To counter the harmful impacts of agricultural chemicals on the environment and human health, there is an increasing demand for safe, eco-friendly, and potent plant-based biopesticides. In this study, we aimed to investigate the antimicrobial effects of ginger essential oil and selected volatile compounds (linalool, eugenol, citral, and cinnamaldehyde [CA]) againstFusarium oxysporumFOX-1. Minimum inhibitory concentrations (MICs) were determined using the mycelium growth inhibition method. The compound CA exhibited the most potent antifungal effect againstF. oxysporumFOX-1 and was selected for further investigation. After treatment with CA at 1/2 MIC or MIC, the spore germination ofF. oxysporumFOX-1 was significantly inhibited at 12 h. Furthermore, microscopic observation revealed that CA treatment resulted in the morphological degradation ofF. oxysporumFOX-1. CA destroyed the cell membrane integrity ofF. oxysporumFOX-1, increasing the relative conductivity and the leakage of intracellular protein, nucleic acids, and malondialdehyde, affecting the integrity and metabolism of the cell membrane. The effects were positively related to CA concentration. Additionally, in vivo experiments with rhizome sections showed that CA significantly reduced the pathogenicity ofF. oxysporumFOX-1. Overall, these findings provide evidence for the potential of using ginger essential oil components as fungicides, offering a basis for future research to develop robust and eco-friendly plant-derived fungicides that serve as a sustainable means to reduce fungus-driven agricultural losses.

Gastrodia elata Bl. Is susceptible to infestation by the fungus Fusarium oxysporum , which can cause severe post-harvest diseases. This paper presents the findings of an investigation into the control and bacteriostatic mechanisms of Lactobacillus curvatus 2768-VOCs on F. oxysporum -infected G. elata . The results of both in vitro and in vivo tests demonstrated that L. curvatus 2768-VOCs exhibited inhibitory effects against F. oxysporum , with inhibition rates of 63.37% and 44.4%, respectively. The results of the in vivo test also demonstrated that the pre-fumigation approach exhibited superior efficacy compared to direct fumigation, with the former showing a preventive effect. Furthermore, it was discovered that VOCs disrupted the integrity of F. oxysporum cell membranes and altered their permeability, resulting in the leakage of cell contents. The fumigation caused damage to the mycelial structure, accompanied by a notable decline in the total lipid and ergosterol content, and an appreciable increase in MDA content. Moreover, the fumigation of VOCs markedly diminished the activities of ATPase and mitochondrial dehydrogenase, thereby disrupting the respiration of F. oxysporum and its energy metabolism. The VOCs were subjected to GC-MS analysis, which revealed that the relative content of acids was the highest, particularly formic acid and glyoxalic acid. The study offers a theoretical basis for the biological control of postharvest diseases in G. elata .

The tubers of Bletilla striata were fermented with Fusarium avenaceum and Fusarium oxysporum . The total phenolic contents (TPCs) and antioxidant and antibacterial activities of the extracts from the fermented products were measured. Additionally, the extracts of both F. avenaceum and F. oxysporum fermented B. striata (FBS) were characterized by UV–vis spectroscopy, Fourier transform infrared (FT-IR) spectroscopy, and nuclear magnetic resonance (NMR) spectroscopy. The results showed that the F. avenaceum and F. oxysporum FBSs possessed higher TPCs and exhibited stronger antioxidant and antibacterial activities ( Bacillus subtilis ) in comparison with original material not co-fermented with microorganisms. Chemical characterization of the two extracts with UV–vis, FT-IR, and 1 H NMR implied that some of the phenolic glycosides may be deglycosylated and that bibenzyl compounds and saccharides may be metabolized in both FBS systems. Microbial fermentation enhances the TPC as well as the antioxidant and antibacterial activities of B. striata . The present study suggests that F. avenaceum and F. oxysporum fermentations were effective in B. striata processing.

