A highly sensitive fluorimetric assay has been developed for the evaluation of glutathione reductase (GR) activity and the screening of its inhibitors by using carbon quantum dots (CQDs) as the signal reporter. The detection mechanism is based on the following facts: (1) the fluorescence of CQDs can be quenched by Hg(II) through the strong CQDs-Hg(II) coordination; (2) GR can catalyze the reduction of oxidized glutathione (GSSG) into reduced glutathione (GSH), so that the fluorescence recovery of CQDs would take place resulting from the strong GSH-Hg(II) interaction; (3) GR can lose its catalytic reduction of GSSG in the presence of its inhibitors, which will inhibit the recovery of the quenched fluorescence of CQDs. The developed CQDs-based fluorimetric method can facilitate the sensitive evaluation of GR activity in the range of 0.10–2.0 mU mL −1 with a detection limit of 0.050 mU mL −1 . In addition, other kinds of enzymes like myoglobin , thrombin, alcohol dehyrogenase, amylase, pepsin, and trypsin could show no significant effects on the evaluation of GR activity. This work may expand the biological applications of CQDs as the fluorescent probes with low cost, easy preparation, and high photostability.

The continued expansion of the fields of photoelectrochemical (PEC) aptasensor has guided the development of photoactive materials that can generate electron-photo pairs to emerge photocurrent. However, only such an improvement has an insatiable appetite for the application of molecular level for PEC aptasensor due to the limited quantity of the bioprobes immobilized on the working electrode. Here, we successfully synthetized SnSe/graphene (SnSe/GR) nanohybrids as a photoactive material with a strong photocurrent signal. The constructed bioprobe, DNA/gold nanoparticles/glutathione disulfide (DNA/AuNPs/GSSG), decreased photocurrent effectively. GSSG, an electron acceptor, was designed as a quencher in the bioprobe to overcome a part of shortcomings of DNA/AuNPs in a way. This was owing to the resonance energy transfer (RET) between AuNPs and SnSe/GR, and GSSG to accept photoelectrons, so that the photocurrent response was obviously decreased comparing to the PEC response of SnSe/GR/CPE. Based on this, probe/SnSe/GR/CPE was built as a signal-on PEC aptasensor to detect endocrine-disrupting chemicals using bisphenol A as model. This signal-on PEC aptasensor showed a linear response in the range of 0.01–7 μM and a limit of detection of 3 nM for bisphenol A. And this method presented a promising prospect for sensitive and accurate analysis of other biological molecules between aptamers and corresponding targets at low levels.

Nanozymes have become one of the key applications in the analysis and sensing of food. However, nanozymes presented have shortcomings that limit their further application, such as complex raw material sources, and high condition requirements for synthesis. Cu-GSSG nanozymes under green preparation conditions were synthesized inspired by the detoxification process of glutathione in living organisms. Cu-GSSG nanozymes were further loaded onto filter paper to construct a smartphone-assisted colorimetric sensor test strip for ascorbic acid (AA) detection. As the excellent peroxidase-like enzymes, Cu-GSSG nanozymes showed low AA detection limit of 2.9 μM with a wide linear range of 10–300 μM. Cu-GSSG nanozymes exhibited excellent anti-interference, accuracy, and biocompatibility performances for reliable detection AA in prickly pear juice and 10 supplements. The intelligent colorimetric sensor platform based on the bio-inspired Cu-GSSG nanozymes showed the great potential in rapid detection and large-scale screening of AA in fruits and supplements.

The efficient delivery of therapeutic genes remains a major challenge in realizing a feasible gene-based treatment. Herein, a versatile oligopeptide, glutathione, was introduced to construct novel non-viral cationic gene vectors. Reduced/oxidized forms of glutathione (GSH/GSSG) and relevant amino acids (Glu, Cys, and Gly) were used to modify low molecular weight PEI through surface modification or crosslinking. These polymers could bind well and condense DNA into spherical nanoparticles, which were stable in the presence of serum. The disulfide bonds within the crosslinked polymer GSSG-PEI may facilitate polymer degradation and DNA release under a reductive environment. In vitro transfection experiments reveal that the modification could largely improve the gene transfection efficiency of low molecular weight PEI, especially in the presence of serum. In HeLa cells, GSSG-PEI could even give up to 150 times higher efficiency than PEI 25 kDa. TEM and serum concentration effect assay also demonstrate the good serum tolerance of the polymers. Flow cytometry results show that GSSG-PEI might induce cellular uptake with higher efficiency than PEI 25 kDa, especially in the presence of serum. Results reveal that GSSG is a good candidate for the crosslinking of small cationic molecules to form polymeric gene vectors with improved transfection efficiency and biocompatibility.

