Gleditsia sinensis saponins (GS) is a biodegradable nonionic surfactant extracted from the plant pericarp. The objective of this study is to evaluate its potential for application in cleaning products in response to resource and environmental changes. Here, high-purity GS (93.92 %) was obtained through the macroporous resin adsorption technique and mixed with sodium lauroyl methylaminopropionate (LMA) to form a Gleditsia sinensis saponins- sodium lauroyl methylaminopropionate (GS-LMA) binary system. The synergistic effect was clarified by calculating the intermolecular interactions and thermodynamic parameters of the mixed micelles. The surface activity, foam stability, and emulsification characteristics were investigated to assess the detergent potential of the GS-LMA mixture. The results indicated that the formation of GS-LMA mixed micelles followed a non-ideal, spontaneous exothermic process. Furthermore, the addition of GS decreased the critical micelle concentration of the mixture while enhancing foam stability (98.34 %) and prolonging the oil–water separation time (103.11 min). The synergistic effects of foaming and emulsification enhanced the cleaning efficiency of the mixtures to 90.60 %. Further studies showed the mixture can reduce oil–water interfacial tension and form stable emulsions in various environments. This work can provide a theoretical basis for a better understanding the synergistic effects of binary surfactants and interfacial science.

Galactomannan comes from a wide range of plant resources and has some biological activities, but its bioavailability is limited due to its large molecular weight and complex structure. In this study, three degradation methods (H 2 O 2 , ultrasound, and β-mannanase) combined with ethanol fractional precipitation (25 %, 50 %, and 75 %) were used to degrade and separate Gleditsia sinensis galactomannans (GSG), and the physicochemical properties and biological activities of GSG after degradation were analyzed. Comprehensive comparison indicates that H 2 O 2 exhibits had a better degradation effect. After 4 h of degradation using 4 % H 2 O 2 , the yield of GSG precipitated with 50 % ethanol was 37.06 % (the yield of undigested GSG is 1.80 %). Simultaneously, the molecular weight (reduced from 225.25 to 36.87 kDa) and viscosity were significantly reduced under this condition, while the solubility was increased. In addition, the low-molecular-weight GSG (LGSG) obtained by 4 % H 2 O 2 /50 % ethanol showed the strongest free radical scavenging activity in vitro . Furthermore, the results of in vivo antioxidant assays showed that LGSG inhibited Aflatoxin B1-induced developmental toxicity by regulating gene expression in the Keap1 / Nrf2 pathway. LGSG also promoted Nrf2 -mediated expression of the lipid metabolism genes ppar-α and cpt1 , while suppressing expression of the fatty acid synthesis genes fas and scd-1 . Therefore, the liver recovered from lipid peroxidation induced nonalcoholic fatty liver disease (NAFLD). The present study introduces a method for green and efficient preparation of LGSG, indicates its potential as a nutritional product.

The self-healing property based on metal-ligand physical coordination is particularly interesting in bio-hydrogel science due to its allowance for multiple local healing events to process. As the most abundant renewable green resource in nature, Gleditsia sinensis galactomannan has great potential as a starting material for functional materials. In this study, the biocompatible Gleditsia sinensis galactomannan and cellulose were firstly chemically modified and then taken as the main constituent for constructing the metal-ligand coordination through an enzyme-regulated strategy. The hydrogel could quickly gelatinize in the surrounding environment, corresponding to the violent exothermic phenomenon, and exhibit extraordinary self-healing behavior. The molecular dynamics simulation of the hydrogel confirmed the more stable coordinated configuration from Fe(III)-chelates than Fe(II)-chelates. The morphology, mechanical property, antibacterial, and cytotoxicity of the prepared hydrogel were also studied. Our results indicated that galactomannan hydrogel based on the metal-ligand networks could balance the kinetic stability and intrinsic healability through the enzyme-induced route, which provide a new perspective in the field of biomaterial applications.