Summary Fusarium oxysporum f. sp. niveum ( Fon ) is a soil-borne fungus causing vascular Fusarium wilt on watermelon; however, the molecular network regulating Fon virulence remains to be elucidated. Here, we report the function and mechanism of nucleotide sugar transporters (Nsts) in Fon . Fon genome harbours nine FonNst genes with distinct functions in vegetative growth, asexual production, cell wall stress response and virulence. FonNst2 and FonNst3 are required for full virulence of Fon on watermelon and FonNst2 is mainly involved in fungal colonization of the plant tissues. FonNst2 and FonNst3 form homo- or hetero-dimers but function independently in Fon virulence. FonNst2, which has UDP-galactose transporter activity in yeast, interacts with FonEro1 and FonPdi1, both of which are required for full virulence of Fon . FonNst2, FonPdi1 and FonEro1 target to endoplasmic reticulum (ER) and are essential for ER homeostasis and function. FonEro1-FonPdi1 module catalyses the dimerization of FonNst2, which is critical for Fon virulence. Undimerized FonNst2 is unstable and degraded via ER-associated protein degradation in vivo . These data demonstrate that FonEro1-FonPdi1 module-catalysed dimerization of FonNst2 is critical for Fon virulence on watermelon and provide new insights into the regulation of virulence in plant fungal pathogens via disulfide bond formation of key pathogenicity factors.

Fusarium is a soil-borne pathogen that poses a serious threat to the quality and yield of hundreds of crops worldwide, particularly tobacco production. Using metabolomics technology, we investigated natural metabolites from disease-conducting soil (DCS) and disease-suppressing soil (DSS) of tobacco rhizosphere as fungicides to control tobacco Fusarium wilt (TFW), which is mainly caused by Fusarium oxysporum. Furthermore, the antifungal mechanisms of these natural metabolites were preliminarily elucidated through various assessments, including antifungal activity determination, chemotaxis effect tests, PI staining experiments, and measurements of extracellular conductivity and protein content. Metabolomics results showed that the DCS with three different disease grades (G1, G5 and G9 groups) had significantly higher levels of 15, 14 and 233 differential rhizosphere metabolites (DRMs) and significantly lower levels of 72, 152 and 170 DRMs compared to the DSS (G0 group). According to KEGG pathway analysis, these DRMs were found to be enriched in the caffeine metabolism, biosynthesis of phenylpropanoids, galactose metabolism and tyrosine metabolism, etc. Linustatin, scopoletin and phenylpropiolic acid were picked out from these DRMs and found to have suppressive activity against F. oxysporum through correlation analysis and antifungal experiments. The three DRMs showed strong inhibitory effects on the growth and spore germination of F. oxysporum at concentrations of 0.5 mM or higher in each test period. Furthermore, F. oxysporum showed a phobotaxis effect against these three DRMs at concentrations as low as 0.25 mM. Finally, we found that the three DRMs had an inhibitory effect on F. oxysporum by destroying the integrity of the cell membrane and increasing the membrane permeability of F. oxysporum. This study firstly reports the inhibition activity of phenylpropiolic acid and linustatin on F. oxysporum, providing a practical and environmentally friendly method for biocontrol of TFW by using natural fungicides.

The volatile organic compounds (VOCs) produced by antagonistic microbes have great antifungal potential against soil-borne fungal pathogens. The VOCs produced by Paenibacillus polymyxa strain WR-2 in the presence of root exudates and organic fertilizer were identified and their effects on the growth and spore germination of Fusarium oxysporum f. sp. niveum were evaluated. The VOCs produced by WR-2 inhibited the growth of F. oxysporum by 38%, 36% and 40% in agar medium, sterilized soil and natural soil, respectively. This inhibitory effect was increased to 60%, 58% and 64% with the addition of organic fertilizer in agar medium, sterilized soil and natural soil, respectively. The addition of root exudates did not affect the production of antifungal VOCs by WR-2. The VOCs produced by WR-2 completely inhibited the germination of F. oxysporum spores. Out of 42 identified VOCs, seven VOCs; benzothiazole, benzaldehyde, undecanal, dodecanal, hexadecanal, 2-tridecanone and phenol were found to inhibit the growth of F. oxysporum . The results of these experiments suggest another significance of using organic fertilizer as a carrier material with the biocontrol agents to control soil-borne fungal pathogens.

