Electrospun nanofibers as controlled release systems for the combined pheromones of Grapholita molesta and Cydia pomonella

Hui-Ming Xiang,Xiao-Hong Wei,Yi-Ran Li,Bo-Jun Zhang,Min Li,Rui-Yan Ma

BACKGROUND As sex pheromones are environmentally friendly and specific, they are often used to monitor and control oriental fruit moths (OFMs). Currently, non-biodegradable polymers are commonly employed as carriers to prepare controlled sex pheromone release systems for plant protection. Electrospinning is a relatively simple technique for preparing biodegradable nanofibers that allows for the controlled release of sex pheromones. This study aimed to develop a biodegradable controlled release system for the combination of OFM pheromone and codlemone via electrospinning, in which codlemone was used as a synergist for OFM pheromone. RESULTS New systems for the controlled release of combined pheromones from OFM and colding moths were developed using electrospun nanofibers from polycaprolactone (PCL), cellulose acetate (CA), and polylactic acid (PLA). In the indoor experiments, the load ratio and release stability of CA nanofibers loaded with combined pheromones (CA-SP) were superior to those of the other two nanofiber types. In the field experiments, among all the treatments, 10 mg of CA-SP was the most attractive to OFMs, and its effective duration was approximately 6 weeks. The optimum storage temperature for all nanofibers was −20 °C. CONCLUSION Electrospun CA-SP nanofibers were biodegradable and environmentally friendly, with stable release. This study presents a new technique that could be beneficial for the monitoring and control of OFMs. © 2025 Society of Chemical Industry.

Reverse chemical ecology guides the screening for Grapholita molesta pheromone synergists

Jiyuan Liu,Tong Zhou,Chaoxia Li,Ruichi Li,Xuan Ye,Zhen Tian

BACKGROUND Pheromone-based management is a leading nonpesticidal strategy among integrated pest management options. Improving the potency of pheromone products by adding synergists would be a practical way to popularize pheromone-based management as well as to reduce pesticide use. RESULTS Using reverse chemical ecology, synergists for Grapholita molesta sex pheromone were screened. Combined molecular docking and in vitro binding assay led to the determination of four potentially active odorants showing high affinity to G. molesta pheromone binding protein 2 (GmolPBP2). Thereafter, the high affinity between Codlemone and GmolPBP2 was further verified by exploration of GmolPBP2–Codlemone interactions. As the only sex pheromone synergist validated by both laboratory behavioral tests and field trapping, Codlemone was used to optimize commercial sex attractants currently used in G. molesta control. The recommended formulation [( Z )-8-dodecenyl acetate:( E )-8-dodecenyl acetate:Codlemone = 95:4:10] was found to trap about five to six times more G. molesta adults than the commercial sex attractant [( Z )-8-dodecenyl acetate:( E )-8-dodecenyl acetate: ( Z )-8-dodecenol = 95:4:1]. CONCLUSION Codlemone is an excellent pheromone synergist that can be potentially sensed by GmolPBP2, which can remarkably improve the potency of G. molesta sex attractants. It is believed that the introduction of reverse chemical ecology would increase the chance of discovering pheromone synergists, promoting the development of more efficacious pheromone products that can be used in controlling G. molesta and beyond. © 2021 Society of Chemical Industry.

Structural insights into the discrepant synergistic activity of Codlemone and (Z)-8-dodecenol towards Grapholita molesta pheromones

