Injectable alendronate-functionalized GelMA hydrogels for mineralization and osteogenesis

Lei Liu,Xiaoyu Li,Xuetao Shi,Yingjun Wang

In a minimally invasive procedure, hydrogels can be injected into an affected area for drug loading and tissue repair. Here, gelatin methacryloyl (GelMA) was modified with alendronate (ALN), and three different alendronate-functionalized GelMA (GelMA-ALN) hydrogels were prepared. The modification greatly improved the swelling ratio, protein adsorption and mineralization. The GelMA-ALN hydrogels significantly promoted the in vitro osteogenic differentiation of hFOB cells as indicated by their higher alkaline phosphatase (ALP) activity, denser mineralization and up-regulated osteogenesis-related genes at both the mRNA and protein levels. Meanwhile, the cells maintained their activity and differentiated into osteoblasts when encapsulated in the GelMA-ALN hydrogels. Alendronate-modified hydrogels have potential for use in the minimally invasive treatment of irregular bone defects.

GelMA - PVAMA hydrogel loaded with MYDGF accelerates wound healing by promoting angiogenesis

Sizhen Huang,Haodi Kou,Zhichao Zheng,Ming Lu,Lihong Wu,Huade Zheng

Angiogenesis within the wound site is a necessary prerequisite for skin tissue regeneration during the process of wound healing. Skin vessels provide adequate nutrients and soluble bioactive factors to the wounded area and remove metabolites. In this study, we prepared a double-cross-linked hydrogel based on GelMA and PVAMA to load Myeloid-derived growth factor (MYDGF). Additionally, we evaluated the healing-promoting effect of the hydrogel using a full-thickness skin injury mouse model. Compared with the control and MYDGF-free hydrogels, the MYDGF-loaded hydrogels significantly increased the percentage of wound healing and promoted granulation tissue proliferation and epidermization. In vitro experiments have shown that MYDGF may augment the migration and angiogenesis of human umbilical vein endothelial cells (HUVECs) by upregulating the expression of phosphorylated ERK and phosphorylated P38. These findings indicate that MYDGF-loaded hydrogels promote wound healing by accelerating angiogenesis.

Preparation and Properties of Partial-Degradable ZrO2–Chitosan Particles–GelMA Composite Scaffolds

Yang Ji,Mengdie Hou,Jin Zhang,Meiqi Jin,Tianlin Wang,Huazhe Yang,Xiaodong Zhang

In the field of bone repair, the inorganic–organic composite scaffold is a promising strategy for mimicking the compositions of the natural bone. In addition, as implants for repairing load-bearing sites, an inert permanent bone substitute composites with bioactive degradable ingredients may make full use of the composite scaffold. Herein, the porous zirconia (ZrO2) matrix was prepared via the template replication method, and the partial degradable ZrO2–chitosan particles–GelMA composite scaffolds with different chitosan/GelMA volume ratios were prepared through the vacuum infiltration method. Dynamic light scattering (DLS) and the scanning electron microscope (SEM) were adopted to observe the size of the chitosan particles and the morphologies of the composites scaffold. The mechanical properties, swelling properties, and degradation properties of the composite scaffolds were also characterized by the mechanical properties testing machine and immersion tests. The CCK-8 assay was adopted to test the biocompatibility of the composite scaffold preliminarily. The results show that chitosan particles as small as 60 nm were obtained. In addition, the ratio of chitosan/GelMA can influence the mechanical properties and the swelling and degradation behaviors of the composites scaffold. Furthermore, improved cell proliferation performance was obtained for the composite scaffolds. Keywords:porous zirconia matrix;chitosan particles;GelMA;composite scaffold

HAMA/GelMA-based hydrogel microneedle patch via DLP 3D bioprinting for high-efficiency ISF extraction

