Biodesulfurization of model oil using growing cells of Gordonia sp. SC-10

Shuiquan Chen,Shuo Sun,Chaocheng Zhao,Qiyou Liu,Meng Zang

The biodesulfurization capacity of Gordonia sp. SC-10 in a biphasic reaction system was investigated in this study. Model oil containing dibenzothiophene was used as the sole sulfur source of bacterium. The results showed that 81.43% of the sulfur in model oil with initial sulfur content of 174 mg/l could be removed within 5 days. Gordonia sp. SC-10 had high desulfurization rate for model oils with low sulfur contents, which would reduce sulfur concentration to below 5 mg/l. Biodesulfurization process might be a potential method to obtain fuels with ultra-low sulfur contents.

Efficient biodesulfurization of diesel oil by Gordonia sp. SC-10 with highly hydrophobic cell surfaces

Shuiquan Chen,Meng Zang,Lin Li,Juntao Chen,Qiyou Liu,Xiwen Feng,Shuo Sun,Chuanwei Zang,Chaocheng Zhao

Biodesulfurization is a promising, environmentally friendly method to desulfurize the organosulfur compounds in crude oils through the highly selective cleavage of carbon-sulfur bonds under mild reaction conditions. In this study, the behavior of a hydrophobic bacterium, Gordonia sp. SC-10, was evaluated in the presence of diesel oils. The biodesulfurization pathways, bacterium hydrophobicity , and cell ultrastructure were investigated, and the capability of strain SC-10 to desulfurize diesel oil was also studied. The results showed that strain SC-10 can desulfurize organosulfur compounds through sulfur-specific desulfurization pathways by producing corresponding hydroxylated products in oil-water polyphasic systems. Mycolic acids consisting of 47 and 58 carbon atoms chains were produced on the cell surface, and inclusion bodies were produced in the cells, both of which may improve the bioavailability of the substrates, thereby enhancing its adaptability to oil-water polyphasic systems. To our knowledge, this is the first report of intracellular inclusion body formation during biodesulfurization. Strain SC-10 also exhibited broad substrate specificity towards organosulfur compounds and effectively desulfurized diesel oils. The sulfur content in real-life diesel oil was reduced from 167.7 mg/L to 19.7 mg/L, with 88.3% of sulfur removed after five days of biodesulfurization. This study showed that Gordonia sp. SC-10 can be effectively used for the biodesulfurization of diesel oils.

Biodegradation of di-n-octyl phthalate by Gordonia sp. Lff and its application in soil

Yangyang Wang,Qiang Ren,Wenhao Zhan,Kaixuan Zheng,Qi Liao,Zhihui Yang,Yansong Wang,Xinling Ruan

A previous isolated Gordonia sp. (Lff) was used to degrade di-n-octyl phthalate (DOP) contamination in both aqueous solution and soil. The influence of temperature, pH, inoculum size, salt content and initial concentration of DOP on DOP degradation by Lff were analysed. The response of soil bacterial community to DOP and Lff was also analysed by Illumina MiSeq sequence method. Results showed that the optimal temperature, pH, inoculum size and salt content were 35oC, 8.0, 5% and <5%, respectively. Under the optimal condition, more than 91.25% of DOP with different initial concentrations (100–2000 mg/L) could be degraded by Lff. Kinetics analysis indicated that biodegradation of DOP by Lff could be described by first-order kinetics (R2 > 0.917) with the half-life (t1/2) changing irregularly between 0.58 and 0.83 d. In addition, Lff enhanced the removal of DOP in soil and alleviated the toxicity of DOP on soil microorganisms. Furthermore, its influence on soil bacterial community is not obvious. These results suggested that Lff was effective in remediating DOP contamination in different environments.