Jiyuan Liu,Tong Zhou,Ruichi Li,Zhen Tian

BACKGROUND Insect pheromone synergists have been widely used to produce potent pheromone products for environment-friendly pest control. Codlemone (Cod) and ( Z )-8-dodecenol (Dod) are two major Grapholita molesta pheromone synergists, with Cod having greater synergism and affinity for G. molesta pheromone binding protein 2 (GmolPBP2). Uncovering structural information key to the different binding affinity of Cod and Dod to GmolPBP2 would gain insights into what causes their synergy activity discrepancy. RESULTS Binding modes of the two synergists in the binding pocket of GmolPBP2 were analyzed and compared by molecular dynamics-based approaches. Although Cod and Dod were stabilized in a similar hydrophobic pocket, their interaction details with GmolPBP2 were divergent due to the extra double bond (C10═C11) in Cod. The C10═C11 improved the hydrophobic interactions of Cod with around residues. Such hydrophobic interaction improvement was also reflected in the raised importance of Phe11 in the GmolPBP2-Cod interaction. Not only that, the increased hydrophobic forces introduced by the C10═C11 changed the CH2–OH orientation in the GmolPBP2-Cod complex, which improved the H-bond interaction. Electrostatic complementarity analysis further indicated the positive role of C10═C11 in optimizing GmolPBP2-Cod interaction. CONCLUSION The C10═C11 is thought to contribute greatly to Cod's stronger synergy as a group key to the higher GmolPBP2-affinity, based on which the improvement directions for Cod and Dod were addressed as well. Our findings will aid in the development and optimization of more effective pheromone synergists, resulting in more effective pheromone-based pest management.

Age-Driven Olfactory Plasticity Regulates Oviposition Behavior in Mated Grapholita molesta Females in Response to Pear Fruit Volatiles

Weina Kong,Hang Chen,Xiaoxue Wei,Changnian Liu,Yi Wang,Xuefeng Hu,Jie Li,Ruiyan Ma,Longlong Liu

Grapholita molesta is a major fruit tree pest. Host plant volatiles act as chemical cues to mediate female oviposition. This study investigated the recognition of ovipositing G. molesta females for pear volatile blends based on age-dependent olfactory plasticity. Results showed that mated females exhibited a distinct reproduction–flight coordination window optimized at 2 days of age. Olfactory gene expression in female antennae depended on the postmating age. Nine odorant receptor (OR) genes were upregulated at 3 vs. 9 days postmating (DPM), with stronger antennal responses to trace-level odorants. RNA interference targeting ORs, especially GmolOR46, reduced antennal responses and oviposition preference at 3 DPM. Mature pears emitted more volatiles than apples, especially after mechanical injury. Females at 3 DPM were attracted to volatiles from intact or injured mature pears, particularly at high concentrations. They increased field moth catches. Our findings reveal that pear volatiles have potential as attractants for G. molesta.

Exposure to benzaldehyde and larval feeding experiences modulate olfactory and oviposition behaviors in female Grapholita molesta (Lepidoptera: Tortricidae)

Yi Wang,Wei-Na Kong,Rui-Wei Shen,Hang Chen,Yu-Song Zhao,Jie Li,Rui-Yan Ma

BACKGROUND Grapholita molesta is an important pest of rosaceous fruit trees. Female oviposition affects larval infestation levels in orchards. This behavior, shaped by larval feeding experiences and female olfactory responses, points to an experience-modulated semiochemical strategy based on host plant secondary metabolites. To elucidate the combined effects of non-volatile and volatile components on female oviposition, we analyzed the volatile profiles of peach, pear, and apple shoots/fruits under intact, mechanically injured, and larval damage states throughout the growing season, along with female oviposition preferences, and compared olfactory responses (electroantennogram (EAG) and wind-tunnel experiments) and oviposition bioassays with benzaldehyde (amygdalin hydrolysate) between females with two larval amygdalin-feeding experiences. RESULTS Female oviposition preference and benzaldehyde emissions, both modulated by host factors, exhibited synchronous fluctuations. In EAG and wind-tunnel experiments, females not fed amygdalin during larval stages exhibited a strong attraction to benzaldehyde at concentrations of 10 and 0.1 μg/μL; however, females from larvae fed amygdalin showed a weaker long-distance orientation but greater responses to benzaldehyde during close-range approach and landing behaviors. Females fed amygdalin during larval stages were more likely to be responsive to lower concentrations of benzaldehyde (0.01, 0.1, and 1.0 μg/μL) for oviposition. In contrast, close-range attraction and oviposition behaviors were stronger for females that developed from amygdalin-fed larvae than those that were not fed amygdalin. CONCLUSION Dietary exposure to amygdalin during the larval stages increased the olfactory responses of females to benzaldehyde. Female oviposition behavior is co-regulated by larval experience derived from amygdalin ingestion and benzaldehyde as a host semiochemical signal. © 2025 Society of Chemical Industry.

