The preparation conditions of Glehniae Radix polysaccharide-iron complex (GLP80-Fe) were optimized and the physicochemical properties were characterized by Ultraviolet-visible spectroscopy (UV-Vis), Fourier Transform Infrared Spectroscopy (FT-IR), X-ray diffraction (XRD), Transmission electron microscope (TEM), Thermogravimetric analyzer, and Differential scanning calorimetry (TGA-DSC), respectively. The stability and reducibility of GLP80 and GLP80-Fe were analyzed, and the monosaccharide composition was determined by gas chromatograph (GC). In vitro antioxidant activity of GLP80 and GLP80-Fe were evaluated by determining the radical (DPPH and hydroxyl radical) scavenging abilities. The results showed that the highest iron content of GLP80-Fe (15.61%±0.04) was obtained at the optimum conditions: reaction temperature was 38.0°C, reaction time was 3.0 h, FeCl3 addition was 2.9 mL and pH was 9. GLP80-Fe was a complex with β-FeOOH as the core and polysaccharides on its surface, and it was stable at pH 3 ~ 14. GC showed that the major monosaccharides were mannose (Man) and galactose (Gal) with a ratio of 1:5.15. In vitro free-radical scavenging activity of GLP80-Fe was significantly better than GLP80. The IC50 of GLP80-Fe against DPPH and hydroxyl radical were 3.785 mg/mL and 1.460 mg/mL, respectively.

Glehniae Radix (GR) is one of the major medicinal materials in China. The global demand for GR, especially in Asian countries, is constantly increasing, and the supply of wild medicinal materials falls short of the demand. Previous studies have shown that the production and processing modes of different value chains (VCs) impact the quality of medicinal materials. After 4 years of field and market research, this study includes interviews with stakeholders in the VCs, integrates different types of VCs, and further analyzes the VCs. GR characteristics were also assessed; the length and upper-middle diameter of the collected samples were measured, and the effective components of the samples were determined to rank the GR samples according to their quality. The effective components were further analyzed by the K-means clustering method. Concomitantly, the local price (the sales price of the place where the medicinal materials are produced) and market price (the sale price of medicinal materials in the market) of GR in Chifeng, Inner Mongolia, and Anguo, Hebei, were documented, and the ARIMA (Autoregressive Integrated Moving Average) method was used to predict the GR price. Ten VCs are summarized in this article. The results showed that the income of the staff at the beginning of the VC is inadequate. Regarding GR origin, Inner Mongolia GR showed higher quality than that of other areas. As a result, the price of medicinal materials is relatively high, which corresponds to the market price of the survey. The forecast results showed that the market price of GR would increase slightly in the future, which could provide reference for the selection of medicinal materials cultivation in the future. Through the study, it was found that the vertical integration in the VCs of GR could guarantee not only the benefit of the growers but also the traceability of the medicinal materials, which further guarantees the quality of the medicinal materials. However, the complex relationship between the cultivation area and the quality of the medicinal materials is not clear, which should be addressed in future research.

Glehniae Radix (GR) primarily produced in Laiyang and Chifeng. It is a functional food with therapeutic and health-promoting effects due to its antioxidant and anti-inflammatory properties. However, the current standards only stipulate authentication criteria, without establishing a comprehensive evaluation framework to systematically enhance the quality control of GR. This study aimed to establish a "chemical-mechanism" quality control framework for GR through a three-tiered strategy: "chemical composition analysis - bioactivity quantification - mechanism network validation." Initially, UPLC fingerprints of 14 GR batches were constructed, and their antioxidant and anti-inflammatory activities were evaluated. Multivariate statistical models combined with activity indices were employed to optimize spectrum-effect relationships using a random forest algorithm, predicting bioactive constituents. Network pharmacology, molecular docking, and dynamics simulations were integrated to elucidate multi-component, multi-target, and multi-pathway mechanisms. Chemometric analysis identified Imperatorin as a marker for quantifying differences between Laiyang and Chifeng origins. Pharmacodynamic assays confirmed GR's antioxidant and anti-inflammatory efficacy. Random Forest-optimized spectrum-effect modeling prioritized falcarindiol and panaxynol as bioactive markers. Network pharmacology predicted GR's regulation of EGFR tyrosine kinase inhibitor resistance and ErbB signaling via targets (EGFR, PTGS2, NFKB1). Molecular docking revealed strong binding affinity (e.g., imperatorin-EGFR: -9.046 kcal/mol), and dynamics simulations confirmed complex stability. Thus, imperatorin, falcarindiol, and panaxynol were prioritized as quality markers. This study establishes a data-driven framework linking GR's chemical composition to antioxidant/anti-inflammatory mechanisms, advancing batch consistency control and precision applications in clinical disorders.