Lujie Zhang,Qinghua Song,Peijing Yang,Zhanqiang Liu

Microneedle (MN) technology is a promising minimally invasive tool for interstitial skin fluid (ISF) extraction. Hydrogel MN patches enable efficient ISF extraction. However, the traditional polydimethylsiloxane (PDMS) photopolymerization method has limitations in morphology control, accuracy, and susceptibility to process parameter interference, affecting MN strength and functional consistency. To address these challenges, we propose a composite hydrogel MN patch based on Digital Light Processing (DLP) 3D bioprinting of hyaluronic acid methacryloyl (HAMA)/gelatin methacryloyl (GelMA) for high-efficiency ISF transdermal extraction. The structure and performance of the MNs were improved by optimizing the bio-ink ratio and printing parameters. Adjusting the ratio of GelMA and HAMA improved the cross-linking structure, optimizing the mechanical properties and swelling performance of the composite hydrogel. Optimizing the light intensity and exposure time ensured consistent MN morphology, resulting in well-defined and evenly distributed needle tips. Controlled UV exposure avoided defects such as uneven cross-linking or brittleness. The results showed that when the HAMA-to-GelMA ratio was 8:2, the light intensity was 9 mW/cm 2 , and the exposure time was 6 s, the MN patches demonstrated excellent microstructures and mechanical properties, with each MN enduring over 1.28 N and achieving a swelling rate above 1200 %. In vitro experiments verified effective penetration and ISF extraction, obtaining 8.5 mg of PBS in 1 min and successfully quantifying glucose concentration. In summary, this study establishes a robust MN patch with superior swelling performance and enhanced ISF extraction efficiency and provides a promising platform for minimally invasive diagnosis and real-time health monitoring.

Predicting rheological properties of HAMA/GelMA hybrid hydrogels via machine learning

Bincan Deng,Sibai Chen,Fernando López Lasaosa,Xuan Xue,Chen Xuan,Hongli Mao,Yuwen Cui,Zhongwei Gu,Manuel Doblare

- Rheological properties are pivotal in determining the printability of biomaterials, directly impacting the success of 3D bioprinted constructs. Understanding the intricate relationship between biomaterial formulations, rheological behavior and printability can facilitate the advancement and rapid development of biomaterials. Herein, we critically measured the rheological properties of hyaluronic acid methacrylate (HAMA)/gelatin methacrylate (GelMA) hybrid hydrogels with varied formulations and generated a dataset to train a machine learning (ML) model. By utilizing four well-known algorithms, we developed the ML model for the viscosity and shear stress of HAMA/GelMA hydrogel mixtures. To improve model interpretability, we further created a multilayer perceptron framed model, known as HydroThermoMLP, by incorporating the Redlich-Kister polynomial as the thermodynamic representation of viscosity of mixtures. To accomplish the MLP learning on limited data, the shared loss function was formulated on the basis of the R-K presentation to guide the joint training process. The established HydroThermoMLP model, while maintaining the same accuracy as Random Forest, produces outputs that adhere to thermodynamic constraints and instill confidence in generalization applications with a simple algorithm informed by the R-K polynomial. It presents a robust predictive ML tool to forecast the viscosity of hybrid hydrogels and direct the design of biomaterials while appropriately abiding by thermodynamic constraints as essential guidelines.

GelMA/κ-carrageenan double-network hydrogels with superior mechanics and biocompatibility

Xueqi Gan,Chen Li,Jiyu Sun,Xidan Zhang,Min Zhou,Yi Deng,Anqi Xiao

Hydrogels are crosslinked hydrophilic polymer networks of high-water content. Although they have been widely investigated, preparing hydrogels with excellent mechanical properties and biocompatibility remains a challenge. In the present work, we developed a novel GelMA/κ-carrageenan (GelMA/KC) double network (DN) hydrogel through a dual crosslinking strategy. The three-dimensional (3D) microstructure of KC is the first network, and covalently crosslinked on the κ-carrageenan backbone is the second network. The GelMA/KC hydrogel shows advantages in physical properties, including higher compression strength (10% GelMA/1% KC group, 130 kPa) and Young's modulus (10% GelMA/1% KC group, 300), suggesting its excellent elasticity and compressive capability. When using a higher concentration of GelMA, the hybrid hydrogel has even higher mechanical properties. In addition, the GelMA/KC hydrogel is favorable for cell spreading and proliferation, demonstrating its excellent biocompatibility. This study provides a new possibility for a biodegradable and high-strength hydrogel as a new generation material of orthopedic implants.