The voltage-gated sodium channel mutation and detoxification enzymes associated with the resistance of Grapholita molesta (Busck) (Lepidoptera: Tortricidae) to lambda-cyhalothrin

Xiaohe Zhang,Sha Su,Caiyun Qiu,Bufei Ma,Zheng Zhao,Xiong Peng,Jaime C. Piñero,Maohua Chen

The oriental fruit moth , Grapholita molesta (Busck) (Lepidoptera: Tortricidae), a major fruit pest, is primarily controlled with pyrethroid insecticides such as lambda-cyhalothrin and resistance has emerged as a serious threat to management. In this study, we compared a lambda-cyhalothrin-resistant strain (RS) with a susceptible strain (SS) of G. molesta . Sequencing of the voltage-gated sodium channel (VGSC) gene revealed the L1014F mutation exclusively in RS individuals, strongly associated with knockdown resistance. Bioassays showed that the RS exhibited a 13.1-fold cross-resistance to deltamethrin and a 6.0-fold cross-resistance to bifenthrin, as well as moderate cross-resistance to chlorantraniliprole, abamectin, and emamectin benzoate. While showing only low or negligible cross-resistance to other insecticides tested. Synergist assays indicated that piperonyl butoxide (PBO) increased lambda-cyhalothrin toxicity by 1.72-fold in RS, implicating cytochrome P450 as key contributors. Biochemical assays confirmed significantly elevated activities of glutathione S-transferase (GST) and P450 in RS relative to SS. Transcriptomic analysis identified 758 differentially expressed genes, including 13 P450s, 5 GSTs, and 2 carboxylesterases (CarEs), with qRT-PCR validating strong up-regulation of CYP333B121 (14-fold), CYP6B230 (15-fold), CYP6AB117 (13-fold), and GSTs2 (5-fold). These findings suggest that the target-site resistance conferred by the L1014F kdr mutation, together with metabolic resistance mediated by overexpressed P450 and GST genes, jointly contribute to the resistance of G. molesta to lambda-cyhalothrin.

Enhancement of attraction to sex pheromone of Grapholita molesta (Busck) (Lepidoptera: Tortricidae) by structurally unrelated sex pheromone compounds of Conogethes punctiferalis (Guenée) (Lepidoptera: Crambidae)

Jianyu Deng,Zhijie Shen,Fumin Wang,Tao Liu,Wenying Hong,Minghua Fang,Linfeng Wo,Sijie Chu

Grapholita molesta (Busck) (the oriental fruit moth; OFM) and Conogethes punctiferalis (Guenée) are both fruit and stem borers with overlapping habitats, occurrences, and outbreak cycles. These two species are in different families and they have completely different sex pheromone components. Here, the effects of the sex pheromone components of C. punctiferalis , ( E )-10-hexadecenal ( E 10-16:Ald) and ( Z )-10-hexadecenal ( Z 10-16:Ald) and their mixture, were evaluated on the sexual communication of OFM by examining electrophysiological (EAG) and behavioral responses. We found that a considerably large amount of E 10-16:Ald or Z 10-16:Ald and a tiny amount of OFM pheromone elicited comparable EAG responses in OFM males, indicating the low antennal olfactory sensitivity of OFM males to the heterospecific pheromone compounds. In two different peach orchard parcels, captures of OFM by their pheromone lures baited in delta traps were increased by at least 1.5 times when OFM pheromone lures were combined with lures that contained 1000 μg of either E 10-16:Ald, Z 10-16:Ald or their mixture. In two other pear orchard parcels, both E 10-16:Ald and Z 10-16:Ald increased captures of OFM in Unitraps in a dose-dependent manner with more than a four-fold increase. Therefore, the enhanced attractiveness of OFM lures by the two interspecific pheromones suggests their potential application to improve mass trapping, population monitoring and mating disruption of OFM.