Inhibiting α-glucosidase activity by secondary metabolites has been confirmed as an effective strategy for treating type 2 diabetes mellitus (T2DM). α-Glucosidase inhibitory activity was determined by different parts of Gleditsia sinensis Lam. (GSL) extracts. The thorn of the GSL methanol extract exhibited the highest activity (IC 50 = 0.21 mg/mL). Eleven potential α-glucosidase inhibitors were detected by Ultra Performance Liquid Chromatography Quadrupole Time of Flight Mass Spectrometry, and fisetin was screened as the dominant compound (IC 50 = 0.211 µM) through virtual screening, correlation analysis, and inhibitory assay. Fluorescence quenching experiment explained that fisetin inhibited α-glucosidase through hydrogen bonds and hydrophobic forces in a reversible and non-competitive mode. Three-dimensional fluorescence and circular dichroism demonstrated that fisetin modified the amino acids’ microenvironment and disrupted the hydrogen-bonded network of α-glucosidase. α-Glucosidase was bounded with fisetin by hydrogen (Thr448, His515, Asp346, and Gln531) and hydrophobic residues (His515, Lys352, and Ala349) simulated by molecular docking. This study points out the fisetin’s inhibitory mechanism of α-glucosidase, indicating that it is a promising natural ingredient in T2DM treatment.

In the context of contemporary health, the integration of functional foods that possess diverse biological activities has been garnering increasing attention. Xylo-oligosaccharide (XOS), as a prominent candidate among prebiotic carbohydrates, have attracted particular interest. In this study, the hydrolysis of Gleditsia sinensis Lam. peel (GSP) was compared using malic acid (MA), glutamic acid (GA), pyruvic acid (PYA), benzoic acid (BA), and acetic acid (AA) for the production of XOS. The results showed that PYA had a potential for low-temperature hydrolysis, yielding 57.74 % XOS and 9.12 % xylose at 130 °C, without harmful by-products. It also had higher selectivity for the degradation of hemicellulose. Significantly, MA demonstrated the highest hydrolysis efficiency, achieving a maximum XOS yield of 69.16 % at 150 °C. The types of organic acids alter the composition and properties of XOS, leading to differences in its biological activity. The XOS produced by glutamic acid (GAX) demonstrated the highest prebiotic activity by enhancing the growth and metabolism of Bifidobacterium adolescentis(B. adolescentis) , which is closely linked to the supportive role of GA. Additionally, the XOS produced by PYA (PAX) and BA (BAX) manifested promising hypoglycemic and antioxidant effects, respectively. This research deepens our understanding of various organic acids pretreatment modes for XOS production patterns and their potential applications in functional oligosaccharides.

Traditional maceration extraction methods for saponins typically involve substantial consumption of organic solvents and long extraction times. To establish an efficient and eco-friendly extraction method for the saponins in Gleditsia sinensis Lam. pods (GSP), deep eutectic solvents (DESs), serving as a sustainable alternative to traditional solvents, have been investigated in conjunction with ultrasound-assisted extraction (UAE) technology. The DES composed with choline chloride and lactic acid in a 1:2 molar ratio was selected based on the highest extraction yield of Gleditsia sinensis Lam. saponins (GSS). The extraction parameters were further optimized using a single factor and response surface methodology (RSM). The maximum GSS extraction yield (183.61 mg/g) was achieved using water content 32%, liquid–solid ratio 35 mL/g, ultrasonic power 427 W, and extraction time 12 min. The structural features of GSS were investigated using ultra-performance liquid chromatography-quadrupole-time-of-flight mass spectrometry, and a total of 16 saponins were tentatively identified. The physicochemical and surface properties of GSS were also investigated. It turns out that GSS solution belongs to pseudoplastic fluid, and the viscosity of GSS solution varied at different pH values. Surface activity analysis showed GSS had a low critical micelle concentration at 0.0161 g/L, and GSS could reduce surface tension to 43.7 mN/m. The foaming and emulsifying evaluations revealed that GSS were an efficient natural surfactant as it could form stable foams and emulsions at low concentrations as compared with Tween 80. Moreover, the antioxidant activities of GSS were studied using antioxidant assay kits, and GSS showed strong capacities in scavenging free radicals. This study reveals the screened DESs coupled with UAE could efficiently separate saponins from GSP, and GSS showed the potential to be applied as natural surfactant or antioxidant in food industry.