Photo-crosslinked bioactive BG/BMSCs@GelMA hydrogels for bone-defect repairs

Yufeng Ai,Fang Dai,Wenfeng Li,Fancheng Xu,Hanwen Yang,Jianxin Wu,Kaiqiang Yang,Li Li,Fanrong Ai,Li Song

The clinical treatments of bone defects remain a challenge. Hydrogels containing bone marrow mesenchymal stem cells (BMSCs) are extensively used to bone regeneration because of excellent biocompatibility and hydrophilicity. However, the insufficient osteo-induction capacity of the BMSC-loaded hydrogels limits their clinical applications. In this study, bio-active glass (BG) and BMSCs were combined with gelatin methacryloyl (GelMA) to fabricate composite hydrogels via photo-crosslinking, and the regulation of bone regeneration was investigated. In vitro experiments showed that the BG/BMSCs@GelMA hydrogel had excellent cytocompatibility and promoted osteogenic differentiation in BMSCs. Furthermore, the BG/BMSCs@GelMA hydrogel was injected into critical-sized calvarial defects, and the results further confirmed its excellent angiogenetic and bone regeneration capacity. In addition, BG/BMSCs@GelMA promoted the polarization of macrophages towards the M2 phenotype. In summary, this novel composite hydrogel demonstrated remarkable potential for application in bone regeneration due to its immunomodulatory, excellent angiogenetic as well as osteo-induction capacity.

DLP-printed GelMA-PMAA scaffold for bone regeneration through endochondral ossification

Jianpeng Gao,Hufei Wang,Ming Li,Zhongyang Liu,Junyao Cheng,Xiao Liu,Jianheng Liu,Xing Wang,Licheng Zhang

Intramembranous ossification (IMO) and endochondral ossification (ECO) are two pathways of bone regeneration. The regeneration of most bone, such as limb bone, trunk bone, and skull base bone, mainly occurs in the form of endochondral ossification, which has also become one of the effective ways for bone tissue engineering. In this work, we prepared a well-structured and biocompatible methacrylated gelatin/polymethacrylic acid (GelMA/PMAA) hydrogel by digital light processing (DLP) printing technology, which could effectively chelate iron ions and continuously activate the hypoxia-inducible factor-1 alpha (HIF-1α) signaling pathway to promote the process of endochondral ossification and angiogenesis. The incorporation of PMAA endowed the hydrogel with remarkable viscoelasticity and high efficacy in chelation of iron ions, giving rise to the activation of HIF-1α signaling pathway, improving chondrogenic differentiation in the early stage, and facilitating vascularization in the later stage and bone remodeling. Therefore, the findings have significant implications on DLP printing technology of endochondral osteogenesis induced by the iron-chelating property of biological scaffold, which will provide an effective way in the development of novel bone regeneration.

3D printing of GelMA/nanohydroxyapatite/melanin nanoparticles composite hydrogel scaffolds for bone regeneration through immunomodulation

Meng Huang,Luyue Zhang,Jiantong Cui,Min Zhang,Zhaoxiang Wang,Shuhao Yu,Fangchong Du,Ziyan An,Lulu Xu,Junkai Cao

Addressing bone tissue defects is a critical challenge in clinical practice, necessitating the development of biomaterials that can orchestrate both immune modulation and tissue regeneration. This study introduces and assesses the immunomodulatory effects and bone repair capabilities of a novel 3D-printed scaffold composed of gelatin methacryloyl (GelMA), nanohydroxyapatite, and melanin nanoparticles (GHM). The GHM scaffolds, characterized by their optimal porosity, viscosity, and mechanical strength, have been shown to effectively direct macrophage polarization from the initial M0 state to the anti-inflammatory M2 phenotype. Concurrently, osteogenic precursor cell lines MC3T3 are stimulated to differentiate into osteoblasts under the influence of macrophage-conditioned medium. In vivo studies using normal mice cranial defect models and macrophage-depleted cranial defect models have demonstrated that GHM scaffolds can attract macrophages to the implantation site, promote their M2 polarization, and consequently, significantly enhance bone formation and effectively treat cranial defects in mice. RNA-sequencing analysis has revealed elevated expression of the Leukemia inhibitory factor (Lif) gene in macrophages treated with GHM, implicating its role in regulating macrophage polarization. These findings underscore the potential of GHM scaffolds as an immunomodulatory biomaterial for bone tissue engineering applications.

