Studies on the Phosphorus-Solubilizing Ability of Isaria cateinannulata and Its Influence on the Growth of Fagopyrum tataricum Plants

Guimin Yang,Can Liu,Lingdi Gu,Qingfu Chen,Xiaona Zhang

I. cateinannulatahas been shown to promote the growth ofF. tataricum. However, whether its growth-promoting capacity is related to its ability to solubilize phosphorus has not been reported. Therefore, in this study, we sought to assess the phosphorus-solubilizing ability of 18 strains ofI. cateinannulataby analyzing their growth in an inorganic phosphorus culture medium. The effects ofF. tataricumon growth and effective phosphorus content were analyzed through field experiments. The results showed that all 18 strains ofI. cateinannulatahad a phosphorus release capacity, with phosphorus solubilization ranging from 5.14 ± 0.37 mg/L to 6.21 ± 0.01 mg/L, and strain 9 exhibited the best phosphorus solubilization effect. Additionally, the field results demonstrated thatI. cateinannulatapositively influenced the growth, root length, and yield ofF. tataricumby increasing the chlorophyll and soluble phosphorus content. This study will provide a material basis and theoretical support for investigating the interaction mechanism betweenI. cateinannulataandF. tataricum.

The cause of germination increases the phenolic compound contents of Tartary buckwheat (Fagopyrum tataricum)

Yue Chen,Yong Zhu,Likang Qin

ABSTRACT In order to study the changes and mechanism of phenolic compounds during Tartary buckwheat germination, the dynamic changes of phenolic compounds were analyzed, and the activities of enzymes that regulate phenolic compound biosynthesis and degradation were monitored. Total phenolics and total flavonoids presented an interesting dynamic trend with the extension of the germination time, and rutin, gallic acid, chlorogenic acid as well as 2,3,4-trihydroxybenzoic acid contents showed an increasing trend during germination period, the contents were up to (2 663.4 ± 61.1), (449.12 ± 5.26), (99.953 ± 7.800) and (50.442 ± 1.477) mg/100 g DW, respectively. Phenylalanine ammonia lyase (PAL) and chalcone isomerase (CHI) are dominant enzymes in regulating phenolic compound biosynthesis, which showed an increasing trend. Rutin degrading enzyme (RDEs) is the key enzyme in regulating phenolic compound degradation, which showed a decreasing trend. These results suggested that germination processing increases the phenolic compound contents of Tartary buckwheat, which may be regulated by the activation of PAL, CHI and the inhibition of RDEs.

Effects of microwave irradiation of Fagopyrum tataricum seeds on the physicochemical and functional attributes of sprouts

Hui Ma,Xianmeng Xu,Shunmin Wang,Junzhen Wang,Simeng Wang

The effects of microwave-preexposing Tartary buckwheat seeds on their germination, including nutritional and functional attributes were investigated. The results showed that microwave irradiation (300 W/50 s) accelerated the germination process. Germination caused a 7.51-fold increase in reducing sugar content and a 69.61% reduction in starch content, accompanied by significantly enhanced amylase activity, which was amplified by 300 W/50 s microwaving. Microwave also enhanced the levels of various amino acids (especially at 300 W/50 s) and fatty acids (especially at 550 W/75 s) during germination. Enhancements of phenylalanine ammonia-lyase (PAL), chalcone isomerase (CHI), and flavonol synthetase (FLS) activities were observed with microwave irradiation, and maximum total flavonoid content (5.15 g/100 g dry weight; 31.78% higher than that of the control) was reached after irradiation at 300 W/50 s. In addition, the antioxidant capacity and the tyrosinase and acetylcholinesterase inhibitory activities were enhanced. Principal component analysis indicated that changes in flavonoids, PAL, CHI, and FLS were highly correlated with antioxidant capacity and enzyme inhibitory activities. Therefore, microwave irradiation is an efficient protocol for enhancing the physicochemical and functional attributes of edible Tartary buckwheat sprouts.

