Proline-rich protein PRPL1 enhances Panax notoginseng defence against Fusarium solani by regulating reactive oxygen species balance and strengthening the cell wall barrier

Linlin Su,Wenyun Li,Xiaohua Chen,Pengcheng Wang,Diqiu Liu

The root rot mainly caused by Fusarium solani is a bottleneck in the cultivation of Panax notoginseng . In this study, we reported a gene encoding a plant cell wall structural protein, P. notoginseng proline-rich protein (PnPRPL1), whose transcription was upregulated by F. solani and induced by some hormone signals. The PnPRPL1 recombinant protein significantly inhibited the growth and conidial germination of the root rot pathogens. Downregulation of PnPRPL1 by RNA interference (RNAi) in P. notoginseng leaves increased the susceptibility to F. solani , whereas overexpression of PnPRPL1 in tobacco ( Nicotiana tabacum ) enhanced the resistance to F. solani . Compared with wild-type tobacco, the PnPRPL1 -overexpressing transgenic tobacco had higher reactive oxygen species (ROS)-scavenging enzyme activities, lower ROS levels, and more lignin and callose deposition. The opposite results were obtained for the P. notoginseng expressing PnPRPL1 RNAi fragments. Furthermore, the PnPRPL1 promoter transcription activity was induced by several plant hormones and multiple stress stimuli. In addition, the transcription factor PnWRKY27 activated the expression of PnPRPL1 by directly binding to the promoter region. Thus, PnPRPL1 , which is positively regulated by a WRKY transcription factor, encodes an antimicrobial protein that also mediates ROS homoeostasis and callose/lignin deposition during the response to F. solani infection.

Antifungal Mechanism of 4-(4-Hydroxyphenyl)-2-butanone Against Fusarium solani

Yifan Chen,Li Zhang,Yan Chen,Wenpei Ge,Hongling Wang,Zhanying Xue,Xinran Cui,Xin Wang,Aimei Liao,Yuansen Hu,Na Liu

Fusarium solani , a widely distributed plant pathogenic fungus, poses a significant threat to various crops due to its complex pathogenic mechanisms and being difficult to control. In this study, LC–TOF–MS analysis identified 4-(4-hydroxyphenyl)-2-butanone (RK) as a metabolite detected in Bacillus amyloliquefaciens strain M1, and a commercially available RK standard was subsequently used to evaluate its antifungal activity against F. solani . Antifungal assays demonstrated that RK effectively suppressed fungal growth. Further physiological and biochemical assays confirmed that RK disrupts the cell membrane and mitochondrial function, leading to intracellular reactive oxygen species (ROS) accumulation. This study offers new perspectives on the antifungal mechanism of RK and offers theoretical support for the development of innovative agricultural disease management strategies.

Immobilization of Fusarium solani Cutinase onto Magnetic Genipin-Crosslinked Chitosan Beads

Zhanyong Wang,Tingting Su,Jingjing Zhao

Genipin was used as a crosslinking agent to prepare magnetic genipin-crosslinked chitosan beads, which were then used as a carrier for immobilizing recombinant cutinase fromFusarium solani(FSC) to obtain immobilized FSC. The optimal temperature for the immobilized FSC was 55 °C, which was 5 °C higher than that of the free enzyme, whereas its optimal pH was increased from 8.0 to 9.0; this indicates that the immobilized FSC had improved pH and thermal stability. After repeated use for 10 cycles, the activity of the immobilized FSC remained at more than 50%; after being stored at 4 °C for 30 days, its activity was still approximately 88%. We also found that the Km of the immobilized FSC was higher than that of the free enzyme. These results indicate that the performance of FSC was improved after immobilization, which is an important basis for the subsequent application of FSC in industrial production. Keywords:immobilization;cutinase;chitosan;genipin

Cepharanthine Inhibits Fusarium solani via Oxidative Stress and CFEM Domain-Containing Protein Targeting

Yuqing Wang,Zenghui Yang,Jingwen Xue,Yitong Wang,Haibo Li,Zhihong Wu,Yizhou Gao

Cepharanthine (CEP) is a natural bisbenzylisoquinoline alkaloid known for its antibacterial, antiviral, and anti-inflammatory activities. Its antifungal effect, however, has not been well studied. In this work, we used machine learning-based virtual screening with Random Forest, Neural Network, and Support Vector Machine models to identify potential inhibitors ofFusarium solani. CEP was selected as a candidate and tested experimentally. The results showed that it inhibited the growth ofFusarium solani,Fusarium proliferatum,Fusarium oxysporum,Alternaria alternata, andBotrytis cinerea. It also reduced the sporulation and spore germination ofFusarium solaniand disrupted its redox balance. Transcriptome analysis showed changes in gene expression related to basic metabolic pathways. Molecular docking suggested that CEP binds to the FsCFEM1 protein, and molecular dynamics simulations confirmed stable binding, with key roles for residues THR748 and LEU950. These results suggest that CEP is a potential bio-based antifungal agent and provide novel insights into its mechanism againstFusarium solani.