Degradation of sex pheromone and plant volatile components by an antennal glutathione S-transferase in the oriental fruit moth,Grapholita molesta Busck (Lepidoptera: Tortricidae)

Guang-Wei Li,Xiu-Lin Chen,Xiang-Li Xu,Jun-Xiang Wu

Insect antennae have a primary function of perceiving and discerning odorant molecules including sex pheromones and host plant volatiles. The assumption that genes highly expressed in the antennae may have an olfactory-related role associated with signal transduction. Here, one delta subfamily glutathione S -transferase (GST) gene (GmolGSTD1) was obtained from an antennal transcriptome of Grapholita molesta . Quantitative real-time polymerase chain reaction results revealed that GmolGSTD1 was mainly expressed in antennae and the expression levels were significantly higher in female antennae than in male antennae. The recombinant GmolGSTD1 (rGmolGSTD1) showed glutathione-conjugating activity toward 1-chloro-2,4-dinitrobenzene (CDNB) as substrates. The pH range for optimal rGmolGSTD1 enzyme activity was 6.0–6.5, and rGmolGSTD1 enzyme activity had maximal peaks at 35–40°C. Spectrophotometric analysis indicated that insecticides had weak inhibitory effects on the activity of rGmolGSTD1 with the inhibitory rates of 28.82% for chlorpyrifos, 22.27% for lambda-cyhalothrin, 18.07% for bifenthrin, 20.42% for acetamiprid, 17.57% for thiamethoxam, 25.67% for metaflumizone, 27.43% for abamectin, and 7.24% for chlorbenzuron. rGmolGSTD1 exhibited high degradation activity to the sex pheromone component (Z)-8-dodecenyl alcohol and the host plant volatile butyl hexanoate with the degradation efficiency of 75.01% and 48.54%, respectively. We speculate that GmolGSTD1 works in inactivating odorant molecules and maintaining sensitivity to olfactory communication of G. molesta .

Sublethal Effects of Abamectin on the Development, Reproduction, Detoxification Enzyme Activity, and Related Gene Expression of the Oriental Fruit Moth (Lepidoptera: Tortricidae)

Su Sha,Jian Chengzhi,Zhang Xiaohe,Fang Sensen,Peng Xiong,Piñero Jaime C,Chen Maohua

Grapholita molesta is one of the most important fruit pests worldwide. Abamectin is a biological pesticide frequently used to control fruit borers like G. molesta in part owing to its translaminar properties. In this study, we characterized the toxicity of abamectin to G. molesta larvae using the diet incorporation method. The sublethal effects of abamectin on the development, reproduction, detoxification enzyme activity, and related gene expression of G. molesta were assessed. The results showed that the LC20 and LC50 values of the insecticide against G. molesta 72 h post-treatment were 1.17 mg L–1 and 5.85 mg L–1, whereas the LC20 and LC50 values 96 h post-treatment were 0.34 mg L–1 and 3.63 mg L–1. When compared to the control, sublethal concentrations of abamectin 1) significantly increased the mortality of the larvae, prepupae, and pupae of G. molesta, 2) prolonged the duration of 3rd to 5th instar larva, prepupal and pupal periods, 3) shortened the longevity of adults, and 4) reduced female fecundity. The enzymatic activity of glutathione S-transferase (GST) varied significantly after exposure to sublethal concentrations of abamectin, but the cytochrome P450 monooxygenases and carboxylesterase activity were not significantly affected. Thirteen of the 25 GST genes were significantly upregulated under different sublethal concentrations of abamectin. The combined findings increase our understanding of the effects of abamectin on G. molesta and the potential role of GSTs in the metabolic interactions of abamectin in this pest, and have applications for more rational and effective use of abamectin to control G. molesta.