Gleditsia sinensis is a major crop for the extraction of triterpenoid saponins, which peel waste is rich in oxygenated functional groups and considered an ideal raw material of modified biochar. In this work, iron oxide/biochar composites were prepared using Gleditsia sinensis peel as raw material and iron oxide. The adsorption capacity of the modified Gleditsia sinensis biochar was significantly enhanced with the highest adsorption rate of 89.5% for methylene blue (MB), while the highest adsorption removal rate for Cr (VI) was close to 70%. The possible adsorption mechanism was investigated by quantitative computational methods combined with density functional theory (DFT) and adsorption thermodynamic and adsorption kinetic studies. The adsorption process of pollutants by biochar is consistent with the Freundlich isotherm model and the quasi-secondary kinetic model, performed spontaneously and absorbs heat. This study prepared a recyclable biomass-based adsorbent, which provides a new option for the high-value utilization of forest residues.

Carboxyl group number (CGN) in hydroxy acids serves as a molecular design parameter. • Lignin condensation is minimized by tuning CGN, preserving its antioxidant structure. • A CGN-dependent steric-electronic synergy mechanism prevents XOS side reactions. • Integrated production of prebiotic XOS, glucose, and antioxidant lignin is achieved. • A green, scalable biorefinery was developed to advance the circular bioeconomy. This study proposes a sustainable biorefinery strategy for converting waste Gleditsia sinensis pods into high-value fine chemicals using hydroxy acids, with the carboxyl group number (CGN) serving as a critical molecular design parameter. The results showed that increasing CGN changes the composition of XOS, increases XOS yield, promotes glucose conversion, and protects lignin structure. The highest xylooligosaccharide (XOS) yield of 62.88% was achieved with citric acid pretreatment (CGN = 3), which reduced furfural formation by 43.8% compared to lactic acid (CGN = 1), resulting in 98.05% glucose conversion. The lignin structure remained largely intact, with a retention rate of 53.04% per 100 Ar of β-O-4′ bonds and phenolic hydroxyl content of 4.36 mmol/g. This preserved lignin structure enhanced antioxidant activity (75.38%) in DPPH radical scavenging compared to BHT. Besides, XOS produced via acid pretreatment at 150 °C for 30 min showed enhanced prebiotic effects, promoting the growth of Bifidobacterium adolescentis by 53% and increasing propionic acid production by 300% relative to commercial fructooligosaccharides. Structural analyses indicated that CGN protects lignin through intramolecular hydrogen bonding and chelation, thereby suppressing condensation. Overall, this environmentally sustainable biorefinery approach efficiently converts agricultural waste into high-value products, supporting advances in the food, feed, and materials sectors. The seeds from Gleditsia sinensis Lam., a common ecologically and economically useful tree, have high economic and nutritional value. The protective effect of polysaccharides from Gleditsia sinensis Lam. seeds (ZJMP) against dextran sulfate sodium-induced colitis in mice was investigated in this study. ZJMP alleviated weight loss, reduced the disease activity index, prevented colon shortening, alleviated colonic tissue damage, and restored goblet cell secretion in colitic mice. Dietary ZJMP reduced proinflammatory cytokine overproduction in the colonic mucosa and serum, which was accompanied by suppression of NO levels and MPO and SOD activities. The addition of ZJMP increased the expression of Muc2 and tight junction proteins. Furthermore, dietary ZJMP partially reversed the alteration of gut microbiota in colitic mice by boosting the abundance of beneficial bacteria like Akkermansia, Lactobacillus, and Christensenella while lowering the abundance of harmful bacteria like Bacteroides, Prevotella, and Mucispirillum. Additionally, the decreased production of short-chain fatty acids in the colitic mice was recovered by ZJMP administration. The findings demonstrated the anti-inflammatory properties and mechanism of dietary ZJMP in the colon, which is essential for the sensible application of ZJMP in the prevention and amelioration of inflammation-related diseases as a nutritional supplement.