Fungal colonization and candidate metabolite-mediated toxicity of Fusarium solani KMZW-1 against adult Bactrocera dorsalis

Huiqin Li,Zhiyi Zhao,Mingqi Wu,Chunlan Shi,Tao Zhu,Xi Gao,Chun Xiao,Mehboob Hussain,Deqiang Qin,Guoxing Wu,Junfu Yu

This study examined the interaction between Fusarium solani KMZW-1 and adult Bactrocera dorsalis, with emphasis on cuticular colonization, adult-stage pathological effects, and candidate metabolite-mediated toxicity. KMZW-1 showed Fusarium-like colony and conidial morphology and colonized mainly tarsal setae, joints, and recessed cuticular regions, where fungal conidia and hyphae progressively accumulated over time. Fungal treatment reduced adult survival in a concentration-dependent manner (P < 0.001) and lowered egg production per female, likely due to reduced male survival and fewer mating opportunities, whereas egg hatchability, larval pupation, and adult emergence of offspring were not significantly affected (P > 0.05). Untargeted LC-MS/MS profiling annotated 3073 metabolites, and candidate secondary metabolites were prioritized using pathway assignment, CAS-confirmed identity, normalized peak-area abundance, reported biological activity and commercial availability. Among coumarin, umbelliferone and podophyllotoxin, podophyllotoxin showed the lowest 24 h LC50 values against adult flies, at 24.33 μg mL−1 for females and 21.03 μg mL−1 for males. These results indicate that KMZW-1 produces a reproducible surface-colonization-associated adult-stage phenotype in B. dorsalis. Podophyllotoxin should be interpreted as a prioritized candidate with confirmed topical toxicity, not as a confirmed in vivo virulence determinant. Further work is required to confirm metabolite identity with authentic standards, quantify metabolite production during infection, determine internal invasion dynamics, and evaluate mode of action and non-target safety.

Antifungal activity and mechanism of chitosan against Fusarium solani caused ginger soft rot during postharvest storage

Lingling Zhang,Ying Yang,Yongxing Zhu,Haijun Hu,Qie Jia,Chong Sun,Xuedong Zhu,Yiqing Liu

Postharvest rot caused by Fusarium solani ( F . solani ) results in enormous economic loss to the global vegetables industry. Chitosan has been shown to be effective in control postharvest diseases of vegetables, but the specific mechanism of chitosan on F . solani has not been studied. In this study, we evaluated antifungal activity of chitosan against F . solani caused soft rot in vitro and in vivo. Antifungal assay results showed that chitosan significantly inhibited the mycelial growth and spore germination of F . solani . Simultaneously, chitosan showed disruptive activity on the mycelial morphology and cell membrane, which were achieved by decreasing intracellular sugars, proteins and lipids content. Furthermore, chitosan treatment significantly decreased tricarboxylic acid (TCA)-related enzymes activity , reducing the number of metabolites in the TCA cycle . UPLC-Triple-TOF/MS-based metabolomics analysis showed that chitosan interferes with fungal metabolic pathways, mainly related to energy metabolism, lipid metabolism and sugar metabolism. Exogenous α-D-glucose and linoleic acid treatment promoted mycelia growth, verifying the sugar catabolism and TCA cycle was involved in the inhibition of chitosan against F . solani . The in vivo study showed that chitosan treatment effectively reduced disease rate and severity of postharvest soft rot through activating the biochemical defense response and modulating the expression of phytopathogenic responses LysM genes. These results contribute to explore the underlying mechanisms of chitosan against F . solani .

Design, Synthesis and Bioactivity of Benzyl Propiolates with Broad-Spectrum Inhibition Activity on Phytopathogenic Fungi

Ruo-Fei Bai,Yi-Wei Wang,Shan-Shan Yang,Yu-Hao Zhang,Bo-Hang Zhou,Cong-Wei Zhou,Le Zhou

