Nanozymes with peroxidase-mimicking activities have been extensively explored for hydrogen peroxide (H 2 O 2 ) detection. However, the conventional nanozyme assay with suspensions suffer the limitations of restricted available surface, loss of short-lived radicals, and slow reaction in non-acidic conditions, thus have a greatly reduced detection sensitivity. We here proposed to overcome these drawbacks by constructing aligned nanoreactor sensor with Fe 3 O 4 nanozymes distributed alongside the nanochannels of anodized aluminum oxide (termed as NR-Fe 3 O 4 ), which allowed for a spatially confined nanozyme reactions ( ca. 5 nm) operated under a filtration reaction mode. We presented the unique merits of NR-Fe 3 O 4 on H 2 O 2 detection compared to Fe 3 O 4 nanozyme suspensions, together with the mechanism discussion about how the radical production and capturing got maximized under the confinement. As a result, our NR-Fe 3 O 4 sensor featured a highly sensitive H 2 O 2 detection with a linear concentration from 10 to 5000 nM (detection limit of 0.9 nM), expanded workable pH to neutral, and enhanced nanozyme stability for at least 8 h continuous run. We further showed the great feasibility of using NR-Fe 3 O 4 sensor in complex water matrices for H 2 O 2 detection, as it allowed for a pore rejection of background organics which in turn protected the internal radicals for a capturing and signal magnification. Our findings highlight the great interest of constructing confinement nanoreactors as an innovative approach of enhancing H 2 O 2 detection sensitivity based on the nanozyme assay.

Sensitive but with simple, inexpensive detection of disease-related biomarkers in real biological samples is of quite necessity for early diagnosis and disease surveillance. We herein first introduced high-activity Fe 3 O 4 nanozyme as signal amplifier to develop an ultrasensitive photoelectrochemical (PEC) immunoassay , which meanwhile has the distinct merits of both simplicity and low cost compared with previously reported enzyme-labeling PEC immunoassays. In the proposal, to illustrate and describe the PEC platform, prostate-specific antigen (PSA, Ag) was used as a target model. Specifically, ZnO nanorods (ZnO-NRs) grown vertically on a bare indium–tin oxide (ITO) electrode was deposited with ZnIn 2 S 4 nanocrystals , producing ZnIn 2 S 4 /ZnO-NRs/ITO photoelectrode as the PEC matrix to modify capture PSA antibody (Ab 1 ). Histidine-modified Fe 3 O 4 (his-Fe 3 O 4 ) nanozyme as signal amplifier was linked with signal PSA antibody (Ab 2 ) to form his-Fe 3 O 4 @Ab 2 conjugate, and was anchored through specific sandwich immunoreaction. The labeling his-Fe 3 O 4 nanozyme acted as a peroxidase to induce the generation of the insoluble and insulating precipitation, resulting in an evident decrease in the photocurrent signal. On account of combined effects of high catalytic efficiency of the his-Fe 3 O 4 nanozyme and excellent PEC properties of the ZnIn 2 S 4 /ZnO-NRs/ITO photoelectrode, ultralow detection limit of 18 fg/mL for target Ag detection was achieved. Besides, as high-activity his-Fe 3 O 4 nanozyme has substituted natural enzyme as signal amplifier, simplicity and low cost of the PEC immunoassay was realized.

As pioneering Fe 3 O 4 nanozymes, their explicit peroxidase (POD)-like catalytic mechanism remains elusive. Although many studies have proposed surface Fe 2+ -induced Fenton-like reactions accounting for their POD-like activity, few have focused on the internal atomic changes and their contribution to the catalytic reaction. Here we report that Fe 2+ within Fe 3 O 4 can transfer electrons to the surface via the Fe 2+ -O-Fe 3+ chain, regenerating the surface Fe 2+ and enabling a sustained POD-like catalytic reaction. This process usually occurs with the outward migration of excess oxidized Fe 3+ from the lattice, which is a rate-limiting step. After prolonged catalysis, Fe 3 O 4 nanozymes suffer the phase transformation to γ-Fe 2 O 3 with depletable POD-like activity. This self-depleting characteristic of nanozymes with internal atoms involved in electron transfer and ion migration is well validated on lithium iron phosphate nanoparticles. We reveal a neglected issue concerning the necessity of considering both surface and internal atoms when designing, modulating, and applying nanozymes.

Cerebral ischemic stroke stimulates excessive reactive oxygen species, which lead to blood–brain-barrier disruption, neuron death, and aggravated cerebral infarction. Thus, it is critical to develop an antioxidant strategy for stroke treatment. Herein, we report a dietary strategy to promote stroke healing using iron oxide (Fe3O4) nanoparticles with intrinsic enzyme-like activities. We find that Fe3O4 nanozymes exhibit triple enzyme-like activities, peroxidase, catalase, and superoxide dismutase, thus potentially possessing the ability to regulate the ROS level. Importantly, intragastric administration of PEG-modified Fe3O4 nanozymes significantly reduces cerebral infarction and neuronal death in a rodent model following cerebral ischemic stroke. Ex vivo analysis shows that PEG-modified Fe3O4 nanozymes localize in the cerebral vasculature, ameliorate local redox state with decreased malondialdehyde and increased Cu/Zn SOD, and facilitate blood–brain-barrier recovery by elevating ZO-1 and Claudin-5 in the hippocampus. Altogether, our results suggest that dietary PEG-modified Fe3O4 nanozymes can facilitate blood–brain-barrier reconstruction and protect neurons following ischemic stroke.

