Effects of Flammulina velutipes polysaccharides on gut microbiota composition and metabolism in vitro fermentation

Su Anxiang,Ma Gaoxing,Ma Ning,Pei Fei,Yang Wenjian,Hu Qiuhui

Flammulina velutipes polysaccharides (FVP) exhibit many biological activities, but the effects on gut microflora and metabolism were still unclear. Here, we explored the composition of FVP, their influence on human gut microflora composition and metabolites. FVP were used to vitro fermentation through human fecal inoculums. In addition, 16S rRNA sequencing were used to assess the effects of FVP on the gut microbiota. The metabolic profiles were investigated using untargeted metabolomics approaches in the LC-MS platform. The results showed that FVP was mainly consisted of glucose, mannose, xylose, fucose and galactose. FVP is shown to increase the relative abundances of Bifidobacteriaceae , as well as Bacteroidaceae and remarkably decrease the numbers of genera Lachnospiraceae coupled with Enterococcaceae . The differential metabolites were identified and mainly involved the metabolism of glycerophospholipid, linoleic acid and synthesis of unsaturated fatty acids. FVP may exhibit biological activity function by regulating gut microflora composition and metabolites.

Molecular Properties of Flammulina velutipes Polysaccharide–Whey Protein Isolate (WPI) Complexes via Noncovalent Interactions

Jiaqi Shang,Minhe Liao,Ritian Jin,Xiangyu Teng,Hao Li,Yan Xu,Ligang Zhang,Ning Liu

Whey protein isolate (WPI) has a variety of nutritional benefits. The stability of WPI beverages has attracted a large amount of attention. In this study,Flammulina velutipespolysaccharides (FVPs) interacted with WPI to improve the stability via noncovalent interactions. Multiple light scattering studies showed that FVPs can improve the stability of WPI solutions, with results of radical scavenging activity assays demonstrating that the solutions of the complex had antioxidant activity. The addition of FVPs significantly altered the secondary structures of WPI, including its α-helix and random coil. The results of bio-layer interferometry (BLI) analysis indicated that FVPs interacted with the WPI, and the equilibrium dissociation constant (KD) was calculated as 1.736 × 10−4M in this study. The in vitro digestibility studies showed that the FVPs protected WPI from pepsin digestion, increasing the satiety. Therefore, FVPs effectively interact with WPI through noncovalent interactions and improve the stability of WPI, with this method expected to be used in protein-enriched and functional beverages. Keywords:Flammulina velutipes;protein–polysaccharide complexes;stability;bio-layer interferometry;in vitro digestibility;binding regions

Modulation of gut microbiota and metabolites by Flammulina velutipes polysaccharides during in vitro human fecal fermentation: Unveiling Bacteroides as a potential primary degrader

Zi Ye,Leilei Yu,Chuan Zhang,Yuhang Gao,Jianxin Zhao,Arjan Narbad,Wei Chen,Qixiao Zhai,Fengwei Tian

Flammulina velutipes , a widely cultivated species of edible fungus , exhibits diverse functional activities attributed to its polysaccharides . In this study, we employed an in vitro model to investigate the impact of F. velutipes polysaccharides (FVP) fermentation on gut microbiota , with a particular focus on Bacteroides . FVP fermentation resulted in the proliferation of microbiota associated with short-chain fatty acid (SCFA) metabolism and suppression of Escherichia-Shigella . Bacteroides emerged as potential primary degraders of FVP, with species-level analysis identifying the preference of B. thetaiotaomicron and B. intestinalis in FVP degradation. Metabolomics analysis revealed significant increases in hypoxanthine and 7-methyladenine contents, with histidine metabolism emerging as the most enriched pathway. B. nordii and B. xylanisolvens exhibited the most influence on amino acid and SCFA metabolism. Understanding the mechanisms by which gut microbiota metabolize FVP can provide valuable insights into the potential of FVP to promote intestinal health and disease prevention.

