Graphene oxide quantum dots (GOQDs) induce behavioral disorders via the disturbance of kynurenine pathway in zebrafish larvae

Shun Deng,Enming Zhang,Yunyang Zhao,Hao Guo,Lan Luo,Song Yi,Bingxin Zheng,Xiaoyuan Mu,Kezhu Yuan,Xuangen Deng,Haibo Rong,Yanbo Ma,Wanping Bian,Hai Shen

The emergence of graphene quantum dots (GQDs) expands the use of graphene derivatives in nanomedicine for its direct therapeutic applications in treating neurodegeneration, inflammation, metabolic dysfunction, and among others. Nevertheless, the biosafety assessment of GQDs remains deficient mostly because of the diverse surface characteristics of the nanoparticles. Our prior work demonstrated that GQDs can induce strong thigmotactic effects in zebrafish larvae over a wide range of concentrations, yet the underlying metabolic mechanisms remain largely unknown. In this study, we conducted a further exploration about graphene oxide quantum dots (GOQDs) for its potential neurotoxic effect on the behaviors of zebrafish larvae by combining neurotransmitter-targeted metabolomics with locomotion analysis. After continuous exposure to a concentration gradient of GOQDs (12.5 - 25 – 50 – 100 - 200 μg/mL) for 7 days, the thigmotactic activities of zebrafish larvae were observed across all exposure concentrations relative to the control group, while the basal locomotor activities, including distance moved and average velocity, were significantly changed by low concentrations of GOQDs. Targeted metabolomics was performed using zebrafish larvae at 7 days post-fertilization (dpf) that were exposed to 12.5 and 200 μg/mL, both of which were found to perturb the kynurenine pathway by regulating the levels of kynurenine, 3-hydroxyanthranilic acid (3-HAA), and quinolinic acid (QA). Furthermore, the thigmotaxis of larval fish induced by GOQDs during exposure could be counteracted by supplementing Ro-61-8048, an agonist acting on kynurenine 3-monooxygenase (KMO). In conclusion, our study establishes the involvement of the kynurenine pathway in GOQDs-induced thigmotaxis, which is independent of the transcriptional modulation of glutamate receptor families.

Aptamer based fluorometric sulfamethazine assay based on the use of graphene oxide quantum dots

He Yanhua,Zhang Bingyan,Fan Zhefeng

The authors have developed a homogeneous “off-on” fluorometric method for the determination of the antibiotic sulfamethazine (SMZ). Aptamer against SMZ was labeled with graphene oxide quantum dots upon which the Graphene oxide quenched the blue fluorescence of the GOQDs. On addition of SMZ, the aptamers will bind SMZ and this will cause the release of GOQDs. As a result, fluorescence will be regenerated. Fluorescence, best measured at excitation/emission wavelengths of 365/455 nm, increases linearly in the 8 pg·mL −1 to 60 ng·mL −1 SMZ concentration range, with a 5 pg·mL −1 detection limit. The method is reliable and was successfully applied to the determination of SMZ in spiked milk samples, with recoveries ranging from 89 to 96% depending on analyte concentration. Graphene oxide quantum dots (GOQDs) were covalently bound to the aptamer (apt) against sulfamethazine (SMZ) and adsorbed on the surface of graphene oxide (GO). This results in quenching of the fluorescence of GOQDs. On addition of SMZ, fluorescence is restored due to the release of GOQD@apt from GO.

Real-time detection of isoprene marker gas based on micro-integrated chromatography system with GOQDs-modified μGC column and metal oxide gas detector

Baoqing Han,Xinyu Zhang,Yan Wang,Wenjuan Wang,Benben Wang,Shuai Li,Hairong Wang,Yuefei Yan,Jiusheng Han,Chuanliu Wang,Congsi Wang

Isoprene is a typical physiological marker that can be used to screen for chronic liver disease. This work developed a portable micro-integrated chromatography analysis system based on micro-electromechanical system technology, nanomaterials technology and embedded microcontroller technology. The system integrated components such as graphene oxide quantum dots modified semi-packed microcolumn, In2O3 nanoflower (NF) gas-sensitive detector and 3D printed miniature solenoid valve group. The effectiveness of the separation effect of the micro-integrated system was verified by gas mixture test; the laws of the influence of carrier gas pressure and column temperature on the chromatographic separation performance, respectively, were investigated, and the working conditions (column temperature 90 °C and carrier gas pressure 7.5 kPa) for system testing were determined. The percentages of relative standard deviation of the peak areas and retention times obtained for the separated gases were in the range of 0.95%–6.06%, indicating the good reproducibility of the system. Meanwhile, the microintegrated system could detect isoprene down to 50 ppb at small injection volume (1 ml). The system response increased with increasing isoprene concentration and was linearly correlated with isoprene concentration (R2 = 0.986), indicating that the system was expected to be used for trace detection of isoprene, a marker gas for liver disease, in the future.