Herein, a highly sensitive and selective ratiometric fluorescence platform was constructed for probing adenosine triphosphate (ATP) for the first time by using magnetic Fe 3 O 4 nanozymes that were modified by ATP-specific aptamer labeled with 7-Amino-4-methyl-3-coumarinylacetic acid (Apt-AMCA) to yield Fe 3 O 4 @Apt-AMCA. It was discovered that the incorporation of Apt-AMCA greatly enhance the peroxidase-like activity of Fe 3 O 4 nanozymes, which might catalyze more o -phenylenediamine (OPD) substrates into fluorescent oxidized OPD (DAP) with a emission of 560 nm. More importantly, upon ATP addition, Apt-AMCA could potentially be released from Fe 3 O 4 @Apt-AMCA because of Apt-target binding and the competition between ATP and Apt. The rational increase in the levels of free Apt-AMCA could thereby be attained in the reaction solutions so as to induce an enhancement of fluorescence at 450 nm. Meanwhile, the desorption of Apt-AMCA from the Fe 3 O 4 @Apt-AMCA surface would decrease the peroxidase-like activity of Fe 3 O 4 @Apt-AMCA, thereby reducing its ability to catalyze OPD into fluorescent DAP and thus decreasing fluorescence at 560 nm. A ratiometric fluorescence method was accordingly formulated to permit the sensitive and selective identification of ATP, featuring a linear dynamic range that ranges from 0.10 μM to 80 μM. Accurate measurements of ATP in practical blood samples were also confirmed, promising a new detection method for ATP analysis that can widely applied in clinical laboratories.

The significance of the tumor microenvironment (TME) in tumor initiation and progression is increasingly acknowledged. Conventional therapeutic approaches face limitations within the complex TME, including the restrictions imposed by hypoxia on photodynamic therapy (PDT) and the deficiency of endogenous H₂O₂ affecting chemodynamic therapy (CDT). In response to the TME's characteristics of high metabolism, hypoxia, and weak acidity, a multifunctional nanosystem MNPs/GOD@CS/IR820, which synergistically integrates CDT and PDT, has been developed. This system can actively accumulate at tumor sites under an external magnetic field and release active components in response to the weakly acidic TME. It mitigates the limitations imposed by hypoxia and endogenous H₂O₂ deficiency on PDT and CDT, respectively, thereby enabling synergistic treatment. Additionally, the system's multimodal imaging capabilities facilitate precise tumor localization and real-time, non-invasive in vivo assessment via fluorescence imaging and MRI. In vitro and in vivo evaluations demonstrate significant antitumor efficacy, effectively inhibiting tumor growth and improving survival rates. By comprehensively addressing the challenges posed by the complex TME and enhancing real-time monitoring capabilities, our nanosystem paves the way for personalized and precise cancer treatment. Statement of Significance This study introduces an innovative MNPs/GOD@CS/IR820 nanosystem that represents a significant advancement in cancer nanomedicine by addressing critical limitations of conventional photodynamic therapy (PDT), particularly in hypoxic tumor microenvironments. By synergistically integrating chemodynamic therapy (CDT) with PDT and incorporating MRI and fluorescence dual-modal imaging capabilities, this multifunctional platform offers enhanced therapeutic efficacy and real-time monitoring. The system's ability to generate oxygen in situ overcomes hypoxia-induced limitations, while its multimodal mechanism of action induces tumor cell apoptosis through multiple pathways. In vitro and in vivo studies demonstrate remarkable antitumor efficacy across diverse cancer types, significantly inhibiting tumor growth and improving survival rates. This comprehensive approach to cancer diagnosis and treatment not only advances precision medicine for targeted, multimodal cancer management but also provides a promising foundation for future clinical applications, potentially transforming cancer treatment strategies and improving patient outcomes.

The development of cell-like nanoreactors with the ability to initiate biocatalytic cascades under special conditions holds tremendous potential for therapeutic applications. Herein, conformationally gated nanoreactors that respond to the acidic microenvironment of infected diabetic wounds were developed by cucur[8]bituril (CB[8])-based supramolecular assembly. The bioinspired nanoreactors exhibit not only self-regulated permeability and selectivity to control internal enzyme activities by substance exchange but also distinct binding specificities toward Gram-positive and Gram-negative bacteria via noncovalent modification with different ligands. The encapsulation of glucose oxidase (GOx), Fe3O4 nanozyme, and l-arginine (l-Arg) into the nanocarriers enables intelligent activation of multienzyme cascade reactions upon glucose (Glu) uptake to produce gluconic acid (GA) and hydrogen peroxide (H2O2), which is further converted into highly toxic hydroxyl radicals (·OH) for selective antibacterial activity. Moreover, acidic H2O2 promotes the oxidization of l-Arg, leading to the release of nitric oxide (NO). Consequently, this nanoreactor provides a multifunctional and synergistic platform for diabetic chronic wound healing by combining enzyme dynamic therapy with NO gas therapy to combat bacterial infections and inflammation under high blood Glu levels.