Studying on the structure-activity relationship of Flammulina velutipes polysaccharides via ultrasonic degradation: Insights into molecular weight, chain conformation, and anti-inflammatory activity

Jingsi Zhang,Xin Chen,Qiping Zhan,Yifan Wang,Keke Meng,Qiuhui Hu,Liyan Zhao

Flammulina velutipes ( F. velutipes ) polysaccharides have been shown to play a crucial role in preventing inflammatory diseases. However, further exploration is needed to understand the specific relationship between their structure and anti-inflammatory activity. In this study, four structurally distinct F. velutipes polysaccharides (FVP2, FVP2U1, FVP2U2, and FVP2U3) were isolated via a previously established ultrasonic degradation model, and the exploration on the impact of chain conformation on their anti-inflammatory activities was focused. The results indicated that FVP2 exhibited a spherical conformation, while FVP2U1 and FVP2U2 displayed flexible chains, and FVP2U3 showed a semi-rigid chain. The study on the impact of four kinds of F. velutipes polysaccharides on inflammatory response in LPS-stimulated RAW264.7 cells revealed that ultrasonic degradation enhanced the anti-inflammatory activity of FVP2. The primary anti-inflammatory mechanism involved the inhibition of NO, TNF-α, IL-1β, and IL-6 release, and reduced transcriptional levels of TLR4, MyD88, NF-κB, and NLRP3. Among the ultrasonically degraded polysaccharides with compliant chain conformations and lower molecular weight demonstrated superior anti-inflammatory activity. These results provide insights into the relationship between molecular weight, chain conformation, and anti-inflammatory activity of F. velutipes polysaccharides, contributing to the potential application of ultrasound in developing highly active anti-inflammatory polysaccharides and related nutritional products.

Characteristic of polysaccharides from Flammulina velutipes in vitro digestion under salivary, simulated gastric and small intestinal conditions and fermentation by human gut microbiota

Anxiang Su,Gaoxing Ma,Minhao Xie,Yang Ji,Xiangfei Li,Liyan Zhao,Qiuhui Hu

Summary In this study, in vitro digestion and fermentation of Flammulina velutipes -derived polysaccharides (FVP) were investigated. It was found that FVP mainly consisted of 48.45% glucose, 15.40% mannose, 14.60% xylose, 11.80% fucose and 9.90% galactose. The -human saliva, simulated gastric and small intestinal juices conditions did not break down the FVP. Based on in vitro fermentation tests, FVP modulated the composition of gut microbiota by elevating the amounts of Bifidobacteriaceae and Bacteroidaceae and reducing the numbers of genera Lachnospiraceae and Enterococcaceae . Meanwhile, FVP affected the synthesis of short-chain fatty acids derived from gut microbiota.

Flammulina velutipes-derived carbon dots for fluorescence detection and imaging of hydroxyl radical

Yu Hou,Jiawei Lin,Yichao Chen,Jianbin He,Zhe Su,Qinhua Zheng,Xiujuan Zhao,Xiaodan Lv,Xiuping Tang,Changren Zhou

Monitoring hydroxyl radical (•OH) fluctuation is of great importance to study some relative pathological processes and to predict early diagnosis of diseases. Efficient •OH-responsive fluorescent sensors based on carbon dots (CDs) have been reported, but most researches have focused on the new strategies for the synthesis and doping of the CDs. Herein, a kind of biomass CDs (F-CDs) with Flammulina velutipes ( F. velutipes ) as the carbon source was prepared by a one-step hydrothermal method without any additional modification. The prepared F-CDs have remarkable sensitivity and selectivity and there is a good linear relationship from 0 to 12 μM with a low detection limit of 95 nM for quantitative •OH assay. With excitation-independent emission, favourable biocompatibility and low toxicity, the F-CDs can penetrate cell membranes as •OH-responsive fluorescent sensors to detect intracellular •OH in A549 cells stimulated by phorbol 12-myristate 13-acetate (PMA) and successfully monitor the •OH concentration levels by the corresponding fluorescence change. Given the combined benefits of the green and eco-friendly approach, the F-CDs show promise as novel theranostics tools for early detection and treatment of related diseases.