Biodegradation of di-(2-ethylhexyl) phthalate by a halotolerant consortium LF

Fangfang Li,Yidan Liu,Diwei Wang,Chaosheng Zhang,Zhihui Yang,Siqi Lu,Yangyang Wang

A halotolerant bacterial consortium capable of degrading di-(2-ethylhexyl) phthalate (DEHP) was enriched from activated sludge. Community analysis revealed that LF contained seven families and seven genera of bacteria. The predominant species was Gordonia sp. (54.93%), Rhodococcus . sp. (9.92%) and Achromobacter sp. (8.47%). The consortium could degrade 93.84% of 1000 mg/l DEHP after 48 h incubation. The optimal temperature and pH for LF to degrade DEHP were 30 °C and 6.0, respectively. LF degraded more than 91% of DEHP with salt concentrations ranging from 0–3%. The inoculum size had great effects on DEHP degradation (incubation time < 24h). LF could degrade high concentrations of DEHP (from 100 to 2000 mg/l) with the degradation ratio above 92% after 72 h incubation. Kinetics analysis revealed that the degradation of DEHP by LF was best fitted by the first-order kinetics when the initial concentration ranged from 100 to 2000 mg/l. The main intermediates (2-ethylhexyl pentyl phthalate, butyl (2-ethylhexyl) phthalate (BEHP), mono-ethylhexyl phthalate (MEHP), mono-hexyl phthalate (MHP), mono-butyl phthalate (MBP)) in DEHP degradation process were identified using gas chromatography–mass spectrometry (GC-MS), and a new complex biochemical pathway was proposed. Furthermore, LF could also degrade dimethyl phthalate (DMP), diethyl phthalate (DEP), di- n -butyl phthalate (DBP), di- n -octyl phthalate (DOP) and phthalic acid (PA).

The Efficient PAE-Degrading Performance and Complete Genome Sequencing of Gordonia sp. LUNF6

Shuanghu Fan,Zihan Feng,Meiting Xu,Zhenxia Shi,Yufeng Zhang,Peipei Zhang,Xiaoqiang Hou

Phthalic acid esters (PAEs) are mainly used as plasticizers and result in serious environmental contamination worldwide. Microbial biodegradation becomes an efficient strategy for PAE elimination. In the current study, the PAE-degrading strainGordoniasp. LUNF6 was isolated from contaminated soil. Strain LUNF6 can efficiently degrade DBP in a wide range of temperatures, pH values, and salinity levels. This strain is also capable of degrading 11 types of PAEs and displays remediation potential in wastewater. The complete genome of strain LUNF6 was sequenced to determine its efficient degradation performance. Its genome comprises a chromosome (3,971,257 bp) and a plasmid (78,813 bp). After gene function annotation, the complete PAE degradation pathway was proposed. The gene of monoalkyl PAE hydrolase MphGd2 was cloned and heterologously expressed. The protein of MphGd2 was purified by infinity chromatography, and we hydrolyzed MBP to produce PA. These results reveal the molecular mechanism of PAE degradation by strain LUNF6, which will contribute to the application of strain LUNF6 and hydrolase MphGd2 in bioremediation.

Biodegradation and metabolic pathway of phenanthrene by a newly isolated bacterium Gordonia sp. SCSIO19801

Zhimao Mai,Lin Wang,Qiqi Li,Yingting Sun,Si Zhang

The bacterium Gordonia sp. SCSIO19801, which could effectively utilize phenanthrene as the sole carbon source, was isolated from the seawater of the South China Sea. Its biodegradation characteristics, whole genome sequence, and biodegradation pathway were investigated. The phenanthrene biodegradation process of Gordonia sp. SCSIO19801 was estimated to be a first-order kinetic model with a k value of 0.26/day. Based on the identification of metabolites, utilization of probable intermediates, and genomics analysis of related genes, the degradation of phenanthrene by Gordonia sp. SCSIO19801 was proposed to occur via the salicylate metabolic pathway. This is the first report of a phenanthrene degradation pathway in Gordonia species. In addition, the Gordonia sp. SCSIO19801 could use other aromatic compounds as the sole source of carbon and energy. These characteristics indicate that Gordonia sp. SCSIO19801 can be utilized for developing effective methods for the biodegradation of petroleum hydrocarbons in marine environments.

