Improving the antioxidant and antibacterial activities of fermented Bletilla striata with Fusarium avenaceum and Fusarium oxysporum

Jian-Wei Dong,Le Cai,Jie Xiong,Xiu-Hua Chen,Wei-Ying Wang,Ni Shen,Bei-Lei Liu,Zhong-Tao Ding

The tubers of Bletilla striata were fermented with Fusarium avenaceum and Fusarium oxysporum . The total phenolic contents (TPCs) and antioxidant and antibacterial activities of the extracts from the fermented products were measured. Additionally, the extracts of both F. avenaceum and F. oxysporum fermented B. striata (FBS) were characterized by UV–vis spectroscopy, Fourier transform infrared (FT-IR) spectroscopy, and nuclear magnetic resonance (NMR) spectroscopy. The results showed that the F. avenaceum and F. oxysporum FBSs possessed higher TPCs and exhibited stronger antioxidant and antibacterial activities ( Bacillus subtilis ) in comparison with original material not co-fermented with microorganisms. Chemical characterization of the two extracts with UV–vis, FT-IR, and 1 H NMR implied that some of the phenolic glycosides may be deglycosylated and that bibenzyl compounds and saccharides may be metabolized in both FBS systems. Microbial fermentation enhances the TPC as well as the antioxidant and antibacterial activities of B. striata . The present study suggests that F. avenaceum and F. oxysporum fermentations were effective in B. striata processing.

Growth boundary of Fusarium graminearum spores as a function of temperature, pH, and H2S based on neural network

Yan Zhu,Yameng Gao,Wenke Wang,Wenjie Kan,Caiguo Tang,Lifang Wu

Fusarium graminearum ( F. graminearum ) poses a substantial threat to global food security, with its impact closely linked to environmental conditions. This study aims to anticipate F. graminearum spore growth across diverse environments and assess alterations in the fungistatic capacity of hydrogen sulfide (H 2 S) gas in response to environmental variations. The experimental data, namely spore germination ratios, are monitored and recorded at hourly intervals across 100 permutations comprising four distinct temperature levels, five varying pH values, and five diverse concentrations of H 2 S. The investigation integrates three environmental factors as input variables, utilizing a feedforward neural network to establish growth boundaries based on spore germination ratios. The accuracy of models at various stages–training, validation, and testing–is evaluated using a confusion matrix. Logistic regression models are employed for effectiveness comparison with neural networks. Results indicate that neural networks successfully predict F. graminearum spore germination events. Notably, H 2 S does not influence the temperature preferences of spores. Temperature has a more substantial impact on the fungistatic ability of H 2 S compared to pH. The study reveals the adaptability of F. graminearum spores to adverse conditions over time. This research delves into temperature, pH, and H 2 S synergistic effects on spore germination, contributing to predictive models for F. graminearum and enhancing our understanding of how environmental factors regulate its growth.

Activity of a SDHI fungicide penflufen and the characterization of natural-resistance in Fusarium fujikuroi

Yanan Sun,Haiping Shi,Chenxin Mao,Jianyan Wu,Chuanqing Zhang

The occurrence of bakanae disease of rice caused by the fungus Fusarium fujikuroi in Zhejiang Province has become increasingly aggravated in recent years, concomitant with the development of resistance to the widely applied fungicides , prochloraz and phenamacril. In this study, the activity of a novel succinate dehydrogenase inhibitor (SDHI) fungicide, penflufen, against different fungi was evaluated in addition to the potential of penflufen in controlling F. fujikuroi infections. Penflufen exhibited good bioactivity against F. fujikuroi , but weak activity against Fusarium spp. and other investigated plant-pathogenic fungi including Colletotrichum spp. In addition to inhibiting mycelial growth , penflufen effectively inhibited F. fujikuroi conidium production. For germination, penflufen could effectively inhibit that of small conidia, but only delay the germination of large conidia. In addition, the sensitivity to penflufen among 100  F. fujikuroi isolates that were collected in areas that were never exposed to SDHIs was determined based on mycelium growth. Sensitivities surprisingly exhibited bimodal distributions, indicating the presence of natural resistance. Cross-resistance was not observed between penflufen in F. fujikuroi and two fungicides that have been extensively applied in field including prochloraz (a DMI) and phenamacril (a 2-cyanoacrylate fungicide), nor with the three SDHIs, fluopyram, benzovindiflupyr, and pydiflumetofen. Additional analysis identified five different point mutations in SDH-A (i.e., at residues 46, 225, 283, 430, and 586) of naturally resistant isolates. These results inform the potential application of the new SDHI fungicide penflufen for managing crop diseases and understanding possible resistance mechanisms among pathogens .

