Cao Xueqin,Ye Ting,Zhou Ling,Liu Cunming,Yang Chun,Zhang Xianwei

Background Increasing evidence has shown that the occurrence and development of various human diseases are closely related to the gut microbiota. We compared the gut microbial communities of human subjects with congenital insensitivity to pain with anhidrosis (CIPA) and healthy controls (HCs) to assess whether fecal microbiota transplantation (FMT) into germ-free mice and mice in acute pain influenced the behaviors of the host. Methods We utilized 16 s rRNA analysis to compare the gut microbial communities of CIPA subjects and HCs and assessed whether FMT into germ-free mice and mice in acute pain influenced the behaviors of the host. Results In a 16 s RNA analysis, the CIPA group had significant decreases in the relative abundance of 11 bacteria, whereas 7 bacteria were significantly increased. In further animal experiments, the transplantation of fecal samples from CIPA patients to healthy mice significantly increased their scores on both the mechanical withdrawal test and the tail flick test; in an acute plantar incision model, scores were also significantly increased on the mechanical withdrawal test at 4 and 5 days after the operation. Moreover, pseudo-germ-free mice receiving fecal bacteria from patients with CIPA took significantly longer to escape and had a significantly longer path length on training days 1, 2, and 5 and also had fewer platform crossings and spent less time in the target quadrant in the probe trial. Conclusions Our results suggest that the gut microbiota in CIPA subjects plays a key role in behaviors. Therapeutic strategies for improving the gut microbiota might alleviate CIPA symptoms.

Design of novel dual-emitting ratiometric fluorescence system based on FMT-CDs and MnO2 NS for sensitive isoniazid detection in urine sample

Xin-Ran Guo,Yan-Xin Guo,Juan Chen,Xin-Yue Chen

In this work, a simple and sensitive ratiometric fluorescence probe to detect isoniazid (INH) was developed on the basis of carbon dots and MnO 2 nanosheets (MnO 2 NS). Nitrogen-doped carbon dots (FMT-CDs) were synthesized by microwave method using formononetin (FMT) in ammonia water. The oxidase-like activity of MnO 2 NS was utilized to oxidize non-fluorescent o -phenylenediamine (OPD) to 2, 3-diaminophenazine (DAP) with orange fluorescence. It was demonstrated that DAP could effectively quench the fluorescence of FMT-CDs through the inner filtering effect (IFE). Due to the reducing nature of INH, MnO 2 NS was decomposed and thus lost its oxidase-like activity after the addition of INH. Therefore, the production of DAP was reduced, and the fluorescence intensity was decreased, while IFE on FMT-CDs was weakened and the fluorescence of FMT-CDs was recovered. Based on the above principle, a sensitive method for the detection of INH was established based on the fluorescence intensity ratio of the two fluorescent substances, FMT-CDs and DAP, and was successfully applied to the detection of INH in human urine samples. The limit of detection (LOD) could reach to 0.16 μM in human urine samples.

Faecal microbiota transplantation-mediated jejunal microbiota changes halt high-fat diet-induced obesity in mice via retarding intestinal fat absorption

Luoyi Zhu,Jie Fu,Xiao Xiao,Fengqin Wang,Mingliang Jin,Weihuan Fang,Yizhen Wang,Xin Zong

Summary Faecal Microbiota Transplantation (FMT) is considered as a promising technology to fight against obesity. Wild boar has leanermuscle and less fat in comparison to the domestic pig, which were thought to be related with microbiota. To investigate the function and mechanism of the wild boar microbiota on obesity, we first analysed the wild boar microbiota composition via 16S rDNA sequencing, which showed that Firmicutes and Proteobacteria were the dominant bacteria. Then, we established a high-fat diet (HFD)-induced obesity model, and transfer low and high concentrations of wild boar faecal suspension in mice for 9 weeks. The results showed that FMT prevented HFD-induced obesity and lipid metabolism disorders, and altered the jejunal microbiota composition especially increasing the abundance of the Lactobacillus and Romboutsia , which were negatively correlated with obesity-related indicators. Moreover, we found that the anti-obesity effect of wild boar faecal suspension was associated with jejunal N6-methyladenosine (m 6 A) levels. Overall, these results suggest that FMT has a mitigating effect on HFD-induced obesity, which may be due to the impressive effects of FMT on the microbial composition and structure of the jejunum. These changes further alter intestinal lipid metabolism and m 6 A levels to achieve resistance to obesity.

