Chemical and biological characterization of naturally aged GCP by using a rigorous sample collection protocol

Meng-ling Peng,Ying-ying Zhan,Li-fen Cao,Anastassiya V. Gadetskaya,Kai-jing Yu,Jing Zhang,Xiao-hui Qiu,Zhi-hai Huang,Wen Xu

Naturally aged GCP (peel of Citrus reticulata ‘ChaChi’) is a traditional medicine and food ingredient in China. Extensive studies have explored how the aging process affects the chemical and biological properties. In the present study, we used a rigorous sample collection protocol in which GCP were annually collected from a single citrus orchard and aged for 1 to 6 years in a sample warehouse for natural aging. A global chemical investigation was conducted using GC–MS and UHPLC-Orbitrap-MS, identifying 37 volatile and 112 non-volatile chemicals. Volatile terpenes and flavonoid glycosides metabolized into other constituents during aging via oxidation and hydrolysis. The contents of 55 major targets and pseudotargeted metabolomics analysis revealed distinct variation trends in polymethoxyflavones (PMFs) and limonoids. Antioxidant assays of volatile and non-volatile extracts indicated that four-year GCP samples exhibit the best antioxidant activity. The accumulation of terpenoid alcohols and sesquiterpenes, but not terpenes, presented significant positive correlations with antioxidant activity. For non-volatile constituents, PMFs and limonoids displayed significant positive correlations with antioxidant activity. RAW 264.7 cell test showed that all samples demonstrated moderate anti-inflammatory effects, and total PMFs correlated positively with anti-inflammatory activity. This study enhances understanding of the aging mechanism of GCP and establishes a rationale for its practical application.

Biomimetic rhCOL17-P334 Conjugate for Enhanced Wound Healing

Yijie Lou,Chunyan Dai,Shuting Feng,Rijuan Jin,Shuangshuang Liu,Zhanyi Zhou,Minjun Wang,Yang Yang,Guoqiao Xu,Jue Hou,Jiayi Chen,Guping Tang,Nan Wang,Hongzhen Bai,Zhe Chen

Wound healing remains a significant global health challenge, affecting millions annually and imposing substantial economic burdens. Most commercially available biomaterials for wound management primarily address external symptoms, including hemostasis, exudation, scarring, and infection. Advanced biomaterials derived from endogenous molecules aim to better replicate the native wound microenvironment, promoting enhanced repair. Since wounds frequently occur on exposed skin, which is vulnerable to UVA radiation and requires protective yet invisible materials, traditional wound care products often lack these essential features. Inspired by natural UV protection mechanisms, a novel bioscaffold is developed using recombinant human collagen XVII (rhCOL17) crosslinked with porphyra-334 (P334) to improve wound healing under UVA exposure. The resulting rhCOL17-P334 conjugate integrates extracellular matrix (ECM)-like cues with UVA-shielding properties provided by P334. This conjugate is used to construct a transparent, injectable hydrogel combining gelatin methacryloyl (GelMA) and rhCOL17-P334 (GCP). GCP significantly inhibits UVA-induced fibroblast senescence and improves wound healing outcomes by targeting integrin α6β4 through rhCOL17. Its transparency facilitates convenient wound monitoring while also addressing the aesthetic requirement for invisibility. By combining UVA shielding with wound repair capabilities, GCP presents a promising platform for advanced wound management.

Discrimination of Citri reticulatae pericarpium (CP) and Citrus reticulata 'chachi' (GCP): Focus on HPTLC, UHPLC techniques combined with machine learning and content differences of three specific flavonoids

Yu-shi Huang,Ya-ling An,Wen-jie Zhao,Chun-qian Song,Zi-jun Tang,Lin Feng,Zhen-wei Li,Xiao-kang Liu,De-an Guo

The global consumption of Citri reticulatae pericarpium (Chenpi, CP) and Citrus reticulata 'chachi' (Guangchenpi, GCP) has been experiencing a steady increase, driven by its extensive applications in healthcare, flavoring, and therapeutic fields. While the two varieties exhibit analogous macroscopic characteristics and therapeutic efficacies, rigorous market differentiation and identification are imperative owing to their distinct botanical origins, qualitative specifications, and especially significant price discrepancy. A comprehensive strategy integrating HPTLC and UHPLC analyses with machine learning was implemented in this research to discriminate between CP and GCP and to conduct a comparative content analysis of three characteristic flavonoids in the two materials. The discrimination results of eight models upon HPTLC data reveal that all models except Tree achieved 100% discrimination accuracy. (2S)-Narirutin, Nobiletin, and Tangeretin, three compounds contributed to the differentiation were quantified and exhibited distinct content differences in CP and GCP samples. This study could provide simple and scientific guidance for the authentication of the two varieties.
