Macropinocytosis in Gracilariopsis lemaneiformis (Rhodophyta)

Haihong Chen,Yiyi Hu,Guanpin Yang,Pingping Li,Jingru Yin,Xiaoqing Feng,Qiong Wu,Jingyu Zhang,Baoheng Xiao,Zhenghong Sui

Macropinocytosis is an endocytic process that plays an important role in animal development and disease occurrence but until now has been rarely reported in organisms with cell walls. We investigated the properties of endocytosis in a red alga, Gracilariopsis lemaneiformis . The cells non-selectively internalized extracellular fluid into large-scale endocytic vesicles (1.94 ± 0.51 μm), and this process could be inhibited by 5-(N-ethyl-N-isopropyl) amiloride, an macropinocytosis inhibitor. Moreover, endocytosis was driven by F-actin, which promotes formation of ruffles and cups from the cell surface and facilitates formation of endocytotic vesicles. After vesicle formation, endocytic vesicles could be acidified and acquire digestive function. These results indicated macropinocytosis in G. lemaneiformis . Abundant phosphatidylinositol kinase and small GTPase encoding genes were found in the genome of this alga, while PI3K, Ras, and Rab5, the important participators of traditional macropinocytosis, seem to be lacked. Such findings provide a new insight into endocytosis in organisms with cell walls and facilitate further research into the core regulatory mechanisms and evolution of macropinocytosis.

Characterization and bioactivities of a novel polysaccharide obtained from Gracilariopsis lemaneiformis

SHI CHEN-SHAN,SANG YA-XIN,SUN GUI-QING,LI TIAN-YE,GONG ZHENG-SI,WANG XIANG-HONG

Gracilariopsis lemaneiformis is a type of red alga that contains seaweed polysaccharide agar. In this study, a novel non-agar seaweed polysaccharide fraction named GCP (short of crude polysaccharide obtained from Gracilariopsis lemaneiformis) was isolated from Gracilariopsis lemaneiformis. Structural analysis showed that GCP shows triple helical chain conformation when dissolved in water and has many branches and long side chains. Also, 1→3 linkage is the major linkage and the sugar structures are galactopyranose configurations linked by β-type glycosidic linkages. Two macromolecular substance fractions (GCP-1 and GCP-2) were purified by DEAE Sepharose Fast Flow column chromatography. Moreover, a splenocyte damage assay and splenocyte proliferation assay were used to analyse the bioactivities of GCP, GCP-1 and GCP-2. It was demonstrated that polysaccharides could protect splenocyte damaged by H2O2; GCP-2 shows a greatest protection rate, that is, 92.8%, which significantly enhanced the splenocyte proliferation, and GCP showed the highest proliferation rate, 9.30%. The results suggested that this type of novel non-agar polysaccharide displayed remarkable antioxidant and immunomodulatory activities and early alkali treatment could decrease the activities. It may represent a potential material for health food and clinical medicines.

Widely targeted metabolomics reveals metabolic patterns during tetraspore formation in the edible seaweed Gracilariopsis lemaneiformis

Deguang Sun,Xueyou Zhou,Desheng Chen,Fengjie Liu,Bo Zhang,Xue Sun,Nianjun Xu

A sustainable food supply is crucial for addressing the food crisis resulting from the increasing global population. Seaweeds are promising food candidates because of their nutritional value and favorable growing conditions, as they do not occupy arable land or require fresh water. Gracilariopsis lemaneiformis (Rhodophyta), known for its ability to produce agar, is attracting increasing attention in the food industry. To develop new G. lemaneiformis germplasms, the four stages tetrasporophyte development and related molecular mechanisms were previously studied, but the metabolite changes that occur during the reproductive process remain unknown. In this study, we employed comprehensive metabolomics technology to examine changes in metabolites during the reproductive process of G. lemaneiformis , and a total of 817 metabolites were found, including 355 differentially accumulated metabolites (DAMs). The results revealed that G. lemaneiformis fulfills its energy requirements during reproduction by increasing glycolysis and TCA cycle flux, while it meets its material requirements by activating amino acid and nucleotide metabolism. Additionally, a significant accumulation of biogenic amines produced by the shikimate pathway may promote the formation of tetraspores, and a combination of flavonoids and phenolic acids effectively preserved the balance of the antioxidant system during the reproductive process of G. lemaneiformis . In this research, we analyzed changes in metabolites during the transition from the nutritional stage to the reproductive stage of G. lemaneiformis , revealing the potential relationship between tetraspore formation and metabolites and providing a theoretical basis for the cultivation of new germplasms of G. lemaneiformis .

1-Aminocyclopropane-1-carboxylic acid mitigates copper stress by decreasing copper transport and inducing jasmonic acid synthesis in Gracilariopsis lemaneiformis

Yueyao Tang,Xiaojiao Chen,Hao Zhang,Xue Sun,Nianjun Xu

Severe marine pollution caused by excessive copper (Cu) has resulted in a great threat to marine biosystems. Gracilariopsis lemaneiformis , the main economically important cultivated macroalgal species of coastal China, is affected by Cu. 1-Aminocyclopropane-1-carboxylic acid (ACC), the direct precursor of ethylene, acts as a potential signaling molecule for plants to defend against heavy metal stress. In the present study, the role of ACC in alleviating Cu stress in G. lemaneiformis was explored by measuring physiological indicators and by performing a comparative transcriptome analysis. Excess Cu reduced photosynthetic rates and relative growth rates, increased Cu accumulation and malondialdehyde levels and induced the antioxidant system in G. lemaneiformis . ACC was found to inhibit the expression of genes encoding Cu transport proteins , greatly decrease Cu accumulation and MDA contents and relieve the inhibition of G. lemaneiformis growth caused by Cu stress. In addition, the expression of most antioxidant synthesis-related genes encoding SOD and APX was downregulated by ACC, and the activity of antioxidant enzymes was reduced. Moreover, ACC stimulated jasmonate acid (JA) synthesis to increase endogenous JA contents and activated pathways related to translation, photosynthesis, and nitrogen metabolism , while ACC did not stimulate the expression of genes involved in ethylene signaling pathways. These findings provide insight into the molecular mechanisms of Cu stress resistance in G. lemaneiformis and benefit the breeding and cultivation of G. lemaneiformis .