As part of our continuing research on propiolic acid derivatives, a series of benzyl propiolate derivatives and analogues were designed, synthesized, and investigated for inhibition activity in vitro and in vivo on phytopathogenic fungi, structure–activity relationship (SAR) and action mechanism. The results showed that most of the compounds had potent and broad-spectrum antifungal activity in vitro at 50 μg/mL. Among the compounds, 2-bromobenzyl propiolate (3h) displayed the highest comprehensive activity with total activity index (TAI) of 4.57 against all the eight test fungi followed by 3-fluorobenzyl propiolate (3c) (TAI = 4.53), superior to positive fungicides thiabendazole (TAI < 4.12) and/or azoxystrobin (TAI < 2.90). Relative to the other fungi, Fusarium solani and Fusarium graminearum revealed higher total susceptibility indexes (TSI) of 11.1 and 8.98, respectively, for all compounds. 3h and 3g (4-chlorobenzyl propiolate) gave the smallest EC50 values of 0.86 and 0.67 μg/mL against F. solani and F. graminearum, respectively, superior or comparable with thiabendazole and azoxystrobin. As a representative, 3h at 200 μg/mL displayed 100% protection on F. solani infections on potatoes over 7 days and high safety for plant growth. In antifungal mechanism, 3h was able to change mycelial morphology, destroy the structures of both hypha and cell membrane, increase the intracellular ROS level, decrease the intracellular mitochondrial membrane potential level, and change the permeability of the cell membrane. The SAR showed that ethynyl is a key group for the activity, and the presence of halogen atoms at the 2-site of the benzene ring can significantly improve the activity in most cases. Thus, benzyl propiolate can be considered a novel antifungal lead compound, while 3h can be considered a promising fungicide candidate to develop new agricultural fungicides.

Influence of monomer composition of poly(butylene succinate-co-hexamethylene succinate)s on their enzymatic hydrolysis by Fusarium solani cutinase

Ting Hu,Yujun Wang,Jingwei Lin,Zhanyong Wang

Poly(butylene succinate) (PBS), poly(hexamethylene succinate) (PHS) and three kinds of poly(butylene succinate-co-hexamethylene succinate)s (P(BS-co-HS)s) were enzymatically hydrolyzed by Fusarium solani cutinase. The results showed that the characteristics of the enzymatic hydrolysis of these polyesters were mainly affected by crystallinity, thermal properties and the BS/HS ratio. The enzymatic hydrolysis rates of the polyesters are as follows: P(BS-co-HS52) ≈ P(BS-co-HS71) > PHS > P(BS-co-HS32) > PBS. Furthermore, with increasing HS content, both the degree of crystallinity (Xc) and melting temperature (Tm) of the polyesters first decreased and then increased. P(BS-co-HS52) and P(BS-co-HS71) had the lowest Xc and the lowest Tm, thus had the highest hydrolysis rate; this shows that the hydrolysis rate is affected by Xc and Tm. The results also showed that BS/HS ratio could affect the physical properties and degradability of polyesters. Thus, it is possible to prepare polyesters with various physical properties and degradability for different applications by adjusting BS/HS ratio. The crystalline and amorphous regions of the polyesters were both hydrolyzed, during which parts of the crystalline regions were converted into the amorphous regions. Finally, we found that the crystal structure and thermal stability of the polyesters were not affected by the enzymatic hydrolysis.

Targeting the Snf1 kinase by cinnamaldehyde induces glucose starvation and oxidative stress in Fusarium solani for the control of sweet potato postharvest decay

Chao Pan,Fajuan Wang,Yanzi Chu,Yahui Xue,Yan Ye,Kunlong Yang,Jinfeng Hua,Ling Shen,Jun Tian

Fusarium root rot, caused by Fusarium solani , is one of the major postharvest diseases of sweet potato. Cinnamaldehyde (CA), a natural product from cinnamon bark, has demonstrated promising antifungal efficacy; however, its mechanism of action, especially regarding fungal sugar metabolism, remains poorly understood. CA above 0.375 g L −1 inhibited F. solani mycelial growth, induced glucose deprivation, and triggered the accumulation of reserve carbohydrates (trehalose and glycogen). Moreover, CA promoted the accumulation of reactive oxygen species and hydrogen peroxide (H 2 O 2 ), protein carbonylation, and DNA fragmentation, indicating that CA caused oxidative damage and led to apoptotic cell death in F. solani . Notably, CA suppressed the function of the metabolic regulator sucrose non-fermenting protein kinase 1 (Snf1) by disrupting its transcription and translation. Genetic knockout of Snf1 increased the inhibition rate of mycelial growth, amplified intracellular glucose scarcity, promoted the accumulation of reserve carbohydrates, accelerated H 2 O 2 accumulation and protein oxidative damage, and ultimately exacerbated the extent of cell apoptosis. Complementarily, activation of Snf1 attenuated the antifungal effect of CA. Moreover, CA reduced rot incidence and better preserved nutritional quality (soluble sugars and starch), particularly in sweet potato roots infected with Snf1 -deletion mutants. These findings reveal that CA exerts its antifungal effect by disrupting Snf1-mediated glucose supply and inducing oxidative stress in F. solani to control sweet potato postharvest decay. This study provides novel insights into the antifungal mechanism of plant essential oils and supports the potential application of CA as an eco-friendly preservative for controlling postharvest spoilage in carbohydrate-rich fresh produce.