Effects of Flammulina velutipes polysaccharide on immune response and intestinal microbiota in mice

Ruiqiu Zhao,Qiuhui Hu,Gaoxing Ma,Anxiang Su,Minhao Xie,Xiangfei Li,Guitang Chen,Liyan Zhao

A crude polysaccharide derived from Flammulina velutipes (FVP), mainly consisting of glucose linked with β-glycosidic bonds, was administered to 6 week-old male ICR mice by gavage for five weeks, to investigate the effects on mice intestinal microbiota and immune response. The pH of the intestinal compartments decreased while short chain fatty acids concentrations increased in FVP-fed groups, in a dose dependent manner. High throughput sequencing revealed the enrichment in the diversity and an alteration in the composition of the fecal microbiota in the FVP fed mice. A significant decrease in the Firmicutes / Bacteroidetes ratio, relative abundances of Lactobacillus and Akkermansia were related to the energy absorption. The relative abundances of Porphyromonadaceae and Bacteroidaceae positively correlated with an increase in serum immunoglobulin and cytokines. This study indicated that this crude polysaccharide from F. velutipes could modulate the gut microbiota and represented a potential capacity of obesity suppression and immune regulation.

Pullulan-stabilized Soybean Phospholipids/Cinnamaldehyde emulsion for Flammulina velutipes preservation

Xue Shao,Ben Niu,Xiangjun Fang,Weijie Wu,Ruiling Liu,Honglei Mu,Haiyan Gao,Hangjun Chen

Fresh mushrooms ( Flammulina velutipes ) are very perishable and easily brown; also they undergo postharvest loss of nutritive constituents. In this study, cinnamaldehyde (CA) emulsion was prepared by using soybean phospholipids (SP) as emulsifier and pullulan (Pul) as stabilizer. The effect of emulsion on the quality of mushroom during storage was also studied. The experimental results indicated that the emulsion obtained by adding 6 % pullulan was found to the most uniform and stable, which is beneficial to its application. Emulsion coating maintained the storage quality of Flammulina velutipes. The incorporation of CA emulsion into the coating system showed a positive effect on inhibiting the accumulation of reactive oxygen species, resulting from improving the effectiveness of delaying active free radical scavenging enzymes . The shelf life of mushrooms coated with emulsion was significantly prolonged, which indicates its potential application in food preservation.

Isolation, characterization and HepG-2 inhibition of a novel proteoglycan from Flammulina velutipes

Hui Xu,Ye Hu,Qiuhui Hu,Jianhui Liu,Anxiang Su,Minhao Xie,Gaoxing Ma,Fei Pei,Alfred Mugambi Mariga,Wenjian Yang

Flammulina velutipes has anti-inflammatory, immunomodulatory, antioxidant and many bioactive properties with high contents of carbohydrate, proteins and fibers. In this study, a novel proteoglycan with polysaccharide complexes and protein chain, named PGD1-1, was isolated from F. velutipes . The structural characteristics of PGD1-1 were then determined, and its anti-proliferation and pro-apoptotic activities against HepG-2 cells were demonstrated in vitro. Results proved that the average molecular weight of PGD1-1 was 32.71 kDa, and the carbohydrate and protein contents were 93.35 and 2.33%, respectively. The protein moiety was bonded to a polysaccharide chain via O-glycosidic linkage. The monosaccharides consisted of d -glucose, D-galactose and D-xylose in a molar ratio of 21.90:2.84:1.00. PGD1-1 significantly inhibited the proliferation of HepG-2 cells by affecting cell lipid peroxidation and nitric oxide production. In addition, PGD1-1 promoted the apoptosis of HepG-2 cells, especially the early apoptosis. These findings proved that PGD1-1 was a novel potent ingredient against the proliferation of HepG-2, which will provide a theoretical basis for the development and utilization of the functional ingredients of the F. velutipes .

Ultrasonic assisted three-phase partitioning extraction and bioactivities validation of polysaccharides from Flammulina velutipes residues

Yao Zhang,Changjiao Zhong,Shiyu Liu,Linna Du,Hongxia Ma,Haipeng Zhang,Yuting Li,Chengguang He,Lili Guan