Visual detection of Fusarium temperatum by using CRISPR-Cas12a empowered LAMP assay coupled with AuNPs-based colorimetric reaction

Yingchun Li,Yaqin Zhang,Fange Kong,Chunxia Wang,Shanshan Chen,Jiasi Wang,Di Wang

Fusarium temperatum ( F. temperatum ) causes maize stalk rot disease, reducing grain yield and producing multiple harmful mycotoxins that threaten food safety and quality. However, current detection methods are limited by the requirement of well-trained personnel, complicated operations and expensive instruments. To address this challenge, gold nanoparticles (AuNPs)-based colorimetric assay using loop-mediated isothermal amplification (LAMP) coupled with CRISPR-Cas12a (Au-CRISPR) was tested for F. temperatum detection. The assay utilizes DNA-modified AuNPs as a colorimetric probe and the results can be readily observed by the naked eye. After optimization, the method can specifically detect as low as 100 copies/μL of target DNA or 10 −8  ng/μL of extracted DNA from F. temperatum . The detection platform allows high-throughput testing by integrating microtiter plates and microplate reader. Furthermore, a portable smartphone-based device was developed to realize point of care (POC) detection of F. temperatum in resource-limited settings. This simple, inexpensive and sensitive detection platform has great potential for the applications in field detection.

Effects of Different Carbon Sources on Fumonisin Production and FUM Gene Expression by Fusarium proliferatum

Yu Wu,Taotao Li,Liang Gong,Yong Wang,Yueming Jiang

Fusarium proliferatumcan infect many crops and then produce fumonisins that are very harmful to humans and animals. Previous study indicates that carbon sources play important roles in regulating the fumonisin biosynthesis. Unfortunately, there is limited information on the effects of carbon starvation in comparison with the carbon sources present in the host of fumonisin production inF. proliferatum. Our results indicated thatF. proliferatumcultivated in the Czapek’s broth (CB) medium in the absence of sucrose could greatly induce production of fumonisin, while an additional supplementation of sucrose to the culture medium significantly reduced the fumonisin production. Furthermore, cellulose and hemicellulose, and polysaccharide extracted from banana peel, which replaced sucrose as the carbon source, can reduce the production of fumonisin byF. proliferatum. Further work showed that these genes related to the synthesis of fumonisin, such asFUM1andFUM8, were significantly up-regulated in the culture medium in the absence of sucrose. Consistent with fumonisin production, the expressions ofFUMgene cluster andZFR1gene decreased after the addition of sucrose. Moreover, these genes were also significantly down-regulated in the presence of cellulose, hemicellulose or polysaccharide extracted from peel. Altogether, our results suggested that fumonisin production was regulated inF. proliferatumin response to different carbon source conditions, and this regulation might be mainly via the transcriptional level. Future work on these expressions of the fumonisin biosynthesis-related genes is needed to further clarify the response under different carbon conditions during the infection ofF. proliferatumon banana fruit hosts. The findings in this study will provide a new clue regarding the biological effect of the fumonisin production in response to environmental stress. Keywords:Fusarium proliferatum;fumonisin biosynthesis;carbon source;environmental stress;gene expressionKey Contribution:F. proliferatumcan produce fumonisins which are harmful to human health. This study aimed to explore the effect in the absence; followed by addition of carbon sources on fumonisin production; which will be beneficial for understanding the infection ofF. proliferatuminto banana fruit.