Modified Tartary buckwheat (Fagopyrum tataricum Gaertn.) starch by gaseous ozone: Structural, physicochemical and in vitro digestible properties

Jingwei Hu,Xiaoping Li,Zhiyuan Cheng,Xin Fan,Zhen Ma,Xinzhong Hu,Guiling Wu,Yanan Xing

The present study investigated the effects of gaseous ozone treatment on the structural, physicochemical and in vitro digestible properties of Tartary buckwheat starch (TBS). The results showed that gaseous ozone treatment increased the detectable carbonyl and carboxyl groups content, improved the amylose content, and decreased pH of TBS . The granule microstructures of native TBS and ozone-treated TBS (OTBS) were characterized by SEM, CLSM and small-angle X-ray scattering (SAXS), showing that OTBS exhibited a reduced granule smoothness and integrity, lessened coloration area of FITC fluorescent as well as decreased crystalline layer and increased amorphous region. FTIR and XRD results revealed that appropriate time of gaseous ozone treatment (2.5 and 7.5 min) enhanced short-range and long-range order structure due to the increased intermolecular oxidation cross-linking, whereas over treatment (15 and 20 min) caused degradation of the starch. With the increasing treatment time, apparent viscosity increased while breakdown viscosity, setback viscosity, swelling power and solubility decreased. OTBS exhibited a higher gelatinization enthalpy (ΔH) and a better viscoelasticity after gelation (6% OTBS gel, w/w ). Additionally, OTBS-2.5 and OTBS-7.5 showed a decreased rapidly digested starch (RDS) content and increased resistant starch (RS) content, indicating that the digestible properties of two oxidized starches was decreased, which provide a feasible way to develop TBS as a new modified starch source.

Calcium supply promotes seed germination in Tartary buckwheat (Fagopyrum tataricum) by mediating amino acid and lipid metabolism to drive osmotic regulation and antioxidant responses

Wang Qiang,Deng Jinyang,Zeng Qingchen,Wang Zhiyan,Yin Mingda,Zhan Chao,Wang Yu,Liang Xiaotian,Xiang Dabing,Zheng Xiaoqin,Huang Jingwei,Liang Chengang,Mei Linsen,Fan Yu,Zou Liang,Wan Yan

Background Tartary buckwheat ( Fagopyrum tataricum ) is predominantly cultivated in arid and semi-arid mountainous regions. However, existing studies predominantly focus on describing the natural metabolic changes and the regulation of stress resistance during the germination of Tartary buckwheat. In contrast, research on the systematic influence of calcium ions on the germination physiology and metabolic networks of plants has yet to be reported. In this study, we elucidated the physiological mechanisms underlying calcium-mediated effects on Tartary buckwheat seed germination using six concentrations of CaCl₂ (0, 1, 2, 3, 4, and 5 g·L⁻¹). Results Exogenous calcium application exhibited a concentration-dependent effect on seed germination, characterized by low-dose promotion and high-dose inhibition. An appropriate calcium supply significantly promoted the accumulation of osmoregulatory substances, including total sugars, reducing sugars, and free amino acids, by enhancing the decomposition of starch and soluble proteins. Furthermore, it facilitated the accumulation of active compounds, such as total phenols, flavonoids, and γ-aminobutyric acid, and enhanced the activities of key enzymes including phenylalanine ammonia-lyase, glutamate decarboxylase, and α-amylase, thereby improving seed antioxidant capacity. Metabolomic analysis revealed that linoleic acid metabolism, D-amino acid metabolism, and phenylalanine metabolism are the core pathways involved in calcium-regulated seed germination. Furthermore, exogenous calcium systematically improved stress resistance and germination capacity by modulating the levels of key metabolites in these pathways, thereby influencing lipid remodeling, nitrogen metabolism, and secondary metabolite synthesis. Conclusions Exogenous calcium effectively promoted the germination of Tartary buckwheat seeds by mediating amino acid and lipid metabolism, thus regulating osmotic balance and the antioxidant defense system. The optimal treatment concentration identified was 3 g·L⁻¹ CaCl₂. This study elucidates the multiple mechanisms through which calcium regulates the germination of Tartary buckwheat seeds at both physiological and metabolic levels, providing a theoretical basis and practical guidance for the rational application of calcium fertilizers in Tartary buckwheat cultivation.