Autophagy is a degradation cellular process which also plays an important role in virus infection . Glutamine is an essential substrate for the synthesis of glutathione which is the most abundant thiol-containing compound within the cells and plays a key role in the antioxidant defense and intracellular signaling . There is an endogenous cellular glutathione pool which consists of two forms of glutathione, i.e. the reduced form (GSH) and the oxidized form (GSSG). GSH serves as an intracellular antioxidant to maintain cellular redox homeostasis by scavenging free radicals and other reactive oxygen species (ROS) which can lead to autophagy. Under physiological conditions, the concentration of GSSG is only about 1% of total glutathione, while stress condition can result in a transient increase of GSSG. In our previous report, we showed that the replication of snakehead fish vesiculovirus (SHVV) was significant inhibited in SSN-1 cells cultured in the glutamine-starvation medium, however the underlying mechanism remains enigmatic. Here, we revealed that the addition of L-Buthionine-sulfoximine (BSO), a specific inhibitor of the GSH synthesis, could decrease the γ- glutamate-cysteine ligase (GCL) activity and GSH levels, resulting in autophagy and significantly inhibition of the replication of SHVV in SSN-1 cells cultured in the complete medium. On the other hand, the replication of SHVV was rescued and the autophagy was inhibited in the SSN-1 cells cultured in the glutamine-starvation medium supplemented with additional GSH. Furthermore, the inhibition of the synthesis of GSH had not significantly affected the generation of reactive oxygen species (ROS). However, it significantly decreased level of GSH and enhanced the level of GSSG, resulting in the decrease of the value of GSH/GSSG, indicating that it promoted the cellular oxidative stress . Overall, the present study demonstrated that glutamine starvation impaired the replication of SHVV in SSN-1 cells via inducing autophagy associated with the disturbance of the endogenous glutathione pool.

Free radical-induced oxidative damage and nitrosative stress have been identified as key factors in neuroinflammation responses after traumatic brain injury (TBI), with which reactive oxygen and nitrogen species (RONS), especially nitrogen signaling molecules, are strongly associated. Here, we prepared ultrasmall carbon dot (CD) by using a simple and facile method. In vitro assessment experiments show that the antioxidative CD exhibits an ultrahigh target-scavenging effect for nitrogen signaling molecules, especially the highly reactive ˙NO and ONOO−. However, CD can only partially eliminate conventional oxygen radials such as O2˙− and ˙OH, indicating CD has a preference for RNS modulation. Moreover, in vitro cell experiments and in vivo mice experiments reveal that CD can reduce the reactive oxygen species (ROS) level and lipid peroxidation, enhance superoxide dismutase (SOD) activity and GSSG level, and further improve the survival rate of neuron cells and TBI mice. These results declare that antioxidative CD could serve as an effective therapeutic for TBI.

Prostate cancer (PCa) is a complex disease with diverse molecular alterations. The aryl hydrocarbon receptor (AhR) is a ligand-activated transcription factor that exhibits pleiotropic roles in PCa, and 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) is a potent ligand for AhR. While targeting ferroptosis is an innovative PCa therapeutic strategy, the impact of AhR on this process remains unclear. This study aimed to investigate the influence of AhR on lipid peroxidation and ferroptosis. Results showed that TCDD activated AhR, as evidenced by increased CYP1A1 expression, leading to reduced cell viability. TCDD caused mitochondria shrinkage, decreased the GSH/GSSG ratio, and elevated the MDA levels and lipid peroxidation. Interestingly, AhR knockdown reversed these effects, similar to the action of ferroptosis inhibitors. Mechanistically, TCDD suppressed nuclear receptor subfamily 4 group A member 1 (NR4A1) expression, in part due to AhR activation. This suppression subsequently led to a reduction in the expression of the NR4A1 downstream target stearoyl-CoA desaturase 1 (SCD1). NR4A1 overexpression counteracted the effects of TCDD. In vivo, TCDD activated AhR, downregulated NR4A1 and SCD1 expression, induced mitochondria shrinkage, and increased the MDA and 4-hydroxynonenal (4-HNE) levels. In summary, TCDD promotes ferroptosis in androgen-dependent PCa via inhibiting the NR4A1/SCD1 axis, in part dependent on AhR activation.

