Structural characterization and in vitro hypoglycemic activity of a glucan from Euryale ferox Salisb. seeds

Wen-Na Zhang,Ri-Na Su,Li-Li Gong,Wei-Wei Yang,Juan Chen,Rui Yang,Ya Wang,Wen-Juan Pan,Yong-Ming Lu,Yan Chen

In this research, a polysaccharide fraction (EFSP-1) was obtained from the seeds of Euryale ferox Salisb. by DEAE sepharose FF and Superdex ™ 75 gel chromatography. The average molecular weight (Mw) of EFSP-1 was 8.75 kDa. Monosaccharides composition analysis indicated that EFSP-1 was a glucan. The structure of EFSP-1 was characterized by analysis of FT-IR, GC–MS and NMR, which indicated that the backbone of EFSP-1 was mainly composed of (1→4)- α -D-Glc p with branches substituted at O-6 and terminated with T- α -D-Glc p . Moreover, the hypoglycemic effect of EFSP-1 was investigated by establishing insulin resistance HepG2 and 3T3-L1 cells. The results showed that EFSP-1 could increase glucose consumption by up-regulating the expression of GLUT-4 via activating PI3K/Akt signal pathway in IR cells. Hence, EFSP-1 could be a potential functional food to ameliorate insulin resistance for diabetes therapy.

Hypoglycemic Effects and Mechanisms of Polyphenols From Myrica rubra Pomace in Type 2 Diabetes (db/db) Mice

Guoli Chang,Siyi Tian,Xinyu Luo,Yannan Xiang,Chenggang Cai,Ruiyu Zhu,Haiying Cai,Hailong Yang,Haiyan Gao

The Myrica rubra pomace polyphenols (MRPP) were used to study their hypoglycemic effects and mechanisms using Type 2 diabetes (T2D) (db/db) mice as the model. The results showed that the oral administration of MRPP (low-dose myricitrin, 50 mg/kg BW by gavage; high-dose myricitrin, 250 mg/kg BW by gavage; and Myrica rubra pomace, 500 mg/kg BW by gavage) for 4 weeks significantly reduced fasting blood glucose, glycated serum protein, serum insulin, and insulin resistance index in mice, as well as the dyslipidemia in mice was improved. MRPP was able to improve the structural morphology of hepatocytes and pancreatic β-cells in diabetic mice. Its hypoglycemic mechanism may involve the upregulation of GLUT-4 and IRS-1 genes expression in the PI3K and AMPK signaling pathways, downregulation of GSK-3β, AMPK, PI3K, and AKT genes expression, and enhancement of the activity of enzymes related to glycogen synthesis and glucose metabolism. Research findings on 16S rRNA suggest that MRPP has the ability to alter the composition of the gut microbiota, impede the growth of harmful bacteria, and foster the growth of beneficial bacteria. Therefore, MRPP was capable of reshaping the specific gut microbial community and supporting its application as a novel supplement in functional foods for the treatment of T2D.

Luteolin improves myocardial cell glucolipid metabolism by inhibiting hypoxia inducible factor-1α expression in angiotensin II/hypoxia-induced hypertrophic H9c2 cells

Jia Wang,Tian Gao,Feng Wang,Jie Xue,Hua Ye,Meilin Xie

Luteolin , a natural flavonoid , can attenuate hepatic lipid accumulation and insulin resistance in obese mice . Therefore, we hypothesized that luteolin may also improve the abnormal glucolipid metabolism of hypertrophic myocardial cells. This study aimed to investigate the effect and possible molecular mechanisms of luteolin. Hypertrophic H9c2 cells were induced by angiotensin II/hypoxia and simultaneously treated with 2 to 8 μg/mL luteolin for 24 h. Luteolin might dose-dependently decrease intracellular total protein, atrial natriuretic peptide , and free fatty acid levels , and increase supernatant glucose levels . Western blot assay showed that luteolin could inhibit the expressions of intracellular hypoxia inducible factor-1α (HIF-1α) and glucose transporter-4 (GLUT-4) proteins, and increase the expressions of intracellular peroxisome proliferator-activated receptor α (PPARα), carnitine palmitoyltransferase-1A (CPT-1A), and pyruvate dehydrogenase kinase-4 (PDK-4) proteins. These findings demonstrate that luteolin can improve abnormal glucolipid metabolism in angiotensin II/hypoxia-induced hypertrophic H9c2 cells, and its mechanisms are related to the inhibition of HIF-1α expression and subsequent modulation of PPARα-mediated target genes, including CPT-1A, PDK-4, and GLUT-4.

Extraction, purification, chemical characterization, and in vitro hypoglycemic activity of polysaccharides derived from Rosa laevigata Michx.

Shuai Chen,Liang Wang,Shuang Rong,Yuyuan Duan,Huizhu Wang

This study aimed to optimize the extraction process of polysaccharides from Rosa laevigata Michx. (RLMP) using an ultrasound-microwave-assisted method and investigate its in vitro hypoglycemic activity. Key factors affecting RLMP yield were identified using a Plackett–Burman design, followed by a Box–Behnken design and response–surface methodology, to determine the optimal extraction conditions. RLMP was purified using DEAE-52 cellulose, yielding two homogeneous fractions: RLMP-1 and RLMP-2. Monosaccharide composition was analyzed by gas chromatography, and structural characterization of RLMP, RLMP-1, and RLMP-2 was performed using FT-IR, SEM, and TEM. Methylation analysis and NMR were used to elucidate the sugar-chain structure of RLMP-1. In vitro hypoglycemic activity analysis showed that RLMP improved the glucose consumption and glycogen synthesis and enhanced the activities of pyruvate kinase and hexokinase in IR-HepG2 cells. Moreover, RLMP significantly increased the activities of antioxidant enzymes, such as CAT, SOD, and GSH-Px and decreased those of ROS and MDA. Western blotting analysis confirmed that RLMP enhances glucose and lipid metabolism and reduces oxidative stress by activating the PI3K/Akt/GLUT-4 signaling pathway, thereby exerting its hypoglycemic effect. These findings suggest that RLMP is a promising candidate for developing novel antioxidant agents or hypoglycemic drugs.
