Preparation, characterization, and anticancer activity evaluation of Ganoderma lucidum polysaccharide/5-fluorouracil conjugate

Wang Ying,Yang YunXia,Zhu Daliang,Zhang Hongmei,Wang Yanqing

Polysaccharide-based drug delivery systems have gained significant attention due to their biocompatibility and potential for targeted therapy. In this study, we utilized Ganoderma lucidum polysaccharides (GLP) as a macromolecular carrier to synthesize polysaccharide drug conjugates by forming ester bonds with 5-fluorouracil (5-FU) derivatives (5-FUAC). This not only improves the solubility and stability of 5-FU but also enhances its targeted delivery to tumor tissues, potentially reducing systemic toxicity. Utilizing Ganoderma lucidum polysaccharides (GLP) as a macromolecular carrier, we successfully synthesized GLP-5-FU conjugates by forming ester bonds with 5-fluorouracil derivatives (5-FUAC). These conjugates were then characterized using spectrum analysis technologies. The drug loading capacities of 5-FU in the GLP-5-FU conjugates were found to be approximately 4.7%. Morphological observations revealed that the GLP-5-FU conjugates had a regular spherical shape. In terms of drug release, 5-FU was found to be released faster at pH 6.8 compared to pH 7.4. To investigate the dynamic viscoelastic behavior of GLP and GLP-5-FU conjugates in water, dynamic rheology analysis was conducted. In terms of cytotoxicity, the GLP-5-FU conjugates exhibited greater inhibition of proliferation compared to 5-FU and GLP alone in HeLa, 786-O, and SKOV3 cells. Furthermore, the GLP-5-FU conjugates showed lower cytotoxicity to 293A cells. Graphical

Ganoderma lucidum Polysaccharide-Decorated Extracellular Vesicle Enables Synergistical Antitumor Immunotherapy

Zhaorong Ouyang,Siyu Li,Ao Zhang,Shu Ye,Wanqiu Ye,Guolei Wen,Tao Wei,Biao Cai,Houli Liu

Ganoderma lucidum polysaccharide (GLP) holds considerable promise for tumor therapy, but its clinical application is limited by its poor tumor-targeting capability. Herein, we report the development of an immunoregulatory nano-bioconjugate formed by conjugating GLPs with low-pH culture medium reprogrammed CT26 tumor cell-derived extracellular vesicle (LTEV). Specifically, 4-carboxybenzeneboronic acid (CPBA), used as an intermediate coupling agent, is conjugated with the amino group of LTEV via its carboxyl group to obtain CPBA-LTEV. GLP is then covalently linked via boric ester linkages between its hydroxyl groups and the boronic acid groups of CPBA. After systemic administration, GLP@LTEV accumulates in homologous tumor tissues derived from the LTEV parent cells, remodeling the immunosuppressive tumor microenvironment (TME) by repolarizing M2-like macrophages towards the M1 phenotype, promoting dendritic cell (DC) maturation, activating cytotoxic T lymphocytes (CTLs), and inhibiting immunosuppressive regulatory T cells (Tregs). The synergism of tumor-targeting delivery and potent immunomodulatory effects in this combined manner therefore significantly inhibits subcutaneous tumor growth and lung metastasis with minimal side effects, providing a novel combination of polysaccharide and nanovesicle for robust tumor immunotherapy.

A Ganoderma lucidum Polysaccharide-Modified Nanoadjuvant Elicits Potent Cellular Immunity for Hepatitis B Vaccine