Cinnamaldehyde Acts as a Fungistat by Disrupting the Integrity of Fusarium oxysporum Fox-1 Cell Membranes

Li-Rong Zhou,Hai-Jun Hu,Jie Wang,Yong-Xing Zhu,Xue-Dong Zhu,Jia-Wei Ma,Yi-Qing Liu

To counter the harmful impacts of agricultural chemicals on the environment and human health, there is an increasing demand for safe, eco-friendly, and potent plant-based biopesticides. In this study, we aimed to investigate the antimicrobial effects of ginger essential oil and selected volatile compounds (linalool, eugenol, citral, and cinnamaldehyde [CA]) againstFusarium oxysporumFOX-1. Minimum inhibitory concentrations (MICs) were determined using the mycelium growth inhibition method. The compound CA exhibited the most potent antifungal effect againstF. oxysporumFOX-1 and was selected for further investigation. After treatment with CA at 1/2 MIC or MIC, the spore germination ofF. oxysporumFOX-1 was significantly inhibited at 12 h. Furthermore, microscopic observation revealed that CA treatment resulted in the morphological degradation ofF. oxysporumFOX-1. CA destroyed the cell membrane integrity ofF. oxysporumFOX-1, increasing the relative conductivity and the leakage of intracellular protein, nucleic acids, and malondialdehyde, affecting the integrity and metabolism of the cell membrane. The effects were positively related to CA concentration. Additionally, in vivo experiments with rhizome sections showed that CA significantly reduced the pathogenicity ofF. oxysporumFOX-1. Overall, these findings provide evidence for the potential of using ginger essential oil components as fungicides, offering a basis for future research to develop robust and eco-friendly plant-derived fungicides that serve as a sustainable means to reduce fungus-driven agricultural losses.

Biocontrol of Fusarium oxysporum-infested Gastrodia elata Bl. by Lactobacillus curvatus 2768-VOCs and mechanism of inhibition

Zhiqiang Yi,Yusen Yue,Jianquan Kan,Zhirong Wang,Sameh Awad,Amel Ahmed Ibrahim,Muying Du

Gastrodia elata Bl. Is susceptible to infestation by the fungus Fusarium oxysporum , which can cause severe post-harvest diseases. This paper presents the findings of an investigation into the control and bacteriostatic mechanisms of Lactobacillus curvatus 2768-VOCs on F. oxysporum -infected G. elata . The results of both in vitro and in vivo tests demonstrated that L. curvatus 2768-VOCs exhibited inhibitory effects against F. oxysporum , with inhibition rates of 63.37% and 44.4%, respectively. The results of the in vivo test also demonstrated that the pre-fumigation approach exhibited superior efficacy compared to direct fumigation, with the former showing a preventive effect. Furthermore, it was discovered that VOCs disrupted the integrity of F. oxysporum cell membranes and altered their permeability, resulting in the leakage of cell contents. The fumigation caused damage to the mycelial structure, accompanied by a notable decline in the total lipid and ergosterol content, and an appreciable increase in MDA content. Moreover, the fumigation of VOCs markedly diminished the activities of ATPase and mitochondrial dehydrogenase, thereby disrupting the respiration of F. oxysporum and its energy metabolism. The VOCs were subjected to GC-MS analysis, which revealed that the relative content of acids was the highest, particularly formic acid and glyoxalic acid. The study offers a theoretical basis for the biological control of postharvest diseases in G. elata .