Dendritic mesoporous silica nanoparticles (DMSNs) have emerged as promising nanocarriers due to their unique three-dimensional structure and tunable pore characteristics. This study investigates the potential of DMSNs to deliver thyme essential oil (TEO) for enhanced antifungal activity against Fusarium oxysporum (F. oxysporum), a major plant pathogen. DMSNs were successfully synthesized and characterized, followed by the encapsulation of TEO within their porous structure. The resulting TEO@DMSNs composites exhibited significant antifungal activity against F. oxysporum, with inhibition rates reaching 70%, indicating an effective crop protection with a dose-dependent effect. The enhanced antifungal efficacy of TEO@DMSNs compared to free TEO is attributed to the sustained release of TEO and the synergistic effect of the nanocarrier itself. In-depth mechanism investigations revealed that TEO@DMSNs likely disrupted the fungal cell membrane, leading to leakage of cellular contents and ultimately cell death. Moreover, DMSNs and TEO@DMSNs were found to be safe for plant growth, demonstrating their potential as environmentally friendly antifungal agents. This study provides valuable insights into the design and development of advanced nanocarriers for targeted drug delivery and disease management in agriculture.

The infection caused by Fusarium oxysporum severely threatens cotton production. Herein, carvacrol-chitosan nanoparticles (CS@CAR) are facilely fabricated using tripolyphosphate as a crosslinker. When the initial mass ratio of chitosan to carvacrol is 1:0.4, the nanoparticles show the best stability and drug loading. Morphological analysis reveals that the CS@CAR nanoparticles possesses a uniform spherical structure with an average diameter of approximately 150 nm. About 51.23 % of total CAR is released within the 48 h in the simulated fungal infection environment. The contact angle of drug-loaded particles decreases by 12.2 and 6.6 ° compares with that of pure water and unloaded particles. The fluorescence intensity of leaves before and after rain erosion is 1.3 times and 1.2 times that of the pure water control group by fluorescence imaging analysis. The bacteriostatic activity of drug-loaded nanoparticles is 6–7 times that of non-drug-loaded nanoparticles. The contents of nucleic acid, protein, and malondialdehyde leaked by spores after treatment with drug-loaded particles are 1.4, 1.5 and 2.3 times those in the non-drug-loaded particle treatment group, respectively. The CS@CAR entirely prevents Fusarium oxysporum from infecting cotton leaves in spore inoculation experiments. The CS@CAR nanoparticles are expected to be used to prevent and control the infection of Fusarium oxysporum on cotton.

Due to their impressive pharmaceutical activities and safety, prenylated flavonoids have a high potent to be applied as medicines and nutraceuticals. Biocatalysis is an effective technique to synthesize prenylated flavonoids. The major concern of this technique is that the microbe-derived prenyltransferases usually have poor regiospecificity and generate multiple prenylated products. In this work, a highly regiospecific prenyltransferase (FoPT1) was found from Fusarium oxysporum . It could recognize apigenin, naringenin, genistein, dihydrogenistein, kampferol, luteolin and hesperetin as substrates and only 6- C -prenylated flavonoids were detected as the products. The catalytic efficiency of FoPT1 on flavonoids was in a decreasing order with hesperetin >naringenin >apigenin >genistein >luteolin >dihydrogenistein >kaempferol. Chalcones, flavanols and stilbenes were not active when acting as the substrates. 5,7-Dihydroxy and 4-carbonyl groups of flavonid were required for the catalysis. 2,3-Alkenyl was beneficial to the catalysis whereas 3-hydroxy impaired the prenylation reaction. Docking studies simulated the prenyl transfer reaction of FoPT1. E186 was involved in the formation of prenyl carbonium ion. E98, F89, F182, Y197 and E246 positioned apigenin for catalysis.