Zhe Zhai,Jiansong You,Xuhan Liu

Background: Aluminum-adjuvanted hepatitis B vaccines effectively prevent primary HBV infection but predominantly favor humoral immunity and have limited capacity to induce strong cellular responses. To improve immune-response quality, we developed aGanoderma lucidumpolysaccharide-modified hybrid nanoparticle adjuvant (GHNP) for hepatitis B surface antigen (HBsAg) vaccination. Methods: Cationic PEG-PCL/DOTAP hybrid nanoparticles (HNPs) were surface-modified withGanoderma lucidumpolysaccharide (GLP) at different mass ratios and characterized by particle size, polydispersity index, zeta potential, and transmission electron microscopy. Antigen uptake, RAW264.7 cell viability, MHC II expression, and TLR2/4-associated TNF-α secretion were evaluated in vitro. C57BL/6 mice were immunized intramuscularly with PBS, HBsAg, HBsAg + Aluminum, HBsAg@HNP, or HBsAg@GHNP6 on Days 0, 14, and 28. HBsAg-specific antibodies, splenocyte activation, proliferation, and cytokine secretion were assessed. Results: GHNP6 showed suitable physicochemical properties, acceptable cytocompatibility at concentrations up to 10 μg·mL−1, and efficient model-antigen uptake. GHNP increased MHC II expression in RAW264.7 cells, and TLR2/4 inhibition reduced nanoparticle-associated TNF-α secretion, supporting involvement of these pathways in immune activation. In vivo, HBsAg@GHNP6 induced robust HBsAg-specific IgG responses and a higher IgG2a/IgG1 ratio than HBsAg@HNP and HBsAg + Aluminum. It also produced the highest splenocyte proliferation after HBsAg restimulation, with lower IL-6 secretion than HBsAg + Aluminum and moderately increased IFN-γ compared with PBS and free HBsAg. Conclusions: GHNP integrates nanoparticle-assisted antigen delivery with GLP-mediated immunomodulation and represents a promising strategy for enhancing cellular immune responses while maintaining humoral immunity in HBsAg vaccination.

Virus-inspired Ganoderma lucidum polysaccharide-functionalized adjuvant with pseudo-zwitterionic interface for potentiated nasal immunization

Xu-Han Liu,Zhe Zhai,Ling-Ling Tao,Nan-Yu Chen,Yong-Xian Cheng

Intranasal vaccination is promising against respiratory infections but hindered by the mucus barrier. To overcome the adhesion-penetration dilemma, we developed a virus-inspired hybrid nanoparticle (GHNP) by functionalizing a cationic polymer/lipid core (HNP) with Ganoderma lucidum polysaccharide (GLP). By tuning the surface modification density of GLP, GHNP achieved balanced mucosal interaction and penetration, with an optimal GLP-to-HNP mass ratio of 1:24. The optimized GHNP(1/24) (∼104 nm, +27 mV) markedly enhanced mucus penetration via its virus-mimetic pseudo-zwitterionic interface. This led to improved antigen uptake under mucin barrier conditions by 79-fold in dendritic cells and 25-fold in macrophages. In a murine intranasal immunization model, the intrinsic immunostimulatory activity of GLP, mediated through TLR2/4 activation, further potentiated the immune response, eliciting robust mucosal and systemic immune responses. Specifically, GHNP(1/24) elevated bronchoalveolar sIgA 1.5-fold and serum IgG 1.6-fold compared to antigen alone, while maintaining a balanced IgG1/IgG2a profile. This work establishes a modular platform that bridges structural biomimetics and carbohydrate immunoengineering, offering a tunable, dual-functional strategy for advanced mucosal vaccine design that surpasses the capabilities of traditional delivery systems which often address either adhesion or immunostimulation alone.

Peroxidase-Like Platinum Clusters Synthesized by Ganoderma lucidum Polysaccharide for Sensitively Colorimetric Detection of Dopamine

Xiang Lai,Yu Han,Jie Zhang,Jinyu Zhang,Weifeng Lin,Zhiwei Liu,Longgang Wang

The sensitive and selective detection of dopamine (DA) is very important for the early diagnosis of DA-related diseases. In this study, we reported the colorimetric detection of DA usingGanoderma lucidumpolysaccharide (GLP) stabilized platinum nanoclusters (Ptn-GLP NCs). When Pt600-GLP NCs was added, 3,3’,5,5’-tetramethylbenzidine (TMB) was rapidly catalyzed and oxidized to blue oxTMB, indicating the peroxidase-like activity of Pt600-GLP NCs. The catalytic reaction on the substrate TMB followed the Michaelis-Menton kinetics with the ping-pong mechanism. The mechanism of the colorimetric reaction was mainly due to the formation of hydroxyl radical (•OH). Furthermore, the catalytic reaction of Pt600-GLP NCs was used in the colorimetric detection of DA. The linear range for DA was 1–100 μM and the detection limit was 0.66 μM. The sensitive detection of DA using Pt-GLP NCs with peroxidase-like activity offers a simple and practical method that may have great potential applications in the biotechnology field. Keywords:dopamine;detection;platinum;colorimetric;Ganoderma lucidumpolysaccharide

