Effects of elevated temperature and copper exposure on the physiological state of the coral Galaxea fascicularis

Meng Cheng,Yong Luo,Xiao-Lei Yu,Lin-Tao Huang,Jian-Sheng Lian,Hui Huang

The co-occurrence of elevated seawater temperature and local stressors (heavy metal contamination) affects the ecophysiology of phototrophic species, and represents a risk to the environmental quality of coral reefs. Therefore, we investigated the effects of both Cu alone and Cu in combination with elevated temperature (ET) on the physiology of the coral Galaxea fascicularis , and measured the parameters related to the photo-physiology and oxidative state. G.fascicularis is one of the dominant coral species in the South China Sea which exhibits strong adaptability to environmental stress. We exposed the common coral species G.fascicularis to a series of environmentally relevant concentrations of Cu at 29 °C (normal temperature, NT) and 32 °C (elevated temperature, ET) for 96 h. Single polyps were used in the experiments, which reduced individual variability when compared to the coral colonies. The results suggested that: i) Cu or ET had significant negative effects on the actual operating ability of photosystem Ⅱ (PSII), but not on the maximal chlorophyll fluorescence in darkness (Fv/Fm). ii) Symbiodiniaceae density was significantly reduced by high Cu concentrations, for Cu-NT and Cu-ET, a high concentration of Cu (40 μg/L) significantly impacted Symbiodiniaceae density, causing a 75.4% and 81.0% decrease, respectively. iii) the content of malondialdehyde (MDA) in coral tissues increased significantly under Cu-ET. iv) a certain range of copper concentration (25–30 μg/L) increased the pigment content of the Symbiodiniacea. Our results indicated that the combined stressors of Cu and ET made the coral tissue sloughed, caused the coral tissue damaged by lipid oxidation, reduced the photosynthetic capacity of the Symbiodiniacea, and led to the excretion of Symbiodiniacea.

Long-term exposure to climbazole may affect the health of stress-tolerant coral Galaxea fascicularis

Chuhan Sun,Yuehua Huang,Alireza Riyahi Bakhtiari,Dongdan Yuan,Yanyu Zhou,Hongwei Zhao

The persistence of coral reefs globally is threatened by various forms of chemical pollution. Climbazole, an azole antibacterial agent extensively utilized in pharmaceuticals and personal care products (PPCPs) in everyday life, has been detected in various environment media and proved to have significant adverse effects on aquatic organism. However, the effects of climbazole on coral remain largely unknown. Therefore, in this study, we conducted a 42-day investigation to examine the effects of varying concentrations of climbazole on Galaxea fascicularis , a stress-tolerant coral species. Our investigations included coral color observations, physiological experiments, and assessments of microbial diversity. The results showed that, after 42 days of exposure, the coral color in the treatment group exposed to 100 μg/L climbazole significantly decreased by one color category on the reference chart (D6 shifted to D5), while there was no change in the control group. This was accompanied by an increase in oxidative stress and a decrease in photosynthetic capacity in coral specimens. Additionally, there was a notable alteration in microbial diversity, resulting in reduced community stability. Elevated levels of climbazole (100 μg/L) stress led to an increased abundance of potentially pathogenic bacteria such as unclassified Erysipelotrichaceae . However, at an environmentally relevant concentration of 1 μg/L, climbazole decreased the photosynthetic efficiency and induced oxidative stress in the stress-tolerant coral G. fascicularis , while not significantly impacting the microbial community diversity of the coral. The findings of our study have important implications for the protection and management of nearshore coral reefs and offer essential data for ecological risk assessment of climbazole.

Proteomic insights into the combined effects of near-threshold bleaching temperature and prometryn exposure on the coral Galaxea fascicularis

Sichen Li,Yanyu Zhou,Haochen Niu,Weilu Kang,Guowei Fu,Bo Hu,Hongwei Zhao

Ocean warming events increasingly co-occur with coastal agrochemical pollution, yet their joint effects on coral holobiont molecular responses remain poorly resolved. We exposed the reef-building coral Galaxea fascicularis to near-threshold warming (30 °C), prometryn (1 μg L −1 ), and their combination for 4 days, and quantified coral host and Symbiodiniaceae protein abundance using TMT-based proteomics to capture early-stage molecular responses. Heat was the primary driver of proteome shifts, consistent with suppressed biosynthesis and reallocation toward maintenance, whereas prometryn imposed additional changes on specific pathways under heat. Under combined stress, symbionts showed a mismatch in which light harvesting increased without a proportional rise in reaction-center/electron-transport capacity, which may elevate excitation pressure and oxidative stress risk. Consistent with this interpretation, photophysiological measurements showed that light-adapted PSII parameters were more responsive than dark-adapted maximum photosystem II quantum yields ( Fv/Fm) during acute exposure, with the combined treatment showing modest but the clearest decline in photochemical performance. In parallel, host stress-associated signaling pathways were enriched, while antioxidant/detoxification functions were reduced in both partners. Together, these proteomic patterns suggest that prometryn exposure may compromise glutathione-based redox buffering capacity, thereby weakening the molecular response of a relatively heat-tolerant coral to near-threshold warming. The present study provides valuable insights into the critical need to prioritize the reduction of PSII herbicide pollution as part of coral reef protection strategies. Our findings support management efforts to reduce coastal PSII herbicide inputs as a local lever that could mitigate bleaching susceptibility under warming.
