Accurate quantification of total antioxidant capacity (TAC) is crucial for evaluating the overall antioxidant activity in biological systems and food products. This is of growing importance in diverse applications such as health risk assessment and nutritional recommendations. Here, we introduce a novel Förster resonance energy transfer (FRET)-based sensor for the determination of TAC. The sensor utilizes an iodine-mediated FRET mechanism, employing BODIPY as the donor and Nile Red as the acceptor, both encapsulated within polystyrene (PS) microspheres. In the presence of iodine, its lipophilicity facilitates adsorption into the PS microspheres, resulting in fluorescence quenching due to the heavy-atom effect. This quenching results in an increased distance between the unquenched donors and acceptors, thereby decreasing FRET efficiency. Conversely, the presence of antioxidants reduces iodine concentration, which increases FRET efficiency. This dynamic allows for the development of a ratiometric fluorescence method for TAC determination. Critically, the sensor demonstrates exceptional photostability and resilience to variations in pH and ionic strength, contributing to its analytical robustness. The proposed method exhibits a linear response to ascorbic acid concentrations ranging from 0 to 50 μM, exhibiting good sensitivity (LOD = 2.5 μM) and selectivity. Additionally, the method has been successfully applied to evaluate TAC in ascorbic acid chewable tablets and lemon juice, demonstrating its potential as a robust and efficient tool for TAC determination.

A Förster resonance energy transfer (FRET)-based biosensor using Cu nanoclusters (NCs) as energy donors and signal magnifiers and MnO 2 nanospheres as both energy acceptors and target recognizers for selective detection of glutathione (GSH) was developed. MnO 2 nanospheres, synthesized through an ultrasonication assisted top down method, can quench the photoluminescence (PL) of Cu NCs through FRET, after forming complex through electrostatic interactions. GSH can recover PL of Cu NCs by triggering the decomposition of complex through digesting MnO 2 nanospheres into Mn 2+ . Additionally, the PL can be further magnified through the host-guest interactions between GSH and Cu NCs, with around 25 times higher than the original intensity and 3 times higher than the initial PL intensity of Cu NCs. Under the optimum conditions, the FRET based sensor can detect GSH in the concentration range from 0 to 140 μM, with a detection limit of 17 μM. Moreover, the proposed method shows a strong anti-interference ability, achieving the sensitive directly detection of GSH in human whole blood samples. Our results provide insights in designing FRET based sensors. Moreover, all the components are nontoxic and biodegradable, demonstrating their potential applications in other fields such as nanomedicine and environmental science.

In the presence of Hg 2+ , a fluorescence resonance energy transfer (FRET) system was constructed between CdSe quantum dots (QDs) (donor) and g-C 3 N 4 (receptors). Nanocomposites of g-C 3 N 4 supported by CdSe QDs (CdSe QDs/g-C 3 N 4 nanosheets) were fabricated through an electrostatic interaction route in an aqueous solution. The nanocomposites were characterized by X-ray photoelectron spectroscopy, X-ray diffraction, Fourier-transform infrared spectroscopy, and transmission electron microscopy. Results showed that the g-C 3 N 4 nanosheets were decorated randomly by CdSe QDs, with average diameter of approximately 7 nm. The feasibility of the FRET system as a sensor was demonstrated by Hg (II) detection in water. At pH 7, a linear relationship was observed between the fluorescence intensity and the concentration of Hg (II) (0–32 nmol/L), with a detection limit of 5.3 nmol/L. The new detection method was proven to be sensitive for detecting Hg 2+ in water solutions. Moreover, the method showed high selectivity for Hg 2+ over several metal ions, including Na + , Mg 2+ , Ca 2+ , Pb 2+ , Cr 3+ , Cd 2+ , Zn 2+ , and Cu 2+ . The CdSe QDs/g-C 3 N 4 nanosheet conjugate exhibited desirable long-term stability and reversibility as a novel FRET sensor. The novel FRET-based fluorescence detection provided an attractive assay platform for quantifying Hg 2+ in complex water solutions.

Exploring sensors based on Förster resonance energy transfer (FRET) systems enables the continuous development of biological sensing technologies. Herein, we report the construction of a FRET sensor with dual-emissive quantum dots (QDs) and meso -tetra(4-sulfonatophenyl) porphine (TSPP). The sensor is composed of mesial green-emissive QDs with a thick silica shell (gQD@SiO 2 ) and circumjacent blue-emissive QDs coated with ultra-thin silica spacer, on which is linked TSPP (bQD@SiO 2 -TSPP). The gQD@SiO 2 endows the sensor with a fluorescent background. Due to the ultra-thin silica spacing, coupled with the superior resonance effect of bQD fluorescence and the Soret-band absorption of TSPP, the FRET efficiency is highly sensitive to the chelation state of TSPP. Relying on the absorbance transition of TSPP complexed with Fe(III), the FRET sensor is applied for ultra-sensitive Fe(III) detection. In aqueous solution, the sensor is demonstrated to linearly detect Fe(III) in the range of 0–1 μM, with a limit of detection (LOD) of 40 nM. More importantly, reliable Fe(III) detection can be achieved via the specific complexation of Fe(III) by TSPP and the ratiometric fluorescent response. As such, the inter-/intra-day precisions in standard samples, as well as the recovery rate in biological matrices, are fully validated. The excellent analytical performance, in combination with the excellent biocompatibility of the FRET sensor, allows semi-quantitative Fe(III) imaging in living cells. Graphical abstract