Ganoderma lucidum Polysaccharide Enzymatic Hydrolysate Suppresses the Growth of Human Colon Cancer Cells via Inducing Apoptosis

Jing hui Bai,Jian Xu,Jian Zhao,Rui Zhang

Ganoderma lucidum is a popular traditional Chinese medicine used in China to improve health. Previous researches have revealed that the polysaccharide from G. lucidum could exert diversity activities, including immunomodulation, antioxidant, and antitumor effects. However, the effect of enzymatically hydrolyzed G. lucidum polysaccharide (EGLP) in colorectal cancer (CRC) progression remains unknown. The present research aimed to investigate the antitumor mechanism of EGLP in human colon cancer cells. For this purpose, the cytotoxic effects of EGLP were measured by the (3-(4,5)-dimethylthiahiazo (-z-y1)-3,5-di-phenytetrazoliumromide (MTT) method. The apoptosis was evoked upon EGLP treatment, which was assayed using flow cytometry. The results indicated that EGLP may induce apoptosis in human colon cancer cell (HCT-116) cells via the upregulation of BCL-2 associated X protein (Bax), phospho-extracellular regulated protein kinases (P-ERK), and cleaved caspase-3 expression and downregulation of B-cell lymphoma-2 (Bcl-2), phospho-serine/threonine kinase 1 (p-Akt1), and cyclo-oxygen-ase (COX-2) expression. The obtained findings indicated EGLP as a new therapeutic agent in fighting CRC.

Preparation and characterization of Ganoderma lucidum polysaccharide-based multifunctional tellurium nanorods to realize combination cancer therapy

Wenhui Liu,Haojing Jiang,Jing Xu,Yuanqiang Guo

Cancer remains a significant global health challenge. Given the limitations of conventional single-treatment modalities and the potential benefits of combination therapies, this study aimed to develop a multifunctional nanoplatform integrating immunotherapy, chemotherapy, and photothermal therapy for cancer treatment. Ganoderma lucidum polysaccharides (GLPs) and luteolin (LU) were employed to regulate the one-dimensional growth of tellurium nanorods, forming distinctive nanorods (GLP-LU-TeNRs) with an aspect ratio of 5.8. GLP-LU-TeNRs demonstrated excellent photothermal performance with a photothermal conversion efficiency ( η ) of 30.9 %. Irradiation with 808 nm near-infrared (NIR) light, GLP-LU-TeNRs inhibited the proliferation of cancer cells, reducing it by up to 22.8 % in vitro (150 μg/mL), while in vivo , it suppressed tumor growth and metastasis by up to 36.1 % and 66.7 %, respectively, compared to the non-laser group (15 μg/mL). GLP-LU-TeNRs markedly induced apoptosis in tumor cells. In vivo experiments demonstrated that GLP-LU-TeNRs inhibited the proliferation and metastasis of cancer cells. GLP not only upregulated nitric oxide (NO) production but also promoted the maturation of dendritic cells (DCs), exhibiting the potential to initiate anti-tumor immune responses. The multifunctional tellurium nanorods developed in this study offer a promising approach to enhance the efficacy of cancer treatment, particularly in cases where single-agent therapy is suboptimal.

Ganoderma lucidum Polysaccharide Peptide Alleviates Cyclophosphamide-Induced Male Reproductive Injury by Reducing Oxidative Stress and Apoptosis

Hang Zhang,Nannan Li,Yukun Zhang,Yue Xu,Feng Lu,Dongmei Lin,Shuqian Lin,Min Li,Baoxue Yang