Confrontation assays and mycotoxin treatment reveal antagonistic activities of Trichoderma and the fate of Fusarium mycotoxins in microbial interaction

Ye Tian,Dianzhen Yu,Na Liu,Yan Tang,Zheng Yan,Aibo Wu

Mycotoxins are toxic fungal metabolites, contaminating cereal grains in field or during processing and storage periods. These environmental contaminants pose great threats to humans and animals’ health due to their toxic effects. Type A trichothecenes, fumonisins and fusaric acid (FA) are commonly detected mycotoxins produced by various Fusarium species. Trichoderma spp. are promising antagonists in agriculture for their activities against plant pathogens , and also regarded as potential candidates for bioremediation of environmental contaminants. Managing toxigenic fungi by antagonistic Trichoderma is regarded as a sustainable and eco-friendly strategy for mycotoxin control. However, the metabolic activities of Trichoderma on natural occurring mycotoxins were less investigated. Our current work comprehensively explored the activities of Trichoderma against type A trichothecenes, fumonisins and FA producing Fusarium species via co-culture competition and indirect volatile assays. Furthermore, we investigated metabolism of type A trichothecenes and FA in Trichoderma isolates. Results indicated that Trichoderma were capable of bio-transforming T-2 toxin, HT-2 toxin, diacetoxyscirpenol and neosolaniol into their glycosylated forms and one Trichoderma strain could bio transform FA into low toxic fusarinol. These findings proved that Trichoderma isolates could manage toxigenic Fusarium via direct competition and volatile-mediated indirect inhibition. In addition, these antagonists possess defensive systems against mycotoxins for self-protection, which enriches our understanding on the interaction mechanism of Trichoderma spp. on toxigenic fungus.

α-Glucosidase inhibitory effect of an anthraquinonoid produced by Fusarium incarnatum GDZZ-G2

Jiahe Fan,Chaoyi Lv,Zhizhou Li,Mengru Guo,Yichen Yin,Hui Wang,Wei Wang,Shiwei Sun

α-Glucosidase is the key enzyme on carbohydrate metabolism, and its bioactive inhibitors are supposed to be an effective therapeutic for type 2 diabetes mellitus. During our continuing study for discovering α-glucosidase inhibitors, a fungus GDZZ-G2 which is derived from a medicinal plant Callicarpa kwangtungensis Chun, exhibited significant inhibition on α-glucosidase. The strain was identified as Fusarium incarnatum by morphological and molecular methods. Further bioassay-guided fractionation result in six known secondary metabolites ( 1 - 6 ). All the compounds except 4 were isolated from F . incarnatum for the first time. Among them, an anthraquinonoid ( S )-1,3,6-trihydroxy-7-(1-hydroxyethyl)anthracene-9,10-dione (compound 1 ) exhibited strong inhibitory effect against α-glucosidase (IC 50 = 77.67 ± 0.67 μΜ), compared with acarbose (IC 50 = 711.8 ± 5 μΜ). An enzyme kinetics analysis revealed that compound 1 was an uncompetitive inhibitor. Besides, docking simulations predicted that compound 1 inhibited α-glucosidase substrate complex by binding Gln322, Gly306, Thr307, and Ser329 through hydrogen-bond interactions. Our findings suggested that compound 1 can be considered a lead compound for further modifications and the development of a new effective drug candidate in the treatment of type 2 diabetes mellitus.

Genome-Wide Identification and Functional Analysis of the bZIP Transcription Factor Family in Rice Bakanae Disease Pathogen, Fusarium fujikuroi

Kehan Zhao,Lianmeng Liu,Shiwen Huang

Fungal basic leucine zipper (bZIP) proteins play a vital role in biological processes such as growth, biotic/abiotic stress responses, nutrient utilization, and invasion. In this study, genome-wide identification of bZIP genes in the fungusFusarium fujikuroi, the pathogen of bakanae disease, was carried out. Forty-four genes encoding bZIP transcription factors (TFs) from the genome ofF. fujikuroi(FfbZIP) were identified and functionally characterized. Structures, domains, and phylogenetic relationships of the sequences were analyzed by bioinformatic approaches. Based on the phylogenetic relationships with the FfbZIP proteins of eight other fungi, the bZIP genes can be divided into six groups (A–F). The additional conserved motifs have been identified and their possible functions were predicted. To analyze functions of the bZIP genes, 11FfbZIPswere selected according to different motifs they contained and were knocked out by genetic recombination. Results of the characteristic studies revealed that theseFfbZIPs were involved in oxygen stress, osmotic stress, cell wall selection pressure, cellulose utilization, cell wall penetration, and pathogenicity. In conclusion, this study enhanced understandings of the evolution and regulatory mechanism of theFfbZIPsin fungal growth, abiotic/biotic stress resistance, and pathogenicity, which could be the reference for other fungal bZIP studies. Keywords:bZIP TF;bakanae disease;Fusarium fujikuroi;phylogenetic analysis;functional analysis