Salidroside Alleviates Furan-Induced Impaired Gut Barrier and Inflammation via Gut Microbiota–SCFA–TLR4 Signaling

Ziyue Wang,Lu Li,Wenliang Li,Haiyang Yan,Yuan Yuan

As a food contaminant that can be quickly absorbed through the gastrointestinal system, furan has been shown to disrupt the intestinal flora and barrier. Investigation of the intestinal toxicity mechanism of furan is of great significance to health. We previously identified the regulatory impact of salidroside (SAL) against furan-provoked intestinal damage, and the present work further explored whether the alleviating effect of SAL against furan-caused intestinal injury was based on the intestinal flora; three models, normal, pseudo-germ-free, and fecal microbiota transplantation (FMT), were established, and the changes in intestinal morphology, barrier, and inflammation were observed. Moreover, 16S rDNA sequencing observed the variation of the fecal flora associated with inflammation and short-chain fatty acids (SCFAs). Results obtained from the LC–MS/MS suggested that SAL increased furan-inhibited SCFA levels, activated the mRNA expressions of SCFA receptors (GPR41, GPR43, and GPR109A), and inhibited the furan-activated TLR4/MyD88/NF-κB signaling. Analysis of protein–protein interaction further confirmed the aforementioned effects of SAL, which inhibited furan-induced barrier damage and intestinal inflammation.

Gut microbiota from NLRP3-deficient mice ameliorates depressive-like behaviors by regulating astrocyte dysfunction via circHIPK2

Zhang Yuan,Huang Rongrong,Cheng Mengjing,Wang Lirui,Chao Jie,Li Junxu,Zheng Peng,Xie Peng,Zhang Zhijun,Yao Honghong

Background Inflammasomes have been found to interact with the gut microbiota, and this effect is associated with depression, but the mechanisms underlying this interaction have not been elucidated in detail. Results The locomotor activity of NLRP3 KO mice was significantly greater than that of their WT littermates, while cohousing and transplantation of the NLRP3 KO gut microbiota avoid the effects of NLRP3 KO on the general locomotor activity at baseline. Meanwhile, transplantation of the NLRP3 KO microbiota alleviated the CUS-induced depressive-like behaviors. The compositions of the gut microbiota in NLRP3 KO mice and WT mice were significantly different in terms of the relative abundance of Firmicutes, Proteobacteria, and Bacteroidetes. Fecal microbiota transplantation (FMT) from NLRP3 KO mice significantly ameliorated the depressive-like behavior induced by chronic unpredictable stress (CUS) in recipient mice. Given the correlation between circular RNA HIPK2 (circHIPK2) and depression and the observation that the level of circHIPK2 expression was significantly increased in CUS-treated mice compared with that in the control group, further experiments were performed. FMT significantly ameliorated astrocyte dysfunction in recipient mice treated with CUS via inhibition of circHIPK2 expression. Conclusion sOur study illustrates the involvement of the gut microbiota-circHIPK2-astrocyte axis in depression, providing translational evidence that transplantation of the gut microbiota from NLRP3 KO mice may serve as a novel therapeutic strategy for depression.

Vitamin A supplementation ameliorates ulcerative colitis in gut microbiota–dependent manner