Chemotherapy is an important factor leading to male infertility. It is crucial to discover safe and effective treatments to prevent male reproductive injury caused by chemotherapy. TheGanoderma lucidumpolysaccharide peptide (GLPP) has multiple pharmacological activities. The purpose of this study was to determine whether GLPP could protect the male sperm production from chemotherapeutic injury using a mouse model, with testicular damage induced by cyclophosphamide (CP). CP (50 mg/kg/day) was injected intraperitoneally into male ICR mice gavaged with different doses of GLPP at certain spermatogenic stages. The experimental results showed that GLPP alleviated the CP-induced reduction in reproductive organ coefficients and sperm parameters and reduced the morphological damage of testicular tissues in a dose-dependent manner. GLPP significantly improved the reproductive index, sperm-related parameters, sex hormone levels, and histological testis architecture at different spermatogenic stages. Furthermore, GLPP significantly increased superoxide dismutase (SOD), glutathione (GSH), catalase (CAT), Nrf2, and HO-1, and decreased malondialdehyde (MDA) and Keap-1 in the testicular tissue, indicating reduced oxidative stress. In addition, GLPP limited CP-induced apoptosis via a reduction in Bax expression and increase in Bcl-2 expression. This study suggests that GLPP plays a protective role in spermatogenesis by reducing chemotherapeutic injury and might be developed into drug for male patients receiving chemotherapy.

pH and glutathione dual responsive nanoparticles based on Ganoderma lucidum polysaccharide for potential programmable release of three drugs

Dan Zheng,Jingyang Zhao,Yinghua Tao,Jing Liu,Luying Wang,Jing He,Jiandu Lei,Kefeng Liu

Nanoparticles have been widely studied as carries of anti-cancer drugs, but designing and preparing smart drug delivery system for precisely controlled drug release remains a major challenge. Herein, a novel Ganoderma lucidum polysaccharides (GLP) based, rutin-carboxyphenyl boronic acid (CPBA)-GLP-dithiodipropionic acid (DPA)-dihydroartemisinin (DHA)/10-hydroxy camptothecin (HCPT) polymeric nanoparticles (RCGDDH NPs) with pH and redox dual-responsive were first presented. The three drugs loaded in RCGDDH NPs can be released programmatically: rutin is released in tumor tissue with acidic microenvironment, DHA and HCPT are released in the redox environment in tumor cells. Interestingly, GLP, as a carrier, has anti-cancer activity and can enhance anti-cancer activity. Size of the RCGDDH NPs prepared is about 98 nm. The in vitro release study shows when under the condition of pH 5.2 and 10 mM reductive glutathione, the amount of HCPT and DHA released from nanoparticles is 2.5 and 2.7 folds higher than the normal physiological condition. In addition, rutin in the nanoparticles can be released in a slightly acidic condition and has an obvious inhibitory action of matrix metalloproteinase MMP-9. Further, in vitro and in vivo experiments indicate that the prepared RCGDDH NPs can effectively kill tumor cells, inhibit tumor growth and cause low side effects. The above results suggest that the pH and redox dual-responsive RCGDDH NPs are a promising candidate for tumor therapy.

Self-Assembled pH-Sensitive Nanoparticles Based on Ganoderma lucidum Polysaccharide–Methotrexate Conjugates for the Co-delivery of Anti-tumor Drugs

Dan Zheng,Jingyang Zhao,Yucheng Li,Liyu Zhu,Mengchen Jin,Luying Wang,Jing Liu,Jiandu Lei,Zhonglong Li

In tumor therapy, polymer nanoparticles are ideal drug delivery materials because they can mask the disadvantages of anti-tumor drugs such as poor solubility in water, high toxicity, and side effects. However, most polymer-based nanoparticles do not themselves have anti-tumor properties. Herein, a novel pH-sensitive nanoparticle drug delivery system based on Ganoderma lucidum polysaccharides (GLPs), which have demonstrated anti-tumor activities, was designed to enable the delivery of methotrexate (MTX) and 10-hydroxycamptothecin (HCPT) to tumor cells, where they could exert synergistic anti-tumor effects. The prepared nanoparticles were irregularly spherical in shape with a uniform particle size of ∼190 nm, and they exhibited a high drug-loading capacity (MTX 21.5% and HCPT 22.6%) and excellent biocompatibility. Moreover, the loaded MTX and HCPT units were rapidly released under acidic conditions within the tumor cells while remaining stable under normal physiological conditions. Meanwhile, compared to free MTX and HCPT, the GLP–APBA–MTX/HCPT nanoparticles presented exhibited better tumor suppressive effects and fewer side effects in vivo, which indicates that they may be an effective anti-tumor treatment strategy.