Long-term monoculture of ginger triggers Fusarium wilt, a disease caused by Fusarium oxysporum f. sp. zingiberi ( Foz ). However, the interaction between autotoxins and pathogens remains poorly understood. This study examined the allelopathic effects of four autotoxins, syringic acid, coumarin, ferulic acid, and 7-hydroxycoumarin, on the growth, reproduction, and virulence traits of Foz . These compounds were previously identified as inhibitors of ginger growth and enzyme activity. The results revealed that the responses of Foz varied, likely because of the structural differences among the autotoxins. Syringic acid significantly inhibited mycelial growth, sporulation, and spore germination, while markedly enhancing the activity of cell wall-degrading enzymes and mycotoxin synthesis, as evidenced by the upregulation of FUB3 and FUB9 . Coumarin demonstrated a pronounced inhibitory effect on biomass production while concurrently stimulating sporulation and mycotoxin synthesis, as indicated by the upregulation of FUB6 and FUB9 . Ferulic acid treatment reduced the activity of cell wall-degrading enzymes and spore germination, while upregulating sporulation and mycotoxin synthesis. A similar pattern was observed with 7-hydroxycoumarin, which exerted a strong inhibitory effect on mycelial growth, biomass production, mycotoxin synthesis, and FUB gene cluster expression, except for FUB1 and FUB3 expression. In conclusion, four autotoxins exhibited diverse defensive roles against Fusarium wilt, whereas the pathogen enhanced its pathogenicity to counter host regulations. This study provides the first evidence of an interaction between ginger autotoxins and Foz .

Tomato wilt disease, caused by Fusarium oxysporum f. sp. lycopersici , is a major soil-borne disease affecting tomato cultivation. In this study, two strains exhibiting significant control effects on F. oxysporum f. sp. lycopersici and tomato growth-promoting effects were isolated from the rhizosphere. Through physiological and biochemical identification, as well as 16S rRNA identification, these strains were identified as Enterobacter quasihormaechei L33 and Klebsiella oxytoca TL2, respectively. And the two strains of bacteria were confirmed to have no antagonistic effect through antagonistic compatibility tests. Notably, when these strains were mixed in a ratio of 5:1, they demonstrated remarkable antifungal activity. The mixed strains induced mycelium swelling, necrosis, and rupture in F. oxysporum f. sp. lycopersici , resulting in a 56.65 % inhibition of mycelial fresh weight. Additionally, they inhibited spore production with an inhibition rate of 787.5 %, and the inhibition rate of the lipopeptide produced reached 93.80 % at 50 mg/mL. Furthermore, in potting experiments, the mixed strains reduced the disease incidence by 36.25 % after 8 weeks. Additionally, compared to the control group, the mixed bacterial strain group enhanced fresh weight, plant height, chlorophyll a and b levels, and root activities by 96.31 %, 18.51 %, 184.48 %, 53.03 %, and 44.31 %, respectively. In addition, mixed strains can inhibit the cell wall degrading enzymes produced by Fusarium oxysporum f sp., induce plant defense enzymes, including increased activity of peroxidase, catalase, and superoxide dismutase, and reduce the decrease in malondialdehyde levels. The mixed strains could colonize tomato plant roots. Overall, these findings suggest a potential biocontrol agent deriving from the mixed strains against tomato wilt disease.

Microbial transformation of glycinonitrile into glycine by nitrile hydrolase is of considerable interest to green chemistry. A novel fungus with high nitrile hydrolase was newly isolated from soil samples and identified as Fusarium oxysporum H3 through 18S ribosomal DNA, 28S ribosomal DNA, and the internal transcribed spacer sequence analysis, together with morphology characteristics. After primary optimization of culture conditions including pH, temperature, carbon/nitrogen sources, inducers, and metal ions, the enzyme activity was greatly increased from 326 to 4,313 U/L. The preferred carbon/nitrogen sources, inducer, and metal ions were glucose and yeast extract, caprolactam, and Cu 2+ , Mn 2+ , and Fe 2+ , respectively. The maximum enzyme formation was obtained when F. oxysporum H3 was cultivated at 30 °C for 54 h with the initial pH of 7.2. There is scanty report about the optimization of nitrile hydrolase production from nitrile-converting fungus.