Bing Pang,Han Jin,Ning Liao,Junjun Li,Chunmei Jiang,Junling Shi

Ulcerative colitis (UC), is a chronic relapsing inflammatory condition of the gastrointestinal track. The purpose of this study is to explore whether Vitamin A (V A ) can treat UC and its mechanisms. A mouse model of UC was established using 3.0% ( w / v ) dextran sodium sulfate (DSS). V A was used to treat UC by intragastric administration of 5000 international unit (IU) retinyl acetate. Fecal microbiota transplantation (FMT) was also used to treat the UC model mice to verify the effect of influenced gut microbiota. The content of short-chain fatty acids (SCFAs) in cecal contents was quantitatively detected by gas chromatography and mass spectrometry. V A supplementation significantly ameliorated UC. 16S rRNA sequencing indicated that V A -treated mice exhibited much more abundant gut microbial diversity and flora composition. Targeted metabolomics analysis manifested the increased production of SCFAs in V A -treated mice. Gut microbiota depletion and FMT results confirmed the gut microbiota–dependent mechanism as that V A relieved UC via regulating gut microbiota: increase in SCFA-producing genera and decrease in UC-related genera. The restore of intestinal barrier and the inhibition of inflammation were also found to contribute to the amelioration of UC by V A . It was concluded that a V A supplement was enough to cause a significant change in gut microbiota and amelioration of UC.

Evaluation of microbial and vancomycin treatments in ulcerative colitis in murine models

Nihal Hasan,Hongyi Yang

Background Despite the number of available therapies for ulcerative colitis (UC), severe side effects and high cost has limited their clinical application. Thus, finding new alternative strategies with minimal side effects is inevitable. Therefore, this study aimed to compare the effectiveness of different therapeutic approaches in DSS-induced colitis. Methods Firstly, we designed oral bio-therapeutic products, Live Bacterial Products (LBP), which include a mixture of fecal bacteria strains isolated from healthy mice and prepared by microencapsulation and freeze-dried techniques. Then we investigated the efficiency of 7 days of freeze-dried FMT, LBP, and vancomycin treatments in DSS-induced colitis. Secondly, we compared the effect of 15 days of microbial therapies (freeze-dried powder of FMT and LBP microcapsules) and seven days of oral vancomycin on the severity of colitis in mice. Furthermore, the levels of IL-1β and TNF-α were measured in serum by ELISA, and the fecal microbiota diversity was analyzed by high-throughput sequencing for all mice groups. Results After seven days of treatments, our results indicated that oral vancomycin reduced the severity of DSS-induced colitis in mice, where weight gain and a decrease in IL-1 β and TNF-α levels were observed in the vancomycin group compared with other treatment groups. While after two weeks of treatment, the LBP microcapsules were able to reduce the severity of colitis. And at the end of the treatment period, weight gain and a decrease in the DAI scores and the levels of IL-1β and TNF-α were noted in the LBP treatment group compared to other treatment groups. By high-throughput sequencing of the 16S rRNA gene, our results showed that while the microcapsules LBP treatment increased the fecal microbial diversity, after vancomycin therapy, most of the fecal microbiota genera and operational taxonomic units (OTUs) were depleted. Conclusion Our results concluded that treatment duration and preparation methods affect the microbial therapies’ efficiency in UC. Furthermore, this study highlighted the negative consequences of oral vancomycin administration on gut health that should be known before using this medication.

Green synthesis, characterization of formononetin mediated AgNPs and its testing for formothion in typical fruit and vegetable samples

Jiahui Ji,Lianggong Zhao,Xiaohua Liu,Huifang Wu,Donghan Wang,Dan Liu,Xinyue Chen,Shilan Feng

In this study, a natural extract from the plant Radix hedysari (ER) was proven to be useful for the green synthesis of silver nanoparticles (AgNPs) and ER-AgNPs in particular. Considering the complexity of plant extracts, high-pressure liquid chromatography (HPLC) was effective at confirming the components of ER, surprisingly, formononetin was found to comprise more than 85% of the total contents. Since formononetin was confirmed to be the primary component of ER, this compound was also used as the basis to synthesize AgNPs designated FMT-AgNPs that was highly pure and of high quality. The FMT-AgNPs functionalized by formononetin were optimized for further study. A simple, fast and sensitive colorimetric method detect formothion was achieved using FMT-AgNPs. A wide linear range from 0.1 μM to 10 μM was established with a low limit of detection (LOD) of 14.6 nM. Since formononetin performed multiple roles during the synthesis, it was confirmed that the mechanism of detection was based on aldol condensation , intermolecular hydrogen bonds and electrostatic attraction between formononetin and formothion. Finally, the green synthesized FMT-AgNPs were further applied in typical fruit and vegetable samples, indicating that FMT-AgNPs were potentially highly useful.