Oxidative damage resulting from weaning stress significantly impacts the growth performance and health status of piglets. Taurine, a dietary antioxidant with diverse functions, was investigated in this study for its protective role against weaning stress-induced oxidative damage and its underlying mechanism. Forty 28-day-old male castrated weaned piglets were randomly assigned to four groups. The control group received the basal diet, while the experimental groups were fed the basal diet supplemented with 0.1, 0.2%, or 0.3% taurine over a 28-day period. In vitro, H2O2 was utilized to induce oxidative damage to the jejunal mucosa of piglets via IPEC-J2 cells. The results demonstrated that taurine supplementation reduced the incidence of diarrhea in piglets compared to that in the control group (p < 0.05); the addition of 0.2 and 0.3% taurine led to increased average daily gain and improved feed conversion efficiency in weaned piglets, showing a linear dose-response correlation (p < 0.05). Taurine supplementation at 0.2 and 0.3% enhanced the activities of serum CAT and GSH-Px while decreasing the levels of serum NO, XOD, GSSG, and MDA (p < 0.05). Moreover, it significantly elevated the levels of GSS, Trx, POD, complex I, mt-nd5, and mt-nd6, enhancing superoxide anion scavenging capacity and the hydroxyl-free scavenging rate in the livers of weaned piglets while reducing NO levels in the liver (p < 0.05). Additionally, 0.2 and 0.3% taurine supplementation decreased serum IL-6 levels and elevated the concentrations of IgA, IgG, and IL-10 in weaned piglets (p < 0.05). The levels of occludin, claudin, and ZO-1 in the jejunum mucosa of weaned piglets increased with 0.2 and 0.3% taurine supplementation (p < 0.05). In IPEC-J2 cells, pretreatment with 25 mM taurine for 24 h enhanced the activities of SOD and CAT; reduced the MDA content; upregulated the mRNA expression of various genes, including ZO-1, occludin, claudin-1, Nrf2, and HO-1; and reversed the oxidative damage induced by H2O2 exposure (p < 0.05). Overall, the findings suggest that the inclusion of 2 and 3% taurine in the diet can enhance growth performance, reduce diarrhea rates, ameliorate oxidative stress and inflammation, and promote intestinal barrier function in weaned piglets.

Marine hydrosphere is in grave danger if extreme weather conditions, such as high temperatures and heat waves, become the norm. The functional characteristics of nano Zn oxides (nZnO) might provide a “booster substance” that helps marine species to endure environmental stress. In the present study, physiological indicators and simulated severe environments were used to evaluate the antioxidative ability of nZnO in marine rabbitfish Siganus fuscescens, with an aim to help them resist extreme environmental stress. Rabbitfish were exposed to different diets with ZnSO4 and nZnO at the optimum requirement level (Zn content of 62.5 μg g−1), and then challenged with a low dissolved oxygen concentration and higher temperature. We demonstrated that dietary nZnO promoted the anti-oxidation defense system of the rabbitfish by improving the activities of the antioxidant enzymes, i.e., catalase (CAT), superoxide dismutase (SOD), and the reduced glutathione/oxidized glutathione (GSH/GSSG) ratio without tissue damage. Compound stressors reduced the median survival duration of the rabbitfish from 4.25 h to 2.5 h (dietary exposure) and 9 h to 5.75 h (dietary nZnO exposure) compared to single environmental stress. nZnO downregulated the HIF gene expression and increased the prolyl hydroxylase (PHD) activity in the liver of the rabbitfish. After 10 h of extreme environmental exposure, the gene expression results indicated that nZnO inhibited the NF-κB/IL-1β/IL-6 pathway and TNF-α/Bcl-2/Bax/caspase 9 pathway and activated the akt/mTOR/elovl5 and elovl8 pathway of the rabbitfish to maintain the resilience of the fish under the extreme environment. These results strongly suggested that nZnO may improve the anti-oxidation ability of fish to resist extreme environments.