3D printing of biomimetic multi-layered GelMA/nHA scaffold for osteochondral defect repair

Jingyi Liu,Liang Li,Hairui Suo,Mengling Yan,Jun Yin,Jianzhong Fu

Currently, osteochondral defects frequently cause limited motion and impaired function of the joint, leading to serious healthcare problems, and it is still very challenging to realize the simultaneous regeneration of subchondral bone with cartilage. In the current study, we designed a tri-layered scaffold and fabricated it using the extrusion-based multi-nozzle 3D printing technology. The bioinks used for 3D printing included a 15% methacrylated gelatin (GelMA) hydrogel for cartilage on top layer, a combination of 20% GelMA and 3% nanohydroxyapatite (nHA) (20/3% GelMA/nHA) hydrogel for interfacial layer, and a 30/3% GelMA/nHA hydrogel for subchondral bone at bottom layer. The water absorption capacity, biodegradation, and mechanical properties of hydrogels and scaffolds were characterized, and in vitro assay with bone marrow mesenchymal stem cells (BMSCs) was performed to indicate the biocompatibility of scaffolds. Based on the results of in vivo repair of rabbit osteochondral defect, the neo-tissues in defects integrated better with the surrounding tissues, the joint surface of the defects was smoother, and more cartilage-specific extracellular matrix and collagen type II were observed using the tri-layered scaffolds. This study not only provides a potential manufacturing method for multi-layered scaffolds, but also is helpful for understanding the regeneration mechanism of cartilage-subchondral bone.

Preparation and antibacterial properties of an AgBr@SiO2/GelMA composite hydrogel

Bo Li,Hong Li,Haocheng Yang,Yue Shu,Kejiang Li,Ke Chen,Wenqian Xiao,Xiaoling Liao

Pure gelatin hydrogels lack antibacterial function and have poor mechanical properties, which restrict their application in wound dressings. In this study, nanosized silver bromide-doped mesoporous silica (AgBr@SiO2) microspheres with hollow structures were prepared by a modified Stober method. The novel microspheres can not only release silver ions to treat bacteria but also release drugs to treat skin wound. Furthermore, AgBr@SiO2 microspheres were modified with propyl methacrylate, incorporated into methacrylated gelatin (GelMA), and crosslinked by UV light to prepare AgBr@SiO2/GelMA dressings consisting of composite hydrogels. The results showed that the AgBr@SiO2 microspheres could enhance the mechanical properties of the hydrogels. With the increase in the AgBr@SiO2 concentration from 0.5 to 1 mg ml−1, the dressings demonstrated effective antimicrobial activity against both Staphylococcus aureus and Escherichia coli. Furthermore, full-thickness skin wounds in vivo wound healing studies with Sprague–Dawley rats were evaluated. When treated with AgBr@SiO2/GelMA containing 1 mg ml−1 AgBr@SiO2, only 15% of the wound area left on day 10. Histology results also showed the epidermal and dermal layers were better organized. These results suggest that AgBr@SiO2/GelMA-based dressing materials could be promising candidates for wound dressings.

Biodegradable double-network GelMA-ACNM hydrogel microneedles for transdermal drug delivery

Wensheng Lin,Shixian Lin,Xingwu Zhou,Fanwen Yang,Zishan Lin,Shiqing Li,Haoyuan Zhang,Yuehan Ouyang,Jieying Zhu,Wei Sun,Dequn Huang,Baojian Liao,Jixiang Zhu

As a minimally invasive drug delivery platform, microneedles (MNs) overcome many drawbacks of the conventional transdermal drug delivery systems, therefore are favorable in biomedical applications. Microneedles with a combined burst and sustained release profile and maintained therapeutic molecular bioactivity could further broaden its applications as therapeutics. Here, we developed a double-network microneedles (DN MNs) based on gelatin methacrylate and acellular neural matrix (GelMA-ACNM). ACNM could function as an early drug release matrix, whereas the addition of GelMA facilitates sustained drug release. In particular, the double-network microneedles comprising GelMA-ACNM hydrogel has distinctive biological features in maintaining drug activity to meet the needs of application in treating different diseases. In this study, we prepared the double-network microneedles and evaluated its morphology, mechanical properties, drug release properties and biocompatibility, which shows great potential for delivery of therapeutic molecules that needs different release profiles in transdermal treatment.