A Highly Sensitive and Rapid Enzyme Biosensor Based on AuNPs/CS@cMWCNTS for Detecting Fusarium Acid

Qiaoning Dang,Bolu Sun,Hongxia Shi,Quhuan Ma,Yuhong Liu,Haoye Zou,Xinlan Wang,Xuanxiu Da,Miao Zhou,Ying Lv,Lin Yang,Xiaofeng Shi

Fusarium acid (FSA) serves as a highly sensitive biomarker for the early warning of bulb rot in Lanzhou lily caused by Fusarium oxysporum infection. Realizing its highly sensitive, accurate, and rapid detection is crucial for the early warning and control of lily bulb rot. Herein, an enzyme biosensor based on the enzyme inhibition principle was developed for the rapid detection of FSA. Firstly, chitosan-functionalized carboxylated multi-walled carbon nanotubes (CS@cMWCNTS) and gold nanoparticles (AuNPs) were modified on a bare electrode by drop coating and electrochemical deposition methods, respectively. Subsequently, dopamine β-hydroxylase (DBH) was firmly immobilized on the electrode surface through the gold-sulfur bond. Because the activity of DBH can be inhibited by FSA, which leads to a noticeable change in the signal response during the conversion of dopamine (DA) to norepinephrine (NA). Under optimized experimental conditions, the sensor exhibits an excellent linear relationship in the concentration range of 1.00 × 10−5 μg ml−1 ∼ 1.00 × 103 μg ml−1 with the correlation coefficient of 0.9856 and the detection limit is 4.60pg ml−1. Additionally, the sensor showed good stability, repeatability (RSD 1.85%), and selectivity. The method was applied to analyze FSA in the extraction from Lanzhou lily with a recovery higher than 97.59% and RSD less than 1.383%. This method enables the highly sensitive and rapid detection of FSA in real samples, and provides scientific basis and technical support for early warning of diseases and accurate implementation of prevention and control strategies in crop planting and production.

Ero1-Pdi1 module-catalysed dimerization of a nucleotide sugar transporter, FonNst2, regulates virulence of Fusarium oxysporum on watermelon

Yizhou Gao,Xiaohui Xiong,Hui Wang,Jiajing Wang,Yan Bi,Yuqing Yan,Zhongye Cao,Dayong Li,Fengming Song

Summary Fusarium oxysporum f. sp. niveum ( Fon ) is a soil-borne fungus causing vascular Fusarium wilt on watermelon; however, the molecular network regulating Fon virulence remains to be elucidated. Here, we report the function and mechanism of nucleotide sugar transporters (Nsts) in Fon . Fon genome harbours nine FonNst genes with distinct functions in vegetative growth, asexual production, cell wall stress response and virulence. FonNst2 and FonNst3 are required for full virulence of Fon on watermelon and FonNst2 is mainly involved in fungal colonization of the plant tissues. FonNst2 and FonNst3 form homo- or hetero-dimers but function independently in Fon virulence. FonNst2, which has UDP-galactose transporter activity in yeast, interacts with FonEro1 and FonPdi1, both of which are required for full virulence of Fon . FonNst2, FonPdi1 and FonEro1 target to endoplasmic reticulum (ER) and are essential for ER homeostasis and function. FonEro1-FonPdi1 module catalyses the dimerization of FonNst2, which is critical for Fon virulence. Undimerized FonNst2 is unstable and degraded via ER-associated protein degradation in vivo . These data demonstrate that FonEro1-FonPdi1 module-catalysed dimerization of FonNst2 is critical for Fon virulence on watermelon and provide new insights into the regulation of virulence in plant fungal pathogens via disulfide bond formation of key pathogenicity factors.