Mushroom residues, especially Flammulina velutipes residues (FVR) rich in bioactive polysaccharides, are under exploited due to inefficient extraction methods conflicting with green chemistry. This study developed an ultrasonic-assisted three-phase partitioning (UAE-TPP) approach to extract crude polysaccharides (cTP-FVRP) from FVR, combined with structural and functional characterization. An integrated optimization strategy combining response surface methodology (RSM), artificial neural network (ANN), and genetic algorithm (GA) identified global optimum conditions (18 min, 1:1 crude extract to n-Butanol, 44 % (NH₄)₂SO₄), yielding cTP-FVRP at 3.49 ± 0.05 %-significantly higher than that of UAE method (2.38 ± 0.04 %). Purified TP-FVRP was a 108.13 kDa non-starch polysaccharide with a triple helix, α-linkages, and 9 monosaccharides, stable under physiological conditions. It exhibits potent antioxidant activity, biocompatibility, and immunomodulatory potential. UAE-TPP reduces solvents while preserving polysaccharide integrity. This green technique enables high-value utilization of FVR, aligning with sustainability goals and providing a scalable model for industrial by-product valorization.

Green ultrasonic-assisted enzymatic extraction of polysaccharides from Flammulina velutipes residues by response surface methodology

Yiting Yang,Xiaoxue Zhang,Jingbo Zhang,Tiezhu Wang,Shuming Liu,Hongxia Ma,Muhammad Inam,Lili Guan

Flammulina velutipes residues, the by-products of Flammulina velutipes industry, cause environmental pollution. The aim of the current work was to evaluate the extraction efficiency of polysaccharides from Flammulina velutipes residues by an ultrasonic assisted snailase enzymatic extraction (UAEE) method, following the principles of green chemistry, mainly solvent and energy saving. The optimized parameters obtained by Response Surface Methodology (RSM) were determined as follows: ultrasonic power at 150 W, extraction time of 90 min, and extraction temperature of 55 °C. The experimental extraction yield (109.54 ± 5.44 mg/g) was closely matched the predicted yield (110.92 mg/g). Polysaccharides from Flammulina velutipes residues (UE-FVRP) had the molecular weight of 170 kDa and a composition comprising 8 monosaccharides, primarily xylose (Xyl, 24.71%), mannose (Man, 17.29%), arabinose (Ara, 11.48%), and galactose (Gal, 15.63%). FT-IR analysis suggested that UE-FVRP likely contained pyranose with α-glycoside bonds. In vitro antioxidant activity assays revealed that UE-FVRP displayed potent DPPH radical scavenging activity and reducing ability. Additionally, UE-FVRP demonstrated the ability to promote the growth of two Lactobacillus strains, indicating its potential as a novel polysaccharide with high in vitro prebiotic properties. Therefore, these findings proposed a novel, green, and efficient extraction method for extracting polysaccharides from Flammulina velutipes residues, highlighting the potential of utilizing Flammulina velutipes residues as a valuable ingredient in both food and feed applications, contributing to the sustainable reuse of agricultural by-products.

Naringenin-loaded Flammulina velutipes chitin-glucan complexes gel system: structural modulation and dual lipid-lowering mechanisms

Simin Feng,Shuqi Liang,Jianyu Yang,Jiahao Yu,Yang Lin,Yizhen Xie,Peilong Sun,Ping Shao

This study developed a synergistic lipid-lowering system using chitin-glucan complex (CGC) from Flammulina velutipes through green preparation and structural optimization. Enzyme-alkali treated CGC3 exhibited superior acetylation (47.85 %), crystallinity (42.29 %), and molecular weight (1.848 × 10 6  Da) through partial glucan removal. The derived hydrogel (gel-CGC3) had a dense porous network (density 2.92 ± 0.09 × 10 −3  g/cm 3 ), excellent mechanical stability, and regular pore structure. Ball milling prolonged naringenin (NAR) release to 900 min (87.5 % longer than block hydrogels) without compromising structural integrity. Meanwhile, the microgel conferred multiple functional advantages to the system (includes CGC gel and NAR loaded), and its porous structuring (powder-to-hydrogel transition) and structural refinement ( via microgel conversion) maximized binding site exposure, enhancing adsorption efficiency for cholesterol micelles (12.42 ± 0.73 mg/g), sodium taurocholate (3.91 ± 0.07 mg/g), and sodium glycopyrrolate (3.51 ± 0.07 mg/g) by exposing more binding sites. The system synergistically combines lipid adsorption and biometabolism, underscoring the significance of structure-function synergy in polysaccharide-based platforms for metabolic disorder intervention.
