The western flower thrips (Frankliniella occidentalis) is a major pest insect in agriculture. However, few insecticides are effective for their control. The recombinant gamma-aminobutyric acid receptor-associated protein (rGABARAP) was examined as a potential target of the monoterpenoids responsible for their insecticidal activities. The insecticidal activity of anethole, linalool, and methyl eugenol (ME) was evaluated in the laboratory. The half-maximum lethal concentration (LC50) of ME against second-instar nymphs of F. occidentalis was 5.5 mg/L using membrane and leaf immersion methods, while that of spinosyn A was 1.0 mg/L. The dissociation constants of ME binding to rGABARAP were 1.30 and 4.22 μmol/L, respectively, according to microscale thermophoresis (MST) and isothermal titration calorimetry (ITC) measurements. A molecular docking study showed interactions between ME and Tyr174 via π–π stacking. The MST and ITC experiments showed loss of specific binding between ME and the rGABARAPY174A mutant. Therefore, Tyr174 is a key amino acid residue of rGABARAP involving ME binding. The results revealed GABARAP as a potential target for the development of monoterpenoid insecticides.

Plant volatile organic compounds (VOCs) are an important link that mediates chemical communication between plants and plants, plants and insects, and plants and natural enemies of insect pests. In this study, we tested the response in the selective behavior of western flower thrips,Frankliniella occidentalis, to the VOCs of kidney bean,Phaseolus vulgarisL., to explore their “attraction” or “repellent” effects regarding their application in integrated pest management (i.e., IPM). The results indicated that 12.7 μL/mL (E, E, E, E)-squalene, 3.2 μL/mL dioctyl phthalate, and 82.2 μL/mL ethyl benzene had a significantly attractive effect on the selective behavior ofF. occidentalis, while 10.7 μL/mL and 21.4 μL/mL 2,6-ditert-butyl-4-methyl phenol had a significantly repulsive effect on the selective behavior ofF. occidentalis, showing thatF. occidentalisresponds differently to specific concentrations of VOCs fromP. vulgarisplant emissions. Interestingly, the three compounds with the specific above concentrations, after being mixed in pairs, significantly attractedF. occidentaliscompared to the control treatment; however, the mixture with the three above compounds had no significant different effect onF. occidentaliscompared to the control treatment. It can be seen that the effect with the mixtures of three kinds of VOCs had the same function and may not get better. Simultaneously, the reasons for this result from the transcription levels of odorant-binding protein genes (OBPs) were determined. There were differences in the types and transcription levels of OBPs, which played a major role in the host selection behavior ofF. occidentalisunder the mixed treatment of different VOCs. It is presumed that there are specific VOCs fromP.vulgarisplants that have a good repellent or attracting effect on the selective behavior ofF.occidentalis, which can be used for the development of plant-derived insect attractants and repellents to serve as IPM in fields. But attention should be paid to the antagonism between plant-derived preparations and VOCs produced by plants themselves after application. Keywords:Phaseolus vulgarisL.;Frankliniella occidentalis;VOCs;attractive and repellant effects;selection behavior

Simple SummaryInsect repellents have been intensively studied due to growing interest in environmentally safe pest control methods. Numerous repellent compounds have been reported to be effective against western flower thrips,Frankliniella occidentalis. However, few studies have been conducted to test the repellent activity and control efficacy of these compounds against thrips in the field. In this study, we investigated the repellent effects of two plant volatiles, α-pinene and eucalyptol, onF. occidentalisusing cage and field experiments. Our results showed that both α-pinene and eucalyptol at high doses significantly reduced the egg-laying ofF. occidentalison host plants in cage experiments. In addition, the application of eucalyptol EC and α-pinene EC reduced the field population ofF. occidentalis, with eucalyptol EC performing better. The combination of aggregation pheromone traps and repellent eucalyptol spray also significantly reducedF. occidentalispopulations in the field. Our results provided effective eco-friendly control methods for the integrated pest management ofF. occidentalis.Western flower thrips,Frankliniella occidentalis(Pergande), is a serious pest of horticultural and agronomic crops. Using plant-released semiochemicals to control thrips is one eco-friendly control method for their management. In this study, to develop repellents and an attractant–repellent push–pull strategy forF. occidentalis, we investigated the effects of α-pinene and eucalyptol on their oviposition on pepper plants in cage experiments and evaluated the control efficiency of repellents on them in pepper fields. The control efficacy of a combination of aggregation pheromone attractants and plant volatile repellents was also investigated. The results showed that spraying eucalyptol (at the dose of 200 μL) and α-pinene (at the dose of 100 μL) significantly reduced the oviposition behavior ofF. occidentalisin cage experiments. Field results showed that the application of eucalyptol and α-pinene could significantly reduce the population ofF. occidentalisin the field, with the highest control efficacy of 80.96% and 66.66%, respectively. Furthermore, a push–pull strategy combining thrips-repellent eucalyptol and aggregation pheromone lures was an effective strategy to suppress a population of thrips in the field, with the highest control efficacy of 81.95%. The repellents and push–pull strategy developed here provided an effective method for the eco-friendly control ofF. occidentalis.

Simple SummaryThe western flower thrips,Frankliniella occidentalis(Pergande) (Thysanoptera: Thripidae) is an economically damaging invasive pest that is difficult to control due to their resistance to insecticides. Utilizing plant-induced defense responses is a new strategy to increase plant resistance toF. occidentalis. A growing number of studies have shown that calcium plays an important role in enhancing plant resistance to pest-induced stresses. In this study, we found that exogenous calcium treatment could alter the composition of volatile compounds in kidney bean plants, repellingF. occidentaliswhile attracting its natural enemy,Orius similisZheng (Hemiptera: Anthocoridae), and reducing the oviposition preference choice ofF. occidentalis. This study provides a theoretical basis for the control ofF. occidentalis.AbstractFrankliniella occidentalisis a destructive pest of horticultural plants, whileOrius similisis a natural enemy of thrips. It has been demonstrated that exogenous calcium could induce plant defenses against herbivore attack. We examined whether CaCl2supplementation altered the volatile emissions of kidney bean plants, which influence the oviposition preference ofF. occidentalis. We also assessed the influence of volatile cues onO. similis. Using Y-tube olfactometer tests, we found that exogenous CaCl2treatment inhibited the selectivity ofF. occidentalisbut attractedO. similis. In addition, CaCl2treatment reduced the oviposition preference ofF. occidentalis. Gas chromatography–mass spectrometry analyses revealed that CaCl2treatment altered the number and relative abundance of the volatile compounds in kidney bean plants and that (E)-2-hexen-1-ol, 1-octen-3-ol, β-lonone, and (E,E)-2,4-hexadienal might be potential olfactory cues. Furthermore, the results of the six-arm olfactometer test indicated that 1-octen-3-ol (10−2μL/μL), β-lonone (10−2μL/μL), and (E,E)-2,4-hexadienal (10−3μL/μL) repelledF. occidentalisbut attractedO. similis. Overall, our results suggested that exogenous CaCl2treatment induced defense responses in kidney bean plants, suggesting that CaCl2supplementation may be a promising strategy to enhance the biological control ofF. occidentalis. Keywords:Frankliniella occidentalis;Orius similis;exogenous calcium;induced defense;herbivore-induced plant volatiles;olfactory response;gas chromatography–mass spectrometry