Phthalate Esters Metabolic Strain Gordonia sp. GZ-YC7, a Potential Soil Degrader for High Concentration Di-(2-ethylhexyl) Phthalate

Tong Hu,Chen Yang,Zhengyu Hou,Tengfei Liu,Xiaotong Mei,Lianbao Zheng,Weihong Zhong

As commonly used chemical plasticizers in plastic products, phthalate esters have become a serious ubiquitous environmental pollutant, such as in soil of plastic film mulch culture. Microbial degradation or transformation was regarded as a suitable strategy to solve the phthalate esters pollution. Thus, a new phthalate esters degrading strainGordoniasp. GZ-YC7 was isolated in this study, which exhibited the highest di-(2-ethylhexyl) phthalate degradation efficiency under 1000 mg/L and the strongest tolerance to 4000 mg/L. The comparative genomic analysis results showed that there exist diverse esterases for various phthalate esters such as di-(2-ethylhexyl) phthalate and dibutyl phthalate inGordoniasp. GZ-YC7. This genome characteristic possibly contributes to its broad substrate spectrum, high degrading efficiency, and high tolerance to phthalate esters.Gordoniasp. GZ-YC7 has potential for the bioremediation of phthalate esters in polluted soil environments. Keywords:PAEs;comparative genomics analysis;DEHP;bioremediation

Synthetic bacterial consortia enhanced the degradation of mixed priority phthalate ester pollutants

Tengfei Liu,Lixiao Ning,Chengyu Mei,Shuang Li,Lianbao Zheng,Pei Qiao,Haixia Wang,Tong Hu,Weihong Zhong

Dimethyl phthalate (DMP), diethyl phthalate (DEP), di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), bis(2-ethylhexyl) phthalate (DEHP), and di-n-octyl phthalate (DOP) are hazardous chemicals listed as priority pollutants that disrupt endocrine systems. According to available reports, these six priority phthalate esters (PAEs) are considered the most polluting; however, no studies have been conducted on the efficient remediation of these PAEs. We therefore designed and constructed a synthetic bacterial consortium capable of the simultaneous and efficient degradation of six priority PAEs in minimal inorganic salt medium (MSM) and soil. The consortium comprised Glutamicibacter sp. ZJUTW, which demonstrates priority for degrading short-chain PAEs; Cupriavidus sp. LH1, which degrades phthalic acid (PA) and protocatechuic acid (PCA), intermediates of the PAE biodegradation process; and Gordonia sp. GZ-YC7, which efficiently degrades long-chain priority PAEs, including DEHP and DOP. In MSM containing the six mixed PAEs (250 mg/L each), the ZJUTW + YC + LH1 consortium completely degraded the four short-chain PAEs within 48 h, and DEHP (100%) and DOP (62.5%) within 72 h. In soil containing the six mixed PAEs (DMP, DEP, BBP, and DOP, 400 mg/kg each; DBP and DEHP, 500 mg/kg, each), the ZJUTW + YC + LH1 consortium completely degraded DMP, DEP, BBP, and DBP within 6 days, and 70.84% of DEHP and 66.24% of DOP within 2 weeks. The consortium efficiently degraded the six mixed PAEs in both MSM and soil. We thus believe that this synthetic microbial consortium is a strong candidate for the bioremediation of environments contaminated with mixed PAE pollutants.

Biodegradation of di-(2-ethylhexyl) phthalate by a newly isolated Gordonia sp. and its application in the remediation of contaminated soils

Yangyang Wang,Wenhao Zhan,Qiang Ren,Shanshan Cheng,Junnan Wang,Xiaoyu Ma,Chaosheng Zhang,Yansong Wang

A bacterial strain ( Gordonia sp. Lff) capable of efficiently degrading di-(2-ethylhexyl) phthalate (DEHP) was isolated from river sludge. The optimal pH and temperature for the degradation of DEHP by Lff were 7.0 and 35 °C, respectively. Lff could degrade high concentrations of DEHP (100–2000 mg/L) with a degradation efficiency of over 91.43%. The DEHP degradation curves fit well with first-order kinetics, with a half-life ranging from 0.598 to 0.746 d. Substrate inhibition analyses showed that the maximum specific degradation rate, half-saturation constant and inhibition constant were 0.8 d −1 , 45.8 mg/L and 462.18 mg/L, respectively. A detailed biodegradation pathway of DEHP was proposed based on GC–MS analysis. Furthermore, Lff could also efficiently degrade DEHP in soils. DEHP or DEHP plus Lff changed the bacterial community in soils, and Lff accelerated the shaping of the bacterial community. To the best of our knowledge, this study is the first to perform a detailed investigation into the biodegradation of DEHP in soil by Gordonia sp. and its effect on the soil bacterial community. These results suggest that Lff is an ideal candidate for the bioremediation of DEHP-contaminated environments.
